7. Preventive antibiotic treatment of calves: emergence of dysbiosis causing propagation of obese state‐associated and mobile multidrug resistance‐carrying bacteria

Abstract

Summary In agriculture, antibiotics are used for the treatment and prevention of livestock disease. Antibiotics perturb the bacterial gut composition but the extent of these changes and potential consequences for animal and human health is still debated. Six calves were housed in a controlled environment. Three animals received an injection of the antibiotic florfenicol (Nuflor), and three received no treatment. Faecal samples were collected at 0, 3 and 7 days, and bacterial communities were profiled to assess the impact of a therapy on the gut microbiota. Phylogenetic analysis (16S‐rDNA) established that at day 7, antibiotic‐treated microbiota showed a 10‐fold increase in facultative anaerobic Escherichia spp, a signature of imbalanced microbiota, dysbiosis. The antibiotic resistome showed a high background of antibiotic resistance genes, which did not significantly change in response to florfenicol. However, the maintenance of Escherichia coli plasmid‐encoded quinolone, oqxB and propagation of mcr‐2, and colistin resistance genes were observed and confirmed by Sanger sequencing. The microbiota of treated animals was enriched with energy harvesting bacteria, common to obese microbial communities. We propose that antibiotic treatment of healthy animals leads to unbalanced, disease‐ and obese‐related microbiota that promotes growth of E. coli carrying resistance genes on mobile elements, potentially increasing the risk of transmission of antibiotic resistant bacteria to humans.

Abstract

In order for AGVs to be able to effectively carry out the tasks assigned to them, it is important to accurately determine their position and orientation in the working space. Having data on the location of an AGV is crucial for the navigation process, and the most commonly used odometry method is unreliable due to errors. To correct these errors, additional measuring systems are used. These systems use a variety of sensors. Some of the most widely used types are laser rangefinders. These sensors are also used in the automatic course correction methodology that is developed and presented in this article. The measurements from laser rangefinders are used to determine the shift of the actual trajectory from the set one, and then to guide the AGV to the previously set course. The developed methodology is experimentally verified on the basis of several dozen test drives. The conducted experimental studies prove the correctness of the developed methodology. The proposed course correction algorithm can be implemented in most working conditions, and guarantees correct passage over the given route.

11. Using bio-adhesive and bio-inert surfaces to maximize biogas production and influence microbial growth in anaerobic digesters

Abstract

The optimisation of biogas digesters is crucial for further development of sustainable energy sources. In this research, an integrative approach was taken to understanding how this problem can be addressed including: seeking a better understanding of protein – surface interactions on the molecular level; larger scale experiments to screen the best materials for use in laboratory scale anaerobic digesters to influence microbial growth and biofilm development as well as analysis of farm-scale data using the ADM1 multiparameter model. The laboratory-scale experiments were undertaken to develop surfaces suitable for studying microbial immobilization. This work, currently using tertiary amines, amine oxides and comparator oligoethylene glycol studies the adsorption of two classic model proteins: lysozyme and fibrinogen using quartz crystal microbalance methods and represents important steps for selecting and exploring surface – protein interactions. The data showed that tertiary amine oxides are more resistant to nonspecific protein adsorption than the corresponding tertiary amines. Heat modified polyurethane foam was used to explore biofilm and planktonic phase microbial populations in a fixed film biogas reactor. After four weeks the foam was analysed by ‘next generation’ 454-sequencing to identify the influence of the supporting materials on microbial population residing in anaerobic digesters. The results revealed that Spirochaetes, Methanobacterium and Methanocorpusculum associated themselves with heat modified polyurethane foams. Finally, data from a farm-scale anaerobic digester (volatile fatty acid concentration, temperature and pH) have been gathered and entered into the ADM1 model, developed by the mathematical modelling group from Lund University, to mimic the behaviour of a laboratory scale 1.5 l reactor and identify improved conditions for methanogens stability. The combination of approaches described above will allow the identification of which parameters will enhance the operation of anaerobic digesters and has identified surfaces that promote adhesion of particular Bacteria and Archaea in order to increase biogas production.

Abstract

The development of the COVID-19 pandemic and the related lockdown had a major impact on vehicle traffic in cities. Based on available data from the selected city of Rzeszow, Poland, it was decided to assess changes in vehicle traffic and the impact of these changes on roadside environmental pollution. As part of the research, data from the first half of 2020 regarding vehicle traffic on selected streets of the city and on the city’s inlet routes were analyzed. For the selected road sections, changes in hourly traffic volume in 2020, compared with 2019, were also determined. With data on traffic volume, an attempt was made to estimate the impact of changes in traffic volume on air pollution in the city. Research on air pollution from motor vehicles was focused on a selected section of a city road that was equipped with an automatic air pollution measurement station located very close to the road. Additionally, at the road intersection and in the vicinity of the measuring station, a sensor was installed in the roadway to count passing vehicles. A preliminary analysis of air pollution data revealed that factors such as wind speed and direction and outside temperature had a large impact on measurement results. To eliminate the influence of these factors and to obtain data concerning only contamination originating from motor vehicles traveling along the road, an appropriate mathematical model of the traffic flow–roadside environment system was built. This model was designed to determine the air pollution in the vicinity of the road generated by traffic flow. The constructed model was verified, and the conditions for its use were determined in order to study the impact of traffic and roadside environment on the level of air pollution from harmful exhaust substances. It was shown that at certain times of the day, especially at low temperatures, other sources of harmful emissions related to home heating play a major role in air pollution in the city.

Abstract

In the case of automated guided vehicles using odometry, a very important issue is to know the actual rolling radius of the wheel used to calculate the position of the vehicle. This radius is not constant. Its changes depend on the elastic deformation of the band layer and wheel slip. The theoretical determination of the value of the radius and the nature of the change is very difficult. For this reason, it was decided to determine the rolling radii by the experimental method. For this purpose, an appropriate test stand was built and the proposed research method was checked. Within the tests, there was also obtained a number of interesting results characterizing the material used to build bands and the ranges of changes rolling radii for the tested material were specified.

Abstract

Metabolic bone disease of prematurity still occurs in preterm infants, although a significant improvement in neonatal care has been observed in recent decades. Dual-energy X-ray absorptiometry (DXA) is the precise technique for assessing bone mineral content (BMC) in preterm infants, but is not widely available.To investigate the clinical and biochemical parameters, including bone metabolism markers as potential predictors of BMC, in preterm infants up to 3 months corrected age (CA).Ca-P homeostasis, iPTH, 25-hydroxyvitamin D, osteocalcin, N-terminal propeptide, cross-linked C-telopeptide and amino-terminal pro C-type natriuretic peptide and the DXA scans were prospectively performed in 184 preterm infants (≤ 34 weeks' gestation) between term age and 3 mo CA. Lower bone mass was defined as BMC below or equal to respective median value for the whole study group, rounded to the nearest whole number.The appropriate quality DXA scans were available for 160 infants (87%) examined at term and for 130 (71%) tested at 3 mo CA. Higher iPTH level was the only independent predictor of lower BMC at term, whereas lower BMC at 3 mo CA was associated both with lower urinary phosphate excretion and higher serum osteocalcin level. ROC analysis showed that iPTH >43.6 pg/mL provided 40% sensitivity and 88% specificity in identification of preterm infants with lower BMC at term. In turn, urinary phosphate excretion (TRP>97% or UP/Cr ≤0.74 mg/mg) and serum osteocalcin >172 ng/mL provided 40% sensitivity and 93% specificity in identification of infants with decreased BMC at 3 mo CA.Serum iPTH might to be a simple predictor of reduced BMC in preterm infants at term age, but urinary phosphate excretion and serum osteocalcin might predict reduced BMC at 3 mo CA. These results represent a promising diagnostic tool based on simple, widely available biochemical measurements for bone mass assessment in preterm infants.
