8. İskemi/Reperfüzyon Hasarı Sonrası İloprost İle Tedavi Edilen Sıçanlarda Tuba Uterina Dokusunun Histopatolojik Değerlendirilmesi

Amaç: Sunulan çalışmada sıçanlarda oluşan I/R hasarı sonrasında tuba uterina dokusu üzerinde antioksidan özelliği olan Iloprostun koruyucu etkisinin olup olmadığının değerlendirilmesi amaçlandı.Gereç ve Yöntemler: Çalışmamızda; 32 dişi Sprag Dawley sıçan kullanıldı. Gruplar sham, torsiyon, detorsiyon ve detorsiyon + ıloprost olarak belirlendi. Sham grubu hariç diğer ratlara torsiyon işlemi yapıldı. Torsiyon grubuna üç saat torsiyon, detorsiyon grubuna 3 saat torsiyon + 3 saat detorsiyon, I/R + ıloprost grubuna 3 saat torsiyon + 3 saat detorsiyon ve detorsiyon başlangıcında 60 dk intravenöz iloprot infüzyonu yapıldı. Histopatolojik değerlendirme için alınan Tuba uterina dokularının histolojik takibi yapıldı. Sonrasında 7 mikron kalınlıkta kesitler alındı ve hematoksilen & Eozin boyası ile boyandı. Alınan kesitler üzerinde ışık mikroskobik inceleme yapıldı.Bulgular: Tuba uterinada yapılan incelemelerde kontrol grubunda herhangi bir histopatolojik bulguya rastlanmadı. Kontrol grubundan farklı olarak Torsiyon grubunda vasküler tıkanıklık ve epitelde vakuolleşme meydana geldi. Detorsiyon grubunda epitelin bütünlüğünün bozulduğu gözlendi. İloprost uygulanan gruba ait görüntülerde hemarojik alanlar dışında herhangi bir histopatolojik duruma rastlanmadı. Bununla birlikte, bütün gruplarda bağ doku, muskuler alanlar ve seroza tabakaları normaldi.Sonuç: Antioksidan ve vazodilatör etkisi nedeniyle tedavi amaçlı kullanılan iloprostun sıçan tuba uterina dokusu üzerinde I/R hasarı sonrasında korucu etkisinin olduğu gözlendi. Bu sonuçlar doğrultusunda organın tekrar fonsiyone edilmesi ile torsiyone olan tarafta ektopik gebelik riskinin azaltılabileceği düşünüldü.

9. Assessment of age- and sex-dependent changes of cerebellum volume in healthy individuals using magnetic resonance imaging

Amaç: Serebellum fossa cranii posterior’da yerleşmiş, motor fonksiyonların koordinasyonu, kas tonusunun düzenlenmesi ve motor öğrenmede; ayrıca hassasiyet ve doğru zamanlamanın sağlanmasına katkıda bulunan beyinde önemli rolü olan bir yapıdır. Bu çalışma ile amacımız öncelikle yaşa ve cinsiyete bağlı olarak serebellum ölçüm skalası oluşturabilmektir.Yötemler: Çalışma Adıyaman Klinik Araştırmalar Etik Kurulu’ndan alınan onay ile Adıyaman Üniversitesi Eğitim ve Araştırma Hastanesi Radyoloji Anabilim Dalı’nın katkılarıyla gerçekleştirilmiştir. Veriler manyetik rezonans görüntüleme yöntemi kullanılarak (80 kadın ve 80 erkek) toplam 160 bireye ait görüntüler üzerinde retrospektif kohort bir çalışma olarak elde edilmiştir. Çalışmaya dâhil edilen bireyler 0-4 yaş, 5-9 yaş, 10-19 yaş, 20-29 yaş, 30-39 yaş, 40-49 yaş, 50-59 yaş ile 60-80 yaş toplam 8 gruba ayrılmış ve her yaş grubu da 10 kadın ve 10 erkek toplam bireylerden oluşmaktadır. Elde edilen verilerin istatistiksel tüm analizleri için SPSS programı kullanılmıştır.Bulgular: Çalışmamızda serebellum hacmi 10-19 yaş kadınlarda 139,23 (11,73) cm3 ve erkeklerde 151,14 (12,30) cm3 idi (P=0,040). 0-4 yaş arası bireylerin serebellum hacmi 105,05 (33,28) cm3 diğer yaş gruplarına göre daha küçük bulundu ve diğer gruplar ile arasında anlamlı bir farklılık vardı (P, Aim: Cerebellum is a structure ingrained in fossa cranii posterior and has a great role in contributing to the coordination of motor functions, regulation of muscle tone, and motor learning, as well as precision and accurate timing. This study primarily aimed to establish a cerebellum measurement scale based on age and sex. Methods: The study was carried out with the approval of Adiyaman Clinical Research Ethics Board and the contributions of Adiyaman University Training and Research Hospital Radiology Department. The data were retrospective cohort study obtained from the magnetic resonance images of 160 individuals (80 female and 80 male). The individuals included in the study were divided into age groups of 0-4, 5-9, 10-19, 20-29, 30-39, 40-49, 50-59 and 60-80 years, each consisting of 10 women and 10 men. All statistical analyses were performed using SPSS program. Results: The cerebellum volume was 139.23 (11.73) cm3 for women and 151.14 (12.30) cm3 for men aged 10-19 years, and the difference was significant (P=0.040). Giriş: Sonzamanlarda diş hekimliği bilimsel anlamda karakteristikdeğerlerini belirlemiştir. Sağlık alanında eğitim veren tüm branşlarda olduğugibi Diş Hekimliği Fakültelerinde öğretimprogramının ilk yılında Anatomi eğitimi verilmektedir. Anatomi eğitimindeöğrencilerin bakış açılarının değerlendirilmesi ve anlaşılması, anatomi eğitimive öğretim yöntemlerinin geliştirilmesinde büyük önem taşımaktadır. Buçalışmada Adıyaman Üniversitesi Diş Hekimliği Fakültesi 1. sınıf öğrencilerininanatomi eğitimi hakkındaki düşüncelerinin değerlendirmesi ve takım çalışmalarıile interaktif eğitimin anatominin öğrenilmesine sağladığı katkıların ortaya çıkarılmasıamaçlanmaktadır. Materyalve Metod:Çalışmaya 2016-2017 eğitim-öğretim yılında AdıyamanÜniversitesi Diş Hekimliği Fakültesi 1. sınıfta Anatomi eğitimi alan toplam 60öğrenci (n=60) dahil edildi. Veriler 29 kapalı uçlu sorudan oluşan bir anketformu kullanılarak geri bildirim yöntemiyle elde edilmiştir. Anketsoruları 5’li Likert ölçeği ile cevaplandırılacak şekilde hazırlamıştır. Eldeedilen veriler IBM SPSS Statistics paket programı ile analiz edilmiştir.Bulgular:Elde edilen sonuçlara göre öğrencilerin, anatominin teorik kısmını çalışırkenen çok ders notlarından, pratik kısmınıçalışırken en çok ders kitabından faydalandıkları tespit edildi. Aldıklarıanatomi eğitiminin teorik kısmı için %88.3 ve pratik kısmı için %85.0memnuniyet düzeyleri olduğu belirlendi. Verilen proje ödevleri ile ilgiliolarak öğrencilerin %71.7’sinin 1-3 hafta içerisinde ödevlerini bitirdiği, %70.0’inin anatomiyi sevmelerini sağladığı,%91.7’sinin o konunun anatomisini öğrenmelerine katkıda bulunduğu ve %73.4’ünün “anatomi çalışmak ve anlamak zordur” önyargısını kırdığı belirlenmiştir.Sonuç:Elde edilen verilerin anatomi ile ilişkili eğitim alternatiflerigeliştirmesinde, mevcut eksikliklerin giderilmesinde ve verilen proje ödevleriile öğrencilerin eğitime aktif katılmasının sağlanmasıyla anatomi’yi daha etkinöğrenmede yararlı olacağını düşünmekteyiz. Ayrıca diş hekimliği fakültelerindede ciddi bir zaman ve emek harcanan anatomi eğitiminde geri bildirim anketleriuygulanarak verilen eğitimlerin daha iyi noktalara gelebileceğine de dikkatçekmiş olmaktayız., Recently, dentistry has determined itscharacteristic values in scientific sense. As in all branches of education in the field of health, Anatomyeducation is given in the first year of curriculum in Faculties ofDentistry. Evaluating and understandingthe perspectives of the students on anatomy education is of great importance inthe development of anatomy education and teaching methods. In this study, weaimed to evaluate the students' thoughts about anatomy education and thecontributions of team work and interactive education to the learning ofanatomy. Materialand Method: Inthis study, 60 first-year students of Adıyaman University Faculty of Dentistrywho took Anatomy lecture in the spring semester of 2016-2017 academic year wereincluded. Data were obtained by using a questionnaireconsisting of 29 closed-ended questions. The questionnaires have been preparedto be answered with a scale of 5 likert. The data were analyzed using the IBMSPSS statistics package program.Results: According to the results, the students usedlecture notes for theoretical part and textbook for practical part mostly. Asatisfaction level of 88.3% and 85 % wasdetected for theoretical part andpractical part respectively. It was determined that 71.7% of the studentscompleted their homework in 1-3 weeks, in 70.0% these homeworks made them likeanatomy, in 91.7% they made contribution to anatomy learning and in 73.4% of the studentes the homeworksbroke the bias of “anatomy is difficult to study and understand”.Conclusion: We believe that the data obtained will be usefulin developing educational alternatives related to anatomy, in overcomingexisting deficiencies and in providing students with assignments and activeparticipation in education. In addition, we have also pointed out that theeducation given in dentistry faculties can reach better points by applyingfeedback questionnaires in anatomy education which needs a lot of time andeffort.

Çalışmamızda, sıçanlarda streptozotocin (STZ) ile oluşan diyabetik pankreas hasarındaki histopatolojik değişiklikler ve bu değişiklikler üzerine caffeic acid phenethyl ester (CAPE)’in etkilerinin incelenmesi amaçlanmıştır.32 adet Wistar albino erkek sıçan 4 eşit gruba ayrıldı. Grup 1: Kontrol, Grup 2: CAPE (10 µmol/kg/ip 20 gün boyunca), Grup 3: Diabetes mellitus (DM)(55 mg/kg/ip tek doz), Grup 4: DM+CAPE. Deney sonunda sakrifiye edilen sıçanların pankreas dokuları çıkarıldı. Rutin histolojik takip işlemlerinden geçirilen dokular parafine gömüldü. Parafin bloklardan alınan 5 µm kalınlığındaki kesitler Hematoksilen-Eozin (H-E) ile boyandıktan sonra histopatolojik skorlama yapıldı. Leica DFC 280 ışık mikroskobu ve Leica Q Win Görüntü Analiz sisteminde incelenen kesitlerin fotoğrafları çekildi. Pankreas dokusunda MDA, GSH düzeyleri ve SOD, CAT aktivitelerine bakıldı. İstatistiksel analizler için SPSS 13.0 programı kullanıldı. Veriler aritmetik ortalama±standart hata olarak ifade edildi. p, In our study, we aimed to evaluate histopathological changes and therapeutic effects of caffeic acid phenethyl ester (CAPE) in streptozotocin (STZ)-induced diabetic pancreas damage in rats.32 male Wistar albino rats were divided into equal 4 groups. At the end of the study, pancreas tissues were removed and routin histological procedures were done. Paraffin blocks were cut at 5 µm and sections were stained with H-E and histopathological damage score was calculated. Sections were examined using a Leica DFC280 light microscope and a Leica Q Win Image Analysis system (Leica Micros Imaging Solutions Ltd., Cambridge, UK). MDA, GSH levels and SOD, CAT activities were measured in pancreatic tissue. Statistical analysis was carried out using the SPSS 13.0 (SPSS Inc., Chicago, III., USA) statistical program. Objective: In recent years, dentistry has determined its characteristic values in scientific sense. Anatomy lectures is availabe in the first year of dentistry education as in all branches of health care related field. The evaluation and understanding of students' perspectives in anatomy education is of great importance in the development of anatomy education and teaching methods. In this study, it is aimed to evaluate the thoughts about anatomy education of first year students of Adıyaman University Faculty of Dentistry and to find out the contribution that they have made to the interactive anatomy learning through team work. Material-Method: 60 students (n = 60) who receive anatomy lecture in the first semester of Adıyaman University Faculty of Dentistry in the academic year of 2016-2017 were included in the study. Data were obtained by using a feedback questionnaire consisting of 29 closed-ended questions. The questionnaires have been prepared to be answered with a scale of 5-point likert. The data were analyzed using the IBM SPSS statistics package program. Results: It is estimated that the students used lecture notes for theoretical part and textbook for practical part mostly. Satisfaction levels of 88.3% and 85% were detected for theoretical part and practical courses respectively. 71.7% of the students completed project assignments within 1-3 weeks. 70.0% of the students included in the study reported that they found this practice contributed to like anatomy courses. While 91.7% of the students emphasized that the homeworks they have contributed to their anatomy learning, 73.4% of the students gave up the idea of 'working anatomy and understanding difficult'. Conclusions: We believe that the data obtained will be useful in developing educational alternatives related to anatomy, in overcoming existing deficiencies and in providing students with assignments and active participation in education. In addition, we have also pointed out that the education given in dentistry faculties can reach better points by applying feedback questionnaires in anatomy education which needs a lot of time and effort. 17. Lamotrigin kullanan epileptik ratların yeni doğan yavrularının karaciğerlerinde oksidatif hasarın incelenmesi

The present study was performed to evaluate the effects of lamotrigine, which is a widely used antiepileptic drug for epilepsy treatment, on oxidative stress parameters in the liver of neonates. Ten adult Wistar-albino rats and twenty newborn of these rats were used in this study. Adult rats weighing 170–250 g were divided into five groups: the pregnant control group, experimental epilepsy group, lamotigine group, lamotrigine + experimental epilepsy group and lamotrigine + epilepsy + folic acid group. An experimental penicillin acute epilepsy model was created in pregnant rats on day 13 of pregnancy. The lamotrigine group was administered lamotrigine at 25 mg/day by intraperitoneal injection starting from the first day of pregnancy. The lamotrigine + folic acid group was administered 400 microgram/kg folic acid and 25 mg/day lamotrigine by intraperitoneal injection. We then waited for the rats to give birth. Immediately after birth, the glutathione, malondialdehyde, glutathione peroxidase and superoxide dismutase levels in the newborn rat liver were measured by spectrophotometry. The lamotrigine and lamotrigine + epilepsy groups showed significantly elevated liver malondialdehide levels, while glutathione peroxidase enzyme activation was significantly decreased compared to the control group. In the group administered folic acid together with lamotrigine, the liver tissue sample for malondialdehide levels showed a significant decrease, while glutathion and glutathion peroxidase levels increased. This study on pregnant rats demonstrated that lamotrigine use can lead to hepatic oxidative damage in newborn rats, and this oxidative tissue damage was prevented by folic acid use., Bu çalışmada amacımız epilepsi tedavisinde sıkça kullanılan antiepileptik ilaçlardan lamotirijinin yalnız veya folik asit ile birlikte gebelikte kullanımının yeni doğan yavruların karaciğerlerinde oksidatif stres parametreleri üzerine etkilerini araştırmaktır. Çalışmamız 170-250 gr ağırlığında 10 adet erişkin dişi Wistaralbino türü rat ve bunlardan doğan 20 yavru rat ile yapıldı. Ratlar; Gebe ratlar kontrol grubu, deneysel epilepsi grubu, lamotrijin grubu, lamotrijin+deneysel epilepsi grubu ve lamotrijin epilepsi+folik asit olarak beş gruba ayrıldı. Gebeliğin 13. Gününde gebe ratlara deneysel penisilin akut epilepsi modeli oluşturuldu. Lamotrijin grubuna gebeliğin ilk gününden itibaren her gün intraperitoneal enjeksiyon ile 25mg/gün dozunda lamotrijin verildi. Lamotrijin+folik asit grubuna her gün intraperitoneal enjeksiyon ile 25mg/gün dozunda lamotrijin ve 400µg/kg folik asit verildi. Daha sonra gebe ratların doğum yapmaları beklendi. Doğumdan hemen sonra yeni doğan ratların karaciğerlerinde glutatyon, malondialdehit, glutatyon peroksidaz ve superoksid dismutaz seviyeleri spektrofotometrik olarak ölçüldü. Lamotrijin ve lamotrijin+epilepsi gruplarına ait biyokimyasal değerler incelendiğinde, karaciğer malondialdehit anlamlı bir şekilde yükseldiği, glutatyon peroksidaz enzim aktivasyonlarının kontrol grubuna göre istatistiksel olarak anlamlı azalmış olduğu görüldü. Folik asit ile birlikte lamotrijin verilen grupta, karaciğer doku örneklerine ait malondialdehit düzeylerinin anlamlı azaldığı, glutatyon ve glutatyon peroksidaz düzeyinin arttığı tespit edildi. Acrylamide (AA) has neurotoxic, mutagenic, and genotoxic effects in humans and experimental animals. Fruit consumption is important for human health, because fruits are the source of many nutrients such as vitamins, minerals, carotenoids, dietary fiber, and phytonutrients. Many agricultural products provide natural melatonin in the diet. At the onset of the study, rats were weighted and randomly divided into four groups each containing 10 rats as follows: group 1: control (fed with normal diet and normal drinking water); group 2: apricot (fed with a daily diet with 5% apricot and normal drinking water); group 3: AA (administered daily acrylamide at 500 µg/kg b.w. via drinking water and fed a normal diet); group 4: apricot-AA (administered daily acrylamide at 500 µg/kg b.w. via drinking water and fed with a diet with 5% apricot). The diet schedule was continued for 12 weeks. At the end of the study, samples of large intestine were collected for biochemical analyses. The highest lipid peroxidation (as malondialdehyde, MDA) levels were observed in the AA groups, but MDA levels decreased significantly (P < 0.05) with apricot intake. Glutathione peroxidase activity in the apricot- AA group was higher than in the other three groups (P < 0.05). Glutathione S-transferase (GST) enzyme activity increased significantly in the AA group as compared with the other groups (P < 0.05). However, GST activity was significantly (P < 0.05) decreased by the apricot-supplemented diet. GST-Pi mRNA levels in the AA group increased significantly (P < 0.05) as compared with the other groups. In conclusion, the results of the current study demonstrated that AA caused large intestine damage and showed the efficiency of apricot in preventing this damage by inhibiting lipid peroxidation and improving antioxidant enzyme activities. The aim of this study is to investigate the effects of sun dried organic apricot (SDOA) (Primus armeniaca L.) supplementation on the daily food consumption of rats. In this study, 120 male and 120 female Sprague Dawley rats were used. They were grouped as 24 rat per group of female and male rats. Group 1 (control)was fed lip with standart rat chow, group 2; 1%, group 3; 2.5%, group 4; 5% and group 5 were fed up with 10% SDOA supplemented chow throughout 16 days. .1/ the beginning of the study the body weight of the male rats was measured as 321±24.6 g (n=120), and the body weight of the female rats was 210±21.4 g (n=120). The daily avarage normal food intake of control group of male and female rats were determined as 24±1.9 g/rot/day and 16±1.1 g/raf/day respectively. Consequently, it can be confidently said that in the ratio of 2.5% ve 5% supplementation of SDOA food on the male rats, 10% suplementation on the female rats have a significant positive effect in their daily food consumption. Human development begins with the formation of zygote by the merging of the male gametocyte (spermatozon) and the female gametocyte (oocyte). The zygote by cell division, cell migration, growth and differentiation, becomes a multi-celled person. This process includes embryonic and fetal periods. Fetus development is under the influence of genetic and environmental factors. Some substance causes malformations in pregnant women which is passed from placenta through fetal circulation to the fetus. In this case it is called teratogenesis. Free oxygen radicals are natural product of energy production processes. Increase of free radicals in the cell and their negative effects on cell functions are defined as oxidative stress. In our studies, we investigated that effect of various medicines on the fetal development of pregnant rat. We made pregnancies on rats in our studies that we have done at different times. Drugs such as cyclophosphamide, doxorubicin, ciprofloxacin phenytoin and lamotrigine were investigated for their effects on fetal developmental effects in pregnant rats. Fetuses were taken by C-section on the 20th day of pregnancy. Thereafter, the tissues suitable for the purpose of each study were taken. These tissues were subjected to histological assessments and biochemical analyzes. Many parameters were also affected such as antioxidant parameters, enzyme levels and levels of malondialdehyde (MDA) (a marker of lipid peroxidation) were affected. In all our studies we observed clearly that the free radical-induced oxidative damage in tissues such as fetal brain and liver. We have observed that free radical damage can be prevented by antioxidant supplementation. In conclusion, the use of antibiotics and chemotherapy drugs in pregnancy negatively affects fetal development through oxidative stress. We think that antioxidant supplementation is important when the use of these drugs is absolutely necessary. [ABSTRACT FROM AUTHOR]
