A method has been developed and implemented for synthesis of radiopharmaceutical based on synthetic peptide octreotate conjugated with bifunctional chelating agent DOTA, labeled by isotopes 210Pb and 212Pb. Researches of the dependence of the labeling efficiency on the synthesis duration and the peptide amount have been performed. It was revealed that labeling is effective (>95%) at the synthesis temperature of 90°C, the synthesis duration of 30 -60 min and the pH values of 4.5-6.5. Investigations have been made of the synthesized complex stability in isotonic solution and in human blood serum at room temperature. It is proven that the complex stability remains unchanged in isotonic solution at the level of >95% during the entire period of the 212Pb half-life (10.64 h), while serum stability is 80-85%. The results of the research allow us to come to a conclusion about 212Pb being a promising nuclide for further investigations and transition to studies of the [212Pb]DOTATATE cytotoxicity. In the long term, it can be regarded as one of the therapeutic instruments for neuroendocrine tumors and other oncological diseases.
