2. Early Prediction and Streamline of Nucleophosmin Mutation Status in Acute Myeloid Leukemia Using Cup-Like Nuclear Morphology.

Abstract

Background and Objectives: With the advent of novel therapies for nucleophosmin gene (NPM1)-mutated acute myeloid leukemia (AML), there is a growing need for the reliable prediction of NPM1 mutations. This study explored the role of cytomorphological features in the early prediction of NPM1-mutated AML. Materials and Methods: Altogether, 212 de novo AML cases with normal karyotypes, diagnosed and treated at a single institution within 5 years (2018–2023), were retrospectively evaluated. A final diagnosis of NPM1-mutated AML, based on the World Health Organization (WHO) integrated criteria, including real-time based identification of NPM1 mutation and normal karyotype, was established in 83/212 (39.15%) cases. Results: Cup-like blasts (CLBs), a cytomorphological feature suggestive of NPM1-mutated AML, were detected in 56/83 (67%) patients. Most cases (44/56, 78.6%) had CLB ≥ 10%. In total, 27 of 83 AML NPM1-mutated patients had no CLB morphology (missed call). Additionally, two of 212 had CLB morphology without confirmed NPM1 mutation (wrong call). The positive/negative predictive values of cytomorphological evaluation for CLB ≥ 10% were 95.7%/75.6%, with sensitivity/specificity of 53%/98.5%, while the accuracy was 80.7%. We noted an increased percentage of CLBs (≥15%) in 77.8% and 50% of patients with AML without and with granulocytic maturation, respectively (the specificity for NPM1 mutation prediction was 100%). CLB was associated with fms-like tyrosine kinase 3 (FLT3) mutation (p = 0.03), but, without statistical significance for CLB ≥ 10% and CLB ≥ 15%. Conclusions: Our investigation confirmed that the morphological identification of CLB at diagnosis represents a reliable and easily reproducible tool for the early prediction of NPM1 mutations, enabling a streamlined genetic work-up for its confirmation. Abstract

This paper documents the results of the PIM.3 (Process Improvement Management) working group in INTACS (International Assessor Certification Schema) supported by the VDA‐QMC (Verband der Deutschen Automobilindustrie/German Automotive Association–Quality Management Center). INTACS promotes Automotive SPICE, which is an international standard that allows process capability assessment of projects, which implement systems that integrate mechanics, electronics, and software including optionally cybersecurity, functional safety, and machine learning. The paper outlines that for the first time since more than 20 years, the INTACS and VDA‐QMC included a process like PIM.3 Process Improvement Management in the scope for the assessor training. Before that, the assessments focused on the management, engineering, and support processes of series projects, while the improvement management has not been trained or assessed. Abstract

Intentions to purchase meat and repeat purchasing patterns will depend on the consumer's perception of intrinsic and extrinsic quality cues. In order to discover consumers' insights into the quality of pork and beef, the main objective of this study was to investigate meat color, marbling, and types of fresh meat defects. A total of 50 samples of meat were analyzed for three quality characteristics on different levels. This study applied a statistical calculation of a loss function invented by Taguchi. The results showed the values of variations in meat color and marbling that present the limits of consumers' tolerance. Based on the obtained results, it was determined that the tolerable level of color variations of beef is 3.61, while for pork, this limit is 2.00. The most preferred marbling percentages were 21% and 5.74% for beef and pork, respectively. On the other hand, the negative effect of defects was the strongest for "dark cutting" for beef and "blood spotting" for pork loins. [ABSTRACT FROM AUTHOR]

Abstract

This study analyzes the influence of two different types of game meat (deer and wild boar) in relation to quality characteristics and oral processing attributes of skinless sausage. The goal of this study was to compare grilled game-meat-based “ćevap” with conventional pork-meat-based samples. Research comprised of color analysis, evaluation of textural components, testing degree of difference, temporal dominance of sensations, calculation of main oral processing attributes, and examination of particle size distribution. The results show that oral processing attributes are similar in between samples and concur with results of the pork-based sample. This confirms the working hypothesis that it is possible to make game-meat-based “ćevap” fully comparable with conventional pork meat products. In parallel, color and flavor characteristics are influenced by the type of game meat in the sample. Most of the dominant sensory attributes that occurred during mastication were game meat flavor and juiciness. © 2023 by the authors.

Abstract

The aim of this study was to investigated the seasonal variation in the nutrient composition of Mytilus galloprovincialis, cultivated in Boka Kotorska Bay, Montenegro, during all seasons in one year. Biometric parameters, meat yield, condition index, proximate composition, minerals, and the lipid and fatty acid compositions of mussels were analyzed. The most significant factors determining these parameters were temperature, food amount and gametogenesis The biometric parameters showed no significant differences during the sampling period. In the spring, meat yields and mussel condition index increased at substantial levels. Low values od meat yield and condition index during cold months have resulted from food shortage and reproductive cycle, when mussels use carbohydrates and protein reserves. The highest amount of protein was detected in mussels harvested in August (10.76%), while the highest amount of lipids was recorded in the winter months (2.11%). Docosahexaenoic acid and eicosapentaenoic acid were the most abundant PUFA. The concentration of metals found in mussels from the study area is within the range of mean values reported in the literature. Our results indicate that the best period for mussels harvesting was during the spring and summer (April and August), opposite the winter months when the mussels were not favorable for harvesting. Abstract

Hepatitis E is considered an emerging human viral disease with a zoonotic nature, and domestic and wild pigs are the main reservoirs of hepatitis E virus (HEV) among animals. Pork liver is the target tissue of this virus. This study aimed to investigate the presence of HEV in commercial pig liver samples. Sixty samples were collected during one year from different retail outlets in Serbia. Furthermore, the collected samples were separated by four seasons, and every season included three months. The presence of HEV in the livers was examined by molecular analysis using RT-qPCR. The overall prevalence of the virus in analysed pig livers was 5%. HEV was detected in three livers, two in the first season and one in the second, while in the third and fourth season, no positive livers were detected. However, there were no statistically significant differences between the surveyed seasons. HEV was quantified in positive livers. Among positive livers, HEV concentrations ranged between 8×101 and 1.9×104 genome copies of the virus per gram. The presence of HEV in commercial pig livers indicates a potential risk for consumers. Appropriate heat treatment of meals during preparation is essential to eliminate the potential risk of developing the illness. © Published under licence by IOP Publishing Ltd.
