Abstract Our previous findings demonstrated that astrocytic HIF‐1α plays a major role in HIV‐1 Tat‐mediated amyloidosis which can lead to Alzheimer's‐like pathology‐a comorbidity of HIV‐Associated Neurocognitive Disorders (HAND). These amyloids can be shuttled in extracellular vesicles, and we sought to assess whether HIV‐1 Tat stimulated astrocyte‐derived EVs (ADEVs) containing the toxic amyloids could result in neuronal injury in vitro and in vivo. We thus hypothesized that blocking HIF‐1α could likely mitigate HIV‐1 Tat‐ADEV‐mediated neuronal injury. Rat hippocampal neurons when exposed to HIV‐1 Tat‐ADEVs carrying the toxic amyloids exhibited amyloid accumulation and synaptodendritic injury, leading to functional loss as evidenced by alterations in miniature excitatory post synaptic currents. The silencing of astrocytic HIF‐1α not only reduced the biogenesis of ADEVs, as well as amyloid cargos, but also ameliorated neuronal synaptodegeneration. Next, we determined the effect of HIV‐1 Tat‐ADEVs carrying amyloids in the hippocampus of naive mice brains. Naive mice receiving the HIV‐1 Tat‐ADEVs, exhibited behavioural changes, and Alzheimer's ’s‐like pathology accompanied by synaptodegeneration. This impairment(s) was not observed in mice injected with HIF‐1α silenced ADEVs. This is the first report demonstrating the role of amyloid‐carrying ADEVs in mediating synaptodegeneration leading to behavioural changes associated with HAND and highlights the protective role of HIF‐1α.

The modeling and the implementation of the embedded switched z-source type cascaded multilevel inverter for photovoltaic (PV) interfaced applications have been presented. With the ability to draw continuous current with an inherent filtering capability, the embedded switched z-source type inverter provides a single-stage conversion with a high output gain which makes it suitable for PV arrays with a low voltage rating. By applying a modular cascading method with a reduced number of H-bridge the multilevel inverter (MLI) is designed for a series-parallel connected PV array. It is controlled using the basic multicarrier PWM technique and synchronized with the grid. With the derived design equations for each mode, its stability has been analyzed and compared for different duty cycles. The developed MLI connected with a PV array has been simulated with the idea of reducing the impact of partial shading by using shorter series of strings, providing a high gain conversion with lower stress across the components. A prototype of the MLI has been tested to give a power rating of 2 kW and the results from both the simulation and the hardware have been discussed.

The present study uses Adenanthera pavonina L. plant extract to synthesize Y/Fe2O3 nanoparticles in a sustainable and efficient manner. Several physicochemical investigations were performed on the synthesized Y/Fe2O3 to evaluate its structure and morphological characteristics. The high crystallinity of Fe2O3 and decreased crystal size of Y/Fe2O3 were revealed by the XRD study. The formulated Y/Fe2O3 was spherical in shape. Y/Fe2O3 has shown potential photocatalytic activity in the degradation of congo red and brilliant green dyes in the presence of visible light. Degradation efficiency is higher than 90% for both dyes. In addition, the antibacterial activity of Y/Fe2O3 was measured against pathogenic strains of Bacillus subtilis, Escherichia coli, Klebsiella pneumoniae, Micrococcus luteus, Proteus mirabilis, Pseudomonas putida, Shigella dysenteriae, and Staphylococcus aureus. Y/Fe2O3 showed promising antibacterial activity against all microorganisms. Micrococcus luteus showed a higher zone of inhibition (equal to 35 mm in diameter) than other microorganisms using Y/Fe2O3. Thus, it is established that the green synthesized Y/Fe2O3 nanoparticles are active catalysts for the photocatalytic degradation of hazardous dyes and have antibacterial properties against different bacterial stains. This study proposes three contributions focused on lung cancer detection and severity level identification. The absence of non-invasive technologies for predicting lung cancer necessitates faster, more efficient, and more accurate classification procedures due to the absence of non-invasive technologies for predicting lung cancer. Creating an automated and intelligent prediction system is crucial for identifying phases and predicting the possibility of a recurrence. The objective is to create an automated detection system for identifying lung cancer using an optimization-focused deep learning model. We develop an adaptive multi-swarm PSO and combine it with the firefly algorithm to determine the ideal weight values for the Wavelet Neural Network (WNN) model. We use the HAPSO-FFA-WNN method to explore problems with multiple optimal solutions. This study evaluated two lung cancer datasets, and the proposed HAPSO-FFA-WNN model achieved 97.58% accuracy for dataset 1 and 98.54% accuracy for dataset 2. The object of the study is to determine the optimal warranty length under free replacement warranty (FRW), pro rata warranty (PRW) and combined warranty policies and the most beneficial warranty scheme to the producer for the product having Pareto life time distribution. A Bayesian approach is used to determine the optimal warranty length based on the general progressive type-II censored data. The optimal warranty is obtained by maximizing the expected utility of the product. A numerical data is presented to exemplify the theory. A simulation study is carried out to check the effect of the hyper parameters on the optimal warranty length and the optimal value of expected utility. From our study we observed that the combined policy gives maximum utility followed by PRW and then by FRW for any choice of the prior parameters. Hence we suggest the producer to adopt the combined policy for such a product.

Summary: RNA binding motif protein 10 (RBM10) is a regulator of alternative splicing in apoptosis and inflammation. We discovered a splicing-independent function of RBM10 critical for the regulation of heart failure (HF). RBM10 is enriched in the heart and associates with Star-PAP (TUT1) to control the expression and 3′ end processing of cardiac mRNAs. The RBM10 RRM2 domain binds the Star-PAP catalytic domain, which directs Star-PAP activity toward polyadenylation. RBM10 binds the pre-mRNA UTR, assembles the Star-PAP complex, and guides this complex specifically to mRNAs encoding anti-hypertrophy regulators. Accordingly, we tested cellular hypertrophy in rat cardiomyoblasts and cardiac hypertrophy (CH) and the subsequent progression to HF in Wistar rat hearts. We demonstrated downregulation of RBM10 during CH and HF. Ectopic re-expression of RBM10 rescued cardiomyocyte hypertrophy. RBM10 depletion evoked a hypertrophic response in H9c2 cells. Our results establish an anti-hypertrophy mechanism mediated by RBM10 in the heart that is directly linked to HF. : Regulation at the mRNA 3′ UTR is a critical mechanism in the heart, but how it is accomplished during hypertrophy or heart failure is poorly understood. Indiscriminate use of antibiotics has led to the emergence of antibiotic‐resistant microbes and the loss of natural flora in aquaculture systems necessitating the ban of many of these chemotherapeutants in aquaculture. Actinobacteria play a profound role in the biogeochemical cycling in the marine environment and represent the principal source of secondary metabolites with antimicrobial property. In the present study, 98 marine‐derived actinomycete isolates were screened for antimicrobial activity against the common aquatic pathogens. A potent actinomycete isolate S26, identified as Streptomyces variabilis based on 16 S ribosomal RNA (rRNA) gene sequencing was then checked for the production of antibiotic in five different fermentation media and the one which showed maximum production was chosen for further study. Optimization of the fermentation medium for secondary metabolite production was carried out by response surface methodology (RSM) using DESIGN EXPERT. The analysis of variance (ANOVA) of the quadratic regression model demonstrated that the model was highly significant for the response concerned that is, antimicrobial activity as evident from the Fisher's F‐ test with a very low probability value [(P model>F) = 0.0001]. Of the 10 different solutions suggested by the software, the most suitable composition was found to be starch, 1.38%; soy powder, 0.88%; ammonium sulfate, 0.16% and salinity, 27.76‰. S. variabilis S26 cultured in the optimized production medium was applied in the Penaeus monodon larval rearing system and the total Vibrio count and survival rate were estimated. S. variabilis S26 treatment showed a significant reduction in vibrios and conferred better protection to P. monodon in culture system compared with control. [ABSTRACT FROM AUTHOR]

Background: People with diabetes have 10 to 30 times higher risk of lower limb amputation than people without the disease. 85% of non-traumatic amputations in diabetic patients begin with a foot ulcer. Therefore, early intervention is necessary to control the symptoms of diabetic peripheral neuropathy and improve balance. Resistance and aerobic training reduce diabetic symptoms by lowering HbA1C levels. In order to restore functional sensibility, improve adaptive functioning, and improve balance, sensory re-education and aerobics are combined. Therefore, the aim of this study is to find out the effect of Sensory re-education and aerobics on sensation and balance among diabetic peripheral neuropathy patients. Purpose: The Purpose of the study is to determine the effects of sensory re-education with aerobic training on sensory perception and balance in Diabetic peripheral neuropathy patients. Material and Method: This study included diabetic individuals of both genders between the ages of 45-60, low PAR-Q risk scores and Toronto clinical neuropathy scores (TCNS) between 6 and 11. A total of 20 participants were randomly selected and received sensory re-education with aerobics for six weeks. Results: Individuals with diabetic peripheral neuropathy experienced statistically significant improvement in TCNS from 9.3±1.25 to 4.5± 1.3, in BBS 48.5 ± 2.70 to 41.7±1.41, in TUG 8.12±1.28 to 6.95 ±1.30, with a P value of < 0.01. Conclusion: Sensory re-education with aerobic training for 6 weeks of duration reported improvement in sensory perception and balance among people with diabetic peripheral neuropathy patients.
