This study aimed to investigate differences and similarities of the pet-owner relationship and management of dogs and cats living in the same household. An online survey was completed by 1270 individuals who owned both dog(s) and cat(s). Most of the respondents owned only one dog (56.4%) but two or more cats (63.1%; p < 0.001). Moreover, cats were acquired earlier and were more likely to be neutered than dogs ( p < 0.001). The neutering rate was also affected by breed ( p < 0.001), age ( p < 0.05), and the number of pets owned ( p < 0.05). Two types of owners were identified according to how they managed their pets: owners who keep cats both indoors and outdoors and dogs indoors (65.3%) and owners who keep cats indoors and dogs outdoors (34.7%). The former group was characterized by a higher percentage of owners younger than 25 years ( p < 0.01), owners with more than one dog or cat, large dogs ( p < 0.001), and intact ( p < 0.04) and mix-breed cats ( p < 0.001). Finally, most respondents described the relationship with their cats (93.0%) and dogs (97.0%) as "loving" or "friendly". Keeping cats indoors ( p < 0.05) and allowing dogs and cats to sleep in the owners' bedroom were associated with a stronger perceived pet-owner bond. Implications for cat and dog welfare are discussed.

As livestock production grows to satisfy the global demand for animal products, understanding public attitudes towards different husbandry systems becomes essential for both animal welfare and socio-economic reasons. This study aimed to investigate people's emotional responses toward pictures of farm animals kept in intensive and extensive husbandry systems, their perception of animal welfare, and their choices as animal product consumers. A questionnaire that included demographic questions and photos of cows, pigs, chickens, and rabbits in both intensive and extensive systems was distributed electronically and physically and completed by 835 respondents. Photos of animals in intensive systems elicited more negative emotions, especially for pigs and rabbits ( p < 0.05), as opposed to extensive systems, which elicited more positive emotions, especially for chickens ( p < 0.001). Higher welfare levels were perceived for extensively farmed animals ( p < 0.001) and for cattle compared to all other species, regardless of the husbandry system ( p < 0.001). The quality of the emotional response was positively associated with welfare perception ( p < 0.001) and negatively associated with the importance given to welfare when purchasing animal products ( p < 0.001). Finally, the emotional response was found to be affected by gender, education, household composition, living area, pet ownership, and eating habits. The implications and limitations of these findings are discussed.

Cell-free miRNAs, called circulating miRNAs (cmiRNAs), can act in a paracrine manner by facilitating a diversity of signaling mechanisms between cells. Real-time qPCR is the most accepted method for quantifying miRNA expression levels. The use of stable miRNA endogenous control (EC) for qPCR data normalization allows an accurate cross-sample gene expression comparison. The appropriate selection of EC is a crucial step because qPCR data can change drastically when normalization is performed using an unstable versus a stable EC. To find EC cmiRNA with stable expression in search and rescue (SAR) working dogs, we explored the serum miRNome by Next-Generation Sequencing (NGS) at T0 (resting state) and T1 immediately after SAR performance (state of physiologically recovered stress). The cmiRNAs selected in the NGS circulating miRNome as probable ECs were validated by qPCR, and miRNA stability was evaluated using the Delta Ct, BestKeeper, NormFinder, and GeNorm algorithms. Finally, RefFinder was used to rank the stability orders at both T0 and T1 by establishing miR-320 and miR-191 as the best-circulating ECs. We are confident that this study not only provides a helpful result in itself but also an experimental design for selecting the best endogenous controls to normalize gene expression for genes beyond circulating miRNAs.

Globally, most people now own a pet. Scales have been developed to understand the impact of pet ownership on people's lives and to measure specific aspects of the owner-pet relationship. The Cat-/Dog-Owner Relationship Scale (C/DORS) is a tool developed to investigate this relationship in both dog and cat owners. The aim of the study was to refine and validate the C/DORS for cat owners in Italian. Exploratory Factor Analysis and Confirmatory Factor Analysis (CFA) were used to determine the most appropriate factor model. Construct validity was confirmed by correlating the C/DORS subscales with the Lexington Attachment to Pets (LAPS) subscales. Results confirmed the original structure of the English version (i.e., three factors: Pet Owner Interaction = POI, Perceived Emotional Closeness = PEC, Perceived Costs = PC) and CFA confirmed the structure of LAPS and C/DORS scales. Cronbach's alpha demonstrated the Italian version of the two scales to have good internal reliability for all domains. Owners of cats living exclusively indoors reported higher scores on POI and PEC compared to indoor/outdoor cats. Owning both cats and dogs was correlated with lower scores on POI, and fewer perceived costs (i.e., PC) of cat ownership. Finally, behaviour problems, not being neutered, and lack of previous experience with cat ownership were associated with lower scores on PC.

This review emphasizes the important role of cross-talk between P53 and microRNAs in physiological stress signaling. P53 responds to stress in a variety of ways ranging from activating survival-promotion pathways to triggering programmed cell death to eliminate damaged cells. In physiological stress generated by any external or internal condition that challenges cell homeostasis, P53 exerts its function as a transcription factor for target genes or by regulating the expression and maturation of a class of small non-coding RNA molecules (miRNAs). The miRNAs control the level of P53 through direct control of P53 or through indirect control of P53 by targeting its regulators (such as MDMs). In turn, P53 controls the expression level of miRNAs targeted by P53 through the regulation of their transcription or biogenesis. This elaborate regulatory scheme emphasizes the relevance of miRNAs in the P53 network and vice versa.

Our research explores serum extracellular circulating miRNAs (ecmiRNAs) involved in dog stress response immediately after the search and rescue (SAR) of missing people. The experimental plan considers four arduous SAR simulations. The SAR dogs are trained by the Alpine School of the Military Force of Guardia di Finanza (Passo Rolle, Italy). The First SAR Trial analyzed dog serum samples at rest time (T0), and immediately after SAR performance (T1) using the miRNome-wide screening next-generation sequencing (NGS). T1 versus T0 NGS results revealed a different expression level of let-7a and let-7f. Subsequently, in a large sample size including: 1st (n = 6), 2nd (n = 6), 3rd (n = 6), and 4th (n = 4) trials, let-7a and let-7f were validated by qPCR. Bioinformatics analysis with TarBase (v.8) and the Diana-mirPath (v.3) revealed a functional role of let-7a and let-7f in the p53 pathway to restore cellular homeostasis. Let-7a and let-7f, highly expressed at T1, could stop MDMs-p53 inhibition inducing the p53 increase in level. In addition, let-7a and let-7f, via p53 post-transcriptional regulation, buffers p53 transcription spikes. During SAR stress, the possibility of p53 preconditioning could explain the phenomenon of "stress hardening" where the tolerance of particular stress increases after preconditioning., (© 2022. This study aimed to assess the heart rate (HR) responses of avalanche SAR dogs and handlers under working field conditions. Thirteen SAR units (dogs and handlers) performed an exercise ( Endurance ) consisting of approximately 5.5 km of rough tracks through deep snow, at an altitude of 1991-2250 m.a.s.l. The exercise was repeated twice for each of the two different tracks. Both handlers and dogs were equipped with a global positioning satellite/heart rate (GPS/HR) system (Polar ® ). Multivariable models were used to evaluate the effects of environmental (i.e., gradient, altitude, track, and time) and intrinsic (i.e., speed, repetition, and breed) factors on changes from baseline HR (Δ%HR). The dog's Δ%HR was greater in the flat and uphill compared with downhill, and increased progressively as the speed increased ( p < 0.001). Moreover, it rose at altitudes above 2100 m.a.s.l. and peaked after 30 min of the Endurance activity ( p < 0.01). These findings indicated that HR monitors could be a valuable tool to contribute to the evaluation of avalanche dogs' fitness in their real working environment. In contrast, the lack of correlation between the dogs' and handlers' HR changes suggests that handlers might not perceive the physical conditions of their dog in real-time. Thus, implementing protocols to monitor avalanche SAR dogs' fitness using a GPS/HR monitoring system could help handlers to tailor the training and workload and to detect the risk factors for physical distress of working dogs.

Overweight is common in cats and has health and welfare implications. This study aimed to assess potential predictive/protective factors for feline overweight associated with owner management and their relationship with cat behavior and welfare. A questionnaire was administered to 197 owners to collect information about cat demographics, management, environment, dietary habits and behavior. The feline Body Mass Index was recorded for each cat. Univariate logistic regression was used to evaluate the association of parameters with overweight cats. Variables with a p -value < 0.10 at univariable analyses were selected for the multivariable model. Most cats were mixed- breed, 1-7 years old and neutered; 51.3% were overweight. Age higher than 7 years, neutering, being alone all day and being stressed were predictive for overweight ( p < 0.05). Conversely, the presence of other animals had a protective effect ( p < 0.05). A general disagreement between owners' perception and clinical evaluation of cats' body condition was a common and significant risk factor for overweight (OR = 8.532, 95% CI = 4.073-17.875; p < 0.001). According to the owners, many veterinarians did not inform them about their cat being overweight nor about the risks ( p < 0.001). This study provides helpful information on the influence of management and environment on cats' body condition and its relationship with cat welfare.

The Cat/Dog-Owner Relationship Scale (C/DORS) can be administered to both dog and cat owners. However, the scale as a whole has never been validated on a sample of dog owners. Furthermore, it has never been translated into Italian. The aim of this study was to translate the C/DORS into Italian, modify its response scale in order to improve the degree of response variability, and test its validity and reliability on a sample of dog-owners. Exploratory factor analysis revealed the same three-factor structure (Perceived Emotional Closeness = PEC, Pet-Owner Interactions = POI, Perceived Costs = PC) as the original English version, although some items had to be removed because of low- or cross-loadings. The validity of the construct was confirmed by confirmatory factor analysis, by the correlations between each of the subscales and the C/DORS total score, and by the correlations with the Lexington Attachment to Pets Scale. Cronbach's α values for each subscale were above acceptable levels. Student owners scored higher on PEC and POI than owners with other occupations. Owners of dogs with behavioural problems scored lower on PEC and higher on PC. Keeping the dog outdoor was associated with lower POI. Finally, pet dog owners scored higher on PEC than AAI dog owners.

Resilience is conceived as a dynamic developmental process involving the achievement of positive adaptation within the context of significant adversity. Resilience is not a unique ability but rather a set of capacities of a system put in place to absorb a disturbance and to reorganize while trying to retain the same function, structure, and identity. This review describes the characteristics and the molecular mechanisms of resilience to understand the core elements of resilience and its indicators. The objectives of this review are: (1) to define some of the leading environmental stressors and clarify the mechanism of vulnerability or resilience outcomes; (2) to clarify some of the prominent epigenetic modulations mediating resilience or vulnerability as a stress response; (3) to highlight the neural mechanisms related to stress resilience since the central nervous system is a highly dynamic structure characterized by an everlasting plasticity feature, which therefore has the opportunity to modify resilience. The review aims to introduce the reader to the concept of resilience seen as an ability acquired in life and not only inherited from birth.

In mammals, serotonin (5-HT) levels depend on the availability of tryptophan (TRP). Low 5-HT concentrations have been linked to behavioural disorders in dogs. This study aimed at investigating possible differences in dogs' serum TRP and 5-HT concentrations according to their behavioural response to a potentially stressful procedure. Thirty-nine physically healthy shelter dogs, 15 females and 24 males, mean age = 5.6 years, were categorized by a certified veterinary behaviourist according to their behavioural response to medical examination and blood collection, in: relaxation, stress signals, tension without growling, tension with growling, escape attempts, and aggression attempts. Extraction and quantification of 5-HT and TRP were performed using a HLPC method. Data were statistically analysed, applying Chi-square and Spearman tests. Results showed no significant difference in TRP (χ 2 = 2.084, p = 0.555) nor 5-HT (χ 2 = 0.972, p = 0.808) serum concentrations among different categories of dogs; however, some categories were underrepresented (relaxation = 20.5%, stress signals = 30.8%, tension without growling = 43.6%, tension with growling = 5.1%, escape attempts = 0%, aggression attempts = 0%). No correlation between serum TRP and 5-HT concentrations was found (ρ = 0.086, p = 0.602). Serum 5-HT levels do not seem to be associated with dogs' behavioural response to a stressful situation nor with serum TRP concentrations. The relationship between serum TRP and 5-HT concentrations and behaviour needs further research.
