Resumo A distrofia muscular de Emery-Dreifuss é uma doença neuromuscular hereditária rara. Suas manifestações começam principalmente na infância. As manifestações mais frequentes são fraqueza muscular progressiva, atrofia que geralmente se inicia na região escápulo-vertebral, estendendo-se posteriormente para a cintura pélvica e rigidez da coluna vertebral. Os pacientes também podem manifestar envolvimento cardíaco como palpitações, síncope, intolerância ao exercício, insuficiência cardíaca congestiva e distúrbios variáveis do ritmo cardíaco. 1 - 3 A presença e a gravidade dessas manifestações podem variar de acordo com o indivíduo e os subtipos da doença. 2 O envolvimento cardíaco é a característica mais preocupante desta doença, havendo alguns relatos da necessidade de transplante cardíaco nesta distrofia. 4 Abstract Emery-Dreifuss muscular dystrophy is a rare hereditary neuromuscular disease. Its manifestations begin primarily in childhood. The most frequent manifestations are progressive muscle weakness, atrophy that usually begins in the scapula-vertebral region, extending later to the pelvic girdle, and spinal stiffness. Patients can also manifest cardiac involvement as palpitations, syncope, exercise intolerance, congestive heart failure, and variable heart rhythm disturbances.1 - 3 The presence and severity of these manifestations can vary according to the individual and the disease’s subtypes. 2 Cardiac involvement is the most worrisome feature of this disease, and there are some reports of the need for heart transplantation in this dystrophy. 4

Orientador: Silvio Roberto Consonni Dissertação (mestrado) - Universidade Estadual de Campinas, Instituto de Biologia Resumo: Em células musculares, a transdução de sinais mecânicos da membrana sarcoplasmática ao núcleo e regulação da arquitetura nuclear são dependentes do complexo de ligação do nucleoesqueleto e citoesqueleto (LINC, do inglês linker of nucleoskeleton and cytoskeleton) ¿ composto pelas proteínas integrais das membranas interna e externa do envoltório nuclear ¿ que conecta e transmite as forças mecânicas do citoesqueleto ao núcleo através do envoltório. Os principais componentes do complexo LINC na membrana externa do envoltório nuclear são as proteínas nesprinas que se ligam ao citoesqueleto e ainda interagem com a porção C-terminal das proteínas domínio-SUN e Emerina na membrana interna do envoltório nuclear. Este conjunto de proteínas interage com as laminas A/C, que constituem a lâmina nuclear em contato com a cromatina. Assim, mutações nos genes que codificam as proteínas laminas A/C e Emerina resultam em doenças de espectro amplo, mas com fenótipos distintos, comumente referidas como laminopatias (ou envelopatias) nucleares, tais como: distrofia muscular de Emery-Dreifuss, distrofia muscular das cinturas e distrofia muscular congênita. A distrofia muscular de Emery-Dreifuss ligada ao X, causada por mutações no gene EMD que codifica a Emerina, clinicamente é caracterizada pelo enfraquecimento do músculo estriado esquelético progressivo, cardiomiopatia dilatada, arritmias cardíacas e contratura dos tendões. Assim, tendo em vista que a Emerina está envolvida em muitos processos celulares e intranucleares por meio de interações com vários parceiros moleculares, nosso estudo buscou compreender o papel da Emerina na biologia dos mioblastos C2C12. Para resolver essa questão, nós usamos o modelo de cultura celular in vitro e tecnologia de silenciamento gênico por shRNA para suprimir a expressão de Emerina e avaliar a morfologia das células C2C12 e expressão de genes relacionados ao citoesqueleto e fatores de transcrição. Nossa hipótese indica que a Emerina, quando ausente ou mutada poderia alterar o funcionamento, morfologia e composição dos mioblastos C2C12. A primeira parte deste estudo demostrou que o silenciamento gênico estabeleceu satisfatoriamente um modelo de estudo corroborado pelos resultados de imunofluorescência, microscopia eletrônica de transmissão, quantificação de área nuclear e taxa de proliferação entre os grupos shGFP (controle) e shEMD. A segunda parte deste estudo demonstrou variação na expressão de genes relacionados diretamente à interação com Emerina e ao citoesqueleto, o que corrobora a participação da Emerina na dinâmica celular de mecanotransdução. Em conclusão, nosso trabalho forneceu dados científicos de morfologia e expressão gênica que permitiram analisar o sucesso do silenciamento gênico e alterações fenotípicas dos mioblastos shEMD em estudo integrado entre alterações no núcleo celular e seus efeitos celulares e moleculares. Como perspectiva futura, por meio do uso deste modelo celular, tem-se a possibilidade de se compreender os efeitos da ausência de Emerina no ciclo celular de mioblastos e efeitos na sua diferenciação Abstract: In muscle cells, the transduction of mechanical signals from the sarcoplasmic reticulum membrane to the nucleus and regulation of nuclear architecture are dependent on the linker of nucleoskeleton and cytoskeleton (LINC) complex - composed of the integral proteins within the inner and the outer nuclear membranes ¿ that connects and transmits the mechanical forces of the cytoskeleton to the nucleus through the nuclear envelope. The main components of the LINC complex on the outer membrane of the nuclear envelope are the Nesprin proteins that bind to the cytoskeleton and still interact with the C-terminal portion of the SUN-domain and Emerin proteins on the inner membrane of the nucleus. This set interacts with lamin A/C, proteins that constitute the nuclear lamin in contact with the chromatin. Thus, mutations in the genes which encode the lamin A/C and Emerin proteins result in broad spectrum diseases, but with distinct phenotypes, commonly referred to as nuclear laminopathies (or envelopathies), such as: Emery-Dreifuss muscular dystrophy, limb-girdle muscular dystrophy and congenital muscular dystrophy. Emery¿Dreifuss muscular dystrophy linked to X, caused by mutations in the EMD gene that encodes Emerin, is clinically characterized by weakening of progressive skeletal striated muscle, dilated cardiomyopathy, cardiac arrhythmias, and tendon contracture. Thus, because Emerin is involved in many cellular and intranuclear processes through interactions with several molecular partners, our study sought to understand the role of Emerin in the biology of C2C12 myoblasts. To address this issue, we used the in vitro cell culture model and shRNA gene silencing technology to suppress emerin expression and to evaluate C2C12 cell morphology and expression of genes related to cytoskeleton and transcription factors. Our hypothesis indicates that Emerin, when it is absent or mutated, could alter the operation, morphology and composition of C2C12 myoblasts. The first part of this study demonstrated that gene silencing satisfactorily established a study model corroborated by the results of immunofluorescence, transmission electron microscopy, nuclear area quantification, and proliferation rate between the shGFP (control) and shEMD groups. The second part of this study demonstrated variation in the expression of genes directly related to the interaction with emerin and the cytoskeleton, which corroborates the participation of Emerin in the cellular dynamics of mechanotransduction. In conclusion, our work provided scientific data on morphology and gene expression that allowed us to analyze the success of gene silencing and phenotypic changes of shEMD myoblasts in an integrated study between alterations in the cell nucleus and its cellular and molecular effects.
