The energy consumption of hospital is increasing year by year, and the energy consumed in HAVC accounts for larger proportion. This paper took the inpatient building in hot summer and warm winter climate zone for research object, established the theory model of cooling and heating load, designed the orthogonal simulation experiments and simulated the building load using DEST. This paper aimed to research the influence of beds number, added beds rate, patient accompany system, visiting institution on the building load. The simulation results indicate when the beds number and added beds rate is larger, the patient accompany system is not strict, the building load is larger. It will be smaller conversely. The important order of the influence factor is as follows: beds number > patient’s accompany system > addition beds rate > visiting institution. Beds number and patient accompany system have a remarkable influence on the building load (P 0.05). This paper proposes using per inpatient bed energy consumption to evaluate energy consumption in inpatient building when patient accompany system of the hospital is the same. It will be more scientific.

Inspired by the traditional Chinese herbal pair of Polygala tenuifolia-Acori Tatarinowii for treating epilepsy, 33 novel substituted cinnamic α-asaronol esters and analogues were designed by Combination of Traditional Chinese Medicine Molecular Chemistry (CTCMMC) strategy, synthesized and tested systematically not only for anticonvulsant activity in three mouse models but also for LDH inhibitory activity. Thereinto, 68–70 and 75 displayed excellent and broad spectra of anticonvulsant activities with modest ability in preventing neuropathic pain, as well as low neurotoxicity. The protective indices of these four compounds compared favorably with stiripentol, lacosamide, carbamazepine and valproic acid. 68–70 exhibited good LDH1 and LDH5 inhibitory activities with noncompetitive inhibition type, and were more potent than stiripentol. Notably, 70, as a representative agent, was also shown as a moderately positive allosteric modulator at human α1β2γ2 GABAA receptors (EC50 46.3 ± 7.3 μM). Thus, 68–70 were promising candidates for developing into anti-epileptic drugs, especially for treatment of refractory epilepsies such as Dravet syndrome.

Amino acid networks (AANs) analysis is a new way to reveal the relationship between protein structure and function. We constructed six different types of AANs based on iron superoxide dismutase (Fe-SOD) three-dimensional structure information. These Fe-SOD AANs have clear community structures when they were modularized by different methods. Especially, detected communities are related to Fe-SOD secondary structures. Regular structures show better correlations with detected communities than irregular structures, and loops weaken these correlations, which suggest that secondary structure is the unit element in Fe-SOD folding process. In addition, a comparative analysis of mesophilic and thermophilic Fe-SOD AANs’ communities revealed that thermostable Fe-SOD AANs had more highly associated community structures than mesophilic one. Thermophilic Fe-SOD AANs also had more high similarity between communities and secondary structures than mesophilic Fe-SOD AANs. The communities in Fe-SOD AANs show that dense interactions in modules can help to stabilize thermophilic Fe-SOD.

Incubation with different concentrations of different carbon, nitrogen and metal ion sources was found to significantly influence Shiraia sp. SUPER-H168 laccase production. Based on the magnitude of the range, the factors were ranked as follows: starch > Cu2+ > yeast powder for laccase production. The optimal combination of factors was 20 g/L starch, 4 g/L yeast powder and 0.6 mM Cu2+. Other fermentation media and conditions were also studied. The maximal laccase yield of Shiraia sp. SUPER-H168 varied from 5,058 to 1.105 × 105 U/L. Shiraia sp. SUPER-H168 laccase DNA and its corresponding full-length cDNA were cloned and characterized; furthermore, the Shiraia laccase gene sequence is reported for the first time. The coding region of Shiraia sp. SUPER-H168 laccase consisted of a 1,848 bp open reading frame encoding a protein of 615 aa with a 21 aa signal peptide sequence. Three introns of 50, 49 and 51 bp in length were located at upstream of the N terminus. The molecular mass (66.7 kDa) and isoelectric point (7.63) were predicted. Additionally, the protein contained six potential N-glycosylation sites (Asn-X-Ser/Thr) and four conserved copper-binding domains. The neighbor joining phylogenetic tree demonstrated that Shiraia sp. SUPER-H168 laccase is an ascomycetes laccase closely related to Phoma sp. UHH 5-1-03 laccase.
