Objective To investigate the influence of transient receptor potential cation channel 6 (TRPC6) on renal tubulointerstitial inflammation in mice with diabetic kidney disease (DKD) and its mechanism. Methods A total of 36 male C57/BL6J mice, aged 6 weeks, were randomly divided into control group (group A), DKD model group (group B), DKD+normal saline intervention group (group C), DKD+mitophagy activator intervention group (group D), DKD+negative control lentivirus transfection group (group E), and DKD+TRPC6 knockdown lentivirus transfection group (group F), with 6 mice in each group. The mice in groups A and B were respectively given intraperitoneal injection of 0.1 mmol/L citrate buffer and 10 g/L streptozotocin (hereinafter referred to as administration); the mice in groups C and D were respectively given normal saline and 10 mmol/L urolithin A by gavage in addition to the treatment in group B; the mice in groups E and F were respectively injected with negative control lentivirus and TRPC6 knockdown lentivirus via the tail vein in addition to the treatment in group B. The levels of fasting blood glucose (FBG), urinary albumin-to-creatinine ratio (ACR), and blood urea nitrogen (BUN) were measured at week 12 after administration; PAS staining was used for the observation and scoring of renal tubular injury; RT-qPCR was used to measure the mRNA expression levels of inflammatory factors [interleukin-1β (IL-1β), monocyte chemoattractant protein-1 (MCP-1), and tumor necrosis factor-α (TNF-α)] in renal tissue; immunohistochemical staining was used to measure the protein expression level of TRPC6 in renal tissue, and Western blotting was used to measure the relative expression levels of TRPC6, LC3B, P62, PINK1, and Parkin in renal tissue; transmission electron microscopy was used to observe the change in the number of mitophagosomes in renal tubular cells of mice. HK-2 cells were divided into high glucose+TRPC6 siRNA transfection+DMSO intervention group (group G, treated with TRPC6 siRNA+35.0 mmol/L glucose+0.06% DMSO) and high glucose+TRPC6 siRNA transfection+mitophagy inhibitor intervention group (group H, treated with TRPC6 siRNA+35.0 mmol/L glucose+12 μmol/L oroxylin A), and then RT-qPCR was used to measure the mRNA expression levels of inflammatory factors (IL-1β, MCP-1, and TNF-α) in cells. Results At week 12 after administration, compared with group A, group B had significantly higher whole blood FBG, ACR, BUN, renal tubular injury score, mRNA expression levels of inflammatory factors, and protein expression levels of TRPC6 and P62 in renal tissue (t=2.77-13.61,P

Energy is the foundation of a strong country and an engine to promote high-quality and sustainable economic development. This study summarizes the experiences of major energy powerhouses worldwide regarding energy development, believing that they have achieved excellent performance in multiple dimensions such as the stable and diversified energy supply, low-carbon and efficient energy consumption, leading innovation in energy technologies, orderly and open energy market, and completeness in energy governance systems. Moreover, this study reviews the advantages and disadvantages of energy development in China and proposes a Chinese path to becoming an energy powerhouse, involving staged objectives, key tasks, and measures. The basic conclusion is that we need to increase the clean utilization of coal resources to exploit the role of coal in ensuring energy security. Meanwhile, it is crucial to improve China’s ability of oil and gas self-sufficiency by strengthening the exploration and development of oil and gas resources and promoting the lacustrine shale oil revolution. It is also important to scale up the renewable energy sources and optimize the layout of clean energy sources such as hydrogen, thereby improving energy security and implementing the carbon reduction goals through multi-energy coordination

Abstract Triboelectric nanogenerators (TENGs) have emerged as a promising approach for generating electricity and providing electrical stimuli in medical electronic devices. Despite their potential benefits, the clinical implementation of TENGs faces challenges such as skin compliance and a lack of comprehensive assessment regarding their biosafety and efficacy. Therefore, further research is imperative to overcome these limitations and unlock the full potential of TENGs in various biomedical applications. In this study, we present a flexible silk fibroin-based triboelectric nanogenerator (SFB-TENG) that features an on-skin substrate and is characterized by excellent skin compliance and air/water permeability. The range of electrical output generated by the SFB-TENG was shown to facilitate the migration and proliferation of Hy926, NIH-3T3 and RSC96 cells. However, apoptosis of fibroblast NIH-3T3 cells was observed when the output voltage increased to more than 20 V at a frequency of 2 Hz. In addition, the moderate electrical stimulation provided by the SFB-TENG promoted the cell proliferation cycle in Hy926 cells. This research highlights the efficacy of a TENG system featuring a flexible and skin-friendly design, as well as its safe operating conditions for use in biomedical applications. These findings position TENGs as highly promising candidates for practical applications in the field of tissue regeneration.

Abstract Lately, carbazole‐based self‐assembled monolayers (SAMs) are widely employed as effective hole‐selective layers (HSLs) in inverted perovskite solar cells (PSCs). Nevertheless, these SAMs tend to aggregate in solvents due to their amphiphilic nature, hindering the formation of a monolayer on the ITO substrate and impeding effective passivation of deep defects in the perovskites. In this study, a series of new SAMs including DPA‐B‐PY, CBZ‐B‐PY, POZ‐B‐PY, POZ‐PY, POZ‐T‐PY, and POZ‐BT‐PY are synthesized, which are employed as interfacial repairers and coated atop CNph SAM to form a robust CNph SAM@pseudo‐planar monolayer as HSL in efficient inverted PSCs. The CNph SAM@pseudo‐planar monolayer strategy enables a well‐aligned interface with perovskites, synergistically promoting perovskite crystal growth, improving charge extraction/transport, and minimizing nonradiative interfacial recombination loss. As a result, the POZ‐BT‐PY‐modified PSC realizes an impressively enhanced solar efficiency of up to 24.45% together with a fill factor of 82.63%. Furthermore, a wide bandgap PSC achieving over 19% efficiency. Upon treatment with the CNph SAM@pseudo‐planar monolayer, also demonstrates a non‐fullerene organic photovoltaics (OPVs) based on the PM6:BTP‐eC9 blend, which achieves an efficiency of 17.07%. Importantly, these modified PSCs and OPVs all show remarkably improved stability under various testing conditions compared to their control counterparts.

Objective To investigate the effect of nicotinamide adenine dinucleotide-dependent deacetylase 1 (Sirt1) on high glucose-induced exosome release from podocytes. Methods Immortalized mouse podocytes MPC5 were divided into six groups: normal glucose group (5.5 mmol/L glucose, group A), high mannitol group (5.5 mmol/L glucose+24.5 mmol/L mannitol, group B), high glucose group (30.0 mmol/L glucose, group C), high glucose+Sirt1-overexpressed lentivirus transfection group (Sirt1-overexpressed lentivirus transfection+30.0 mmol/L glucose, group D), high glucose+negative lentivirus transfection group (negative lentivirus transfection+30.0 mmol/L glucose, group E), and high glucose+exosome secretion inhibitor group (GW4869+30.0 mmol/L glucose, group F). Western blot was used to analyze the expression levels of Nephrin, Podocin, Sirt1, CD63, CD81, and Alix in each group. Real-time quantitative polymerase chain reaction was used to analyze the expression level of Sirt1 mRNA in D and E group. The morphology of podocyte exosomes was observed by a transmission electron microscope. The particle size and concentration of exosomes were determined by nanoparticle tracking analysis. Results The results of RT-qPCR showed that the relative expression of Sirt1 mRNA was significantly increased in group D compared with that in group E (t=14.580,P0.05). Conclusion Loss of Sirt1 in high glucose-treated podocytes promotes exosome secretion and podocyte injury.

Instruments for assessing secondary students' conceptual understanding of core concepts in biology are needed by educational practitioners and researchers alike. Most instruments available for secondary biology (years 9 to 12) focus only on highly specific biological concepts instead of multiple core concepts. This study describes the development of a 25-item instrument designed to fill this gap, the High School Biology Concept Inventory (HS-BCI). The HS-BCI not only assesses student knowledge of key biological concepts but also alternative conceptions. Using Rasch theory, the initial instrument was constructed from a pool of 61 instrument items using test results from 1015 students. The final 25-item instrument was validated with 1955 students. The results provide reliability and validity evidence for the HS-BCI. The findings suggest that it can be utilized to assess both conceptual knowledge and alternative conceptions.

Point clouds are essential 3D data representations utilized across various disciplines, often requiring point cloud completion methods to address inherent incompleteness. Existing completion methods like SnowflakeNet only consider local attention, lacking global information of the complete shape, and tend to suffer from overfitting as the model depth increases. To address these issues, we introduced self-positioning point-based attention to better capture complete global contextual features and designed a Channel Attention module for adaptive feature adjustment within the global vector. Additionally, we implemented a vector attention grouping strategy in both the skip-transformer and self-positioning point-based attention to mitigate overfitting, improving parameter efficiency and generalization. We evaluated our method on the PCN dataset as well as the ShapeNet55/34 datasets. The experimental results show that our method achieved an average CD-L1 of 7.09 and average CD-L2 scores of 8.0, 7.8, and 14.4 on the PCN, ShapeNet55, ShapeNet34, and ShapeNet-unseen21 benchmarks, respectively. Compared to SnowflakeNet, we improved the average CD by 1.6%, 3.6%, 3.7%, and 4.6% on the corresponding benchmarks, while also reducing complexity and computational costs and accelerating training and inference speeds. Compared to other existing point cloud completion networks, our method also achieves competitive results.

This study explores parents' views of science as a family sociocultural background that influences how parents support children's science talk as they engage in a science activity together at home. We focus on Indonesian families as they have distinct sociocultural characteristics that may yield different parent-child interactions than their Western counterparts in informal science learning settings. A microethnography approach and thematic discourse analysis are employed to capture the parent-child science talk and interactions. Findings show that Indonesian parent--child interactions are directive and collectivist in nature. Parents tend to lead learning at home, and the other family members voluntarily participate. Through their science talks, Indonesian parents engaged children in the science activity using explanatory talk, corrective feedback, and real-world connections. Throughout the interactions, the parents emphasized particular science knowledge and practices based on their views of science. We present three cases where the parents viewed science as a hypothesis-testing practice, as knowledge related to everyday phenomena, and as an inference-making process. Their talks and support for children's learning varied due to these different views of science. The study adds to the limited literature on parent-child interactions in informal science learning in non-Western contexts.

We investigate students' eye movements when they solve sequential and simultaneous synthesis physics problems. In sequential synthesis problems, multiple events occur chronologically, whereas in simultaneous synthesis problems, multiple events take place concurrently. We captured students' eye fixations on each problem diagram and recorded their verbal descriptions of problem-solving approaches. Results indicate that for sequential synthesis tasks, students fixated heavily and frequently on regions of a diagram where information is unknown or on empty space related to future events. Further, most students could invoke all relevant principles and intermediate variables to solve the sequential problems. However, for simultaneous synthesis tasks, students prioritised aspects of the diagram where information is given in the problem statement. Moreover, they mostly invoked only one fundamental principle to solve the problems by either using the values given in the statement or identifying one relevant intermediate variable. These findings may be explained by the differences in the spatiotemporal occurrence of the multiple events in the two types of synthesis problem. The chronological events in sequential synthesis tasks seem to support students' sensemaking, but the concurrent events of simultaneous synthesis tasks do not.
