A simple UV spectrophotometric analytical method has been developed for simultaneous estimation of Ivabradine HCl and Bisoprolol used in the treatment of angina pectoris. Modied simultaneous equation method was developed with double beam UV-Visible spectrophotometer using water as a solvent. The method was found to be linear in the range of 4-16 µg/ ml of Bisoprolol and 08-32 µg/ ml of Ivabradine. Percentage recovery for bisoprolol and Ivabradine by this method was found in the range of 96.43-100.60% and 95.80-100.10 %, respectively. The method was validated with respect to accuracy, precision, specicity and robustness as per ICH guideline. The method was applied for the estimation of laboratory prepared mixture and was found to be accurate and precise.

A second-order derivative spectrophotometric method is developed and validated for the simultaneous estimation of Azilsartan Medoxomil and Cilnidipine in Tablet dosage form. The determination of Azilsartan Medoxomil in the presence of Cilnidipine was achieved by measuring the second-order derivative signals at 231 nm, while the second-order derivative signal at 223 nm was measured for the determination of Cilnidipine. The linear dynamic ranges were 4–20 μg /ml for Azilsartan Medoxomil and 1–5 μg/ml for Cilnidipine, the calibration graphs' correlation coefficient was 0.9998, their n-value was 5, their precision (%RSD) was 1.43%, and their accuracy (0.9%) was satisfactory. The detection limits were found to be 2.46 and 1.57 μg/ml for Azilsartan Medoxomil and Cilnidipine, respectively. A precise, accurate and sensitive high-performance thin-layer chromatographic method has been developed and validated for the simultaneous determination of montelukast and bilastine in a tablet dosage form which is used in the treatment of allergic rhinitis. The proposed method used aluminum plates pre-coated with silica gel 60F254 as the stationary phase and acetonitrile–ethyl acetate–ammonia (4:6:0.1, V/V) as the mobile phase for separation purpose. Densitometric detection was carried out at 282 nm in ultraviolet. Compact spots of montelukast and bilastine were obtained at RF 0.53 and 0.27, respectively. Linear responses were shown by the detector over the range of 100‒500 ng/band for montelukast and 200‒1000 ng/band for bilastine. The proposed method was validated as per the International Council for Harmonisation guidelines. The method was applied for the estimation of both drugs in tablet formulation and their synthetic mixture.

A specific, accurate, precise, and reproducible liquid chromatographic method has been developed and validated for the estimation of acamprosate calcium and baclofen in combination. The separation was achieved using stationary phase Phenomenex C18 column (150 mm× 4.6 mm.) in isocratic mode, with mobile phase containing 0.05 M potassium dihydrogen orthophosphate buffer (pH 7) : acetonitrile (10:90 V/V), at a flow rate of 1.0 mL/min and effluents were monitored at 210 nm. The retention time of acamprosate calcium and baclofen were found to be 1.9 min and 5.3 min, respectively. The linearity for acamprosate calcium and baclofen were in the range of 2-64 µg/mL and 1.2- 38.4 µg/mL, respectively. The method was validated as per ICH guideline. The recoveries of Acamprosate calcium and baclofen were found in the range of 98.90 - 100.13 % and 98.60 -100.02 %, respectively. The objective of the present investigation is to enhance the dissolution and flow properties of raloxifene hydrochloride (RXH), a biopharmaceutical classification system class II drug. Melt dispersion of RXH with polyethylene glycol (PEG) 6000 was prepared by the fusion method. The melt dispersion was then adsorbed onto a porous adsorbent, Neusilin, by the melt adsorption method. Response surface methodology was employed to establish the design space for formulation variables such as the ratio of RXH to PEG 6000 in melt dispersion and amount of porous adsorbent to melt dispersion. Differential scanning calorimetry, scanning electron microscopy, X-ray diffraction, Fourier-transform infrared spectroscopy, and accelerated stability techniques were utilized to characterize formulations. Negative Gibbs free energy values indicated spontaneous solubilization of RXH in PEG 6000. The time required for 80% of drug release from optimized formulation was20 min compared with plain RXH. Accelerated stability studies confirmed the stabilization of amorphous melt dispersion in nanopores (nanoconfinement) of inorganic silicate Neusilin. Melt dispersion, adsorbed on porous carriers, is a promising technique to improve the dissolution characteristic as well as flow properties of drug molecules.

A sensitive, accurate, precise, and robust high-performance thin-layer chromatography (HPTLC) method has been developed for the simultaneous quantification of empagliflozin, linagliptin, and metformin HCl in bulk and synthetic mixture. The method applied HPTLC aluminum plates pre-coated with silica gel 60 F254 as the stationary phase along with n-butanol:water:glacial acetic acid (6:3:1, v/v) as the mobile phase. The Rf values of empagliflozin, linagliptin, and metformin HCl were found to be 0.73, 0.52, and 0.33, respectively. Densitometric analysis was carried out in the absorbance mode at 223 nm. The method was obtained linear in the range of 0.1–0.7 μg/band for empagliflozin, 0.05–0.35 μg/band for linagliptin, and 10–70 μg/band for metformin HCl, respectively. From synthetic mixture, the percentage (%) amounts of all three drugs were found to be more than 98% by the proposed HPTLC method. The method was validated as per the International Conference on Harmonisation (ICH) guideline Q2(R1).

A sensitive and precise high-performance thin-layer chromatographic (HPTLC) method has been developed for the simultaneous estimation of mirabegron and solifenacin succinate in combination. The method employed HPTLC aluminum plates pre-coated with silica gel 60 F254 as the stationary phase and methanol—ethyl acetate—triethylamine (8:2:0.1, V/V) as the mobile phase. The RF value was observed to be 0.76 and 0.56 for mirabegron and solifenacin succinate, respectively. Densitometric analysis was carried out in the absorbance mode at 222 nm. The method was linear in the range of 2–5.5 µg per band for mirabegron and 0.4–1.1 µg per band for solifenacin succinate. An accurate, sensitive, reproducible and precise stability indicating thin layer chromatographic method and UV-Visible spectroscopic method was established for the amelioration and validation of neuroprotective agent edaravone in bulk and its pharmaceutical dosage form. Edaravone injection is used to treat amyotrophic lateral sclerosis (ALS, Lou Gehrig's disease; a condition in which the nerves that control muscle movement slowly die, causing the muscles to shrink and weaken). Edaravone injection is in a class of medications called antioxidants. The high Performance thin layer chromatographic method was developed with proposed chromatographic condition with mobile phase containing n-Hexane: ethyl acetate (7:3, v/v). Accomplishment of UV–Visible spectroscopic determination was done at wavelength maxima of 245 nm using methanol as a solvent. The linearities were in the range of 1-9 µg/ml for UV-Visible spectroscopic method and 100-1400 ng/band for TLC method, respectively. Validation of proposed method has been accomplished with respect to linearity, accuracy, precision, specificity and robustness. For acid and alkali hydrolysis, chemical oxidation, dry heat degradation and photolytic degradation conditions were performed using stock solution of EDA, and quantification which has been achieved by proposed TLC method. Owing to sensitivity, promptness and accuracy of methods, we rely on both intended methods which will be useful for the regular quality control analysis and quantification of drug in bulk and pharmaceutical dosage form.

The proposed sensitive, precise, and accurate reverse phase-liquid chromatography (RP-LC) method is used for simultaneous estimation Amlodipine besylate, Rosuvastatin calcium and Fimasartan potassium trihydrate in bulk and synthetic mixture which is under phase 3 trial. An accurate and cost effective isocratic approach is employed by using Phenomenex C-18 column (250 mm × 4.6 mm, 5 μm). A method was developed and validated by focusing on ICH Q2 (R1) guideline parameters, efficient separation of all three drugs were obtained by optimised solvent mixture of acetonitrile and 0.02 M potassium dihydrogen phosphate buffer (49:51, v/v) pH 3.0 adjusted with 1% O—phosphoric acid used as mobile phase, flow rate was maintained 1 ml/min and analysis was carried out by using PDA detector at common wavelength 242 nm. The linearity ranges were found to be 1–7 μg/mL for Amlodipine besylate with correlation coefficient (r2) 0.9966, 2–12 μg/mL for Rosuvastatin calcium with correlation coefficient (r2) 0.9998 and 6–36 μg/mL for Fimasartan potassium trihydrate with correlation coefficient (r2) 0.9990, respectively. The proposed LC method has potential qualitative as well as quantitative applications for simultaneous estimation of Amlodipine besylate, Rosuvastatin calcium and Fimasartan potassium trihydrate in bulk and synthetic mixture.

A new, accurate and precise high performance thin layer chromatographic (HPTLC) method has been developed for simultaneous analysis of metformin hydrochloride and teneligliptin in tablet formulation. The chromatographic separations were achieved on TLC plates precoated with silica gel G60 F254 and the plates were developed with 0.5% ammonium acetate-n propanol-ammonia (4.5:5:0.5, v/v/v) as mobile phase. Detection and evaluation of chromatograms were performed densitometrically at 252 nm. The retention factor of metformin hydrochloride and teneligliptin was found to be 0.2 and 0.63, respectively. The method was found to be linear in the range of 100-1200 ng per spot and 200-2400 ng per spot for metformin and teneligliptin, respectively. The method was validated with respect to precision, accuracy, specificity and robustness as per ICH guidelines Q2 (R1) and the results were found to be within the acceptance criteria. The method was successfully applied for the analysis of drugs in combined tablet dosage form.

The method employed HPTLC aluminum plates pre-coated with silica gel 60 F254 as the stationary phase, while the solvent system was toluene‒methanol‒ethyl acetate‒ammonia (6.5:2.5:1:0.1, v/v). The RF values were observed to be 0.43 and 0.60 for clozapine and aripiprazole, respectively. Densitometric analysis was carried out in absorbance mode at 218 nm. The method was linear in the range of 200–1600 ng per band for clozapine and 100-800 ng per band for aripiprazole. The stress degradation study was performed, and it was found that clozapine was susceptible to acid hydrolysis, base hydrolysis, and photolytic degradation study. Aripiprazole was susceptible to oxidative stress degradation study. The method was validated and applied successfully for the estimation of both drugs in the synthetic mixture.

A precise and accurate HPTLC method for simultaneous estimation of pioglitazone HCl (PIO) and alogliptin benzoate (ALO) in synthetic mixture has been developed. The stationary phase selected was Pre-coated TLC plates (silica gel G60 – F254 aluminium sheet) and the mobile phase selected was n-butanol : water : acetic acid (6 : 3 :1 V/V/V). The separated spots were densitometrically analyzed in absorbance mode at 270 nm. The Rf value of PIO and ALO were found to be 0.70 and 0.39, respectively. The calibration curve of PIO and ALO were plotted in the range of 400 – 1600 ng/band, which showed the linear correction coefficient (r2) 0.9971 for PIO and 0.998 for ALO, respectively. The stock solutions of both the drugs were subjected to acid and alkali hydrolysis, chemical oxidation, dry heat degradation and photo degradation. The degraded product peaks were well resolved from the pure drug peak with significant difference in their Rf values. Stressed samples were assayed using developed HPTLC method. The method was validated as per ICH guideline Q2 (R1) and applied successfully for determination of both drugs in synthetic mixture.

A sensitive, selective and precise high performance thin layer chromatographic method has been developed and validated for the quantification of Brexpiprazole in bulk drug and in pharmaceutical dosage form. The method employed HPTLC aluminum plates (pre-coated with silica gel 60 F254) as stationary phase while n-butanol was used as mobile phase. The Rf value of Brexpiprazole was observed to be 0.38. The densitometric analysis was carried out in absorbance mode at 215 nm. The linear regression analysis data for the calibration plots showed a good linear relationship for Brexpiprazole over a concentration range of 200–1,600 ng band−1. The limit of detection and limit of quantification for Brexpiprazole was found to be 66 and 200 ng band−1. To find out the possible degradation pathway, forced degradation studies were performed. The stock solutions of Brexpiprazole (1,000 μg mL−1) were subjected to acid and alkali hydrolysis, chemical oxidation, dry heat degradation and photo degradation. The drug was found to be susceptible to acid and alkali hydrolysis, chemical oxidation, photo degradation and dry heat. The degraded product peaks were well resolved from the pure drug peak with significant difference in their Rf values. Stressed samples were analyzed using developed HPTLC method. The proposed method was validated with respect to linearity, accuracy, precision and robustness. The method was successfully applied to the estimation of Brexpiprazole in marketed formulation and determination of content uniformity of tablet formulation. Statistical analysis showed that the method is repeatable, selective, and precise.

A simple, sensitive and precise high performance thin layer chromatographic (HPTLC)method has been developed for the estimation of ondansetron (OND) and ranitidine (RAN) in combination. The method was employed on thin layer chromatography (TLC) and aluminium plates were precoated with silica gel 60 F254 as the stationary phase, while the solvent system was methanol. The Rf values were observed to be 0.5 ± 0.02, and 0.3 ± 0.02 for OND and RAN, respectively. The separated spots were densitometrically analyzed in absorbance mode at 299 nm. This method was linear in the range of 25-300 ng/band for OND and 50-600 ng/band for RAN. The limits of detection for OND and RAN were found to be 3.47 and 1.83 ng/band, respectively. The limits of quantification for OND and RAN were found to be 10.53 and 5.55 ng/band, respectively. The proposed method was validated with respect to linearity, accuracy, precision and robustness. The method was successfully applied to the estimation of OND and RAN in combined dosage form.

A simple, accurate and precise first order derivative spectrophotometric method was developed for simultaneous determination of Metolazone (MET) and Ramipril (RAM) in their combined pharmaceutical dosage form. MET and RAM were quantified using first derivative responses at 285nm and 212nm respectively. Methanol was taken as a common solvent. A simple, sensitive, precise high-performance thin-layer chromatographic (HPTLC) method has been developed for estimating irbesartan and amlodipine besylate in combination. It involves TLC aluminium plates pre-coated with silica gel 60-F254 as the stationary phase and a solvent system of chloroform/toluene/methanol/acetic acid (6/2.5/1.5/0.5, v/v/v/v). The Rf values were 0.57 ± 0.02 for irbesartan and 0.30 ± 0.02 for amlodipine besylate. The separated spots were analysed densitometrically in absorbance mode at 244 nm. The results were linear in the range 50–500 ng per band for irbesartan and 400–900 ng per band for amlodipine besylate, with limits of detection of 14.4 ng per band and 12.7 ng per band, respectively. The limits of quantification were 47.4 ng per band for irbesartan and 42.0 ng per band for amlodipine. After irbesartan and amlodipine stock solutions were subjected to acid and alkaline hydrolysis, chemical oxidation, dry heat degradation and photodegradation, the peaks for degraded product were well resolved from that for pure drug, with significant differences in their Rf values when assayed by the HPTLC method. The method was validated with respect to linearity, accuracy, precision and robustness and was used successfully to estimate irbesartan and amlodipine besylate in a synthetic mixture.

Background Aspirin combination is prescribed for its thrombolytic activity where gastric ulceration is the major side effect of aspirin which can be prevented by combining it with proton pump inhibitor omeprazole. Present study describes development of analytical method for the estimation of aspirin and omeprazole in combination. Objective The aim of the present study was to develop and validate chromatographic method for simultaneous analysis of aspirin and omeprazole. Methods Isocratic, reversed phase stability indicating liquid chromatographic method was developed for the simultaneous determination of Aspirin and Omeprazole in combination. The separation was achieved on a Thermo Scientific Hypersil ODS (250 x 4.6 mm, 5 µm) column, kept at ambient temperature, using acetonitrile: methanol: 0.05 M phosphate buffer (40:5:55; pH 4 adjusted with 0.1% tri ethyl amine) as a mobile phase at a flow rate of 1 mL/min and UV detection was performed at 225 nm. Results The retention time was found to be 3.9 min for aspirin and 5.3 min for omeprazole. The method was observed to be linear in the range of 2 - 80 µg/mL for aspirin and 1 - 40 µg/mL for omeprazole, respectively. The proposed method was validated as per ICH guidelines Q2 (R1). The developed RP- HPLC method was successfully applied for the simultaneous estimation of aspirin and omeprazole in the presence of degradation products of both the drugs. Conclusion The present study describes liquid chromatographic method for the estimation of aspirin and omeprzole in combination. The method can be used for the analysis of stability samples and routine quality control samples.

A simple, sensitive, and precise high-performance thin-layer chromatographic method has been developed for the estimation of torsemide and amiloride hydrochloride in the pharmaceutical dosage form. Thin-layer chromatographic (TLC) aluminum plates precoated with silica gel 60 F254 were used as the stationary phase, while chloroform-methanol-ammonia (7.5: 3.5: 1, v/v) as mobile phase was used. The RF values observed were 0.46 ± 0.01 and 0.24 ± 0.01 for torsemide and amiloride hydrochloride, respectively. The densitometric analysis was carried out in absorbance mode at 286 nm. The method was linear in the range of 100–600 ng spot−1 for torsemide and 50–300 ng spot−1 for amiloride hydrochloride. The method was validated as per the International Conference on Harmonization (ICH) guidelines. The limit of detection and limit of quantitation were found to be 19.94 ng spot−1 and 100 ng spot−1, respectively, for torsemide. The limit of detection and limit of quantitation were found to be 12.86 ng spot−1 and 50 ng spot−1, respectively, for amiloride. The proposed method was successfully applied to the estimation of torsemide and amiloride hydrochloride in the pharmaceutical dosage form.

The RF values were observed to be 0.27 ± 0.02, 0.41 ± 0.03, and 0.61 ± 0.02 for paracetamol, dicyclomine hydrochloride, and mefenamic acid, respectively. The separated spots were densitometrically analyzed in absorbance mode at 290 nm. The method was linear in the range of 300–1800 ng band−1 for paracetamol, 400–2400 ng band−1 for dicyclomine hydrochloride, and of 150–1500 ng band−1 for mefenamic acid. The method was validated with respect to accuracy, precision, specificity, sensitivity, and robustness. The limits of detection for paracetamol, dicyclomine hydrochloride, and mefenamic acid were found to be 35, 109, and 42 ng band−1, respectively. The limits of quantification for paracetamol, dicyclomine and mefenamic acid were found to be 116, 360, and 140 ng band−1, respectively. The method was successfully applied to the estimation of all three drugs in combined tablet dosage form.

Simple, accurate, precise, reproducible, requiring no prior separation, and economical procedures for simultaneous estimation of benazepril hydrochloride (BEN) and Hydrochlorothiazide (HCT) in tablet dosage form have been developed. Simultaneous equation method employs for estimation of both drugs in methanol at 240 nm and 270 nm as two analytical wavelengths. BEN and HCT at their respective λmax (240 nm and 270 nm) show linearity in a concentration range of 2–12 μg/mL and 4–14 μg/mL. Recovery studies for BEN are 100.0–100.6% and 99.8–100.0% for HCT in the case of simultaneous equation method confirming the accuracy of the proposed method. The proposed method is recommended for routine analysis since it is rapid, simple, accurate and also sensitive and specific.

A sensitive, selective, and precise high-performance thin-layer chromatographic method has been developed and validated for the simultaneous determination of melitracen hydrochloride and flupentixol dihydrochloride both as a bulk drug and in formulation. The method employed HPTLC aluminium plates pre-coated with silica gel 60F-254 as the stationary phase while the solvent system was methanol: toluene: ammonia (8: 2: 0.3, v/v/v). The Rf values of melitracen hydrochloride and flupentixol dihydrochloride were observed to be 0.79 ± 0.08 and 0.88 ± 0.05, respectively. The densitometric analysis was carried out in absorbance mode at 270 nm. The linear regression analysis data for the calibration plots showed a good linear relationship for melitracen hydrochloride and flupentixol dihydrochloride over a concentration range of 1000–8000 ng/spot and 50–400 ng/spot, respectively. An isocratic, reversed phase-liquid-chromatographic assay method was developed for the quantitative determination of ibuprofen and famotidine in combined-dosage form. A Brownlee C18, 5 μm column with mobile phase containing water : methanol : acetonitrile (30 : 60 : 10, v/v/v) was used. The flow rate was 1.0 mL/min, and effluents were monitored at 264 nm. The retention times of ibuprofen and famotidine were 4.9 min and 6.8 min, respectively. The linearity for ibuprofen and famotidine was in the range of 2–20 μg/mL and 0.1–10 μg/mL, respectively. The proposed method was validated with respect to linearity, accuracy, precision, specificity, and robustness. The method was successfully applied to the estimation of ibuprofen and famotidine in combined dosage form.

A simple, accurate, and precise HPTLC method has been developed for the identification and quantification of DES. The developed method is simple, less time-consuming, cost-effective, and sensitive compared to reported chromatographic method [10–14]. The method was successfully validated in accordance with ICH guidelines. A Brownlee C-18, 5 μm column having 250×4.6 mm i.d. in isocratic mode, with mobile phase containing 0.025 M potassium dihydrogen phosphate: methanol (10 : 90, v/v; pH 7.3) was used. The flow rate was 1.0 mL/min, and effluents were monitored at 230 nm. The retention times of melitracen hydrochloride and flupentixol dihydrochloride were 7.75 min and 5.50 min, respectively. The linearity for melitracen hydrochloride and flupentixol dihydrochloride were in the range of 0.5–60 μg/mL. The recoveries obtained for melitracen hydrochloride and flupenthixol dihydrochloride was 99.81–100.77% and 99.42–100.12%, respectively. Both the drugs were subjected to acid and alkali hydrolysis, chemical oxidation, and dry heat degradation and photodegradation.
