11. Species composition, abundance pattern of Bemisia tabaci and its co-inhibitant on eggplants under different chemical fertilization condition

Abstract

This study assesses the interaction, relationship and recorded several species of insectacarines pest on the underside of eggplant leaves. Variation of infestation levels by a community of insect-acarines detected on the leaves namely Whitefly Bemisia tabaci, aphids Myzus persicae, thrips Thrips palmi, mite and spider mite Tetranychus cinnabarinus caused serious damage at the initial stage of plant growth with flowering and fruiting stage. Different effects by fertilizer levels were observed affected the presence of insect-acarines and dominancy of major pest. The result also showed that all the insectspest was colonized competitively on the leaves for every sampling date due to treatments effect. Whitefly was found to be the most dominant on eggplants which resulted in more than 50% from other insect pest composition, particularly at higher fertilizer levels. Some occasions described that the predator-prey relationship has been identified as a major role in the habitat competition. There was an obvious trend of competitor pest of whitefly such as aphids’ population was found higher than whitefly in all treatments. Generally, this study suggests that the dominant species and the first invader probably could suppress or mediate the other competitor significantly once they occupied on the same quality of host plants.

Abstract

Abstract Background Mosquito-borne viruses are transmitted to human hosts via blood-feeding behavior of female mosquitoes. Female mosquitoes seek a host to take blood meals (host-seeking behavior). In order to prevent virus infections, it is important to understand how they modulate host-seeking behavior. Dopamine (DA) in the central nervous system acts as a neuromediator that regulates a variety of behaviors in insects. In female mosquitoes, host-seeking behavior increases when DA levels in the head decline after emergence. However, it remains unclear whether DA directly modulates host-seeking behavior in female mosquitoes. The aim of this study was to examine whether changes in DA levels in the head affects host-seeking activity in the adult female mosquito Aedes albopictus (Ae. albopictus). Findings We compared host-seeking behavior in one group of emerging female adults treated with l-β-3,4-dihydroxyphenylalanine (l-DOPA), the precursor of DA, (l-DOPA group), with that in an untreated control (control group) after confirming elevation of head DA in l-DOPA group by using high-performance liquid chromatography. The content of head DA in l-DOPA group significantly remained higher than that in controls on all days examined. The host-seeking activity in the control group showed a gradual increase over the 6-day experimental period. In contrast, there was no such increase in the host-seeking activity in the l-DOPA group. Therefore, the host-seeking activity of l-DOPA group was significantly lower than that of the controls between day 3 and 6 post-emergence. Conclusion Our results indicate that elevation of DA level reduces host-seeking activity in adult female mosquito Ae. albopictus.

28. Contribution of public places in proliferation of dengue vectors in Penang Island, Malaysia

Abstract

Objective: To determine abundance, distribution and diversity of potential breeding container habitats of the dengue vectors in public places including schools, restaurants, mosques and parks in southwest areas of Penang Island, Malaysia. Methods: Premises at restaurants, schools, parks and mosques were surveyed simultaneously and inspected visually for container habitats and production of immature mosquitoes from March 2015 to March 2016. Abundance (mean ± SE) of breeding containers between sites was compared using One-way ANOVA. Independent sample ttest was used to compare total number of Aedes albopictus (Ae. albopictus) and Aedes aegypti (Ae. aegypti) surveyed. Results: The surveyed locations yielded a total of 3741 breeding containers and 19537 immature mosquitoes from four areas. Concurrent artificial and natural containers produced 78.4% immature Ae. albopictus and 6.3% Ae. aegypti mosquitoes in wet season, with 14.2% Ae. albopictus and 1.1% Ae. aegypti mosquitoes in dry season. Artificial containers accounted for 98.1% of the total containers recorded, with restaurants being the most productive locations (8 012) and schools being the least productive (2 234). Conclusions: It was concluded that public places are good sources of potential container habitats of Aedes mosquitoes in Penang Island, Malaysia and Ae. albopictus has exclusively replaced the home-grown Ae. aegypti even in urban areas. Therefore, treatment of artificial containers in such locations is critical in Aedes mosquito control campaigns during dengue outbreaks.

Abstract

Infectious diseases, especially, diarrhoea, are responsible for high mortality rates in developing countries. Zinc supplementation shows beneficial effects against such diseases, but the mechanism of action is poorly understood. Here, we examined whether zinc supplementation can improve mucosal innate immunity through induction of antimicrobial peptide secretion from intestinal epithelial cells. Zinc was found to induce secretion of the antimicrobial peptide LL-37 from Caco-2 cell in a dose (0.63 ± 0.09 ng/mL and 0.54 ± 0.06 ng/mL at 20 μM and 50 μM respectively) and time dependent manner. LL-37 secretion increased immediately (1 h) after exposure to 20 μM Zn (0.29 ± 0.04 ng/mL), which continued up to 48 h of exposure (0.58 ± 0.05 ng/mL). Zinc induces the phosphorylation of ERK and p38 MAP kinase and regulates LL-37 secretion through these MAP kinases. Zinc supplementation may have beneficial effects on mucosal innate immunity via secretion of LL-37.

デングウイルスを子孫に垂直伝播可能なヤブカ類では,雌雄いずれも本ウイルスに感受性をもつ.デングウイルスの感染により,多くのタンパク質の機能が損なわれるという報告がある.しかし,これらタンパク質は,最初の感染部位である中腸組織から,いずれも分離されていない.我々は,2次元ゲル電気泳動を用いて,デング熱媒介ヒトスジシマカ成虫の体の大きさと性別に関連した中腸タンパク質の特徴を調べた.雌蚊体の大小比較では,一群のタンパク質が両者に共通に見られ,その量は大きな雌蚊に多く観察された.また,これらとは異なる一群のタンパク質が,体の大きな雌蚊の中腸に特異的に認められた.蚊の雌雄間においても中腸タンパク質に明確な違いが認められた.つまり,雄の中腸で認められた一群のタンパク質は,雌の中腸においても同様に認められたが,その生産量は雄よりも多かった.さらに,これらのタンパク質に加えて,雌の中腸内でのみ特異的に産生される一群のタンパク質が観察された.本報告は,以上の結果から,雌蚊が持つ生理的機能との関連について考察した.In Aedes mosquito, which can vertically transmit the dengue virus to offspring, both sexes can be receptive to the virus. A number of proteins are being reported to impair the viral infection, but none of these have been isolated from the site of initial infection, the midgut. Using a two-dimensional (2-D) gel electrophoresis, we categorized proteins of the midgut from the dengue vector Ae. albopictus (Diptera: Culicidae) with regard to size and sex. The comparison of profiles from small and large females revealed that a set of proteins was identified as specific to large female's midguts. The comparison of profiles for males and females showed clear differences as well. A set of proteins identified in male midguts were produced in much higher quantities in females than in males. In addition, many other proteins were identified as being specifically produced only in female midguts. These results are discussed in the context of female functions.

Abstract

This study was conducted to isolate and identify lactobacilli from larval and adult midgut of wild Aedes aegypti (Ae. aegypti) to find candidate bacteria for paratransgenic control. Characterization of the bacterial symbionts was done using Gram staining, motility test, catalase test, and biochemical tests, among others, and the morphological features were confirmed using a standard scheme that simplifies the identification of lactic acid bacteria. A total of 174 strains were isolated and identified, 135 strains from larval midgut and 39 strains from adult midgut (mean ± SE, 4.00 ± 0.72; P = 0.00). The isolated species were confirmed to be Lactobacillus fermentum, L. casei, L. acidophilus, L. viridescens, L. brevis and L. gasseri. It can be concluded that Ae. aegypti has the potential of harboring the cultivable bacterial symbionts. In conclusion, the isolated species were nominated for paratransgenic control, particularly L. fermentum, being found in large numbers from both larval and adulxt midgut. [ABSTRACT FROM AUTHOR]

39. Mortality and Repellent Effects of Coffee Extracts on The Workers of Three Household Ant Species.

Abstract

Coffee consists of a variety of chemical compounds that has not been documented to have resistance on insects. Hence, this research was conducted to study the impact of coffee extracts impregnated in gel bait towards survival and feeding behaviour of Tapinoma indicum (ghost ant), Pheidole megacephala (big-headed ant) and Monomorium pharaonis (Pharaoh ant) (Hymenoptera: Formicidae). The three coffee species used were Coffea arabica, Coffea canephora and Coffea liberica. The coffee extracts were obtained using Soxhlet extraction method, diluted to 0.01%, 0.05% and 0.10% concentration, and eventually impregnated into two sets of gel bait at with the first set (Set I) sugar solution and the second set (Set II) with distilled water. The overall results indicated that Coffea arabica gave highest mortality on all three ant species and higher concentration of extracts showed higher ant mortality in most bioassays. The higher mortality in lower concentration bioassays was probably due to their lower repellency percentages. Furthermore, Set I bioassays had higher mortality as the sugar used act as food attractant. T. indicum was the most susceptible species.
