Abstract Endophytes play a pivotal role in protecting host plants from both biotic and abiotic stresses, promoting the production of active components (AC) and plant growth. However, the succession of the endophyte community in Eucommia ulmoides (E. ulmoides), particularly the community assembly and function, has not been extensively investigated. In this study, we employed high-throughput sequencing and bioinformatics tools to analyze endophyte diversity across different tree ages, parts, and periods. We examined the population differences, correlations, community assembly mechanisms, and functional roles of these endophytes. Functional predictions via PICRUSt2 revealed that most endophytic fungal functions were linked to biosynthesis, with significant differences in biosynthetic functional abundance across parts and periods. In contrast, the metabolic activity of endophytic bacteria remained stable across different periods and parts. Correlation analysis further confirmed a strong positive relationship between ACs and certain endophytic fungi. Among them, the fungal phyla Ascomycota and Basidiomycota were identified as key contributors to the metabolism of chlorogenic acid (CA), while Aucubin was significantly positively correlated with several endophytic bacteria. These findings provide valuable insights into the functional roles and community assembly mechanism of E. ulmoides endophytes, as well as their symbiotic relationships.

Abstract In eukaryotes, the nuclear membrane that encapsulates genomic DNA is composed of an inner nuclear membrane (INM), an outer nuclear membrane (ONM), and a perinuclear space. SUN proteins located in the INM and KASH proteins in the ONM form the SUN-KASH NM-bridge, which functions as the junction of the nucleocytoplasmic complex junction. Proteins containing the SUN domain showed the highest correlation with differentially accumulated proteins (DAPs) in the wheat response to fungal stress. To understand the characteristics of SUN and its associated proteins in wheat responding to pathogen stress, here we investigated and comprehensive analyzed SUN- and KASH-related proteins among the DAPs under fungi infection based on their conserved motifs. In total, four SUN proteins, one WPP domain-interacting protein (WIP), four WPP domain-interacting tail-anchored proteins (WIT), two WPP proteins and one Ran GTPase activating protein (RanGAP) were identified. Following transient expression of Nicotiana benthamiana, TaSUN2, TaRanGAP2, TaWIT1 and TaWIP1 were identified as nuclear membrane proteins, while TaWPP1 and TaWPP2 were expressed in both the nucleus and cell membrane. RT-qPCR analysis demonstrated that the transcription of TaSUN2, TaRanGAP2 and TaWPP1 were strongly upregulated in response to fungal infection. Furthermore, using the bimolecular fluorescence complementation, the luciferase complementation and a nuclear and split-ubiquitin-based membrane yeast two-hybrid systems, we substantiated the interaction between TaSUN2 and TaWIP1, as well as TaWIP1/WIT1 and TaWPP1/WPP2. Silencing of TaSUN2, TaRanGAP2 and TaWPP1 in wheat leaves promoted powdery mildew infection and hyphal growth, and reduced the expression of TaBRI1, TaBAK1 and Ta14-3–3, indicating that these NM proteins play a positive role in resistance to fungal stress. Our study reveals the characteristics of NM proteins and propose the preliminary construction of SUN-WIP-WPP-RanGAP complex in wheat, which represents a foundation for detail elucidating their functions in wheat in future.

Large-volume presses (LVPs) providing large volumes, liquid media, deformation capability, jump compression, and in situ measurements are in great demand for high-pressure research, particularly in the fields of geoscience, condensed matter physics, material science, chemistry, and biology. A high-pressure and high-temperature (HPHT) platform with different LVP subsystems, both solid-state and liquid environments, and nonequilibrium subsystems, has been constructed at the Synergetic Extreme Condition User Facility, Jilin University. This article describes the construction of the different subsystems and provides an overview of the capabilities and characteristics of the different HPHT subsystems. A large sample volume (1000 mm3) at 20 GPa is achieved through the use of a belt-type apparatus in the solid-state subsystem. HPHT conditions (1.8 GPa and 1000 K) are realized in the liquid subsystem through the use of a piston–cylinder-type LVP with optical diamond windows for in situ spectroscopic measurements. A maximum pressure jump to 10.2 GPa can be reached within 20 ms in the nonequilibrium subsystem with the use of an improved bladder-pressurization jump press. Some typical results obtained with different LVPs are briefly reviewed to illustrate the applications and advantages of these presses. In summary, the platform described here has the potential to contribute greatly to high-pressure research and to innovations in high-pressure technology.

Abstract High‐frequency oscillatory activity in cognition‐related neural circuits during wakefulness consistently induces the growth of dendritic spines and axonal terminals. Although these structural changes are essential for cognitive functions, it is hypothesized that if these newly expanded structures fail to establish functional connections, they may become superfluous. Sleep is believed to facilitate the reduction of such redundant structures to maintain neural homeostasis. However, the mechanisms underlying this pruning process during sleep remain poorly understood. In this study, that melatonin type 3 receptors (MT3Rs) are selectively expressed in the stellate neurons of the medial entorhinal cortex (MEC) is demonstrated, an area where high melatonin levels are detected during sleep. Activation of MT3Rs during sleep initiates the shrinkage of dendritic spines in stellate neurons by downregulating neural network activity and dephosphorylating synaptic proteins in the MEC. This process is disrupted when MT3R expression is knocked down or when MT3Rs are blocked during sleep. Notably, interference with MT3Rs in the MEC during sleep impairs the acquisition of spatial memory but does not affect object memory acquisition following sleep. These findings reveal novel molecular mechanisms involving melatonin and MT3Rs in the regulation of dendritic spine shrinkage during sleep, which is crucial for the acquisition and consolidation of spatial memory.

Under the dual-carbon goals, enhancing the green development level of logistics industry and realizing its low-carbon transformation are important issues that need to be solved urgently. Amidst the continuous escalation in the total energy consumption of the national logistics industry, the Beijing-Tianjin-Hebei (BTH) region has exhibited a favorable descending trajectory in this respect. It is necessary to investigate the underlying reasons. Based on the panel data from 2012 to 2021, the DEA and Malmquist index are employed to analyze the low-carbon logistics efficiency of the BTH region from both static and dynamic perspectives. Furthermore, the inefficiency analysis is conducted to identify the deficiencies of low-carbon logistics industry in this region. Results show that (1) from the static perspective, the development of low-carbon logistics industry in the BTH region is relatively unbalanced. Compared to Tianjin and Hebei, Beijing's low-carbon logistics efficiency is significantly lower, becoming the focal area for attention; (2) from the dynamic perspective, technological progress is the main reason for the fluctuation of total factor productivity in the BTH region and a constraining factor for further improvements; (3) from the results of inefficiency analysis, the forthcoming emphasis on low-carbon logistics in Beijing should be on optimizing the number of logistics practitioners, transportation efficiency, and energy efficiency. Economic output and energy efficiency are relatively vulnerable aspects in Tianjin and Hebei, respectively, warranting due consideration. The research results of this paper have important practical implications for better developing low-carbon logistics in the BTH region and leveraging its leading role nationwide.

Both overall survival (OS) and disease-specific survival (DSS) are significant when determining a patient’s prognosis for breast cancer (BC). The effect of DSS-related microRNAs on BC susrvival, however, is not well understood. Here, we spotted differentially expressed miRNAs (DEMs) in the TCGA database of BC DSS, identified eight DSS-related miRNAs, and constructed a risk model. AUC values at 1, 3, and 5 years were 0.852, 0.861, and 0.868, respectively, indicating a risk model’s excellent prognostic prediction ability. Then, we validated miRNA roles in BC OS and finally defined miR-551b as an independently prognostic miRNA in BC. According to function analysis, miR-551b is strongly linked with the emergence and spread of cancer, including protein ubiquitination, intracellular protein transport, metabolic pathways, and cancer pathways. Moreover, we confirmed the low expression of miR-551b in BC tissue and cells. After miR-551b inhibition or overexpression, cell function was either dramatically increased or diminished, respectively, indicating that miR-551b could regulate BC proliferation, invasion, and migration. In conclusion, we thoroughly clarified BC-related miRNAs on DSS and OS and verified miR-551b as a crucial regulator in the development and prognosis of cancer. These results can offer fresh ideas for BC therapy.

Abstract Depression is a common chronic psychiatric illness, which is resistant to medical treatments. While melatonin may alleviate certain depression symptoms, evidence for its efficacy against core symptoms is lacking. Here, we tested a mechanism whereby melatonin rescues the behavioral outcomes of the chronic unpredictable mild stress (CUMS) mouse model of depression. CUMS mice showed depressive behaviors to tail suspension, open field behavior, and sucrose preference test, and cognitive dysfunction in the Morris water maze. Impairments in these measures were relieved by melatonin treatment. Moreover, CUMS mice had impaired glymphatic function across the sleep-wake cycle due to the astrocytic loss and disturbance of circadian regulation of the polarized expression of aquaporin-4 (AQP4) water channels in perivascular astrocytes. EEG results in CUMS mice showed a reduced total sleep time and non-rapid eye movement (NREM) sleep, due to sleep fragmentation in the light phase. CUMS mice lost the normal rhythmic expressions of circadian proteins Per2, Cry2, Bmal1, Clock, and Per1. However, the melatonin treatment restored glymphatic system function and the polarization of AQP4, while improving sleep structure, and rectifying the abnormal expression of Per2, Bmal1, Clock, and Per1 in CUMS mice. Interestingly, Per2 expression correlated negatively with the polarization of AQP4. Further studies demonstrated that Per2 directed the location of AQP4 expression via interactions with the α-dystrobrevin (Dtna) subunit of AQP4 in primary cultured astrocytes. In conclusion, we report a new mechanism whereby melatonin improves depression outcomes by regulating the expression of the circadian protein Per2, maintaining the circadian rhythm of astrocytic AQP4 polarization, and restoring glymphatic function.

To investigate the influence of moisture on the exothermic properties of coal oxidation, this study investigated the variation in thermal effect and radical parameters in the oxidation process of coal under different moisture content. This was achieved through thermogravimetric–differential thermal experiments and electron spin resonance experiments. Additionally, the study analyzed the impact of free radicals on the characteristics of the thermal effect of coal at different oxidation stages using the product–moment correlation method. The results indicate that the moisture content of 8% is a critical point for promoting or inhibiting the oxidation reaction of coal. Below this threshold, it promotes the oxidation reaction, while above it, it plays an inhibitory role. The most significant promotional effect was observed at 8% moisture content, which increased both the weight loss and heat release by 8.61% and 1567.04 J/g, respectively, while also significantly enhancing free radical species and complexity. Conversely, when the moisture content of coal reached 20%, there was a notable inhibition effect, with a reduced weight loss and exothermic capacity by 4.94% and 2705.03 J/g, respectively, along with decreased free radical species and complexity. The free radical species and heat effect parameters in coal showed a strong correlation at all stages of low-temperature oxidation, thus indicating that g-factor can characterize the coal oxidation process to some extent.

Lactic acid bacteria (LAB) has the potential to antagonize harmful microorganisms such as molds, which is expected to be safe and stable biological antimicrobial agents of the next generation. In present study, a strain ST3.5 with inhibiting mold activity was screened and obtained, which was identified as Lactiplantibacillus plantarum (L. plantarum) based on 16S rRNA sequencing. And its characteristics of acid resistance, bile salt resistance, inhibition of pathogenic bacteria, and drug resistance were analyzed. Furtherly, the cell-free supernatant (CFS) of L. plantarum ST3.5 was treated with acid, heat and enzyme, which the main antifungal substances were analyzed. The content of organic acids in supernatant was analyzed by high performance liquid chromatography (HPLC) and the destruction of mold mycelium was observed by scanning electron microscope (SEM). Meanwhile, the biological control experiment was carried out with corn kernels as an actual sample. The results showed that L. plantarum ST3.5 had good acid resistance and weak bile salt resistance. The strain was resistant to antibiotics (e.g., gentamicin and kanamycin) and sensitive to antibiotics (e.g., ampicillin and chloramphenicol), which could inhibit the growth of pathogenic bacteria. Analysis of organic acid found that the content of lactic acid in the CFS was the highest (22.02±0.23) g/L, followed by citric acid (4.99±0.04) g/L and acetic acid (3.67±0.06) g/L. The SEM results demonstrated the CFS could damage mold hyphae. Additionally, the biocontrol experiment confirmed that the CFS of L. plantarum ST3.5 could inhibit the mold residing growth of the corn surface. In conclusion, L. plantarum ST3.5 can inhibit the growth of mold, which can be used to develop biological control agents to minimize mold pollution and ensure food safety.

Science popularization work on earthquake prevention and disaster reduction should keep pace with the times, play a greater role in public opinion guidance, and take more responsibility in public opinion governance. In this paper we discuss that science popularization of earthquake prevention and disaster reduction plays an important role in guiding public opinion from two aspects. First, the hotspots of public opinion provide an opportunity for science popularization of earthquake prevention and disaster reduction. Second, the hotspots of public opinion provide contents for science popularization of earthquake prevention and disaster reduction. I put forward the public opinion guidance strategy of science popularization of earthquake prevention and disaster reduction from three aspects. First, reasonably set the science popularization agenda and take the initiative to guide public opinion; Second, consciously guide public opinion by means of new media communication technology; The third is to play a good “combination boxing” to form the greatest resultant force for public opinion guidance. In the hotspots of public opinion, we should actively do a good job of science popularization of earthquake prevention and disaster reduction, innovate communication forms and methods, skillfully set the agenda, cover the whole time and space, and participate in the whole society, build a benign public opinion field for the exchange of information between the government, the media and the public, enhance the public’s risk awareness and disaster prevention ability, and truly build a people’s defense line for risk prevention and control.

To thoroughly understand the profile of phenolic phytochemicals in kidney bean seeds cultivated in a cold region, the extractions, contents, antioxidant activities, compositions of free and bound phenols in the seed coat and cotyledon, and also relevant color attributes, were investigated. The results indicated that ultrasound-assisted extraction was an efficient method for free phenols. The bound phenols in seed coat and cotyledon were released more efficiently by alkali-acid and acid-alkali sequential hydrolysis, respectively. Under the optimized extractions, total phenols (TPC), flavonoids (TFC), and anthocyanins (TAC) ranged in 7.81–32.89 mg GAE/g dw, 3.23–15.65 mg RE/g dw, and 0–0.21 mg CE/g dw in the whole seeds of the five common kidney beans. There was a big difference in phenolic distribution between red and white seeds. From whole seed, the phenols in the four red cultivars mainly existed in free state (78.84%) and seed coat (71.56%), while the phenols in the white ‘Sark’ divided equally between free (51.18%) and bound (48.82%) states and consisted chiefly in cotyledon (81.58%). The correlation analyses showed that the antioxidant activities were significantly and positively correlated with TPC and TFC. The phenolic attributes were closely associated with the color of the seed coat. Red seeds had higher total contents of phenols than white seeds. TAC had a positively significant correlation with redness. Brightness and yellowness showed a negatively significant correlation with TPC, TFC, and antioxidant capacities, which were necessarily linked with redness degree and spot in red seeds. The spotted red ‘Yikeshu’ with the most outstanding performance on phenolic attributes was selected to analyze phenolic compounds with UHPLC-QE-MS. Among the 85 identified phenolics, 2 phenolic acids and 10 flavonoids were dominant. The characteristic phenolics in free and bound states were screened in both seed coat and cotyledon, respectively. The available information on the phenolic profile may expand the utilization of kidney beans as a nutritional ingredient in the food industry.

Trajectory outlier detection is a crucial task in trajectory data mining and has received significant attention. However, the distribution of trajectories is tied to social activities, resulting in extreme unevenness among regions. While existing methods have demonstrated excellent performance in regions with sufficient historical trajectories, they frequently struggle to detect outliers in regions with limited trajectories. Unfortunately, this issue has not received much attention, leaving a gap in the current understanding of trajectory mining. To deal with this problem, we in this paper propose a model called TTOD that can effectively detect outliers in regions with sparse data by transferring knowledge among regions. The main idea is to learn a feature mapping function that maps the global feature space of auxiliary regions to the target region’s specific feature space. To achieve this, we adopt a VAE-based model called the Global VAE to learn the global feature space in auxiliary regions by modeling the trajectory patterns with Gaussian distributions. Then, we propose a Specific-region VAE that serves as the mapping function to learn the target feature space. Additionally, considering the data drift of feature distributions among regions, we introduced an additional pattern synthesis layer, named the De-drift Layer, to diversify the target feature space, thus addressing the pattern missing issue caused by the gap of feature distributions between the auxiliary regions and the target regions. Then the target feature space can be well studied and applied to detect outliers. Finally, we conduct extensive experiments on two real taxi trajectory datasets and the results show that TTOD achieves state-of-the-art performance compared with the baselines.

In order to quantitatively evaluate the sustainability of bus low-fare policy, this study establishes a sustainability evaluation index system for bus low-fare policy by using the PSR (Pressure–State–Response) model. Based on the matter–element extension model and entropy weight method, the sustainability evaluation model of bus low-fare policy is constructed. Finally, taking Shenzhen as an example, this study compares and analyzes the sustainability changes of bus fare policy in 2006, 2012 and 2016. The results show that the sustainability of the bus fare policy does not depend on the attribute of the fare (profit-making fares or public welfare fares): the sustainability of bus low-fare policy is closely related to the supporting fiscal subsidy system. Compared with the cost regulation subsidy system, the quota subsidy system is more conducive to the sustainability of bus low-fare policy. This study provides a decision-making reference for the sustainable development of urban bus low-fare policy.

IntroductionAlzheimer’s disease (AD) is a progressive neurodegenerative disease that results in cognitive impairment and is often accompanied by anxiety. In this study, we investigated whether the activation of VTAVgat neurons could reduce anxiety in APP/PS1 mice. We hypothesized that acute social defeat stress (SDS) would lead to anxiety in APP/PS1 mice, and that the activation of VTAVgat neurons would alleviate this anxiety.MethodsWe exposed APP/PS1 mice to acute SDS and assessed anxiety using the open field test and elevated plus-arm test. Activated VTAVgat neurons was tested by cfos staining. Sleep quality was detected using electroencephalogram after SDS or non-SDS procedure. Sleep duration, sleep latency, and non-rapid eye movement (NREM) percentage were analyzed. VTAVgat neurons were chemogenetically activated by deschloroclozapine.ResultsOur results showed that acute SDS led to anxiety in APP/PS1 mice, as evidenced by increased anxiety-related behaviors in the open field and elevated plus-arm tests. Activation of VTAVgat neurons by SDS led to an increase in sleep duration, primarily due to a decrease in sleep latency and an increase in NREMs. However, the quality of sleep was poor. Chemogenetical activation of VTAVgat neurons improved sleep quality and relieved SDS-induced anxiety. Furthermore, the anxiety state correlated negatively with sleep duration and NREM percentage and correlated positively with theta power density in APP/PS1 mice.DiscussionOur study provides evidence that the activation of VTAVgat neurons alleviates SDS-induced anxiety in APP/PS1 mice, suggesting that poor sleep quality may exacerbate anxiety in AD. These findings may have important implications for the treatment of anxiety in AD, as targeting VTAVgat neurons could be a potential therapeutic approach.

Abstract Though effective in theoretical simulation, the established traffic control models and optimization algorithms will result in model mismatch or even control strategy failure in actual application. However, they are commonly adopted in traffic signal control research, resulting in the unavailability of many exceptional control algorithms in practice. Simulation should function as a bridge between theoretical research and actual application, allowing the gap between the two to be communicated and made up for. However, an effective connection between the two has yet to be established to enable simulation methods in existing traffic control research. To this end, we designed and developed a simulation platform for "Online Application—HILS (Hardware-in-the-Loop Simulation)—Practice" integration over traffic signal control. In this paper, the architecture and characteristics of the integrated simulation platform were described. Besides, the function of each module of the platform was detailed, followed by listing simulation examples for six complex scenarios, with the active control scenario being selected for simulation comparison analysis. The findings demonstrated extensive road network simulation with the integrated simulation platform, multidimensional control variables, control strategies with support, as well as stable and reliable operation. It can be used to verify several sorts of traffic control simulation with variable dimensions.

Oligosaccharides are widely added to infant formula milk powders as prebiotics for intestinal health. Using in vitro fermentation conditions, this study investigated the fermentation characteristics of fructooligosaccharides (FOS) and galactooligosaccharides (GOS) with infant fecal inoculum. The changes in short-chain fatty acids (SCFAs) and lactate production were monitored by HPLC. The contents of SCFAs and lactate increased significantly in the GOS and FOS + GOS groups. Furthermore, the metagenomic analysis revealed that microbial communities were distinctively different. Most bacteria were significantly positively correlated with SCFAs production and negatively correlated with pH, particularly Bifidobacterium, Enterococcus, Weissella and Streptococcus. More importantly, the relative abundance of most carbohydrate-active enzymes (CAZymes, EC:2.3.1.54, EC:2.3.1.9 and EC:2.8.3.8, etc.) were significantly higher in the GOS and FOS + GOS groups. These results suggested that FOS and GOS had a significant regulatory effect on the composition and metabolism of the intestinal microbiota, further promoting the production of SCFAs and lactate.

Background: Haploidentical hematopoietic stem cell transplantation (haplo-HSCT) based on granulocyte colony-stimulating factor plus anti-thymocyte regimens (‘Beijing Protocol’) provides a salvage treatment for patients of acquired severe aplastic anemia (SAA) in China. However, graft- versus -host disease (GVHD) is a major impediment of haplo-HSCT due to human leukocyte antigen disparity. Recently, haplo-HSCT combined with umbilical cord blood (UCB) (haplo-cord HSCT) is performed in clinical trials to potentially reduce the risk of severe GVHD. Nevertheless, studies comparing GVHD in pediatric patients receiving haplo and haplo-cord HSCT for SAA are limited. Objective: The objective of this study was to investigate the impact of UCB co-infusion on GVHD in pediatric patients receiving haplo-HSCT for SAA. Design: We conducted a retrospective study of 91 consecutive SAA children undergoing haploidentical transplantation based on the ‘Beijing Protocol’ with or without co-infusion of UCB in our center. Methods: All patients received uniform non-myeloablative conditioning and GVHD prophylaxis. We compared baseline characteristics and transplant outcomes between the haplo ( n = 35) and haplo-cord ( n = 56) recipients. Results: All 91 patients achieved hematopoietic recovery from haploidentical donors, with a higher incidence of peri-engraftment syndrome observed with the haplo-cord group as compared with the haplo group (75.0% versus 48.6%, p = 0.029). Notably, the haplo-cord group showed a lower incidence of II–IV acute GVHD (aGVHD) than the haplo group (16.1% versus 42.9%, p = 0.002). Observed incidences of chronic GVHD (cGVHD) and moderate to severe cGVHD in the haplo-cord group were also lower than that in the haplo group (25.6% versus 51.3%, p = 0.019; 16.2% versus 41.3%, p = 0.016, respectively). Haplo-cord HSCT was identified as the only factor associated with a lower incidence of II–IV aGVHD and cGVHD in multivariate analysis. However, no differences were observed between the two groups for infections and survival outcomes. Conclusion: Our data indicated that co-infusion of UCB in ‘Beijing Protocol’-based haplo-HSCT may be effective for reducing the risk of severe GVHD in SAA children.

Lactic acid bacteria (LAB) can produce a vast spectrum of antifungal metabolites to inhibit fungal growth. The purpose of this study was to elucidate the antifungal effect of isolated Weissella cibaria BYL4.2 on Penicillium chrysogenum, the antifungal activity of W. cibaria BYL4.2 against P. chrysogenum was evaluated by the superposition method, results showed that it had obviously antifungal activity against P. chrysogenum. Studying the probiotic properties of BYL4.2 and determining it as beneficial bacteria. Furtherly, different treatments were carried out to characterize the antifungal activity of cell-free supernatant (CFS) produced by W. cibaria BYL4.2, and it was shown that the CFS was pH-dependent, partly heat-sensitive, and was not influenced by proteinaceous treatment. The CFS of W. cibaria BYL4.2 was analyzed by high-performance liquid chromatography (HPLC) and found the highest content of lactic acid. Screening of metabolic markers by a non-targeted metabolomics approach based liquid chromatography-mass spectrometry (LC-MS). The results speculated that organic acid especially detected D-tartaric acid was the main antifungal substance of CFS, which could cause the down-regulation of metabolites in the ABC transporters pathway, thereby inhibiting the growth of P. chrysogenum. Therefore, this study may provide important information for the inhibitory mechanism of W. cibaria BYL4.2 on P. chrysogenum, and provide a basis for further research on the antifungal effect of Weissella.

Subarachnoid hemorrhage (SAH) is an important public health concern with high morbidity and mortality worldwide. SAH induces cell death, blood−brain barrier (BBB) damage, brain edema and oxidative stress. As the most abundant cell type in the central nervous system, astrocytes play an essential role in brain damage and recovery following SAH. This review describes astrocyte activation and polarization after SAH. Astrocytes mediate BBB disruption, glymphatic–lymphatic system dysfunction, oxidative stress, and cell death after SAH. Furthermore, astrocytes engage in abundant crosstalk with other brain cells, such as endothelial cells, neurons, pericytes, microglia and monocytes, after SAH. In addition, astrocytes also exert protective functions in SAH. Finally, we summarize evidence regarding therapeutic approaches aimed at modulating astrocyte function following SAH, which could provide some new leads for future translational therapy to alleviate damage after SAH.

Background and purposeThe relationship between sleep duration and stroke are inconclusive in China, especially in those individuals with metabolic syndrome. We aimed to investigate the association between sleep duration and incident stroke in participants with metabolic syndrome or its specific components in China.Materials and methodsData were taken from the 2011 and 2015 waves of China Health and Retirement Longitudinal Study (CHARLS). Habitual sleep duration (≤6, 6∼8 [reference], >8 h), daytime napping (0, 1∼60 [reference], and >60 min) were determined by self-reported questionnaires. Metabolic syndrome was defined by blood assessment and biomarkers combined with self-reported doctors’ diagnosis. Incident stroke was determined by reported stroke from 2011 to 2015 wave. Cross-sectional and longitudinal associations between sleep and (incident) stroke at baseline and 4-year follow-up period were tested among the population with metabolic syndrome and its components.ResultsA U-shaped relationship was observed between sleep duration and stroke in cross-sectional analysis. Sleep ≤ 6 h/night had a greater risk of incident stroke (hazard ratio [HR] 1.65; 95% confidence interval [CI] 1.04–2.61) compared with sleep 6∼8 h/night. And the HR of stroke was 1.62 (95%CI, 1.03–2.53) for sleep < 7 h/day compared to 7∼9 h/day. These associations were more evident in the female and individuals aged 45–65 years. Furthermore, the effect of short sleep duration on incident stroke was different in each component of metabolic syndrome, which was more pronounced in participants with elevated blood pressure. And a significant joint effect of sleeping ≤ 6 h/night and no napping on risk of stroke was observed (HR 1.82, 95%CI 1.06–3.12).ConclusionShort sleep duration was an independent risk factor for incident stroke, especially among females, individuals aged 45–65 years, or those with some components of metabolic syndrome, such as hypertension. Napping could buffer the risk of short sleep duration on incident stroke.

Buckwheat has beneficial effects on human intestinal health, which is often compounded with wheat to make food. Therefore, the effect of cereals mixture via in vitro fermentation on gut microbes and short-chain fatty acids (SCFAs) were investigated in this study. The mixture of wheat and tartary buckwheat (WT) produced more lactate and acetate, and the mixture of wheat and sweet buckwheat (WE) produced more propionate and butyrate. Compared with wheat (WA), the relative abundance of some beneficial bacteria significantly increased, such as Sutterella in WT and Faecalibacterium in WE. Cereals mixture also affected the expression of functional genes, involved in metabolic pathways and carbohydrate-active enzymes (CAZymes) that modulated SCFAs generation. This study provides new insights into the effects of sweet and tartary buckwheat on intestinal function, which is beneficial to applying both types of buckwheat in practical.

Horizontal wells are more likely than vertical wells to have enlarged wellbore sections due to borehole instability. However, there is scarce research on borehole cleaning of horizontal wells with enlarged wellbore sections. In this paper, we establish a horizontal wellbore model with a breakout enlargement section using field borehole diameter data. We used the three-dimensional computational fluid dynamics (CFD) method and the Realizable k-ε turbulence model with the Euler–Euler approach to simulate the effects of the drilling fluid circulation return speed and the spinning speed of the drill pipe on the cutting movement of conventional horizontal wells and horizontal wells with a breakout enlargement section. The simulation results demonstrate that increasing the drilling fluid circulation return speed and the spinning speed of the drill pipe does not significantly improve the hole cleaning impact for horizontal wells with a breakout enlargement section. We analyzed the effects of the enlargement ratio, ellipticity, and principal axis orientation on the borehole cleaning effect of horizontal wells with a breakout enlargement section. The results show that the cleaning impact is better when the enlargement ratio is lower; moreover, the ellipticity is larger and the principal axis orientation is perpendicular to the gravity direction. This paper fills a gap in the existing theory of hole cleaning in horizontal wells and provides a theoretical basis for improving the hole cleaning effect in actual drilling processes.

Rivers play a key role in regulating urban ecology, which can improve urban climate while slowing the heat island effect. As one of embodied energy in the field of ecology, emergy theory can be used to quantitatively evaluate the ecological characteristics of a system. This will help to further explore urban ecological sustainability in this article. In this study, four ecological riverbank reconstruction projects have been executed to restore the ecology along the banks of the Jinchuan River in Nanjing, China, which focus on the key river–lakeside and waterfront space in the main urban area. The LCA–emergy–carbon emission method was applied through a series of indicators, including emergy indexes and carbon emission indicators. It is important to distinguish prior research, and few have utilized this approach on urban waterways and waterscapes. The results illustrate that the reconstruction system has obvious improvement significance to the whole river ecology. This change can also be seen when using LCA–emergy analysis. In a 20-year cycle, the emergy of the material production stage and maintenance phase account for a major emergy share, followed by the construction stage, transportation process, and design process stage. The sustainability (ESI indicator) has been improved after carrying out the reconstruction projects. By choosing water and gravel as the primary material, the carbon emission can be reduced. The water treatment process accounts for the vast majority of carbon emissions. Secondly, gravel also plays an important role in carbon emission. Finally, an improved measure (clean energy reuse) was conducted to enhance the ecology of the reconstruction projects and obtained a significant ecological sustainability boost.

Metabolic pathway analysis of Bifidobacterium adolescent (B. adolescentis) grown on either xylobiose and xylotriose (X2/X3) or xylopentaose (X5) and identifying key regulatory-related genes and metabolites from RNA-seq and UHPLC system was performed. Compared with X5, X2/X3 highly promoted the growth of B. adolescentis. Also, the transcriptome analysis showed that a total of 268 differentially expressed genes (DEGs) of B. adolescentis cultured with X2/X3 and X5 were screened, including 163 upregulated and 105 downregulated genes (X2/X3 vs. X5), which mainly were ABC transporters. Furthermore, the qRT-PCR results of 16 DGEs validated the accuracy of the RNA-seq data. Meanwhile, metabolomics analysis showed that 192 differential metabolites noted on MS2 included 127 upregulated and 65 downregulated metabolites; mainly, metabolites were amino acids and organic acids. The abundance difference of specific genes and metabolites highlighted regulatory mechanisms involved in utilizing different polymerized xylooligosaccharides by B. adolescentis.

Background. AML (acute myeloid leukemia) is a common hematological malignancy in children with poor treatment effects and poor prognosis. Recent studies have shown that as a novel BRD4 (bromodomain containing 4) PROTACs (proteolysis targeting chimeras) degrader, GNE-987 can slow down the growth of various tumors and increase apoptosis, with promising clinical prospects. However, the function and molecular mechanism of GNE-987 in AML remain unclear. This study is aimed at investigating the therapeutic effect of GNE-987 on AML and its underlying mechanism. Methods. The association between BRD4 and AML was assessed by studying public databases. After GNE-987 was added to AML cells, cell proliferation slowed down, the cycle was disturbed, and apoptosis increased. Western blotting was used to detect BRD2 (bromodomain containing 2), BRD3 (bromodomain containing 3), BRD4, and PARP (poly ADP-ribose polymerase) proteins. The effect of GNE-987 on AML cells was analyzed in vivo. RNA-seq (RNA sequencing) and ChIP-seq (chromatin immunoprecipitation sequencing) validated the function and molecular pathways of GNE-987 in processing AML. Results. BRD4 expression was significantly elevated in pediatric AML samples compared with healthy donors. GNE-987 inhibited AML cell proliferation by inhibiting the cell cycle and inducing apoptosis. BRD2, BRD3, and BRD4 were consistent with decreased VHL (Von Hippel Lindau) expression in AML cells. In an AML xenograft model, GNE-987 significantly reduced the hepatosplenic infiltration of leukemia cells and increased the mouse survival time. Based on analysis of RNA-seq and ChIP-seq analyses, GNE-987 could target multiple SE- (super-enhancer-) related genes, including LYL1 (lymphoblastic leukemia 1), to inhibit AML. Conclusions. GNE-987 had strong antitumor activity in AML. GNE-987 could effectively inhibit the expression of SE-related oncogenes including LYL1 in AML. Our results suggested that GNE-987 had broad prospects in the treatment of AML.

To adapt to the intelligent management of traffic flow in modern aviation, this study improves the algorithm according to the data exercise method and establishes the mathematical model and vector model of intelligent motion coordinates. Moreover, according to the simulation test method, this study analyzes the six-degree-of-freedom equation of motion in the coordinate system of the civil aviation aircraft and uses the simulation method to regard the civil aviation as a rigid body with a certain mass and mass distribution. In addition, this study builds an intelligent model based on the demand analysis, and according to the actual demand, this study combines the simulation method to analyze the structure of the intelligent management model of air traffic flow. Finally, this study uses the experimental analysis method to verify the effect of the model proposed in this study and combines the experimental research to verify that the model proposed in this study has a certain effect.

In high-frequency (HF) radar systems, transient interference is a common phenomenon that dramatically degrades the performance of target detection and remote sensing. Up until now, various suppression algorithms of transient interference have been proposed. They mainly concentrate on the skywave over-the-horizon radar on the basis of the assumption that the interference is sparse in a coherent processing interval (CPI). However, HF surface wave radar (HFSWR) often faces more complex transient interference due to various extreme types of weather, such as thunderstorm and typhoon, etc. The above algorithms usually suffer dramatic performance loss when transient interference contaminates the enormously continuous parts of a CPI. Especially for the compact HFSWR, which suffers from severe beam broadening and fewer array degrees of freedom. In order to solve the above problem, this study developed a two-dimensional interference suppression algorithm based on space-time cascaded processing. First, according to the spatial correlation of the compact array, the statistical samples of the main-lobe transient interference are estimated using a rotating spatial beam method. Next, an adaptive selection strategy is developed to obtain the optimal secondary samples based on information geometry distance. Finally, based on a quadratic constraint approximation, a precise estimation method of the optimal weight is developed when the interference covariance matrix is singular. The experimental results of simulation and measured data demonstrate that the proposed approach provides far superior suppression performance.

(1) Background: Achieving harmonious human–land relations is one of the key objectives of sustainable urban–rural development, and the degree of decoupling of permanent population levels from changes in construction land use is an important factor in related analyses. Due to the existence of huge urban–rural differences, rethinking China’s human–land relations from the perspective of integrating urban and rural areas is of great value for the advancement of high-quality urban–rural development. (2) Methods: By studying the lower reaches of the Yangtze and Yellow Rivers of China, and based on data from the second and third national land surveys of China, this paper analyzes the spatio-temporal evolution of urban and rural population, construction land use, and human–land relations from 2009 to 2019 using exploratory spatial data analysis (ESDA) and a decoupling model; in addition, this paper proposes a differentiated zoning management strategy and establishes a new framework that integrates evolutionary patterns, human–land relations, spatial effects, and policy design. (3) Results: The geographic distribution patterns of urban and rural population and construction land use remained stable over time, with high levels of spatial heterogeneity, agglomeration, and correlation. Changes in urban and rural population levels and construction land use are becoming increasingly diversified and complex, with both increases and reductions existing side by side. Based on a Boston Consulting Group matrix, the evolution patterns of urban and rural population and construction land use are divided into four types, referred to as star-cities, cow-cities, question-cities, and dog-cities. Over the time period examined in this paper, the spatial autocorrelation of urban land evolution patterns turned from negative to positive; however, that of rural land, as well as those of urban and rural population evolution patterns, were statistically insignificant. Urban human–land relations are coordinated, in general, and are mostly in a state of either weak decoupling or expansive coupling. In contrast, rural human–land relations are seriously imbalanced, and most of them are in a state of strong negative decoupling. Human–land relations are dominated by regressive changes in urban areas but remain unchanged in rural areas. Cold- and hot-spot cities are concentrated in clusters or in bands, forming a core-periphery structure. The formation and evolution of the decoupling relationship between construction land use and permanent population are the results of multiple factors, including urbanization, industrialization, globalization, and government demand and policy intervention. The interaction effects between different factors show bifactor enhancement and nonlinear enhancement, with complex driving mechanisms and large urban–rural differences. It should be highlighted that the influence intensity, operation mechanism, and changes in the trends for different factors vary greatly. Urbanization rate, gross domestic product, and government revenue are key factors that exert a strong direct driving force; international trade, foreign direct investment, and per capita GDP are important factors, while the remaining factors are auxiliary factors that remain heavily dependent on interaction effects. (4) Conclusions: To further transform human–land relations from imbalanced to coordinated, we divide the study area into four area types based on the concept of urban–rural community: urban and rural intensive policy areas, urban intensive policy areas, rural intensive policy areas, and urban and rural controlled policy areas. Furthermore, we put forward suggestions on the differentiated management of land use for the four types of policy areas.

BS Sufu is a fermented food that is made by mixed black soybeans and soybeans. Microbial communities and metabolites play an important role for the final product. We characterized microbial diversity of BS Sufu during fermentation by high-throughput DNA sequencing. Meanwhile, volatile compounds were investigated by solid-phase microextraction (SPME) coupled with gas chromatography–mass spectrometry (GC-MS). The results showed that bacterial diversity was higher than that of fungi in BS Sufu. We found the existence of bacterial and fungal core communities, including Enterococcus, Enterobacter, Rhizopus, and Monascus. Network analysis indicated that bacterial and fungal communities maintain positive and negative interactions, which are important to shape the resident microbial communities in Sufu. In addition, 17 free amino acids (FAAs) were detected at the post-fermentation stage, and umami amino acid mainly contributed to taste of BS Sufu. Furtherly, a total of 79 volatile constituents in BS Sufu, including nine alcohols, 31 esters, and four aldehydes, form synergistically the unique odor of Sufu. Additionally, the correlations between microbiota and metabolites were analyzed. Our results suggested that these microbial taxa and metabolites contribute to the taste and flavor of BS Sufu. This study provided information for analysis of BS Sufu at different fermentation periods in terms of the microbial diversity and metabolites, and this information was important to understand the properties of mixed soybeans Sufu.

Since establishment of cadre E-learning of China Earthquake Administration，its work has been widely recognized within the seismic system. However，there are also many issues we’ve seen during these years. The system needs to be re-integrated and optimized in accordance with the real operating conditions，so as to serve the students more effectively.

Background: The precise allocation and efficient use of industrial land are necessary for the development and optimization of urban production space; however, the mismatches between urban industrial land consumption and the growth of manufacturing are becoming more serious and has become the primary obstacle to sustainable urban development. Methods: Based on a combination of the Boston Consulting Group matrix, spatial mismatch model, decoupling index, GIS, and Geodetector tools, this paper conducts an empirical study on the Yangtze River Delta region in an attempt to reveal the spatio-temporal evolution of the mismatch between urban industrial land changes and the growth of manufacturing and provide a basis for spatial planning and land management in the new era. Results: The distribution of urban industrial land is characterized by high heterogeneity and agglomeration, the coexistence of expansion and contraction, and increasingly complex and diversified changes. Gross domestic product, government revenue, the added value of tertiary industry, and government investment in science and technology indicate that the goal orientation and scale effect of economic growth play a decisive role in the allocation of urban industrial land and that the influence of industrial structures and technological innovation is rapidly increasing. The interaction between the different factors is a bifactor enhancement, for example, land used for logistics and storage, utilities, commercial and other services, and the import and export trade, which have a strong synergistic enhancement effect. The mismatches between urban industrial land changes and the growth of manufacturing are still within a reasonable degree but there is an increasing number of cities with negative mismatches, making it necessary to implement a differentiated spatial adjustment and management policy. Conclusions: Compared with the mismatches of mobile resources such as labor, finance, and capital, the mismatches of immovable land resources have an increasing impact with more serious consequences and it is harder to make optimizations and corrections. However, the academic community has limited knowledge about land resource mismatches. By quantitatively assessing the mismatches between industrial land consumption and the growth of manufacturing in YRD cities, this paper argues that the mismatches can be rectified through spatial and land use planning and suggests the establishment of a zoning management and governance system to achieve the optimal allocation of urban industrial land resources through the implementation of a “standard land + commitment system” and industrial land protection lines.

Background/Aims: Our previous studies demonstrated that a novel long non-coding RNA, CYP4B1-PS1-001, was significantly downregulated in early diabetic nephropathy in vivo and in vitro, and CYP4B1-PS1-001 overexpression could inhibit the proliferation and fibrosis of mouse mesangial cells (MMCs). However, the underlying mechanism of the CYP4B1-PS1-001-mediated regulation of proliferation and fibrosis in diabetic nephropathy remains undetermined. Methods: RNA-protein pull-down assay, RNA-binding protein immunoprecipitation, and mass spectrometry were used to investigate CYP4B1-PS1-001 interacted with the upregulated protein nucleolin (NCL). siRNA method was applied to knockdown NCL in MMCs, the interaction between CYP4B1-PS1-001 and NCL were determined by Western blot analysis and RT-qPCR. The effect of CYP4B1-PS1-001 in the regulation of NCL was detected by cycloheximide (CHX) and ubiquitination assays. Results: We found that CYP4B1-PS1-001 interacts with NCL, and CYP4B1-PS1-001 inhibits the proliferation and fibrosis of MMCs depending on interaction with NCL. Furthermore, degradation of CYP4B1-PS1-001-associated NCL was mediated by a ubiquitin proteasome-dependent pathway. Conclusion: Our study provides evidence that CYP4B1-PS1-001 regulates the ubiquitination and degradation of NCL and thereby plays a critical role in the proliferation and fibrosis of MMCs, indicating that CYP4B1-PS1-001 and NCL may be promising prognostic biomarkers and molecular targets for the treatment of diabetic nephropathy.

Abstract Staphylococcus aureus can cause different types of diseases from mild skin infections to life-threatening sepsis worldwide. Owing to the emergence and transmission of multidrug-resistant strains, developing an impactful immunotherapy especially vaccine control approach against S. aureus infections is increasingly encouraged and supported. S. aureus manganese transport protein C (MntC), which is a highly-conserved cell surface protein, can elicit protective immunity against S. aureus and Staphylococcus epidermidis. In this study, we evaluated the humoral immune response and CD4+ T cell-mediated immune responses in a mouse peritonitis model. The results showed that MntC-specific antibodies conferred an essential protection for mice to reduce invasion of S. aureus, which was corroborated via the opsonophagocytic killing assay and passive immunization experiment in mice, and moreover MntC-induced Th17 played a remarkable part in preventing S. aureus infection since the MntC-induced protective immunity decreased after neutralization of IL-17 by antibody in vivo and the Th17 adoptive transferred-mice could partly resist S. aureus challenge. In conclusion, we considered that the MntC-specific antibodies and MntC-specific Th17 cells play cooperative roles in the prevention of S. aureus infection.

In general, the biomass raw materials burned by biomass power plants generally have the characteristics of variable fuel types, high moisture content, and high volatile content. In this paper, a 130 t/h biomass circulating fluidized bed (BCFB) model was established on the MWorks platform with Modelica language. The influence of biomass type changes on operation parameters, the corresponding steady-state characteristics, and the dynamic characteristics of the BCFB were carried out. The temperature corresponding to the combustion of pine was overall higher than that of the other fuels, and the flue gas from the combustion of pine had the highest concentration of SO2, up to 520.49 mg/Nm3. The flue gas from the combustion of pure cotton sticks had the highest concentration of NO, up to 254.34 mg/Nm3. The changes of fuel type and moisture content all have a great influence on the operation of BCFBs. The emission of pollutants was not only related to the element content of fuel, but also closely related to the furnace temperature. The fuel moisture content also indirectly affects the pollutant emission concentration and the steam-water system.

Abstract Heterogeneous artificial core plate models with low permeability are designed, made and evaluated based on similarity theory of heterogeneous reservoir with low-permeability physical simulation by artificial core plate model; then, simulative experiments for seepage flow patterns can be carried out. Pressure data are obtained by pressure transducers symmetrically arranged in artificial core plate models to study on the seepage flow patterns in heterogeneous reservoirs with low permeability. Experimental results show that the pressure gradient around injection and production points is high, and the pressure gradient of diagonal corner is very low. The distribution of pressure gradient changes as plane heterogeneity of artificial core plate models changes. The higher permeability increases the spread range of pressure, but the enhancement of heterogeneity has a negative effect on pressure transmission at the same time. The effect of permeability is greater than the negative impact of heterogeneous when the overall permeability of plate model is at a very low level. Non-seepage flow section becomes smaller with the increase in permeability, and the proportion of quasi-linear seepage flow section which is more conducive to fluid flow raise as seepage flow section becomes larger.

This study analyses the radar fluctuating target detection in G(0)-distributed clutter using dynamic programming – track before detect (DP-TBD) algorithm. G(0)-distributed clutter is often used to fit the complex circumstances. The authors use chi-square distribution to model the fluctuating targets. The log-likelihood ratio (LLR) is taken as the scoring function of DP-TBD. This study makes contribution to simplify the double integral into single integral in LLR and applies numerical integration with variable resolution to evaluating the LLR. Simulations prove the simplified integral equals the original integral and using LLR in DP-TBD is able to improve the performance of detecting fluctuating targets in G(0)-distributed clutter.

A novel polar format algorithm is proposed for spiral trajectory. The synthetic aperture radar (SAR) signal model of spiral trajectory platform is firstly established, then the sector-by-sector phase compensation method of spiral trajectory SAR is proposed. With the descending of the flight platform, the slant range also decreases, thus the sector-by-sector phase compensation is required to correct the quadratic terms of the Range Cell Migration to expand the effective imaging range and eliminate the defocus of the target at the edge of the scene. The 2D-NUFFT is used to transform the 2D wavenumber domain instead of the traditional nearest interpolation method, which decreases the calculation. Finally, the algorithm is validated by simulation.

Circular SAR (CSAR) as a new kind of SAR imaging mode has the advantage of long-time observation. For the moving target, the corresponding parameters of the moving target can be calculated by using the position information from the apparent coordinates of the moving target in these CSAR sub-aperture images. The CSAR sub-aperture imaging algorithms include BP and PFA, and the existing parameter estimation model is established on those well-focused and localised images. However, they account for a substantial computation. Here, an improved algorithm for parameter estimation based on CSAR sub-aperture images obtained by PFA is proposed. The wave-front phase error compensation is abandoned, and a new parameter estimation model is established based on PFA images with geometric positioning error. The computational complexity of the whole process of the algorithm is reduced, and the practicability is improved. The point target simulation shows that the method is effective and accurate.

For stationary targets, circular synthetic aperture radar (CSAR) imaging has tended to be mature and perfect. However, for moving targets, due to the long synthetic aperture and the variation of slant plane with the observation angle, the motion compensation is more complicated and has no effective approach. Based on the latest moving target trajectory reconstruction method in CSAR (2015), the polar format algorithm can be improved to image the moving targets when the motion parameters have been estimated. First, the authors placed the imaging reference point on the target to track it, and then compensated for the phase error caused by the incident angle changing and the wavefront curvature. The simulation results of point show that the algorithm is effective and accurate.

The broadened first-order sea clutter in shipborne high frequency surface wave radar (HFSWR), which will mask the targets with low radial velocity, is a kind of classical space–time coupled clutter. Space–time adaptive processing (STAP) has been proven to be an effective clutter suppression algorithm for space-time coupled clutter. To further improve the efficiency of clutter suppression, a STAP method based on a generalized sidelobe canceller (GSC) structure, named as the auxiliary channel STAP, was introduced into shipborne HFSWR. To obtain precise clutter information for the clutter covariance matrix (CCM) estimation, an approach based on the prior knowledge to auxiliary channel selection is proposed. Auxiliary channels are selected along the clutter ridge of the first-order sea clutter, whose distribution can be determined by the system parameters and regarded as pre-knowledge. To deal with the heterogeneity of the spreading first-order sea clutter, an innovative training samples selection approach according to the Riemannian distance is presented. The range cells that had shorter Riemannian distances to the cell under test (CUT) were chosen as training samples. Experimental results with measured data verified the effectiveness of the proposed algorithm, and the comparison with the existing clutter suppression algorithms showed the superiority of the algorithm.

It is known that the data acquisition and processing system plays an important role in radar target detection system. In order to meet the requirements of real-time processing and accurate transmission of echo signals in high-frequency ground-wave radar (HFGWR) systems, a new acquisition and transmission framework utilizing the designed acquisition card based on the PCIe (peripheral component interconnect express) has been designed and is presented in this paper. The Xilinx FPGA (Field-Programmable Gate Array) chip Kintex7-XC7K325T is adopted as a hardware carrier in acquisition card. The hardware’s composition, analog front-end circuit, the DMA (Direct Memory Access) transmission, FPGA structure, ADC (Analog-to-Digital Converter) chip, and performance test of this card are showed and discussed. Currently, the acquisition card has been accomplished and applied in the practical system of HFGWR.

In engineering construction, the accurate estimation of the investment cost can provide a reasonable basis for the formulation and decision-making of the construction scheme. In this paper, the wind-storage combined operation power station is taken as the research object, the investment cost estimation model is established, and the combined operation mode is analysed to obtain the annual power generation. Finally, according to the investment cost and annual power generation, the cost per unit of electricity model is established. In the past, wind farm and pumped storage power station are usually studied as two independent individuals, but in practical application, there are power conversion, regulation and control operations between them, which are interrelated and interactive. This paper regards the two as a whole and analyses their annual power generation under different cooperative operation modes. The research results of this paper can provide a theoretical basis for the preliminary cost estimation of engineering construction, so as to better determine the construction scheme of power plant.

Due to the motion of the platform, the spectrum of first-order sea clutter will widen and mask low-velocity targets such as ships in shipborne high-frequency surface-wave radar (HFSWR). Limited by the quantity of qualified training samples, the performance of the generally used clutter-suppression method, space–time adaptive processing (STAP) degrades in shipborne HFSWR. To deal with this problem, an innovative training sample acquisition method is proposed, in the area of joint domain localized (JDL) reduced-rank STAP. In this clutter-suppression method, based on a single range of cell data, the unscented transformation is introduced as a preprocessing step to obtain adequate homogeneous secondary data and roughly estimated clutter covariance matrix (CCM). The accurate CCM is calculated by integrating the approximate CCM of different range of cells. Compared with existing clutter-suppression algorithms for shipborne HFSWR, the proposed approach has a better signal-to-clutter-plus-noise ratio (SCNR) improvement tested by real data.
