The pathogenesis of type 2 diabetes (T2D) involves dysfunction in multiple organs, including the liver, muscle, adipose tissue, and pancreas, leading to insulin resistance and β cell failure. Recent studies highlight the significant role of extracellular vesicles (EVs) in mediating inter-organ communication in T2D. This review investigates the role of EVs, focusing on their presence and biological significance in human plasma and tissues affected by T2D. We explore specific EV cargo, such as miRNAs and proteins, which affect insulin signaling and glucose metabolism, emphasizing their potential as biomarkers. Pancreatic islet isolation is critical for type 2 diabetes research. Although -omics approaches have shed light on islet molecular profiles, inconsistencies persist; on the other hand, functional studies are essential, but they require reliable and standardized isolation methods. Here, we propose a simplified protocol applied to very small-sized samples collected from partially pancreatectomized living donors. Islet isolation was performed by digesting tissue specimens collected during surgery within a collagenase P solution, followed by a Lympholyte density gradient separation; finally, functional assays and staining with dithizone were carried out. Isolated pancreatic islets exhibited functional responses to glucose and arginine stimulation mirroring donors' metabolic profiles, with insulin secretion significantly decreasing in diabetic islets compared to non-diabetic islets; conversely, proinsulin secretion showed an increasing trend from non-diabetic to diabetic islets. This novel islet isolation method from living patients undergoing partial pancreatectomy offers a valuable opportunity for targeted study of islet physiology, with the primary advantage of being time-effective and successfully preserving islet viability and functionality. It enables the generation of islet preparations that closely reflect donors' clinical profiles, simplifying the isolation process and eliminating the need for a Ricordi chamber. Thus, this method holds promises for advancing our understanding of diabetes and for new personalized pharmacological approaches.

Objective: Our goal is to evaluate the effects of heat and ultraviolet (UV) irradiation on P3 facial respirator microstructure., Intervention: P3 facial filters were exposed to dry heat and UV sterilization procedures., Methods: P3 facial filter samples underwent a standardized sterilization process based on dry heat and UV irradiation techniques. We analyzed critical parameters of internal microstructure, such as fiber thickness and porosity, before and after sterilization, using 3D data obtained with synchrotron radiation-based X-ray computed microtomography (micro-CT). The analyzed filter has two inner layers called the "finer" and "coarser" layers. The "finer" layer consists of a dense fiber network, while the "coarser" layer has a less compact fiber network., Results: Analysis of 3D images showed no statistically significant differences between the P3 filter of the controls and the dry heat/UV sterilized samples. In particular, averages fiber thickness in the finer layer of the control and the 60° dry heated and UV-irradiated sample groups was almost identical. Average fiber thickness for the coarser layer of the control and the 60° dry heated and UV-irradiated sample groups was very similar, measuring 19.33 µm (±0.47), 18.33 µm (±0.47), and 18.66 µm (±0.47), respectively. There was no substantial difference in maximum fiber thickness in the finer layers and coarser layers. For the control group samples, maximum thickness was on average 11.43 µm (±1.24) in the finer layer and 59.33 µm (±6.79) in the coarser layer. Similarly, the 60° dry heated group samples were thickened 12.2 µm (±0.21) in the finer layer and 57.33 µm (±1.24) in the coarser layer, while for the UV-irradiated group, the mean max thickness was 12.23 µm (±0.90) in the finer layer and 58.00 µm (±6.68) in the coarser layer. Theoretical porosity analysis resulted in 74% and 88% for the finer and coarser layers. The finer layers' theoretical porosity tended to decrease in dry heat and UV-irradiated samples compared with the respective control samples., Conclusions: Dry heat and UV sterilization processes do not substantially alter the morphometry of the P3 filter samples' internal microstructure, as studied with micro-CT. The current study suggests that safe P3 filter facepiece reusability is theoretically feasible and should be further investigated.

Bioprinting is an emerging additive manufacturing technique which shows an outstanding potential for shaping customized functional substitutes for tissue engineering. Its introduction into the clinical space in order to replace injured organs could ideally overcome the limitations faced with allografts. Presently, even though there have been years of prolific research in the field, there is a wide gap to bridge in order to bring bioprinting from "bench to bedside". This is due to the fact that bioprinted designs have not yet reached the complexity required for clinical use, nor have clear GMP (good manufacturing practices) rules or precise regulatory guidelines been established. This review provides an overview of some of the most recent and remarkable achievements for skin, heart, pancreas and cartilage bioprinting breakthroughs while highlighting the critical shortcomings for each tissue type which is keeping this technique from becoming widespread reality.

The preparation and properties of novel ruthenium carbon-rich complexes [(Ph-C≡C-) 2-n Ru(dppe) 2 (-C≡C-bipyM(hfac) 2 ) n ] (n = 1, 2; M = Cu II , Mn II ; bipy = 2,2'-bipyridin-5-yl) characterized by single-crystal X-ray diffraction and designed for molecular magnetism are reported. With the help of EPR spectroscopy, we show that the neutral ruthenium system sets up a magnetic coupling between two remote paramagnetic Cu II units. More specifically, these copper compounds are unique examples of bimetallic and linear heterotrimetallic compounds for which a complete rationalization of the magnetic interactions could be made for exceptionally long distances between the spin carriers (8.3 Å between adjacent Cu and Ru centers, 16.6 Å between external Cu centers) and compared at two different redox states. Surprisingly, oxidation of the ruthenium redox-active metal coupling unit (MCU), which introduces an additional spin unit on the carbon-rich part, leads to weaker magnetic interactions. In contrast, in the simpler parent complexes bearing only one paramagnetic metal unit [Ph-C≡C-Ru(dppe) 2 -C≡C-bipyCu(hfac) 2 ], one-electron oxidation of the ruthenium bis(acetylide) unit generates an interaction between the Cu and Ru spin carriers of magnitude comparable to that observed between the two far apart Cu ions in the above corresponding neutral trimetallic system. Evaluation and rationalization of this coupling with theoretical tools are in rational agreement with experiments for such complex systems.

With the help of EPR spectroscopy, we show that the diamagnetic [Ru(dppe)2(-C≡C-R)2] system sets up a magnetic coupling between two organic radicals R, i.e., two nitronyl nitroxide or two verdazyl units, which is stronger than that of related platinum organometallic systems. Surprisingly, further oxidation of the ruthenium redox-active metal coupling unit (MCU), which introduces an additional spin unit on the carbon-rich part, leads to the switching off of this interaction. On the contrary, in simpler complexes bearing only one of the organic radical ligands [C6H5-C≡C-Ru(dppe)2-C≡C-R], one-electron oxidation of the transition metal unit generates an interaction between the two spin carriers of comparable magnitude to that observed in the above corresponding neutral systems.

A new series of ruthenium organometallic carbon-rich complexes, exhibiting fast electron transfer kinetics combined to a low oxidation potential, was synthesized for self-assembled monolayer (SAM) formation on gold surfaces. The molecules consist of highly conjugated ruthenium(II) mono(σ-arylacetylide) or bis(σ-arylacetylide) complexes functionalized with different bridge units with specific (protected) anchoring groups that possess high affinity for gold, such as thiol, carbodithioate, and isocyanide. Single component and mixed SAMs were prepared and fully characterized by wettability studies, infrared reflection absorption spectroscopy (IRRAS), X-ray photoelectron spectroscopy (XPS), and electrochemical analyses. By applying the Laviron's formalism, fast electron transfer kinetics (≈10(4) s(-1)) were found at the derived self-assemblies while no significant effect could have been evidenced with variation of the bridging unit and of the anchoring moiety. Interestingly, a hexyl aliphatic spacer in the bridging unit with a thiol group and dilution with suitable nonelectroactive thiols lead to better SAM organization and packing, in comparison with undiluted complexes with shorter spacers. Such features make these compounds suitable alternatives to the widely used ferrocene center as redox-active building blocks for reversible charge storage devices.

We describe how the association between an ytterbium ion and a ruthenium carbon-rich complex enables the first switching of the near-IR Yb(III) luminescence by taking advantage of the redox commutation of the carbon-rich antenna.

Association between cationic and anionic heptamethine dyes led to the formation of a new organic salt (3) displaying exceptional light-harvesting properties in the NIR spectral range and having amphoteric redox character. Preliminary results of molecular bulk heterojunction solar cells based on the title compound 3 and [60]PCBM as the only active layer reveal this new dye as a promising light-harvesting material for photovoltaics.

The concept of cyanine has been successfully extended to an anionic heptamethine dye featuring tricyanofuran (TCF) moieties in terms of structure, reactivity, and photophysical properties. Importantly, absorption and emission are red-shifted compared to its classical cationic analog without any cost in terms of thermal stability. In addition to its "cyanine" behavior, this molecule exhibits further redox properties: oxidation and reduction led to the reversible formation of radical species whose absorption is in marked contrast with that of cyanines.
