Abstract

Weather and climate patterns play an intrinsic role in societal health, yet a comprehensive synthesis of specific hazard–mortality causes does not currently exist. Country-level health burdens are thus highly uncertain, but harnessing collective expert knowledge can reduce this uncertainty, and help assess diverse mortality causes beyond what is explicitly quantified. Here, surveying 30 experts, we provide the first structured expert judgement of how weather and climate directly impact mortality, using the UK as an example. Current weather-related mortality is dominated by short-term exposure to hot and cold temperatures leading to cardiovascular and respiratory failure. We find additional underappreciated health outcomes, especially related to long-exposure hazards, including heat-related renal disease, cold-related musculoskeletal health, and infectious diseases from compound hazards. We show potential future worsening of cause-specific mortality, including mental health from flooding or heat, and changes in infectious diseases. Ultimately, this work could serve to develop an expert-based understanding of the climate-related health burden in other countries.

Abstract

From early age in the human brain occurs plasticity process that influences its development. The functioning of the brain is governed by its neuronal connectivity and the synaptic dynamics of these connections. A neuron, over thousands of synapses, can receive a large number of inputs and produce different outputs leading to the consolidation and integration of memory. Synaptic plasticity is the set of experience-dependent changes in neuronal pathways that support acquired habits. It is the ability of the nervous system to reshape connectivity between neurons, changing the functional and structural organization of neuronal circuits that allows us to adapt to the multiple and continuous changes in the environment and leading to processes such as cognitive development and the ability to learn. Synaptic plasticity is mainly due to short- and long-term mechanisms. Short-term synaptic plasticity refers to changes in synaptic strength that occurs very quickly (from one-thousandth of a second to 5 minutes) and are temporary and decay over minutes (maximum 30 minutes). Long-term synaptic plasticity is defined by a long-lasting, activity-dependent change in synaptic efficacy, last from hours up to a lifetime (from 30 minutes to weeks, months, and years) and is thought to constitute the basis of learning and memory. A significant difference occurs in the nature of the change; short-term plasticity adds only a functional change, whereas long-term plasticity causes not only functional but also structural changes. Aside from genetic factors and metabolic processes, brain development is mediated also by environmental factors. Interaction with the environment plays a key role in the development and growth of neural networks and neuroplasticity. 28. Label-free multiphoton microscopy of intracellular lipids using Coherent anti-Stokes Raman Scattering (CARS)

Abstract

Coherent Antistokes Raman Scattering (CARS) microscopy has emerged in the last decade as a powerful multiphoton microscopy technique to rapidly image lipid droplets (LDs) label-free with intrinsic three-dimensional spatial resolution in cells. In this thesis I investigate and compare the ability of hyperspectral CARS and dual-frequency/differential CARS (D-CARS) to enable the chemical specificity required to distinguish lipids of different chemical composition. In hyperspectral CARS a series of spatially-resolved images are acquired over a frequency range thus proving high chemical specificity. In D-CARS two vibrational frequencies are simultaneously excited and probed, and the resulting sum and difference CARS intensities are detected by a fast and efficient single photomultiplier. This results in a higher image speed than hyperspectral CARS and in an improved image contrast against the nonresonant CARS background with a straightforward data analysis. D-CARS and hyperspectral CARS techniques were applied to LDs in model and cellular systems. In model systems made by agarose gel, droplets of pure lipids with different degree of unsaturation (number of carbon-carbon double bonds in the fatty acyl chain) were used as test sample to compare Raman spectra with CARS spectra, and measure D-CARS images at specific chemically-selective wavenumbers. Building from this knowledge, cytosolic droplets induced by loading fatty acids to the culture media of human adipose-derived stem cells (ADSCs) were distinguished in composition both in fixed cells and in living cells during differentiation into adipocytes. Furthermore, the application of a in-house developed Hyperspectral Image Analysis (HIA) software on hyperspectral data provided spatial distributions and absolute concentrations for the chemical components of the investigated specimens. In particular quantitative information was extracted about the concentration of pure neutral lipid components within cytosolic LDs, and changes over time were inferred in living ADSCs according to the type of pure fatty acid added to the culture media.

Abstract

The increasing popularity of cruise tourism has led to the need for effective planning and management strategies to enhance the city tour experience for cruise passengers. This paper presents a deep reinforcement learning (DRL)-based planner specifically designed to optimize city tours for cruise passengers. By leveraging the power of DRL, the proposed planner aims to maximize the number of visited attractions while considering constraints such as time availability, attraction capacities, and travel distances. The planner offers an intelligent and personalized approach to city tour planning, enhancing the overall satisfaction of cruise passengers and minimizing the negative impacts on the city's infrastructure. An experimental evaluation was conducted considering Naples's fourteen most attractive points of interest. Results show that, with 30 state variables and more than 19 ∗ 10 12 possible states to be explored, the DRL-based planner converges to an optimal solution after only 20,000 learning steps. Abstract

Tuberous sclerosis complex (TSC) is an autosomal dominant disorder characterized by a multisystemic involvement. In TSC, reduced function of TSC1 and TSC2 genes products (hamartin and tuberin, respectively) leads to an hyperactivation of the mechanistic target of rapamycin (mTOR) pathway and to a consequent cell growth dysregulation. In TSC patients, neurological and neuropsychiatric manifestations, especially epilepsy and neuropsychiatric comorbidities such as autism or intellectual disability, represent the most disabling features. In particular, epilepsy occurrs up to 80% of patients, is often drug resistant and is frequently associated with neurological impairment. Due to the burden of this morbidity, different treatment strategies have been proposed with the purpose to make patients epilepsy free, such as the use of different antiepileptic drugs like vigabatrin, carbamazepine, valproic acid, and levetiracetam.
