The study focuses on the physico-chemical characteristics of the wild fruit Meyna laxiflora Robyns (Alu) and fruit powder, which was gathered from the Konkan region of Maharashtra. Alu fruit powder is made by using a drying aid called maltodextrin (MD). Powder samples (MP1, MP2, MP3, MP4 and MP5) were made using different amounts of MD (0.5, 1.0, 1.5, 2.0 and 2.5 g). Preliminary phytochemical screening of fruit pulp and powder showed the presence of some phytoconstituents such as reducing sugar, saponin, flavonoids, and terpenoids, whereas alkaloids, tannins, steroids and protein were absent in both. The parameters of the powder are directly influenced by the amount of MD, as evidenced by the variations in bulk density, total soluble solids, pH, specific gravity and the proximate among the powder samples. High solubility was found in the sample with a medium of MD. The highest of particle distribution was found in MD-added samples (MP2 (1 g), MP3 (1.5 g), MP4 (2 g)) whereas the raw (MP0) showed the least. Sensory analysis was done using 9-point hedonic scale, which revealed that MP2, MP3, and MP4 were highly acceptable. This study assumed that fresh Alu fruit, pulp and in the form of powder can be useful for the development of new products.

One of the most difficult issues in the juice industry is to manufacture juices where processing processes minimise the impact on the native characteristics of the fruits. In this study, high-pressure technology was used on whole apple fruits in order to evaluate the effect on the juice production. Two varieties, cv. Limoncella and cv. Pink Lady, were considered. Preliminarily, the fruits were subjected to different pressures, and histological as well as pomological measurements were taken in order to identify the best treatment condition, which was established to be 600 MPa for 3 min. Juice samples were then characterised by measuring the colour, viscosity, total antioxidant capacity (TAC), and total phenolic content (TPC). The storage colour stability of the juices for both varieties showed not significant L* values between the untreated and pre-treated fruits. Juices obtained from pre-treated fruits had a viscosity significantly higher than that obtained from untreated ones. Interestingly, the TPC of high-pressure processing (HPP) pre-treated juice resulted in being significantly higher compared to the untreated ones. The HPP pre-treatment can be considered as a commercial application to modulate some quality standards for apple juice production.

In this study color, texture, starch–pectin, total antioxidant capacity, microbial count, and microstructure of HPP-treated Violina pumpkin cubes were evaluated. Samples were treated at six different pressures (100 to 600 MPa–HPP100 to HPP600) for 3 min. Moisture, total soluble solids, and pH showed no significant differences between untreated (UNTR) and treated samples. Pumpkin tissue showed great structural modifications as changes in cell size and shape, cell wall damage, increased cell wall thickness, cell detachment and dehydration, and calcium ions deposition mainly from HPP300 to 600. UNTR samples showed the highest value of maximum and minimum cell elongation, perimeter segment, and a more regular cell wall thickness whereas HPP600 showed the lowest values for all these parameters. A noticeable difference was observed in HPP600 samples, with a difference in terms of color (ΔE 11.3 ± 1.9) and hardness (87.4 ± 27.8 N) compared to the UNTR ones (194.9 ± 37.9 N) whereas treatments at other pressures changed less markedly the color and texture. HPP200 could ensure a higher amount of starch and pectin availability while HPP200 and HPP400 showed the highest total antioxidants capacity. High-pressure treatment from HPP400 to 600 gave the highest destruction of microorganisms but negatively influenced the structural quality as well as texture and microstructure.

Thin-layer convective drying of plantain banana was performed at four different temperatures from 50 to 80 °C, with slice thicknesses from 2 to 8 mm. The drying curves, fitted to seven different semi-empirical mathematical models, were successfully used to fit experimental data (R2 0.72–0.99). The diffusion approach had better applicability in envisaging the moisture ratio at any time during the drying process, with the maximum correlation value (R2 0.99) and minimum value of x2 (2.5×10−5 to 1.5×10−4) and RMSE (5.0 ×10−3 to 1.2×10−2). The Deff, hm, and Ea values were calculated on the basis of the experimental data, and overall ranged from 1.11×10−10 to 1.79×10−9 m2 s−1, 3.17×10−8 to 2.20 ×10−7 m s−1 and 13.70 to 18.23 kJ mol−1, respectively. The process energy consumption varied from 23.3 to 121.4 kWh kg−1. The correlation study showed that the drying temperature had a close correlation with hm value and sample hardness. A significant (p < 0.05) increase in hardness of dried plantain banana was observed at 80 °C compared to the other temperatures. Additionally, the sample hardness and process energy consumption were more positively correlated with the thickness of the samples.

BACKGROUND AND OBJECTIVES: Smart cities are technology-driven urban agglomerations. Sustainability, competitiveness, and quality of life are the three cardinal principles for smart cities. The available studies specific to smart cities are related to smart technologies, innovations, smart infrastructure, etc., and limited studies are conducted from citizens’ perspectives. The present study aims to assess the impact of governance, general well-being, also called ease of living of citizens, and citizens’ perception of smart cities on their quality of life in mid-sized smart cities of India. The innovation and novelty of present study is that it attempts to fill the research gap in studies on smart cities by adopting the citizen-centric approach rather than infrastructure and technology-centric approach in developing countries. METHODS: Three Indian mid-sized smart cities, Pune, Nagpur, and Indore were identified for the study. For the purpose of this study, a few hypotheses were developed and a structured questionnaire was prepared from the literature review. The data was collected from Pune, Nagpur, and Indore smart cities and it was analyzed by performing factorial analysis and Structural Equation Modelling using appropriate statistical package software. FINDINGS: The findings from the statistical analysis validated the hypotheses. The results of the study indicate a strong positive impact of citizens’ perception of smart cities on quality of life (Standardized Estimates = 0.66) while governance (Standardized Estimates = 0.11) and ease of living (SE=0.46) presented low to medium impact relationship with citizens’ quality of life. CONCLUSION: The findings of this study concludes that citizens’ perception has highest influence on their quality of life. Considering these findings in the context of mid-sized smart cities, this study significantly contributes in existing research on tools and indicators of quality-of-life assessment in urban centers specifically in developing countries. Valproic acid (VPA) is one of the HDAC inhibitors used for the treatment of neurological disorders and hematological malignancies. Its role in self-renewal and proliferation of hematopoietic stem cells (HSCs) is well studied, but little is known about its involvement in regulating megakaryopoiesis and thrombopoiesis. In this study, we evaluated the role of VPA in megakaryopoiesis by using MEG-01, a megakaryoblast cell line. Our results show that VPA treatment differentiates MEG-01 cells to megakaryocytes (MK) and platelet-like particles. It was confirmed by augmented expression of MK and PLT-specific markers, higher ploidy, and PLT functionality. We assessed the molecular events underlying megakaryopoiesis. In the present study, we found an upregulation of Notch3 and its downstream target PDGFR-β upon VPA treatment. The direct role of Notch3 in megakaryopoiesis has not yet been studied. PDGFR-β is known to control actin organization during vascular smooth muscle cell differentiation. The actin cytoskeleton plays important role during proplatelet and PLT formation. We found an upregulation of Rac/Cdc42 GTPase and its downstream effectors that are the key players during actin polymerization events. We speculate that VPA induces PLT formation through Notch-3 signaling that in turn modulates actin polymerization that is one of the crucial steps necessary for thrombopoiesis. These studies were also confirmed with knockdown of Notch3 in MEG01 by using ShRNA approach as well as with apheresis-derived CD34+ cells. Altogether, these findings provide an evidence for a novel role of Notch3 in regulating platelet formation.

This study is aimed at evaluating the quality of strawberry nectar following high-pressure processing (HPP) at different pressure levels (400, 500, and 600 MPa for 3 minutes) and thermal pasteurization (TT) at 75°C for 15 s. Key parameters include total colour difference (Δ E ∗), aparent viscosity, colour analysis via CIELAB coordinates, and pH. Additionally, residual enzymatic activity (RA%) of polyphenol oxidase (PPO) and peroxidase (POD) was measured. Strawberry nectar (12°Brix) was prepared and subjected to processing, with thermal treatment in 50 mL tubes and HPP in 40 mL polyethylene bags. Samples were stored under refrigeration during 21 days, with microbial analysis conducted to assess microbiological stability during storage. TT exhibited a more pronounced reduction in total aerobic microbial count (TAMC) compared to HPP, which showed no detectable yeast and mould. Apparent viscosity varied among samples, with HPP at 600 MPa displaying a significant increase of 52.1% compared to the control. The pseudoplastic behavior, characterized by n values lower than 1, was consistent across all treatments. TT led to a reduction in L ∗ brightness value during storage, whereas HPP-treated samples underwent colour changes over time with Δ E ∗ of 3.1 for 400 MPa, 3 minutes, at 21 days. For PPO and POD, the 600 MPa treatment exhibited the highest inhibition (44% and 4% RA, respectively). This comprehensive study provides insights into how different HPP processing conditions affect the quality attributes, enzymatic activity, and microbiological stability of strawberry nectar during production and refrigerated storage. Stem cells have peculiar property to self-renew and differentiate. It is important to control their fate in safe and effective ways for their therapeutic use. The mediators of essential polyunsaturated fatty acids (PUFAs) namely Arachidonic acid (AA) and Docosahexanoic acid (DHA) are known to play a role in haematopoiesis via various metabolic pathways [1]. However the direct effect of purified AA or DHA on haematopoiesis has not been well investigated yet. We have reported that oral administration of PUFAs enhanced haematopoiesis in mice [2]. Signaling Leukocyte Antigen Molecule (SLAM) (CD48−CD150+) phenotype consists of pure population of haematopoietic stem cells (HSCs). Herein we observed higher percentage of SLAM (CD48−CD150+) phenotype in the bone marrow (BM) cells of mice fed with AA or DHA compared to PBS fed control mice. Data from engraftment study depicts that BM from AA/DHA-fed mice showed higher absolute number of donor cells in recipient mice compared to control. The enhanced hematopoiesis observed in AA/DHA-fed mice was returned to normal when the mice were kept on normal diet for six weeks (after ten days of oral feeding). We confirmed GCMS (Gas Chromatography-Mass Spectroscopy) retention times of AA and DHA by co-injecting fatty acid extract from AA or DHA fed mice with purified AA or DHA standards respectively. Representative flow cytometry profile of Lin−Sca-1+c-kit+(LSK) cells showed higher expression of CXCR4 protein and ligands of Wnt, Notch1 signaling in BM of AA/DHA-fed mice.
