Abstract

The effect of diethylcarbamazine (DEC) treatment on the concentration of histamine (H) and some other biogenic amines namely dopamine (DA), norepinephrine (NE), epinephrine (E) and 5-hydroxytryptamine (5-HT) has been investigated in different brain regions and some peripheral tissues of normal and Acanthocheilonema viteae infected adult Mastomys natalensis. During Acanthocheilonema viteae infection histamine level increased significantly in cerebral hemispheres, mid-brain hypothalamus, lungs. The levels of monoamines were unaffected. Histamine level significantly increased in all the organs studied, except adrenal glands, three hours after diethylcarbamazine (DEC) treatment on normal as well as filaria-infected mastomys. Enhancement in histamine level of tissues after DEC treatment was not responsible for development of allergic reactions in infected animals. Abstract

During the last nine years, CDRI, Lucknow, has been working in close collaboration with WRAIR, Washington and AFRIMS, Bangkok for standardization of experimental models for screening of new blood schizontocidal, radical curative (anti-relapse) and causal prophylactic agents and the pre clinical efficacy trials of the compounds synthesized and developed by US Army antimalarial drug development programme and the CDRI. This joint biomedical collaborative research programme has led to the establishment of reliable anti-malarial screening models. The reproducibility of the following antimalarial screens at CDRI has been validated using standard drugs and it is encouraging to point out that our results with the standard drugs are in close agreement with the data obtained at AFRIMS, Bangkok and WRAIR, Washington. more...

46. Effect of Andrographolide on Monkey Hepatocytes against Galactosamine Induced Cell Toxicity : An In-Vitro Study.

Abstract

Andrographolide, the active constituent, isolated from the plant Andrographis paniculata showed concentration dependent (1-100microg/mL) activity against galactosamine (GALN 400 microg/mL) induced acute injury in isolated monkey hepatocytes, following 48 hours of the incubation at 370 C. The effect of andrographolide was measured on viability (trypan blue exclusion and oxygen uptake tests), biochemical marker enzymes - aspartate amino transaminase (AST), alanine amino transaminase (ALT) and alkaline phosphatase (AlP) and bile contents (bile salts and bile acids). It significantly increased the percent viability of cells and was capable of preserving 90-100% cell integrity. The cellular leakage of enzymes and bile contents following incubation with GALN was significantly modulated by andrographolide treatment. Its activity was found superior to that of silymarin, a standard hepatoprotective compound, tested simultaneously for comparison.
