Anticorpos monoclonais (AcMs) produzidos com uma amostra brasileira do herpesvírus bovino tipo 5 (BHV-5) foram utilizados na adaptação e padronização de técnicas rápidas para o diagnóstico de infecções pelo BHV-1 e BHV-5. A detecção de antígenos virais por imunofluorescência em células descamativas (IFCD) foi comparada com o isolamento viral, em secreções nasais de 16 bezerros, sendo 6 inoculados com o BHV-1 e 10 com o BHV-5. De 203 amostras testadas, 182 (89,6%) apresentaram resultados concordantes nos dois testes (143 positivas; 39 negativas). Comparando-se com o isolamento, a IFCD apresentou sensibilidade de 96,6%, especificidade de 70,9%, e precisão de 89,6%. Os AcMs também foram utilizados para detectar antígenos do BHV-5 em impressões frescas do cérebro de bezerros acometidos de enfermidade neurológica e em células de cultivo utilizadas na técnica de soroneutralização (SN), permitindo a obtenção dos resultados em 24 horas. A técnica, denominada de soroneutralização rápida (SNR), apresentou sensibilidade de 97,3%; especificidade de 95,5% e precisão de 96,2% em comparação com a SN tradicional; e sensibilidade de 94,7%, especificidade de 97,3% e precisão de 96,1% em comparação com um ELISA comercial. Esses resultados demonstram que esses AcMs podem ser muito úteis para uso em técnicas rápidas de diagnóstico de infecções suspeitas de BHV-1 ou de BHV-5, sobretudo em situações de surto, quando a tomada de decisões adequadas depende de um diagnóstico rápido e confiável.

Methylmalonic acidemias consist of a group of inherited neurometabolic disorders caused by deficiency of methylmalonyl-CoA mutase activity clinically and biochemically characterized by neurological dysfunction, methylmalonic acid (MMA) accumulation, mitochondrial failure and increased reactive species production. Although previous studies have suggested that nitric oxide (NO) plays a role in the neurotoxicity of MMA, the involvement of NO-induced nitrosative damage from inducible nitric oxide synthase (iNOS) in MMA-induced seizures are poorly understood. In the present study, we showed a decrease of time spent convulsing induced by intracerebroventricular administration of MMA (2 micromol/2 microL; i.c.v.) in iNOS knockout (iNOS(-/-)) mice when compared with wild-type (iNOS(+/+)) littermates. Visual analysis of electroencephalographic recordings (EEG) showed that MMA injection induced the appearance of high-voltage synchronic spike activity in the ipsilateral cortex which spreads to the contralateral cortex while quantitative electroencephalographic analysis showed larger wave amplitude during MMA-induced seizures in wild-type mice when compared with iNOS knockout mice. We also report that administration of MMA increases NOx (NO(2) plus NO(3) content) and 3-nitrotyrosine (3-NT) levels in a greater extend in iNOS(+/+) mice than in iNOS(-/-) mice, indicating that NO overproduction and NO-mediated damage to proteins are attenuated in iNOS knockout mice. In addition, the MMA-induced decrease in Na(+), K(+)-ATPase activity, but not in succinate dehydrogenase (SDH) activity, was less pronounced in iNOS(-/-) when compared with iNOS(+/+) mice. These results reinforce the assumption that metabolic collapse contributes for the secondary toxicity elicited by MMA and suggest that oxidative attack by NO derived from iNOS on selected target such as Na(+), K(+)-ATPase enzyme might represent an important role in this excitotoxicity induced by MMA. Therefore, these results may be of value in understating the pathophysiology of the neurological features observed in patients with methylmalonic acidemia and in the development of new strategies for treatment of these patients.

Contamination of cell cultures with adventitious viruses may pose serious risks for virology diagnosis, research and vaccine production. This article reports the selection and characterization of three cell lines resistant to bovine viral diarrhea virus (BVDV), a major contaminant of cell cultures. The resistant cells were obtained from canine (MDCK), swine (PK-15) and rabbit (RK-13) parental cell lines by expanding and cloning cells that survived lytic infection with BVDV. All three selected cell lines were resistant to three standard BVDV strains and to 10 field isolates as demonstrated by immunofluorescence for viral antigens and by co-cultivation with susceptible cells. Inoculation of selected cells with BVDV (10 TCID50/cell) resulted in frequencies of infection of <10(-5) (MDCK-R, PK-15R) and 3.3 x 10(-4) (RK-13R). Comparing to the parental cell lines, these cells were >10,000-, >20,000- and 600-fold less susceptible to BVDV, respectively. Inoculation of resistant cells with BVDV in the presence of polyethylene-glycol increased the frequency of infection in the order of >437-, >346- and 87-fold, respectively, indicating that resistance is probably due to a block in viral entry. Nevertheless, each resistant cell line retained the susceptibility to three other viruses that replicate in the parental cells. Thus, these cells may be useful for many virology purposes, with a very low risk of BVDV contamination.
