Cattle of polled phenotype is convenient for breeders, as it decreases the risk of animals being hurt and ensures safety of workers. We developed the system for editing cattle genome using CRISPR/Cas9 which will allow production of animals with polled phenotype genetically based on any cattle breed without changing its main phenotypic traits.

Bovine leukemia virus (BLV) causes enzootic leukemia - a chronic infectious disease occurring against the background of embedding the virus in the genome of B-lymphocytes and leading to malignization, invasion of tumor cells in organs and the formation of tumors. The disease is common in the United States, Japan, and Asia. In Russia, up to 30% is infected with BLV. Moreover, there is evidence of the presence of antibodies to the BLV virus in some groups of people, and the relationship between BLV and cancer in humans is widely discussed. All this indicates an urgent need to study BLV and create breeds resistant to it. The development of approaches to solving this problem is complicated by the fact that the receptor through which the infection is carried out is still unknown. Recently, it has been suggested that the virus penetrates the animal's lymphocytes using the CD209 molecule. In this paper, we propose a genome editing system based on CRISPR/Cas9 to get a knockout for this gene. We assume that animals obtained using the presented genome editing system will be resistant to infection with the bovine leukemia virus.

Obesity is a significant metabolic disorder associated with excessive fat accumulation and insulin resistance. In this study, we explored a gene therapy approach to treat obesity in agouti mice using adeno-associated viruses (AAVs) carrying PRDM16, FoxP4, or Follistatin (FST) genes, which are known to promote the browning of white adipose tissue. Mice treated with AAVs encoding PRDM16, FoxP4, or FST genes showed a reduction in body weight (10–14%) within the first three weeks after administration, compared to the control groups. A lipidomic analysis of the adipose tissue revealed a dramatic reduction in triacylglycerol (TAG) species with low carbon numbers (40–54 acyl carbons) in treated mice. [ABSTRACT FROM AUTHOR]

Introduction: In this review, the analysis of technologies for obtaining biologically active proteins from various sources is carried out, and the comparative analysis of technologies for creating producers of biologically active proteins is presented. Special attention is paid to genetically modified animals as bioreactors for the pharmaceutical industry of a new type. The necessity of improving the technology of development transgenic rabbit producers and creating a platform solution for the production of biological products is substantiated. The advantages of using TrB for the production of recombinant proteins: The main advantages of using TrB are the low cost of obtaining valuable complex therapeutic human proteins in readily accessible fluids, their greater safety relative to proteins isolated directly from human blood, and the greater safety of the activity of the native protein. The advantages of the mammary gland as a system for the expression of recombinant proteins: The mammary gland is the organ of choice for the expression of valuable recombinant proteins because milk is easy to collect in large volumes. Methods for obtaining transgenic animals: The modern understanding of the regulation of gene expression and the discovery of new tools for gene editing can increase the efficiency of creating bioreactors for animals and help to obtain high concentrations of the target protein. Atherosclerosis is a lipid-driven, chronic inflammatory disease that leads to plaque formation at specific sites of the arterial tree. Being the common cause of many cardiovascular disorders, atherosclerosis makes a tremendous impact on morbidity and mortality rates of cardiovascular diseases (CVDs) in countries with higher income. Animal models of atherosclerosis are utilized as useful tools for studying the aetiology, pathogenesis and complications of atherosclerosis, thus, providing a valuable platform for the efficacy testing of different pharmacological therapies and validation of imaging techniques. To date, a large variety of models is available. Pathophysiological changes can be induced in animals by either an atherogenic diet or genetic manipulations.
