For purposes of description, the cells of the Malpighian tubules of the grasshopper may be divided into basal, intermediate, and apical zones. The basal zone of the cell contains what appears to be an elaborate infolding of the cell membrane at the base. This condition results in the basal cell cytoplasm being divided into many compartments. The compartments contain mitochondria that are often arranged in rows. Other small bodies which possess relatively dense outer borders and less dense cores were observed within the compartments. These bodies are unidentified. The brush border of the apical zone contains a multitude of vertically arranged protoplasmic processes. Stages were found which suggest that the filamentous mitochondria migrate from the cell body to the base of the protoplasmic processes, where they enter them and move apically. Some of the mitochondria were observed at the very tips of the processes where they enlarge producing an accompanying bulging of the tips. This condition is interpreted as a stage in the pinching off of the mitochondria-laden tips of the protoplasmic processes.

The dictyosome (Golgi body) in the secondary spermatocyte of the cricket appears in electron micrographs as a duplex structure composed of (a) a group of parallel double-membraned lamellae and (b) a group of associated vacuoles arranged along the compact lamellae in a chain-like fashion. This arrangement of ultramicroscopic structure for the dictyosomes is strikingly comparable to that described for the Golgi apparatus of vertebrates. Accordingly, the two are considered homologous structures. Associated with the duplex structure of the dictyosomes is a differentiated region composed of small vacuoles. This is thought to represent the pro-acrosome region described in light microscope preparations. In the spermatid the dictyosomes fuse, giving rise to the acroblast. Like the dictyosomes, the acroblasts are made up of double-membraned lamellae and associated vacuoles. In addition, a differentiated acrosome region is present which, in some preparations, may display the acrosome vacuole and granule. Both the dictyosomes and acroblasts are distinct from mitochondria.

The effect of x-irradiation on grasshopper spermatogenesis was studied with the aid of light and electron microscopy. The insects were irradiated at the second instar prior to the presence of maturation stages and observed at the last instar and imago stages. Dosages of 100 to 600 roentgens were found to retard the differentiation of the nucleus and mitochondrial nebenkern in spermatids. Evidence is presented that irradiation causes a curtailment and disorganization in the differentiation of the nebenkern from mitochondria. The above doses also induced the formation of supernumerary centrioles, flagellar filaments and acrosomes; nuclear disorganization as well as pycnosis and fragmentation also occur. The nucleus appears to be drawn toward each radiation-induced supernumerary acrosome, with consequent multipolarity of the nucleus. Induction of a set of flagellar filaments is seen only where the centriolar structure is in contact with the nucleus. Details are given of an organelle, heretofore not described, that is composed of anastomosed and interwoven cytoplasmic strands.

1. Observations on the morphological changes of mitochondria preparatory to the formation of the nebenkern, as well as changes within the nebenkern, are reported. 2. Mitochondria enlarge and divide during the meiotic divisions. 3. The mitochondria of the spermatid elongate, become filamentous, form a crescent, and partially encircle the nucleus. 4. Nodes which develop on either end of the crescent become entwined as they move toward each other. 5. The two nodes coalesce to form a filamentous or early type nebenkern which is described by others as chromophilic. 6. Internal rearrangement and partial dissolution of the filaments result in the development of the late or chromophobic nebenkern which separates into two distinct bodies. 7. The nebenkern moieties send out processes toward the centrosome, and after making contact, elongate and occupy part of the space between the tail filaments and sheath of the spermatozoon.
