The hydrodynamics of karst terrain are highly complex due to the diverse fractures and reservoirs within limestone formations. The time delay between rainfall events and subsequent flow into reservoirs exhibits significant variability. However, these hydrological processes are not easily visualized in karst topography. Subsurface geophysics, specifically 2D time-lapse electrical resistivity tomography (ERT), provides an effective method for studying the relationships between hydrological and geophysical features. In our research, we adopted ERT in the Karst Critical Zone (KCZ) to visualize specific karstic zones, including cave galleries, water storage reservoirs, wetting fronts, soil layers, and potential preferential flow paths down to a depth of 20 m. To capture spatial and seasonal variations in resistivity, we presented a comprehensive approach by combining sixteen inversion models obtained between February 2020 and September 2022 above the Villars Cave in SW-France-a well-known prehistoric cave. We used a multi-dimensional statistical technique called Hierarchical Agglomerative Clustering (HAC) to create a composite model that divided the synthetic ERT image into eight clusters representing different karst critical zones. The ERT image clearly visualized the cave gallery with high resistivity values that remained consistent throughout the seasons. Our analysis revealed a close seasonal relationship between water excess and resistivity variations in most infiltration zones, with time delays increasing with depth. The karst reservoirs, located at significant depths compared to other clusters, displayed sensitivity to changes in water excess but were primarily affected by fluctuations in water conductivity, particularly during summer or dry periods. Cyclic di-AMP is an essential signalling molecule in Gram-positive bacteria. This second messenger regulates the osmotic pressure of the cell by interacting directly with the regulatory domains, either RCK_C or CBS domains, of several potassium and osmolyte uptake membrane protein systems. Cyclic di-AMP also targets stand-alone CBS domain proteins such as DarB in Bacillus subtilis and CbpB in Listeria monocytogenes. We show here that the CbpB protein of Group B Streptococcus binds c-di-AMP with a very high affinity. Crystal structures of CbpB reveal the determinants of binding specificity and significant conformational changes occurring upon c-di-AMP binding. Several studies in humans indicate that low birth weight predisposes individuals to obesity in later life. Despite the constant increase in prevalence of obesity in the canine population and the major health consequences of this affection, few investigations have been carried out on the association between birth weight and the development of overweight in dogs. The purpose of the current study was to examine the association between birth weight and some other neonatal characteristics and overweight at adulthood in a population of purebred Labrador dogs. Information was collected about the sex, age, neuter status, birth weight, and growth rates (between 0-2 days and 2-15 days of age) in 93 Labrador dogs raised under similar environmental conditions until two months old. The body condition scores (BCS, scale of 1-9) of these dogs at adulthood were recorded, with BCS equal to or greater than 6 classified as overweight. Dogs were split into two groups based on the median birth weight in the population: lower than the median (LTM) and higher than the median (HTM). A logistic regression model was applied to analyse associations between the general characteristics of the dogs (sex, age, neuter status), early life parameters (birth weight, growth rates) and overweight at adulthood. Birth weight was the only early-life parameter found to be associated with overweight (p value = 0.032) with a prevalence of overweight of 70% among the dogs with LTM birth weight vs. 47% in dogs born with HTM birth weight. Overweight was also associated with age and neuter status (p value = 0.029 and 0.005 respectively). Our results suggest that, as in humans, dogs with the lowest birth weights are more likely to become overweight at adulthood. More studies are needed to further examine this relationship and to explore the underlying mechanisms. Objective: Vancomycin, a commonly used antimicrobial, has a narrow therapeutic index; therefore, Therapeutic Drug Monitoring (TDM) is required. Although the Electronic Medical Record (EMR) may improve patient care, without appropriate optimization, it can contribute to incorrectly drawn vancomycin levels. For medication administration, nurses utilize the Medication Administration Record (MAR) for medication administration documentation and medication workflow guidance. Therefore, we hypothesized creating a MAR level order which would be incorporated into this already established medication workflow may improve the rate of correctly drawn vancomycin levels., Materials and Methods: This was a multicenter, retrospective, pre-and post-intervention study which evaluated the effect of a Medication Administration Record (MAR) level order within the EMR on the correct timing of vancomycin level collection. Vancomycin levels were classified into pre-and post-intervention groups. The primary endpoint was the rate of incorrectly drawn levels, defined as a level being drawn early, a level being drawn late, a level drawn while infusing, or a missed level., Results: A total of 1353 vancomycin levels were assessed, and 628 levels met inclusion criteria. Of the levels eligible for inclusion, 331 were in the pre-intervention period and 297 were in the post-intervention period. Levels in the post-intervention group utilizing the vancomycin MAR level order were less likely to be missed or drawn at an incorrect time (11.1 % vs 36 %, P < 0.01) and were less likely to require rescheduling (3.4 % vs 8.5 %, P < 0.01)., Conclusion: Utilization of a vancomycin MAR level order was associated with a significant decrease in incorrectly drawn vancomycin levels., (Copyright © 2020 Elsevier B.V. Central venous catheter (CVC) vascular access is common among patients on hemodialysis. CVC use carries a substantial risk of central line-associated bloodstream infections (CLABSIs), costly events that place patients at a high risk of mortality. Our hospital and dialysis organization developed a cooperative strategy to reduce the rate of CLABSI among hospitalized patients on hemodialysis with a CVC. The program included the use of training and reporting tools to guide hospital staff with CLABSI prevention, as well as leadership committees to oversee the process. Fourteen CLABSIs were reported in the 17-month period prior to the implementation of the program, while no new CLABSIs occurred in the 30 months following implementation of the program. This prevention program effectively reduced the frequency of CLABSIs. Aim: As with many chronic diseases, most French diabetes services for patients with type II diabetes now have a structured reception approach including collective educational sessions in the form of targeted workshops, thematic days, or even programs with several sessions. The aim of our study is to understand the interlocutory dynamics favorable to the adoption of new cognitions and attitudes during these sessions., Methods: We studied the verbal exchanges of nine collective nutrition education sessions (five focusing on knowledge input, and four implementing behavioral simulation). A total of 76 patients with type II diabetes (per group of six to 13 participants) and 12 group leaders (nurses, dieticians or nutritionists) contributed to this study, based on audio recording, and analysis of these sessions lasting two hours on average (with between 4345 and 8592 words per session). The nine corpora were the subject of two types of content analysis: lexical and dialogic, with the N'vivo 8 software., Results: The results of this study suggest very precisely several ways to improve the content of the informational contributions of these educational sessions. The study of their process also makes it possible to distinguish between self-disclosure and social support in communication between participants. Finally, this study checks the effectiveness of the different types of framing of health messages in the discourse of the group leaders to facilitate the adhesion of the participants., Conclusion: In conclusion, it appears that the reaction spontaneously verbalized by the participants during the course of the educational situation, in these sessions, can be considered as an interesting indicator of their impact. A 9-year-old female rose-breasted cockatoo ( Eolophus roseicapilla ) was presented for a humeral fracture. At presentation, the bird was severely lethargic and obese. On physical examination, an open right humeral fracture, healed left ulnar fracture, and intertarsal joint swelling were present. Results of hematologic testing and biochemical analysis revealed severe leukocytosis with heterophilia and increased creatine kinase and aspartate aminotransferase activities consistent with musculoskeletal lesions. Radiographs confirmed a right humeral fracture and showed severe polyostotic lytic and expansile lesions of the appendicular skeleton, as well as an enlarged hepatic silhouette. Surgical repair of the fracture was attempted, but the bird died during the procedure. Postmortem examination revealed severe bone deformities involving the fractured humerus, both ulnas, and the left tibiotarsus. Histologic findings were consistent with multiple intraosseous lipomas of the long bones and severe hepatic lipidosis. Cyclic nucleotides are universally used as secondary messengers to control cellular physiology. Among these signalling molecules, cyclic di-adenosine monophosphate (c-di-AMP) is a specific bacterial second messenger recognized by host cells during infections and its synthesis is assumed to be necessary for bacterial growth by controlling a conserved and essential cellular function. In this study, we sought to identify the main c-di-AMP dependent pathway in Streptococcus agalactiae, the etiological agent of neonatal septicaemia and meningitis. By conditionally inactivating dacA, the only diadenyate cyclase gene, we confirm that c-di-AMP synthesis is essential in standard growth conditions. However, c-di-AMP synthesis becomes rapidly dispensable due to the accumulation of compensatory mutations. We identified several mutations restoring the viability of a ΔdacA mutant, in particular a loss-of-function mutation in the osmoprotectant transporter BusAB. Identification of c-di-AMP binding proteins revealed a conserved set of potassium and osmolyte transporters, as well as the BusR transcriptional factor. We showed that BusR negatively regulates busAB transcription by direct binding to the busAB promoter. Loss of BusR repression leads to a toxic busAB expression in absence of c-di-AMP if osmoprotectants, such as glycine betaine, are present in the medium. In contrast, deletion of the gdpP c-di-AMP phosphodiesterase leads to hyperosmotic susceptibility, a phenotype dependent on a functional BusR. Taken together, we demonstrate that c-di-AMP is essential for osmotic homeostasis and that the predominant mechanism is dependent on the c-di-AMP binding transcriptional factor BusR. The regulation of osmotic homeostasis is likely the conserved and essential function of c-di-AMP, but each species has evolved specific c-di-AMP mechanisms of osmoregulation to adapt to its environment.

Cyclic di-AMP (c-di-AMP) is a bacterial signaling nucleotide synthesized by several human pathogens. This widespread and specific bacterial product is recognized by infected host cells to trigger an innate immune response. Detection of c-di-AMP in the host cytosol leads primarily to the induction of type I interferon via the STING-cGAS signaling axis, while being also entangled in the activation of the NF-κB pathway. During their long-standing interaction, host and pathogens have co-evolved to control c-di-AMP activation of innate immunity. On the bacterial side, the quantity of c-di-AMP released inside cells allows to manipulate the host response to exacerbate infection by avoiding immune recognition or, at the opposite, by overloading the STING-cGAS pathway., (Copyright © 2017 Elsevier Ltd.
