Tetronic acid, a five-membered heterocyclic moiety present in various natural products, has emerged as a significant building block for many pharmaceutically active compounds. In this study, a novel protocol for the synthesis of the bis-tetronic acid 3,3′-((4-methoxyphenyl)methylene)bis(4-hydroxyfuran-2(5H)-one) (3) via a domino Knoevenagel–Michael reaction is presented. The natural deep eutectic solvent L-proline/glycerol 1:2 molar ratio was utilized as a solvent and catalyst, while the reaction was further promoted via microwave irradiation, providing the desired product in high yield (83%). The solvent was successfully recycled and reused up to three times.

Background: In this study, two chalcone analogs were synthesized through in silico and experimental methods, and their potential to inhibit the lipoxygenase enzyme, which plays a role in the inflammation pathway, was assessed. Specifically, this study is a continuation of previous research in which chalcone derivatives were synthesized and characterized. Objectives/Methods: In the current work, we present the re-synthesis of two chalcones, with a focus on their docking studies, NMR analysis, and dynamic simulations. The structure of each chalcone was elucidated through a combination of Nuclear Magnetic Resonance (NMR) and Density Functional Theory (DFT). The substituent effect on the absorption spectrum of the two chalcone derivatives was studied. Results: A “LOX–chalcone” complex, predicted by docking studies, was further examined using molecular dynamics (MD) simulations to evaluate the stability of the complex. After fully characterizing the “LOX–chalcone” complexes in silico, the atomic details of each chalcone’s interaction with LOX-1 and 5-LOX were revealed through Saturation Transfer Difference (STD) NMR (Nuclear Magnetic Resonance). Finally, their selectivity profile was investigated against human 15-LOX-1 and general Lipoxidase activity. Conclusions: The in silico methods suggest that chalcones could be promising lead compounds for drug designs targeting the LOX enzyme.

A hybrid alginate hydrogel–chitosan nanoparticle system suitable for biomedical applications was prepared. Chitosan (CS) was used as a matrix for the encapsulation of lavender (Lavandula angustifolia) essential oil (LEO) and Mentha (Mentha arvensis) essential oil (MEO). An aqueous solution of an acidic Natural Deep Eutectic Solvent (NADES), namely choline chloride/ascorbic acid in a 2:1 molar ratio, was used to achieve the acidic environment for the dissolution of chitosan and also played the role of the ionic gelator for the preparation of the chitosan nanoparticles (CS-NPs). The hydrodynamic diameter of the CS-MEO NPs was 130.7 nm, and the size of the CS-LEO NPs was 143.4 nm (as determined using Nanoparticle Tracking Analysis). The CS-NPs were incorporated into alginate hydrogels crosslinked with CaCl2. The hydrogels showed significant water retention capacity (>80%) even after the swollen sample was kept in the aqueous HCl solution (pH 1.2) for 4 h, indicating a good stability of the network. The hydrogels were tested (a) for their ability to absorb dietary lipids and (b) for their antimicrobial activity against Gram-positive and Gram-negative foodborne pathogens. The antimicrobial activity of the hybrid hydrogels was comparable to that of the widely used food preservative sodium benzoate 5% w/v.

A ligand-free approach for the Suzuki-Miyaura cross coupling reaction using Natural Deep Eutectic Solvents (NaDES) towards coumarin analogs is described. A model reaction between the synthetically prepared 3-(4-acetyloxy-phenyl)-6-bromo-4-methyl-coumarin (3b) and phenylboronic acid was performed in five different NaDES as well as in pure glycerol, using two inorganic bases and palladium catalysts. The reaction proceeded smoothly in Choline Chloride/Glycerol (ChCl/Gly) and Betaine/Glycerol (Bet/Gly) NaDES at 90 °C in 24 h, affording the desired product in high yields up to 95%. The combination of K2CO3, Pd(OAc)2 and ChCl/Gly NaDES provided optimum yields and high purity of the desired compounds, while the solvent was successfully recycled and reused up to two times. The developed methodology is applicable to boronic acids bearing various substituents. The formation of palladium nanoparticles in the reaction mixture was observed, and the size of the nanoparticles was associated with the reaction yield. In addition, in all the glycerol-based NaDES, an effective removal of the acetyl group of the acetyloxy–coumarin analogs was observed; thus, it is noteworthy that the Suzuki–Miyaura coupling and the deacetylation reaction were achieved in one pot. The ten novel coumarin derivatives synthesized were structurally characterized using 1D and 2D NMR spectroscopy and were tested for their cytotoxicity against the A431 squamous cancer cell line, presenting significant activity.

The present study focuses on the synthesis of the natural product 4-methyl-umbelliferone (4-MU, hymecromone), the preparation, characterization, and biological activity evaluation of 4-MU inclusion complexes with β-cyclodextrin (β-CD), as well as their incorporation into pharmaceutical tablets. The inclusion complexes (ICs) were characterized using DLS, SEM, TGA as well as FT-IR, UV-vis, and NMR spectroscopies. The release profile of 4-MU from the β-CD-4-MU ICs was studied in three different pH: 1.2 (aqueous hydrochloric acid), 7.4, and 6.8 (phosphate-buffered solutions), to simulate the stomach, physiological, and intestine pH, respectively. The ICs were incorporated in pharmaceutical tablets which were prepared by direct compression and were characterized for their mechanical properties. The optimal composition of 4-MU as the active pharmaceutical ingredient (API) and excipients was determined using design of experiment (DoE), and the dissolution studies were performed at pH 1.2 at 37 ± 0.5 °C. The sustained release profile of the pharmaceutical tablets showed a delayed burst release effect at 20 min (20% drug release) compared to that of the ICs at the same time interval (70%). The results indicated that the kinetic model describing the release profile of 4-MU from the ICs and tablets is the Higuchi model, while the release mechanism is swelling and diffusion, as was indicated by the Korsmeyer–Peppas kinetic model. The optimization analysis revealed that the optimum composition contains x1 = 150.95 mg of β-CD-4-MU ICs, x2 = 82.65 mg of microcrystalline cellulose, and x3 = 12.40 mg of calcium phosphate.

In this work, a greener approach for the extraction of Greek propolis using ultrasound-assisted extraction method in combination with Natural Deep Eutectic Solvents (NADES) is presented. Propolis is a natural material of outmost interest as it possesses various biological and pharmacological activities and is therefore used for the manufacturing of extracts useful to various fields, such as pharmaceutics, cosmetics etc. Herein, five NADES were task-specifically selected as appropriate extraction solvents since they provide important assets to the final NADES-extracts, comparing to the conventionally used organic solvents. The screening study of the prepared solvents indicated the NADES L-proline/D,L-Lactic acid as the most effective medium for the raw propolis extraction due to the extract’s high total phenolic content as well as its’ significantly higher antioxidant activity. Then, the extraction using the selected NADES, was optimized by performing Experimental Design to study the effect of extraction time, propolis-to-solvent ratio and the %NADES content in the NADES-water system. All the extracts were characterized regarding their antioxidant activity and total phenolic content. The optimum NADES-extract as well as an extract derived by extraction using a conventional hydroethanolic solution were further characterized by performing LC/MS/MS analysis. The results showed that the NADES-extracts composition was similar or superior to the hydroethanolic extracts regarding the presence of valuable phytochemicals such as apigenin, naringenin etc. A disadvantage that is usually mentioned in the literature regarding the extractions using NADES is that the extracted bioactive compounds cannot be easily separated from the NADES in order to obtain dry extracts. However, this drawback can be converted to an asset as the task-specifically designed NADES that are used in this study add value to the end product and the optimum as-obtained NADES-extract has been successfully incorporated in a cosmetic cream formulation. In this work, The antioxidant activity and organoleptic characteristics of the cream formulation were also determined.

Introduction: The identification of chemical compounds that interfere with SARS-CoV-2 replication continues to be a priority in several academic and pharmaceutical laboratories. Computational tools and approaches have the power to integrate, process and analyze multiple data in a short time. However, these initiatives may yield unrealistic results if the applied models are not inferred from reliable data and the resulting predictions are not confirmed by experimental evidence.Methods: We undertook a drug discovery campaign against the essential major protease (MPro) from SARS-CoV-2, which relied on an in silico search strategy –performed in a large and diverse chemolibrary– complemented by experimental validation. The computational method comprises a recently reported ligand-based approach developed upon refinement/learning cycles, and structure-based approximations. Search models were applied to both retrospective (in silico) and prospective (experimentally confirmed) screening.Results: The first generation of ligand-based models were fed by data, which to a great extent, had not been published in peer-reviewed articles. The first screening campaign performed with 188 compounds (46 in silico hits and 100 analogues, and 40 unrelated compounds: flavonols and pyrazoles) yielded three hits against MPro (IC50 ≤ 25 μM): two analogues of in silico hits (one glycoside and one benzo-thiazol) and one flavonol. A second generation of ligand-based models was developed based on this negative information and newly published peer-reviewed data for MPro inhibitors. This led to 43 new hit candidates belonging to different chemical families. From 45 compounds (28 in silico hits and 17 related analogues) tested in the second screening campaign, eight inhibited MPro with IC50 = 0.12–20 μM and five of them also impaired the proliferation of SARS-CoV-2 in Vero cells (EC50 7–45 μM).Discussion: Our study provides an example of a virtuous loop between computational and experimental approaches applied to target-focused drug discovery against a major and global pathogen, reaffirming the well-known “garbage in, garbage out” machine learning principle.

The efficiency of natural compounds as stabilizers against thermo‐mechanical degradation during the reprocessing of a post‐consumer recycled polypropylene (PCR‐PP) is herein investigated. For this purpose, natural active ingredients are extracted from spent coffee residue (SCR). Different methods of extraction are tested and all extracts are subjected to characterization methods in order to identify the most promising extract in terms of antioxidant properties, which is then reproduced on larger scale and added to a PCR‐PP at a content of 1.0 wt%. The stabilized compound undergoes five cycles of extrusion, after which it is compared to a nonstabilized PCR‐PP and a PCR‐PP stabilized with a commercial synthetic primary antioxidant, which have both undergone the same reprocessing. Consequently, the reprocessing of the nonstabilized PCR‐PP resulted in degradation and extrudates of higher melt flow rate (MFR) and lower thermal stability, that is, reduced oxidative induction time (OIT) and onset temperature (OOT). Contrariwise, with the natural compounds embodiment, the degradation was found to be suppressed, resulting in stable MFRs and higher OITs and OOTs, imitating the behavior of the PCR‐PP with the typical synthetic antioxidant. In conclusion, natural compounds from SCR may sufficiently serve as high value‐added antioxidants for the recycling of a PCR‐PP via remelting‐restabilization. [ABSTRACT FROM AUTHOR]

Nanostructured multi-principal element alloys (MPEAs) have been explored as next-generation engineering materials due to unique mechanical and functional properties which have significant advantages over traditional dilute alloys. However, the practical applications of nanostructured MPEAs are still limited due to the lack of scalable processing approaches to prepare a large quantity of nanostructured MPEAs, as well as lack of an efficient pathway for high-throughput discovery of better functional nanostructured MPEAs within their vast compositional space. Here we tackle these challenges by presenting an integrated approach by combining direct-ink-writing-based additive manufacturing, solid-state sintering, and chemical dealloying to manufacture hierarchically porous MPEAs. The hierarchical structure is comprised of macro- and micro-scale pores introduced via extrusion printing and polymer decomposition during sintering, as well as nanoscale pores formed via chemical dealloying. The macro- and micro-scale pores allow efficient dealloying of a large mass of material as the diffusion length that the corroding medium must penetrate remains at the scale of the ligaments formed after sintering (∼10 μ m), despite the large volume of the 3D-printed samples. In addition, this integrated approach enables versatile control of the alloy composition via precisely tuning the ratio of elemental powders in the starting ink, thus offering a pathway for high-throughput discovery of novel functional MPEAs. As a case study, multiscale macro/micro/nanoporous NiFeMn MPEAs with three different compositions were investigated as catalysts to reduce the overpotential of oxygen evolution reaction (OER), where NiFeMn-based electrocatalysts display composition-dependent performance such that the overpotential measured at a current of 0.5 A g ^−1 for OER increases in the order of Ni _58 Fe _29 Mn _13 ⩽ Ni _64 Fe _26 Mn _10 < Ni _76 Fe _18 Mn _6 . This introduced manufacturing process offers new opportunities for scalable fabrication and rapid screening of nanostructured multi-component complex alloys.

In this work, the privileged scaffold of 4-hydroxy-2quinolinone is investigated through the synthesis of carboxamides and hybrid derivatives, as well as through their bioactivity evaluation, focusing on the ability of the molecules to inhibit the soybean LOX, as an indication of their anti-inflammatory activity. Twenty-one quinolinone carboxamides, seven novel hybrid compounds consisting of the quinolinone moiety and selected cinnamic or benzoic acid derivatives, as well as three reverse amides are synthesized and classified as multi-target agents according to their LOX inhibitory and antioxidant activity. Among all the synthesized analogues, quinolinone–carboxamide compounds 3h and 3s, which are introduced for the first time in the literature, exhibited the best LOX inhibitory activity (IC50 = 10 μM). Furthermore, carboxamide 3g and quinolinone hybrid with acetylated ferulic acid 11e emerged as multi-target agents, revealing combined antioxidant and LOX inhibitory activity (3g: IC50 = 27.5 μM for LOX inhibition, 100% inhibition of lipid peroxidation, 67.7% ability to scavenge hydroxyl radicals and 72.4% in the ABTS radical cation decolorization assay; 11e: IC50 = 52 μM for LOX inhibition and 97% inhibition of lipid peroxidation). The in silico docking results revealed that the synthetic carboxamide analogues 3h and 3s and NDGA (the reference compound) bind at the same alternative binding site in a similar binding mode.

The extraction of valuable phytochemicals from natural sources is an important and constantly evolving research area. Zingiber officinale Roscoe (ginger) contains high amounts of bioactive phytochemicals, which are desirable due to their significant properties. In this work, the ability of different natural deep eutectic solvents (NaDESs) to serve as green solvents for the preparation of high added value extracts from ginger is explored, in combination with ultrasound assisted extraction. The method was optimized by applying a response surface methodology using the NaDES Bet/La/W (1:2:2.5). Three independent variables, namely the extraction time, ultrasound power and NaDES-to-dry-ginger ratio, were investigated by employing a 17-run three-level Box–Behnken Design (BBD) in order to study the correlation between the extraction conditions and the quality of the obtained extracts. The optimum conditions (in order to achieve simultaneously maximum total phenolic content and antioxidant activity), were found to be 23.8 min extraction time, 60 Watt and NaDES/ginger 25:1 w/w. In the optimum conditions the DPPH radical scavenging ability of the extracts was found to reach IC50 = 18.16 mg/mL after 120 min, whereas the TPC was 20.10 ± 0.26 mg GAE/g of dry ginger. The green methodology was also compared with the extraction using conventional solvents. All the obtained extracts were evaluated for their antioxidant activity and their total phenolic content, while the extract derived by the optimum extraction conditions was further investigated for its ability to bind to calf thymus DNA (ctDNA).

In this study, silicon phthalocyanine dichloride (SiCl2Pc) was successfully encapsulated in β-cyclodextrin (β-CD) and hydroxy-propyl-β-cyclodextrin (HP-β-CD) using the kneading method. Dynamic Light Scattering (DLS) demonstrated complexes of various hydrodynamic diameters with moderate stability in aqueous solutions. Their structural characterization by Infrared Spectroscopy (FT- IR) indicated that a part of phthalocyanine is located inside the cyclodextrin cavity. Both photophysical and photochemical studies showed that phthalocyanine's encapsulation in cyclodextrins increased its aqueous solubility. The photodynamic studies against A431 cancer cell line indicated that the complexes are more effective than pure SiCl2Pc. Pure SiCl2Pc's photodynamic effect is characterized as dose-dependent, whereas both complexes presented a biphasic dose-response photodynamic effect. For the highest energy dose of 3.24 J/cm2, pure SiCl2Pc induced mild cell toxicity. SiCl2Pc-β-CD complex was the most promising photosensitizer, exhibiting the highest photodynamic effect when irradiated at 2.16 J/cm2. [ABSTRACT FROM AUTHOR]

Mg-ion batteries represent a promising alternative to Li-ion batteries but face challenges with Mg dendrite formation on Mg metal anodes. Conventional solid alloy anodes prevent Mg dendrite formation but are prone to failure due to significant volume changes during (de)magnesiation. "Self-healing" electrodes composed of liquid alloys can undergo reversible liquid–solid phase transformations during (de)magnesiation, thereby "healing" themselves from the volume expansion-induced material degradation, offering a promising solution. Here, we employ a liquid eutectic Ga–In alloy as a conceptual "self-healing" Mg-ion battery anode with a 246 mA h gGaIn−1 theoretical capacity. This anode demonstrates an unprecedented cycle life of 2000 cycles at the C-rate of 1C, retaining 91% capacity (225 mA h g−1 post-cycled capacity) at 25 °C. We elucidate the morphology evolution and storage mechanism during (de)magnesiation using in situ wide-angle X-ray scattering and cryogenic focused ion beam scanning electron microscopy. P2-type Na2/3[Ni1/3Mn2/3]O2 (NNMO) is a promising Na-ion battery cathode for its high operation voltage, high gravimetric capacity, and low critical element content. Current methods to make NNMO, such as sol–gel, solid-state reaction, and spray pyrolysis, are not environmentally friendly, as they often use acetate and nitrate metal salt precursors, which release undesirable gaseous byproducts such as CO2 and HNO3 when these metal salts are heat-treated at high temperatures. Decarbonizing our industries and the transportation sector requires not only using batteries to store intermittent renewable energy, but also decarbonizing the synthesis methods to make key components of these batteries, i.e., the electrode materials. Here, we demonstrate an environmentally responsible synthesis of NNMO by direct conversion of metallic Ni powder with various particle sizes (10 nm, 100 nm, 1 μm, 5 μm), a process that only releases O2 and H2O as byproducts, as opposed to the harmful gases released when nickel salts are used. The resulting NNMO made from ≈1 μm commercial Ni powder exhibits an excellent cycling performance as a Na-ion battery cathode. When cycled between 2-4 V vs. Na/Na+, the material exhibits a rate-capability of ≈83 mA h g−1 at 10C (i.e., > 96% theoretical capacity) and ≈ 78 mA h g−1 at 20C (i.e., >90% theoretical capacity), together with a remarkable stability of 93% capacity retention after 500 cycles at 10C. When cycled between 1.3–4 V vs. Na/Na+, the material shows a high initial capacity of ≈134 mA h g−1 at 2C, with a capacity retention of 57% after 500 cycles. [ABSTRACT FROM AUTHOR]

In this work, the preparation of inclusion complexes, (ICs) using magnesium phthalocyanine (MgPc) and various cyclodextrins (β-CD, γ-CD, HP-β-CD, Me-β-CD), using the kneading method is presented. Dynamic light scattering (DLS) indicated that the particles in dispersion possessed mean size values between 564 to 748 nm. The structural characterization of the ICs by infrared spectroscopy (FT-IR) and nuclear magnetic resonance (NMR) spectroscopy provides evidence of the formation of the ICs. The release study of the MgPc from the different complexes was conducted at pH 7.4 and 37 °C, and indicated that a rapid release (“burst effect”) of ~70% of the phthalocyanine occurred in the first 20 min. The kinetic model that best describes the release profile is the Korsmeyer–Peppas. The photodynamic therapy studies against the squamous carcinoma A431 cell line indicated a potent photosensitizing activity of MgPc (33% cell viability after irradiation for 3 min with 18 mW/cm2), while the ICs also presented significant activity. Among the different ICs, the γ-CD-MgPc IC exhibited the highest photokilling capacity under the same conditions (cell viability 26%). Finally, intracellular localization studies indicated the enhanced cellular uptake of MgPc after incubation of the cells with the γ-CD-MgPc complex for 4 h compared to MgPc in its free form.
