Closed circuit reverse osmosis (CCRO) and forward osmosis-RO (FO-RO) were evaluated at a pilot scale to generate additional permeate from RO concentrate – achieving a recovery of 61% for CCRO and 35% for FO-RO – at a full-scale advanced water purification facility. This study assessed permeate water quality, suitability of the permeate for treatment by an ultraviolet-advanced oxidation process (UV-AOP), and cost/footprint for a conceptual 10- or 20-mgd system. Both technologies demonstrated inorganic, organic, and microbiological constituent removal suitable for blending with primary RO permeate. Virus challenge testing with MS coliphage demonstrated ≥3.7-log removal by both technologies. Pilot-scale UV/hydrogen peroxide AOP treatment of CCRO or FO-RO permeate yielded similar performance (∼1.4-log N-nitrosodimethylamine removal and ∼0.5-log 1,4-dioxane removal) as the full-scale UV-AOP that treats the RO permeate from the purification facility. The estimated full-scale total unit cost (capital plus operation and maintenance costs) of product water produced by the two technologies was estimated to range from $0.91 to $1.12 per cubic meter, depending on the design flow rate of RO concentrate treated, and is estimated to be similar between the two technologies given the +50%/–30% expected accuracy of the Class 5 cost estimate. Long-term landscape evolution is controlled by tectonic and climatic forcing acting through surface processes. Rivers are the main drivers of continental denudation because they set the base level of most hillslopes. The mechanisms of fluvial incision are thus a key focus in geomorphological research and require accurate representation and models. River incision is often modeled with a stream power model (SPM) based on the along-stream evolution of drainage area and channel elevation gradient but can also incorporate more complex processes such as threshold effects and statistical discharge distributions, which are fundamental features of river dynamics. Despite their importance in quantitative geomorphology, such model formulations have been confronted with field data only in a limited number of cases. Here we investigate the behavior of stochastic-threshold incision models across the southeastern margin of the French Massif Central, which is characterized by significant relief and the regular occurrence of high-discharge events. Our study is based on a new dataset combining measurements of discharge variability from gauging stations, denudation rates from 34 basins from 10Be cosmogenic radionuclide (CRN) concentration measurements in river sediments, morphometric analysis of river long profiles, and field observations. This new dataset is used for a systematic investigation of various formulations of the SPM and to discuss the importance of incision thresholds. Denudation rates across the SE margin of the Massif Central are in the 20–120 mm kyr−1 (equivalent to mm/ka in the figures) range, and they positively correlate with slope and precipitation. However, the relationship with the steepness index is complex and supports the importance of taking into account spatial variations in parameters (D50, discharge variability k, runoff) controlling the SPM. Overall, the range of denudation rate across the margin can mainly be explained using a simple version of the SPM accounting for spatially heterogeneous runoff. More complex formulations including stochastic discharge and incision thresholds yield poorer performances unless the spatial variations in bedload characteristics controlling incision thresholds are taken into account. Our results highlight the importance of the hypotheses used for such a threshold in SPM application to field studies and notably the impact of actual constraints on bedload size.

Abstract Exotic species are often vilified as “bad” without consideration of the potential they have for contributing to ecological functions in degraded ecosystems. The red-eared slider turtle (RES) has been disparaged as one of the worst invasive species. Based on this review, we suggest that RES contribute some ecosystem functions in urban wetlands comparable to those provided by the native turtles they sometimes dominate or replace. While we do not advocate for releases outside their native range, or into natural environments, in this review, we examine the case for the RES to be considered potentially beneficial in heavily human-altered and degraded ecosystems where native turtles struggle or fail to persist. After reviewing the ecosystem functions RESs are known to provide, we conclude that in many modified environments the RES is a partial ecological analog to native turtles and removing them may obviate the ecological benefits they provide. We also suggest research avenues to better understand the role of RESs in heavily modified wetlands.

ObjectivesProteases are key mediators of pain and altered enteric neuronal signalling, although the types and sources of these important intestinal mediators are unknown. We hypothesised that intestinal epithelium is a major source of trypsin-like activity in patients with IBS and this activity signals to primary afferent and enteric nerves and induces visceral hypersensitivity.DesignTrypsin-like activity was determined in tissues from patients with IBS and in supernatants of Caco-2 cells stimulated or not. These supernatants were also applied to cultures of primary afferents. mRNA isoforms of trypsin (PRSS1, 2 and 3) were detected by reverse transcription-PCR, and trypsin-3 protein expression was studied by western blot analysis and immunohistochemistry. Electrophysiological recordings and Ca2+ imaging in response to trypsin-3 were performed in mouse primary afferent and in human submucosal neurons, respectively. Visceromotor response to colorectal distension was recorded in mice administered intracolonically with trypsin-3.ResultsWe showed that stimulated intestinal epithelial cells released trypsin-like activity specifically from the basolateral side. This activity was able to activate sensory neurons. In colons of patients with IBS, increased trypsin-like activity was associated with the epithelium. We identified that trypsin-3 was the only form of trypsin upregulated in stimulated intestinal epithelial cells and in tissues from patients with IBS. Trypsin-3 was able to signal to human submucosal enteric neurons and mouse sensory neurons, and to induce visceral hypersensitivity in vivo, all by a protease-activated receptor-2-dependent mechanism.ConclusionsIn IBS, the intestinal epithelium produces and releases the active protease trypsin-3, which is able to signal to enteric neurons and to induce visceral hypersensitivity.

A self-complementary graph is a graph isomorphic to its complement. A set S of vertices in a graph G is a restrained dominating set if every vertex in V(G) \ S is adjacent to a vertex in S and to a vertex in V(G) \ S. The restrained domination number of a graph G is the minimum cardinality of a restrained dominating set of G. In this paper, we study restrained domination in self-complementary graphs. In particular, we characterize the self-complementary graphs having equal domination and restrained domination numbers.

Casein Kinase 2 (CSNK2) is an extremely pleiotropic, ubiquitously expressed protein kinase involved in the regulation of numerous key biological processes. Mapping the CSNK2-dependent phosphoproteome is necessary for better characterization of its fundamental role in cellular signalling. While ATP-competitive inhibitors have enabled the identification of many putative kinase substrates, compounds targeting the highly conserved ATP-binding pocket often exhibit off-target effects limiting their utility for definitive kinase-substrate assignment. To overcome this limitation, we devised a strategy combining chemical genetics and quantitative phosphoproteomics to identify and validate CSNK2 substrates. We engineered U2OS cells expressing exogenous wild type CSNK2A1 (WT) or a triple mutant (TM, V66A/H160D/I174A) with substitutions at residues important for inhibitor binding. These cells were treated with CX-4945, a clinical-stage inhibitor of CSNK2, and analyzed using large-scale triple SILAC (Stable Isotope Labelling of Amino Acids in Cell Culture) quantitative phosphoproteomics. In contrast to wild-type CSNK2A1, CSNK2A1-TM retained activity in the presence of CX-4945 enabling identification and validation of several CSNK2 substrates on the basis of their increased phosphorylation in cells expressing CSNK2A1-TM. Based on high conservation within the kinase family, we expect that this strategy can be broadly adapted for identification of other kinase-substrate relationships.

Abstract Background Parents play a pivotal role in adolescent sexual health and Human Papillomavirus (HPV) vaccination. Nurses are on the frontlines of healthcare and play a critical role in promoting HPV vaccination and parent-child sexual health communication. We enhanced the Families Talking Together (FTT) parent-based sexual health curriculum to include adolescent vaccinations herein, FTT + HPV, and trained student nurses to provide a strong HPV vaccination and parent-child sexual health communication endorsement. Methods Using a randomized attention-controlled trial design, we examined the efficacy of FTT + HPV among 519 parents and their 11–14 year old youth recruited from medically underserved communities between 2015 and 2018. Participants were recruited from 22 after-school programs (e.g., Boys and Girls Clubs) and 19 charter schools. For parents, we examined protective factors including parent-child sexual health communication and parental involvement. For youth, we examined sexual health knowledge, parent-child sexual health communication, and parent-child connectedness. To assess HPV vaccination initiation and completion, we searched IMMTRAC immunization registry records for 85% of youth and used parental report for youth without registry records. Abstract Assessments of ecosystem service and function losses of wetlandscapes (i.e., wetlands and their hydrological catchments) suffer from knowledge gaps regarding impacts of ongoing hydro-climatic change. This study investigates hydro-climatic changes during 1976–2015 in 25 wetlandscapes distributed across the world’s tropical, arid, temperate and cold climate zones. Results show that the wetlandscapes were subject to precipitation (P) and temperature (T) changes consistent with mean changes over the world’s land area. However, arid and cold wetlandscapes experienced higher T increases than their respective climate zone. Also, average P decreased in arid and cold wetlandscapes, contrarily to P of arid and cold climate zones, suggesting that these wetlandscapes are located in regions of elevated climate pressures. For most wetlandscapes with available runoff (R) data, the decreases were larger in R than in P, which was attributed to aggravation of climate change impacts by enhanced evapotranspiration losses, e.g. caused by land-use changes.

Geography and associated hydrological, hydroclimate and land-use conditions and their changes determine the states and dynamics of wetlands and their ecosystem services. The influences of these controls are not limited to just the local scale of each individual wetland but extend over larger landscape areas that integrate multiple wetlands and their total hydrological catchment – the wetlandscape. However, the data and knowledge of conditions and changes over entire wetlandscapes are still scarce, limiting the capacity to accurately understand and manage critical wetland ecosystems and their services under global change. We present a new Wetlandscape Change Information Database (WetCID), consisting of geographic, hydrological, hydroclimate and land-use information and data for 27 wetlandscapes around the world. This combines survey-based local information with geographic shapefiles and gridded datasets of large-scale hydroclimate and land-use conditions and their changes over whole wetlandscapes. Temporally, WetCID contains 30-year time series of data for mean monthly precipitation and temperature and annual land-use conditions. The survey-based site information includes local knowledge on the wetlands, hydrology, hydroclimate and land uses within each wetlandscape and on the availability and accessibility of associated local data. This novel database (available through PANGAEA https://doi.org/10.1594/PANGAEA.907398; Ghajarnia et al., 2019) can support site assessments; cross-regional comparisons; and scenario analyses of the roles and impacts of land use, hydroclimatic and wetland conditions, and changes in whole-wetlandscape functions and ecosystem services.
