Abstract

Polarized ATR spectroscopy is a powerful method for studying the molecular orientation of thin films and in particular of alpha helical polypeptides. It consists of the determination of the anisotropic optical constants of the studied system, the simulatiom of the polarized ATR spectra for various orientations of the molecular axes, and the comparison of the simulated and experimental spectra.

Abstract

Optical constants of LB CdAr monolayers have been determined using experimental Fourier transform infrared spectroscopy. The optical constants by Kramers-Kronig relations were found to exhibit and interdependent relationship. This method has been found to be effective for ultrathin films and is limited by the quality of the experimental spectra. Eight and 10 CdAr monolayers deposited on calcium fluoride, zinc selenide and silicon substrates were analyzed to determine the in-plane refractive index.

Abstract

Exocytosis depends on cytosolic domains of SNARE proteins but the function of the transmembrane domains (TMDs) in membrane fusion remains controversial. The TMD of the SNARE protein synaptobrevin2/VAMP2 contains two highly conserved small amino acids, G100 and C103, in its central portion. Substituting G100 and/or C103 with the β-branched amino acid valine impairs the structural flexibility of the TMD in terms of α-helix/β-sheet transitions in model membranes (measured by infrared reflection-absorption or evanescent wave spectroscopy) during increase in protein/lipid ratios, a parameter expected to be altered by recruitment of SNAREs at fusion sites. This structural change is accompanied by reduced membrane fluidity (measured by infrared ellipsometry). The G100V/C103V mutation nearly abolishes depolarization-evoked exocytosis (measured by membrane capacitance) and hormone secretion (measured biochemically). Single-vesicle optical (by TIRF microscopy) and biophysical measurements of ATP release indicate that G100V/C103V retards initial fusion-pore opening, hinders its expansion and leads to premature closure in most instances. We conclude that the TMD of VAMP2 plays a critical role in membrane fusion and that the structural mobility provided by the central small amino acids is crucial for exocytosis by influencing the molecular re-arrangements of the lipid membrane that are necessary for fusion pore opening and expansion.

Abstract

The molecular organization in monolayers of deuterated cadmium arachidate deposited on solid metallic and dielectric substrates or spread at the air/water interface is investigated using different techniques of Fourier transform infrared spectroscopy. For metals, the polarization modulated reflectivity shows that the molecules have a high molecular order and are oriented quasi perpendicularly with respect to the surface. The in-plane organization is determined by complementary transmission experiments on CaF2 plates. The CD2 bending vibration in mono- and multilayer Langmuir–Blodgett stacks reveals the hexagonal packing of a single monolayer irrespective of the hydrophilic or hydrophobic character of the substrate. Probing the stack organization using selective monolayer deuteration in hydrogenated Langmuir–Blodgett films shows that the first deposited monolayer of an odd-numbered stack keeps its hexagonal structure while additional bilayers adopt an orthorhombic packing induced by the strong polar head/head interactions. However a monolayer may have an orthorhombic arrangement either if it is deposited on an orthorhombic commensurable substrate or if it is at a low temperature in which case intermolecular interactions are strengthened. Finally, differential reflectivity allows a single monolayer spread at the air/water interface to be studied. It is shown that such a Langmuir film is arranged in a two-dimensional hexagonal packing with its alkyl chains oriented perpendicularly with respect to the surface and its COO- polar groups symmetrically anchored at the interface. © 1996 American Institute of Physics. Abstract

We have been studying the behavior of several globular food proteins at the air-solution interface and their capacity to stabilize aqueous foams. Our aim is to understand the relation of foaming ability to the properties of the adsorbed protein layer, and to understand the relation of the properties of the adsorbed protein layer to the structural and physicochemical characteristics of the adsorbed protein(s). We used a combination of techniques providing insight in the adsorption kinetics (ellipsometry, surface tension), in the interfacial shear rheology and in the molecular conformational rearrangements of adsorbed proteins (polarization-modulation infrared reflection-absorption spectroscopy, PM-IRRAS). We compared proteins with distinctive molecular features, native and chemically or physically modified forms of one protein, and studied the interfacial behavior of proteins in binary solutions. We found that depending on the physicochemical adsorption conditions, different proteins form qualitatively different interfacial layer, as regards monolayer or multilayer adsorption, or the interfacial shear rheology. Strikingly, very minor structural modifications prior to adsorption to the air-solution interface could lead to dramatic changes in the interfacial behavior, in parallel to dramatic changes in the foaming ability. We also showed that in the case of binary solutions of oppositely charged proteins, a clear co-adsorption occurs, leading to very high surface concentrations and to deeply modified interfacial film properties, which cannot be extrapolated from the addition of individual behaviors. In addition, preliminary results about the osmotic pressure of bulk, very highly concentrated protein solutions suggest that in interfacial studies, due to the protein crowding close to the interface, intermolecular interactions could be of prominent importance in the understanding of interfacial properties.

Abstract

International audience; For transmission FT-IR spectroscopy of ultrathin films, we have developed a new double beam approach based on the technique of polarization modulation. Simultaneous recording of sample and ref. interferograms makes detection insensitive to atm. (CO2, H2O) and substrate absorptions. In situ spectra of ultrathin films are obtained in few minutes with high signal-to-noise ratio (S/N).
