In the wild-type strains of the homobasidiomycete Schizophyllum commune microtubules were totally depolymerized by low concentrations of nocodazole, while high concentrations of benomyl only modified the structure of microtubule cytoskeleton. In the nocodazole-tolerant mutant strain NT30 the microtubule cytoskeleton remained partly functional at a nocodazole concentration which demolished the microtubules in the wild-type strains. The continuation of apical growth for several hours in the wild-type strain without cytoplasmic microtubules indicated that microtubules are not the major elements in hyphal extension growth. However, the irregular branching of the treated apical cells both in the nocodazole-sensitive and -tolerant strain suggested that an intact microtubule cytoskeleton is needed for maintaining the direct extension of the leading hyphae at the colony edge. In the nocodazole-sensitive strain growth in the absence of polymerized microtubules frequently led to the death of the apical cells even when the drug was removed. In the tolerant strain the nuclear divisions continued in spite of nocodazole, but the uninucleate hyphal compartments became multinucleate. This probably resulted from poor segregation of nuclei and septation of hyphae at telophase, which indicated that these processes might be dependent on proper polymerization of cytoplasmic microtubules in higher fungi. The different electrophoretic mobility of the beta-tubulin from the NT30 strain and its parental strains suggested that the tolerance of the NT30 to nocodazole could be due to a mutation in a beta-tubulin encoding gene.

The beta-tubulin (beta Tub)-encoding gene (tub-2) of Schizophyllum commune is the first tubulin gene isolated, cloned and sequenced from higher filamentous fungi (homobasidiomycetes). The S. commune tub-2 gene is organized into nine exons and eight introns. The introns vary from 48 to 107 nt in length, and are distributed throughout the gene. The tub-2 exons code for a protein of 445 amino acids (aa), which shows great homology with beta Tubs of filamentous ascomycetes, plants, and animals, but less homology with yeasts. The codon usage of tub-2 from S. commune is biased, as it is in most beta Tub-encoding genes of filamentous fungi. The S. commune beta Tub shows a conserved aa sequence in the C-terminal domain, which is suggested to interact with microtubule-associated proteins in animals. In contrast, the S. commune beta Tub deviates from most known beta Tubs by having a Cys165 residue, which might be significant for the insensitivity of S. commune haploid strains to the antimicrotubule drug, benomyl. In tub-2 of different haploid strains, sequence polymorphisms occur in the 5' and 3' flanking regions. The expression of tub-2 is high in young mycelium, which has a high number of extending apical cells, but decreases with the aging of the mycelium. No significant difference in the hybridization signal intensity for the tub-2 transcripts was recorded either during intercellular nuclear migration at early mating, or in mycelia with a mutation in the B mating-type gene.(ABSTRACT TRUNCATED AT 250 WORDS)

Three monoclonal alpha-tubulin antibodies YL 1/2 (Kilmartin et al., 1982), 6-11B-1 (Piperno and Fuller, 1985) and DM1A (Blose et al., 1984) were used in indirect immunofluorescence (IIF) microscopy of the microtubule (MT) cytoskeleton in tobacco (Nicotiana tabacum) pollen tubes. The majority of pollen tube MTs contain tyrosinated alpha-tubulin recognized by YL 1/2. Acetylated alpha-tubulin revealed by 6-11B-1 was detected in the generative cell and in the kinetochore fibers, in polar spindle regions, and in the cell plate of the phragmoplast during generative cell division. In addition, small fragments of acetylated microtubules were seen in the older parts of the pollen tube grown on a taxol medium. The interaction of pollen tube MTs with mAb 6-11B-1 suggested that acetylation of alpha-tubulin correlates well with the putative arrays of stable MTs.

The ecological importance of different factors generating variation in the photosynthetic rate of a pine seedling (Pinus sylvestris) under nonstressing conditions are evaluated. The data consists of 1,300 observations of the photosynthetic rate and environmental factors possibly affecting photosynthesis. Statistical analysis of the data gave the following results: 1. light solely explained about 90% of the variance of photosynthetic rate during a day, 2. light and temperature together explained about 94% of it and 3. the other factors studied (leakage from the cuvette, different spectral sensitivity of photosynthesis and the monitoring apparatus, high transpiration rate, circadian rhythm and inhibition of photosynthesis by carbohydrates) were not found to be significant from the ecological point of view, since they explain less than 0.5% of the variance.

The ultrastructure of the vegetative cell walls of the charophyte Chara aspera Willd was studied with TEM. Thallus cells, rhizoid bulbil and rhizoidal node cells were investigated. The internodal cells transverse walls contained plasmodesmata. The longitudinal walls of the internodal cells were uniform, fibrillar, with two thin structurally distinct layers with different structure facing the cytoplasm. The outermost layers of internodal, cortical and rhizoid bulbil cells were composed of randomly orientated fibrils. The longitudinal walls of the cortical cells were helicoidal in structure. In the rhizoid bulbil cell walls, six different layers could be distinguished, but their occurrence seemed to depend on the fixation, staining and cutting procedures. A middle lamella and osmophilic deposits were found in the wall between rhizoidal node cells. The cytoplasmic structure of the internodal and cortical cells was not found to differ from other species of Chara. Charasomes were observed only in cortical cells.

The three groups of proteolytic inhibitors present in resting barley grains, namely, trypsin inhibitors, Aspergillus-proteinase inhibitors, and inhibitors of endogenous proteinases, occur in both the embryo and the two endosperm tissues. There are pronounced quantitative differences, however. The three inhibitor activities in the embryo are, respectively, 6-, 0.1-, and 6-fold of those in the endosperm.During germination at 20° all inhibitor activities disappear from the endosperms in 4-5 days. Young rootlets and coleoptiles contain inhibitors of trypsin and Aspergillus proteinase, but these disappear after 4-5 days' germination. However, the trypsin inhibitor content per seedlings remains roughly constant through the whole period. The Aspergillus-proteinase inhibitors, in contrast, exhibit a pronounced increase of activity per seedling.No inhibitor activities were detected in leaves and roots at later stages of growth.The trypsin inhibitor which we have earlier purified from resting grains occurs exclusively in the two endospermal tissues and is immunologically entirely different from the trypsin inhibitors present in embryos and young seedlings.

The effects of canavanine, α,γ-diaminobutyric acid, homoarginine and lathyrine on the germination of pollen and on in-vitro growth of pollen tubes were studied in the following species: Lathyrus niger, L. silvestris, Vicia unijuga, Pisum sativum and Cicer arietinum.The effects of these non-protein amino acids depended on their quantity and on the plant species. Every amino acid had a promoting effect on germination and growth at some concentration in some species. Inhibition or promotion of pollen germination and pollen-tube growth were usually parallel. The stronger influence of some amino acid on growth than on germination may be due to slow penetration of the acid into the cell.Homoarginine and lathyrine had a promoting effect at all concentrations in L. niger, a species in which these amino acids occur naturally. In most other species they had, if anything, a very slight inhibitory effect, α,γ-Diaminobutyric acid and canavanine had the strongest inhibitory effects on the species studied. It seems possible that these amino acids are antimetabolites of common amino acids.It is obvious that non-protein amino acids can form effective hybridization barries, although the conditions in nature are more complex than in vitro. The ability to synthesize a new amino acid may therefore be of evolutionary significance in the isolation of new species and genera.
