Objective To analyze the effects of preoperative oral 10% glucose based on enhanced recovery after surgery (ERAS) concept on insulin receptor β-subunit (InsRβ) protein level and insulin resistance in postoperative gastric cancer patients. Methods Eighty patients with gastric cancer undergoing surgical treatment in Xianning Central Hospital from March 2020 to March 2022 were randomly divided into routine group and ERAS group according to perioperative treatment (n=40, each) . The routine group patients were treated with conventional perioperative treatment, while the ERAS group patients were treated with the concept of ERAS (including oral administration of 500 mL of 10% glucose solution 2 hours before surgery) . The surgical related indicators, InsRβ, blood glucose, C-reactive protein (CRP) , insulin, homeostasis model assessment insulin resistance index (HOMA-IR) , complications and patients' satisfaction were compared between two groups. Results There was no significant difference in operation time and intra-operative bleeding between two groups (P＞0.05) . The first exhaust time, the fluid intake time and postoperative hospital stay in ERAS group significantly shortened compared with those in routine group (P＜0.01) . At 1 and 3 days after surgery, InsRβ protein level decreased in both groups and was statistically higher in ERAS group than that in routine group (P＜0.05) . The levels of blood glucose, CRP, insulin and HOMA-IR increased in two groups and were significantly lower in ERAS group than those in routine group (P＜0.05) . Compared with postoperative 1 and 3 days, the expression of InsRβ protein increased in both groups of patients 7 days after surgery and was higher in ERAS group than that in routine group (P＜0.05) , and the levels of blood glucose, CRP, insulin and HOMA-IR decreased in two groups and were lower in ERAS group than those in routine group (P＜0.05) . Compared with routine group, the incidence of complications in ERAS group was slightly lower without significant difference (2.50% vs 12.50%, χ2=2.882, P=0.089) , and postoperative satisfaction was slightly higher without significant difference (97.50% vs 85.00%, χ2=2.505, P=0.114) . Conclusion Preoperative oral administration of 10% glucose based on ERAS concept can effectively alleviate postoperative insulin resistance, improve InsRβ protein level and reduce postoperative hospital stay, which are conductive to the postoperative rehabilitation of the patients with the gastric cancer.

Objective To investigate the laboratory and clinical characteristics of acute lymphoblastic leukemia (ALL) with e1a3 BCR-ABL fusion protein. Methods The clinical data, and results of bone marrow cytology, flow cytometry, karyotyping, fluorescence in situ hybridization (FISH) and real-time fluorescence quantitative PCR of 2 ALL cases with e1a3 BCR-ABL fusion protein admitted in our department were collected and analyzed. The related literature was reviewed. Results Both cases presented ALL phenotype. FISH test showed BCR-ABL positive, and RQ-PCR (p190, p210, p230) negative. Finally, the rare BCR-ABL fusion gene test was added and the 2 patients were diagnosed as rare e1a3 BCR-ABL positive ALL. One patient received TKI treatment and had achieved complete remission for more than 4 months. Another patient did not receive TKI and died in longer than 1 month after diagnosis. Conclusion Multi-technique joint detection for fusion gene is helpful for the detection and diagnosis of ALL with e1a3 BCR-ABL fusion protein, and TKI therapy is helpful to improve the prognosis.

The phytohormone abscisic acid (ABA) plays a central role in regulating stomatal movements under drought conditions. The root-derived peptide CLAVATA3/EMBRYO SURROUNDING REGION-RELATED 25 (CLE25) moves from the root to shoot for activating ABA biosynthesis under drought conditions. However, the root-to-shoot translocation of root-derived ABA and its regulation of stomatal movements in the shoot remain to be clarified. Here, we reveal that the ABA transporter ATP-binding cassette subfamily G member 25 (AtABCG25) mediates root-to-shoot translocation of ABA and ABA-glucosyl ester (ABA-GE) in Arabidopsis (Arabidopsis thaliana). Isotope-labeled ABA tracer experiments and hormone quantification in xylem sap showed that the root-to-shoot translocation of ABA and ABA-GE was substantially impaired in the atabcg25 mutant under nondrought and drought conditions. However, the contents of ABA and ABA-GE in the leaves were lower in the atabcg25 mutant than in the wild type (WT) under nondrought but similar under drought conditions. Consistently, the stomatal closure was suppressed in the atabcg25 mutant under nondrought but not under drought conditions. The transporter activity assays showed that AtABCG25 directly exported ABA and ABA-GE in planta and in yeast (Saccharomyces cerevisiae) cells. Thus, we proposed a working model in which root-derived ABA transported by AtABCG25 via xylem mediates stomatal movements in the shoot under nondrought conditions but might exhibit little effect on stomatal movements under drought conditions. These findings extend the functions of AtABCG25 and provide insights into the long-distance translocation of ABA and its role in stomatal movements. DUSPs, a diverse group of protein phosphatases, play a pivotal role in orchestrating cellular growth and development through intricate signaling pathways. Notably, they actively participate in the MAPK pathway, which governs crucial aspects of plant physiology, including growth regulation, disease resistance, pest resistance, and stress response. DUSP is a key enzyme, and it is the enzyme that limits the rate of cell metabolism. At present, complete understanding of the DUSP gene family in cotton and its specific roles in resistance to Verticillium wilt (VW) remains elusive. To address this knowledge gap, we conducted a comprehensive identification and analysis of four key cotton species: Gossypium arboreum, Gossypium barbadense, Gossypium hirsutum, and Gossypium raimondii. The results revealed the identification of a total of 120 DUSP genes in the four cotton varieties, which were categorized into six subgroups and randomly distributed at both ends of 26 chromosomes, predominantly localized within the nucleus. Our analysis demonstrated that closely related DUSP genes exhibited similarities in terms of the conserved motif composition and gene structure. A promoter analysis performed on the GhDUSP gene promoter revealed the presence of several cis-acting elements, which are associated with abiotic and biotic stress responses, as well as hormone signaling. A tissue expression pattern analysis demonstrated significant variations in GhDUSP gene expression under different stress conditions, with roots exhibiting the highest levels, followed by stems and leaves. In terms of tissue-specific detection, petals, leaves, stems, stamens, and receptacles exhibited higher expression levels of the GhDUSP gene. The gene expression analysis results for GhDUSPs under stress suggest that DUSP genes may have a crucial role in the cotton response to stress in cotton. Through Virus-Induced Gene Silencing (VIGS) experiments, the silencing of the target gene significantly reduced the resistance efficiency of disease-resistant varieties against Verticillium wilt (VW). Consequently, we conclude that GH_A11G3500-mediated bispecific phosphorylated genes may serve as key regulators in the resistance of G. hirsutum to Verticillium wilt (VW). This study presents a comprehensive structure designed to provide an in-depth understanding of the potential biological functions of cotton, providing a strong foundation for further research into molecular breeding and resistance to plant pathogens. Objective To explore the clinical value of a novel CD45-negative cell sorting technology in sorting and enriching tumor plasma cells in multiple myeloma (MM). Methods A total of 60 newly diagnosed MM patients from 3 hospitals in Chongqing from July 2018 to December 2019 were recruited in this study. Bone marrow samples of 3~4 mL were taken from each patient and divided into 2 parts. One part (direct detection group) was directly detected by flow cytometry and FISH, and the other part (CD45 sorting group) was subjected to the novel CD45-negative cell sorting, followed by flow cytometry and FISH. The proportion of plasma cells in bone marrow, detection rate of cytogenetic abnormalities and risk stratification were compared in every patient before and after sorting. Results The proportion of plasma cells in bone marrow samples was (14.5±13.8)% and (51.5±30.3)%, respectively, in the direct detection group and CD45 sorting group, with that of the latter group 7.69 times higher than that of the former group (P＜0.001). Direct FISH indicated 17 cases of cytogenetic abnormality, while FISH after CD45 sorting found 39 patients having cytogenetic abnormalities (P＜0.001). The positive rates of IgH rearrangement, 13q14 deletion, 1q21 amplification and 17p deletion were all increased significantly after sorting. According to mSMRAT3.0 standard, only 28.3% of the patients were identified at high risk before sorting, but the number was increased significantly after sorting (accounting for 53.3%, P=0.002). Conclusion The new CD45 negative cell sorting technology is simple and easy to operate, and can improve the detection rate of cytogenetic abnormalities and accurately carry out risk stratification for guiding treatment in MM patients. Thus, this technology is worthy of promotion in primary hospitals.

The ETV6::PDGFRB fusion gene is commonly reported in chronic myelomonocytic leukemia with eosinophilia, yet patients with ETV6::PDGFRB presenting myeloid and lymphoid neoplasms successively have not been reported. Here, we report the first case of a 35-year-old man with myeloid and lymphoid neoplasms harboring an ETV6::PDGFRB fusion gene who demonstrated poor response to imatinib. The patient was diagnosed with an ETV6::PDGFRB fusion gene myeloid neoplasm on initial diagnosis at our hospital. After 5 months of treatment with imatinib, he was diagnosed with T-cell lymphoblastic lymphoma. ETV6::PDGFRB turned negative after increasing the dose of imatinib, but enlarged superficial lymph nodes reappeared the following year. Notably, the patient exhibited a worse response to imatinib treatment. This study describes this rare case and speculates on a possible mechanism. [ABSTRACT FROM AUTHOR]

To prediction seed quality through the agronomic traits and fiber properties, 27 upland cotton varieties were as materials. 10 agronomic traits, 3 seed quality traits and 5 fiber quality traits of 27 varieties were evaluated in 2 years. The relationship between agronomic traits and fiber properties with the quality traits of seed were analyzed by variance analysis, correlation analysis, principal component analysis and cluster analysis. The results showed that oil content was negatively correlated with lint content and positively correlated with fruit branch number. There was a significant negative correlation between gossypol content and leaf area. 6 principal components were extracted by principal component analysis, with a total contribution rate of 78.755%. The 3rd and 5th principal components were cottonseed by-product factors, with a contribution rate of 20.470%. Further analysis showed that the number of fruit branches, lint percentage and leaf area were closely related to cottonseed by-products. The above results laid a foundation for the study of their internal relationship with cottonseed oil content, protein and gossypol, and provided theoretical reference for the selection of special cotton.
