Objective. Gout is a chronic disease that causes inflammatory arthritis, which is closely related to urate accumulation induced by a disorder of uric acid metabolism and the consequent deposition of monosodium urate crystals. Dendrobium loddigesii Rolfe is an herbal medicine that has been used in some traditional Chinese medicine formulae in the treatment of gout. This study aimed to explore and verify the antigout activity of Dendrobium loddigesii extract (DLE) on alleviating the hyperuricaemia of mice and the acute gouty arthritis of rats. Methods. An animal model of hyperuricaemia was established using potassium oxonate (PO). We analysed the expression of uric acid transporter mRNA in the kidney in the hyperuricaemic mice after treatment with DLE. Simultaneously, a monosodium urate crystal-induced acute gouty arthritis rat model was used to evaluate the effects of DLE, according to the level of ankle swelling, as well as the protein levels of inflammatory receptors and cytokines, as assayed by WB and ELISA. Results. DLE alleviated hyperuricaemia in mice and inhibited acute gouty arthritis in rats (P<0.05). Meanwhile, DLE regulated the levels of uric acid transporters mRNA transcripts, including mouse organic anion transporter 1 (mOAT1), organic anion transporter 3 (mOAT3), urate transporter 1 (mURAT1), and glucose transporter 9 (mGLUT9) in the kidney (P<0.05), suggesting that DLE promoted uric acid metabolism. Furthermore, DLE significantly suppressed the protein levels of TLRs, MyD88, and NF-κB in the ankle joint’s synovium (P<0.05), and the serum levels of IL-1β, IL-6, and TNF-α were also reduced, which demonstrated the anti-inflammatory effects of DLE. Conclusion. DLE alleviates hyperuricaemia by regulating the transcription level of uric acid transporters in the kidney. It also inhibits acute gouty arthritis by inhibiting the pathway of TLRs/MyD88/NF-κB in the ankle joint’s synovium. The findings of the present study imply that DLE alleviates gout by promoting uric acid metabolism and inhibiting inflammation related to the TLRs/MyD88/NF-κB pathway.

The stems of Dendrobium loddigesii, a Chinese herb, are often used to treat diabetes and its polar extract is rich in shihunine, a water-soluble Orchidaceae alkaloid, but little is known about the anti-diabetes effects and mechanism of shihunine. This study investigated the anti-diabetic effect of a shihunine-rich extract of D. loddigesii (DLS) based on 3T3-L1 cells and db/db mice. The underlying mechanisms were primarily explored using Western blot analysis and immunohistochemical staining. The 3T3-L1 cell experiments showed that DLS can reduce the intracellular accumulation of oil droplets as well as triglycerides (p &lt, 0.001) and promote the 2-[N-(7-nitrobenz-2-oxa-1,3-diazol-4-yl)amino]-2deoxyglucose (2-NBDG) uptake of 3T3-L1 cells (p &lt, 0.001). The animal experiments confirmed that after 8 weeks of DLS treatment, the body weight, fasting blood sugar, and serum lipid levels of mice were significantly lowered, and the oral glucose tolerance test and serum insulin level were significantly improved compared to the no-treatment diabetes mellitus group. Further histomorphology observation led to the conclusion that the quantities of islet cells were significantly increased and the increase in adipose cell size was significantly suppressed. The immunohistochemical test of pancreatic tissue revealed that DLS inhibited the expression of cleaved cysteine aspartic acid-specific protease 3 (cleaved caspase-3). Western blot experiments showed that DLS had agonistic effects on adenosine monophosphate (AMP)-activated protein kinase phosphorylation (p-AMPK) and increased the expression levels of peroxisome proliferator-activated receptor &alpha, (PPAR&alpha, ) and glucose transporter 4 (GLUT4) in liver or adipose tissues. These data suggest that the shihunine-rich extract of D. loddigesii is an anti-diabetic fraction of D. loddigesii. Under our experimental condition, DLS at a dose of 50 mg/kg has good anti-diabetic efficacy.

In the present study, we investigated the whitening effects of DL under various conditions withB16F10 melanoma cells. The DL extract inhibited melanin contents and tyrosinase activity in a dose-dependent manner,compared with untreated group. Treatment of the DL extract effectively suppressed the α-MSH-stimulated melanin forma-tion, tyrosinase activity and dendrite outgrowth. Moreover, the α-MSH-induced mRNA expressions of tyrosinase-relatedprotein-1 (TRP-1), tyrosinase-related protein-2 (TRP-2), microphthalmia-associated transcription factor (MITF) and pro-tein expression of tyrosinase were significantly attenuated by DL treatment. Dendrobium is one of the three largest genera in the Orchidaceae and is distributed throughout various habitats. We investigated photosynthesis in seven Dendrobium species and cultivars by comparing their leaf δ13C values, titratable acidity, and CO2 exchange in well-watered and drought-stressed conditions. In addition, the leaf thickness and mesophyll succulence index (Sm) were measured in well-watered conditions. Our results indicate that Dendrobium loddigesii is a typical obligate (or constitutive) CAM plant because the leaf δ13C values were −14.47 and −14.66‰ in both conditions, respectively. Others showed the leaf thickness of 0.31–0.89 mm and their δ13C values ranged from −25.68 to −28.37‰. These are not the CAM plants but they could not be classified as obligate C3 or C3/CAM intermediate plants. Dendrobium crepidatum and Dendrobium fimbriatum were further identified as the obligate C3 plants because the net CO2 uptake was positive during daytime and negative during nighttime in both conditions. In contrast, Dendrobium chrysotoxum, Dendrobium nobile, and D. nobile ‘V1’ and ‘V4’, showed no positive net CO2 uptake and low ΔH+ values during nighttime under well-watered conditions, indicating C3 photosynthesis. However, they showed the positive net CO2 uptake and large ΔH+ values during nighttime after drought-stress (21 or 28 days without H2O), indicating CAM photosynthesis. Therefore, these four species and cultivars were identified as C3/CAM intermediate (inducible or facultative) plants. In brief, obligate CAM, C3/CAM intermediate, and obligate C3 plant types all exist in the section of Dendrobium. To the best of our knowledge, this is the first report of the obligate C3 plants in Dendrobium, and these diverse photosynthetic pathways may explain their varied environmental adaptations.

Dendrobium loddigesii Rolfe is an endangered perennial herb with ornamental and medicinal value. Due to habitat deterioration and human over-exploitation, it has suffered a significant decline in abundance. Determining the level of genetic diversity and pattern of population genetic structure of this species would be helpful for its conservation and management. In this paper, sequence-related amplified polymorphism (SRAP) markers were applied to assess the level and pattern of genetic diversity in seven populations of D. loddigesii. Seventeen SRAP primer combinations generated a total of 231 clear amplification bands encompassing 187 (80.95%) polymorphic bands. A high level of genetic diversity was detected (PPB = 80.52%, H = 0.2743, I = 0.4113) at the species level. There was a moderate genetic differentiation (Gst = 0.304) among populations. Two main clusters were detected by cluster analysis using the unweighted pair-group method with arithmetic average (UPGMA). Mantel test revealed that no significant positive correlation was found between genetic distances and geographic distances (r = 0.2302; P > 0.05). Recommendations for conservation of the endangered species resources are proposed.

Nine pairs of polymorphic chloroplast microsatellite primers were developed for Dendrobium officinale Kimura et Migo, an endangered herb. Levels of polymorphism were tested across a total of 55 individuals from four natural populations (12–15 individuals per population). Allele numbers varied from two to four per locus, while the number of haplotypes ranged from four to six per population. Transferability of the nine polymorphic chloroplast microsatellite primers was checked on an additional set of 51 Dendrobium individuals (belonging to 17 different species). Three markers could be transferable to all the species tested, while the remaining six markers successfully cross-amplified in most species tested. Moreover, polymorphism of the nine chloroplast microsatellite primers was tested across Dendrobium moniliforme (L.) Sw. and Dendrobium loddigesii Rolfe. All of them were polymorphic in D. moniliforme, while seven of which were polymorphic in D. loddigesii. These polymorphic chloroplast microsatellite primers developed for D. officinale will be a useful tool for the study of genetic diversity, population genetic structure, evolution of D. officinale and establishment of effective conservation strategies.

Endophytic fungi are ubiquitously distributed in orchids and have a great impact on the host plant. The diversity of endophytic fungi in the medicinal orchid Dendrobium loddigesii Rolfe was investigated and their bioactivities in microbe and plant growth were explored here. Endophytic fungi were identified by using morphological and molecular biological methods. Antimicrobial activity was determined by a standard disk assay. Activity in promoting plant growth was confirmed by root inoculation of endophytic fungi in seedling tray and pot experiments. Overall, 48 isolates were isolated from D. loddigesii and identified to belong to 18 genera, with Fusarium and Acremonium being the most dominant populations. A total of 17 isolates belonging to 9 genera were screened for their antimicrobial activity, and Fusarium spp., 8 of the 17 isolates, was also the dominant population. In the seedling tray experiment, two isolates, one of Fusarium named DL26 and the other of Pyrenochaeta named DL351, were shown to enhance plant growth in alder bark–humus medium, and the latter displayed weak activity against Bacillus subtilis (As 1.308) and Aspergillus fumigatus (As 3.2910). In the pot experiment, after inoculation of DL26 and DL351, five out of seven media were fit for plant-endophyte symbionts. Medium #1 of red brick fragments and sphagna was optimal in accelerating plant growth. In conclusion, a great diversity of endophytic fungi in D. loddigesii was first confirmed in a considerable proportion of antimicrobial isolates. Furthermore, two endophytes exhibited the ability to enhance plant growth although their activities were influenced by the growth media.

As a widely used medicinal plant, Dendrobium loddigesii Rolfe is always a possible target for fraudulent labeling. The identification of D. loddigesii is generally difficult from its morphological and chemical appearance only. In order to develop a convenient and efficient identification method for D. loddigesii, six pairs of diagnostic ARMS primers were designed based on nrDNA ITS sequences of D. loddigesii and eleven adulterants. The results showed that one diagnostic primer pair (FJB-04-forward, FJB-04-reverse) could be used to authenticate D. loddigesii by generating a fragment of 353 bp at annealing temperatures from 48 degrees C to 62 degrees C while the other diagnostic primer pair (FJB-03-forward, FJB-03-reverse) took on the same effect at annealing temperatures from 49 degrees C to 55 degrees C. This points out the potential of ARMS analysis for authentication of D. loddigesii.

As an invaluable herb, Dendrobium loddigesii is widely used in Chinese medicinal field. In order to develop a convenient and efficient identification method and investigate the genetic diversity and structure of this species, twelve microsatellite loci were isolated from two microsatellite-enriched libraries. Twenty-six individuals from Baise population were analysed. The observed and expected heterozygosities ranged from 0.174 to 0.617 and from 0.182 to 0.608, respectively. These microsatellite loci characterized from D. loddigesii will contribute to research on the individual authentication, population structure, genetic diversity and conservation of this species and its similar species.

GC-MS method combined with FTIR techniques by the analysis of polysaccharide was applied to evaluate the similarity between wild (W) and tissue-cultured (TC)Dendrobium huoshanense(DHS),Dendrobium officinale(DO), andDendrobium moniliforme(DM) as well as 3 wildDendrobiumspp.:Dendrobium henanense(DHN),Dendrobium loddigesii(DL), andDendrobium crepidatum(DC). Eight monosaccharides involving xylose, arabinose, rhamnose, glucose, mannose, fructose, galactose, and galacturonic acid were identified in the polysaccharide from eachDendrobiumsample while the contents of the monosugars varied remarkably across origins and species. Further similarity evaluation based on GC-MS data showed that thercorvalues of different origins of DHS, DO, and DM were 0.831, 0.865, and 0.884, respectively, while thercorvalues ranged from 0.475 to 0.837 across species. FTIR files of the polysaccharides revealed that the similarity coefficients between W and TC-DHS, DO, and DM were 88.7%, 86.8%, and 88.5%, respectively, in contrast to the similarity coefficients varying from 57.4% to 82.6% across species. These results suggested that the structures of polysaccharides between different origins of the investigatedDendrobiumsmight be higher than what we had supposed.

Objective To find out the relativity among starch quantity, polysaccharides content and total alkaloid content of Dendrobium loddigesii. Method Microscopy-counting process was applied to starch quantity statistics, sulfuric acid-anthrone colorimetry was used to assay polysaccharides content and bromocresol green colorimetry was used to assay alkaloid content. Pearson product moment correlation analysis, Kendall's rank correlation analysis and Spearman's concordance coefficient analysis were applied to study their relativity. Result and conclusion Extremely significant positive correlation was found between starch quantity and polysaccharides content, and significant negative correlation between alkaloid content and starch quantity was discovered, as well was between alkaloid content and polysaccharides content.

The MeOH extract of the stem of Dendrobium loddigesii was found to inhibit the aggregation of rabbit platelets induced by arachidonic acid and collagen. Two active compounds, moscatilin [1] and moscatin [2], were isolated. Moscatilin diacetate [3] also exhibited antiplatelet aggregation activity.

isolation and structure determination of the new compounds, loddigesiinols A—D and evaluation of their effects against NO production and DPPH radical scavenging

New phenanthrenes, loddigesiinols A (1) and B (7), and stilbenes, loddigesiinols C (8) and D (9), were isolated from 80% ethanol extract of Dendrobium loddigesii ROLFE (Orcharedceae) along with known compounds, including five phenanthrenes, three stilbenes, two lignans, and three sterols. Their structures were determined by spectroscopic studies. These compounds were evaluated for their inhibitory activities against nitric oxide (NO) production and 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging. Phenanthrenes (1-3, 7) and dihydrophenanthrenes (4-6) showed significant inhibitory activity against NO production. In particular, 2 showed stronger inhibitory activity against NO production than 5 and 6. This suggested that this phenanthrene is a stronger inhibitor of NO production than dihydrophenanthrene. The new phenanthrenes, loddigesiinols A (1) and B (7), significantly inhibited NO production with IC(50) values of 2.6 and 10.9 muM, respectively. One new dihydrostilbene, loddigesiinol D (9), showed weak inhibitory activity against NO production with an IC(50) value of 69.7 microM, but the other new dihydrostilbene, loddigesiinol C (8), did not show this activity at 100 microM.

Loci were developed for the following species: Acanthopagrus schlegeli, Anopheles lesteri, Aspergillus clavatus, Aspergillus flavus, Aspergillus fumigatus, Aspergillus oryzae, Aspergillus terreus, Branchiostoma japonicum, Branchiostoma belcheri, Colias behrii, Coryphopterus personatus, Cynogolssus semilaevis, Cynoglossus semilaevis, Dendrobium officinale, Dendrobium officinale, Dysoxylum malabaricum, Metrioptera roeselii, Myrmeciza exsul, Ochotona thibetana, Neosartorya fischeri, Nothofagus pumilio, Onychodactylus fischeri, Phoenicopterus roseus, Salvia officinalis L., Scylla paramamosain, Silene latifo, Sula sula, and Vulpes vulpes. These loci were cross-tested on the following species: Aspergillus giganteus, Colias pelidne, Colias interior, Colias meadii, Colias eurytheme, Coryphopterus lipernes, Coryphopterus glaucofrenum, Coryphopterus eidolon, Gnatholepis thompsoni, Elacatinus evelynae, Dendrobium loddigesii Dendrobium devonianum, Dysoxylum binectariferum, Nothofagus antarctica, Nothofagus dombeyii, Nothofagus nervosa, Nothofagus obliqua, Sula nebouxii, and Sula variegata. This article also documents the addition of 39 sequencing primer pairs and 15 allele specific primers or probes for Paralithodes camtschaticus.

Shihu, Herba Dendrobii, is the dry stems of Dendrobium loddigesii Rolfe., D. fimbria-turn Hook. var. oculatum Hook., D. chrysanthum Wall., D. candidum Wall. ex Lindl. or D. nobile Lindl. (Orchidaceae) and is officially listed in the Chinese Pharmacopoeia. It can be collected throughout the year. Dendrobium stems are used in traditional Chinese medicine as a tonic to improve the digestive function after recovery from ailments.
