Böbrek yetmezliği ülkemizde ve tüm dünyada önemli bir sağlık sorunudur. Bu nedenle iskemik akut böbrek yetmezliği için etkin tedavi yöntemleri geliştirilmelidir. İskemi, hücrede enerji düzeyinin düşmesine ve toksik metabolitlerin dokuda birikmesine yol açarak hücre disfonksiyonu ve sonrasında hücre ölümüne kadar gidebilen bir dizi biyokimyasal reaksiyonu başlatır. Özellikle reperfüzyon sonrası ortama gelen nötrofiller ve açığa çıkan medyatörlerin zararlı etkileri de eklenince organ veya doku ölümü kaçınılmaz olmaktadır. Bu tez çalışmasında Wistar albino türü erkek sıçanda oluşturulan böbrek iskemi-reperfüzyon hasarında poliferatif, nekrotik ve apoptotik hücre oranları belirlendi ve telomerik devamlılığı araştırıldı. Tez kapsamında ilk olarak sıçan sol böbrek dokularında 60 dakika boyunca iskemi oluşturuldu ve sağ böbrekler kontrol grubu olarak kullanıldı. Bu çalışmaya paralel olarak iskemi öncesi antioksidan ajan enjekte edilen sıçanlarda aynı işlemler tekrar edilerek kullanılan NAC antioksidan ajanın apoptoz üzerine etkisi belirlendi. Telomerik restriksiyonel fragmantasyon (TRF) yöntemi ile telomerik tekrar devamlılığı apoptoz ilişkisi incelendi. Kidney failure is an important health issue in both our country and the world. Therefore effective treatment strategies need to be emerged to cure acute kidney failure. Ischemia initiates a series of biochemical reactions that may end up with cellular dysfunction and ultimate cell death by causing to diminish cellular energy level and accumulation of toxic subtances. Tissue and organ death is therefore an inevitable result due to added harms exerted by neutrophils, which arose following reperfusion and liberated mediators. This thesis covered the estimation of the percentage apoptotic cell death and telomeric maintenance in kidney ischemia/reprfusion model to be established in Wistar albino rats. First of all, ischemia was made in rat left kidney and the irght was was used as a control. And then the presence of the apoptotic cell was investaigated in reperfused kidney. On the other side, antioxidant agent was injected to rats before ischemia and the effect of N-acetylcystein was tested on the apoptosis as a antioxidant agent. Finally, the realtionship between apoptosis and telomeric repeat persistance was examined using telomeric restrictional fragmentation (TRF) method. 62
