8. Development of an efficient process to extract C‐phycocyanin from marine cyanobacterial strains and evaluation of its bioactive potential.

Abstract

BACKGROUND: Phycocyanin (PC), a natural blue photosynthetic pigment found in cyanobacteria, rhodophyta and cryptophyta has numerous applications in biotechnology and the food industry. The phycocyanin extraction method has a significant impact on its bioactivities, purity index, cost and potential applications; thus, the development of an efficient extraction strategy continues to be a primary research goal. RESULTS: This study evaluated the efficacy of different methods for the extraction of cyanobacterial phycocyanin (C‐PC) from three marine cyanobacterial isolates, namely Arthrospira platensis, Phormidium sp. and Pseudoscillatoria sp. wet biomass. The pretreatment step of soaking biomass in extraction buffer for 60 min, followed by a combination of freeze–thaw (one cycle; 2 h) and sonication (2 min) treatment was found to be the most effective extraction strategy, producing the highest C‐PC yield (16.19 ± 0.34%, 15.89 ± 0.61% and 15.56 ± 0.69% dry cell weight for strains A. platensis, Phormidium sp. and Pseudoscillatoria sp., respectively). Furthermore, a biomass‐to‐solvent ratio of 0.02 g mL−1 was optimal to attain food‐grade purity ((A620/A280) ≥ 0.7). Bioactivity evaluation of crude and purified C‐PC exhibited its potential as antioxidant compounds and as an antimicrobial agent with significant inhibitory effects against various pathogenic microorganisms. CONCLUSION: The study proposed an efficient and cost‐effective C‐PC extraction process for cyanobacteria and highlighted the potential of C‐PC as a valuable bioactive compound with various applications. © 2024 Society of Chemical Industry (SCI). Abstract

Carotenoids are hydrophobic pigments produced exclusively by plants, fungi, and specific microbes. Microalgae are well suited for the production of valuable carotenoids due to their rapid growth, efficient isoprenoid production pathway, and ability to store these compounds within their cells. The possible markets for bio-products range from feed additives in aquaculture and agriculture to pharmaceutical uses. The production of carotenoids in microalgae is affected by several environmental conditions, which can be utilized to enhance productivity. The current study focused on optimizing the extraction parameters (time, temperature, and extraction number) to maximize the yield of carotenoids. Additionally, the impact of various nitrogen sources (ammonia, nitrate, nitrite, and urea) on the production of lutein and loroxanthin in Scenedesmus obliquus was examined. To isolate the carotenoids, 0.20 g of biomass was added to 0.20 g of CaCO3 and 10.0 mL of ethanol solution containing 0.01% (w/v) pyrogallol. Subsequently, the extraction was performed using an ultrasonic bath for a duration of 10 min at a temperature of 30 °C. This was followed by a four-hour saponification process using a 10% methanolic KOH solution. The concentration of lutein and loroxanthin was measured using HPLC–DAD at 446 nm, with a flow rate of 1.0 mL/min using a Waters YMC C30 Carotenoid column (4.6 × 250 mm, 5 μm). The confirmation of carotenoids after their isolation using preparative chromatography was achieved using liquid chromatography–tandem mass spectrometry (LC–MS/MS) with an atmospheric pressure chemical ionization (APCI) probe and UV–vis spectroscopy. In summary, S. obliquus shows significant promise for the large-scale extraction of lutein and loroxanthin. Abstract

Commercial carotenoids of green microalgae have become significant especially for their applications in the cosmetic, pharmaceutical, food and feed industries. Effects of physical and chemical parameters on carotenoid contents in isolated microalgal species have been investigated. The variables of shaking rate, nitrogen concentration and light intensity affect biomass production and the synthesis of carotenoids in the green microalgae were investigated using the statistical design by Box-Behnken (BBD) employing Response Surface Methodology (RSM). Furthermore, the optimized cultivation conditions using BBD for Chlorella vulgaris, Ankistrodesmus convolutus, Dunaliella saline, Tetraselmis striata were determined using the spectrophotometric method to enhance carotenoid concentration. A. convolutus within the green algae was detected with the highest carotenoid concentration. The optimum conditions results indicated that the growth of A. convolutus (0.55 mg/L) and production of total carotenoids (25.1138 mg/g biomass) were found at the stirrer rate of 100 rpm under the light intensity of 100 mu E/m(2)s, and in the nutrient component of 8.82 mM NaNO3. These conditions were validated experimentally for total carotenoid yield (24.13 mg/g biomass). After that the production was performed in a flat-plate photobioreactor with a volume of 6L based on the optimized conditions and the carotenoid profile was defined by HPLC-DAD using standards such as violaxanthin, astaxanthin and beta-carotene. Abstract

The aim of the study was to investigate the influence of physical conditions such as aeration rate (1, 3, 5 L/min) as well as chemical conditions including sodium nitrite (NaNO2), urea (CH4N2O) and ammonium chloride (NH4Cl) on the biomass productivity and fucoxanthin concentration of A. capitellata. The optimum cultures were cultivated in f/2 medium using sodium nitrate (NaNO3) in 2 L bubbling bottle photobioreactors under the light intensity of 100 µE/ m2s with aeration rate of 2 L/min. All the bottles were then incubated at 22.0±2°C, under the light intensities of 300 µE/m2s with three different airflow rates of 1, 3, 5 L/min for 16 days. And then, culture medium was prepared with three different nitrogen sources to achieve higher biomass productivity. During the production of A. capitellata, the maximum specific growth rate of 0.166 day-1, which conformed to the doubling time of 4.166 day, was achieved at the light intensity of 300 µE/m2s with an aeration rate of 1 L/min when sodium nitrate was used. Chlorophyll-a and fucoxanthin contents were also at the highest level in the same light intensity. Dry biomass amount reached the maximum level of 0.66±0.17 g/L in case of NaNO2. In this study, it was defined that the airflow rate of 1 L/min, the light intensity of 300 µE/m2s and sodium nitrate (NaNO3) were the optimum values not only for the growth of A. capitellata cells but also for the production of biomass and a higher fucoxanthin concentration.

Abstract

Astaxanthin is one of the most attractive carotenoid in the cosmetic, food, pharmaceutical, and aquaculture industries due to its strong bioactive properties. Among the various sources, several algae species are considered as rich sources of astaxanthin. Downstream processing of algae involves the majority of the total processing costs. Thus, elimination of high energy involved steps is imperative to achieve cost-effective scale in industry. This study aimed to determine operation conditions for astaxanthin extraction from wet Haematococcus pluvialis using microwave-assisted extraction. The isolated astaxanthin extract was evaluated for cytotoxicity on human lung cancer cells. The microwave-assisted extraction process at 75 °C under the power of 700 Watt for 7 min gave the highest astaxanthin yield (12.24 ± 0.54 mg astaxanthin/g wet cell weight). Based on MTT cell viability and Hoechst 33342 nuclear staining assays on A549 lung cancer cells, astaxanthin inhibited cell growth in dose- and time-dependent manners, where IC50 value was determined as 111.8 ± 14.8 µg/mL and apoptotic bodies were observed along with positive control group at 72 hr. These results showed that the treatment with astaxanthin extracted from wet H. pluvialis by microwave-assisted extraction exhibited anti-cancer activity on lung cancer cells indicating a newly potential to be utilized in industry. Abstract

The present study was conducted to evaluate the antioxidant and antimicrobial activity of methanol, dichloromethane and hexane extracts, as well as the essential oils of brown algae (Phaeophyta) Colpomenia sinuosa, Dictyota dichotoma, Dictyota dichotoma var. implexa, Petalonia fascia and Scytosiphon lomentaria. The essential oil of the macroalgae was obtained by steam distillation and analyzed by GC and GC/MS. The antioxidant activity of the algal extracts was determined using the procedures of inhibition of β-carotene bleaching and ABTS+ methods. The antioxidant effects of the extracts were compared with those of commercial antioxidants, such as butylated hydroxytoluene (BHT), butylated hydroxyanisol (BHA) and α-tocopherol. The hexane extracts of D. dichofoma var. implexa had a higher phenolic content than the other extracts. The dichloromethane extract of S. lomentaria was found to be more active in the decolorization of ABTS+ than the other extracts and generally the dichloromethane extracts were more active than the methanol and hexane extracts. Antimicrobial activities of the extracts were assessed against Gram (+) and Gram (-) bacteria and one yeast strain by the disk diffusion method. According to the results, the dichloromethane extracts generally showed more potent antimicrobial activity than the methanol and hexane extracts at concentrations 1.5 and 1.0 mg/disk.

İzmir’in Gümüldür bölgesinden alınan su örneğinde, seyreltme ve dökme plaka yöntemleri kullanılarak mikroalg izolasyonu yapılmıştır. Işık mikroskobuyla morfolojik olarak değerlendirilen türün Chlorella sp. olduğu saptanmıştır. Moleküler yöntemlerle mikroalg DNA’sı izole edilerek 16S ve 18S rRNA gen bölgeleri PCR’da çoğaltılmıştır. Bu dizinin sekanslanması ve filogenetik olarak değerlendirilmesi sonucu Chlorella sorokiniana olduğu belirlenmiştir. Aksenik C. sorokiniana elde etmek için santrifüj ile yıkama, antibiyotik ile muamele, agar ortamında büyütme ve tek hücre izolasyonu gibi farklı yöntemler kullanılarak aksenikleştirme işleminden başarılı sonuçlar elde edilmiştir. Fototrofik C. sorokiniana’ dan elde edilen biyokütle (0,19 g L-1) ve spesifik büyüme hızı (0,78 gün-1), miksotrofik C. sorokiniana’ dan elde edilen biyokütle (0,31 g L-1) ve spesifik büyüme hızı(1,3 gün-1), heterotrofik C. sorokiniana’ dan elde edilen biyokütle (0,6 g L-1) ve spesifik büyüme hızı (2,52 gün-1) olarak belirlenmiştir. 37. Evaluation of different scale-up strategies for Haematococcus pluvialis cultivation in airlift photobioreactors.

Abstract

Large scale algal biomass production can be very challenging due to the potential issues of sustainability, environmental ethics, and economic concerns. A strategic approach to the transition from the laboratory to the industrial scale allows the prediction of process characteristics, design and analysis of large scale systems, and reduction of extra costs. In this study, a scale-up procedure that considered different approaches was carried out by selecting the Haematococcus pluvialis as a model organism. Three scale-up parameters (constant mixing time (tm), volumetric power consumption rate (P/V), and oxygen mass transfer coefficient (kL a)) were tested for biomass production in a 2-L airlift photobioreactor and they were compared with those obtained from a 1-L aerated cultivation bottle. Among three strategies, the maximum cell concentration, 4.60±0.20×105 cells/mL, was obtained in a constant volumetric power consumption rate experiment. Also, total carotenoid amount showed similar changes with the cell concentration and reached the maximum concentration of 2.02±0.11 mg/L under constant P/V experiment. However, the cultivation bottle presented the highest biomass amount of 0.62 g/L and specific growth rate of 0.38 day-1 of all of the photobioreactors. This result might be attributed to the low aeration rates or improper configuration of the system, which created a non-homogenous culture medium and led to ineffective mass transfer. The species was identified as Chlorella sp. through morphologic evaluation under light microscope. Microalgae DNA was isolated through molecular methods and 16S and 18S rRNA gene regions were amplified by means of PCR. As a result of sequencing and phylogenetic evaluation of the gene regions, it was determined that the microalgae was Chlorella sorokiniana. Different methods such as centrifuge washing, antibiotic treatment, growth on agar and single cell isolation used to obtain axenic C. sorokiniana yielded successful results. Biomass and specific growth rate of phototrophic C. sorokiniana (0.19 g L-1 and 0.78 days-1, respectively), mixotrophic C. sorokiniana (0.31 g L-1 and 1.3 days-1, respectively) and heterotrophic C. sorokiniana (0.6 g L-1 and 2.52 days-1, respectively) were determined. According to the findings obtained, the biomass productivity of axenic C. sorokiniana under different culture conditions can be ordered as: heterotrophy > mixotrophy > phototrophy.

39. A novel subcritical fucoxanthin extraction with a biorefinery approach

Abstract

Eco-friendly, cost efficient and effective extraction methods have become significant for the industries applying zero waste principles. The two main objectives of this study were; to examine fucoxanthin extraction from wet Phaeodactylum tricornutum using subcritical fluid extraction and to characterize the residual biomass in order to determine the potential application areas. The highest fucoxanthin yield of 0.69 ± 0.05 mg/g wet cell weight was achieved using methanol with solvent-to-solid ratio of 200:1 at 120 rpm, 20 MPa pressure and at 35 °C for 60 min by subcritical extraction. Microscopy images showed that most of the cells were disrupted and intracellular components were effectively released. Based on the results of energy dispersive spectroscopy, biomass contained a mixture of organic molecules including mainly carbon (57–72%), oxygen (26–41%), magnesium (0.6–1.4%) and silica (0.4–1%) (wt%). These results make the residual biomass a potential candidate for various areas such as bioenergy, material sciences and sensor technologies.

Abstract

Microalgae with their improved growth rates and accumulation of high-value-added products make their commercial production attractive. Among them, lutein, which is a carotenoid, plays a very important role due to its various applications in the food and pharmaceutical industry. Induction of its biosynthesis can be triggered by various stress conditions like light. In this study, three different light intensities (50,150 and 300 µmol photons/m²s) and aeration rates (1, 3, and 5 L/min) were utilized to induce the lutein biosynthesis and biomass productivity in Scenedesmus obliquus. Lutein was isolated by preparative chromatography using a semiprep C30 column (10 × 250 mm, 5µm) and its confirmation was made by LC-MS/MS. According to the results, Scenedesmus obliquus synthesized the maximum lutein (8.01 ± 0.1 mg/g) with biomass productivity of 1.698 g/L at 150 µmol photons/m²s light intensity using 3 L/min as aeration rate. To the best of the authors' knowledge, this was the first study that the lutein was isolated by preparative chromatography using semiprep C30 carotenoid column with a simple and rapid separation, which can be used as a reference methodology for the isolation of other carotenoids. Objective: In our study, we aimed to compare the hematological efficacy of intravenous and oral forms of iron treatment in clinically stable patients with moderate postpartum anemia.Material and Method: One hundred and eighty postpartum cases, who had pre-discharge hemoglobin evaluation levels between 8-9.5 g/dl, were allocated into two groups. The 1st group was treated with oral fe +2 (n = 127, 70.6%), and the other group was treated with intra venous ferric carboxymaltose (n = 53, % 29.4). Group 1 received oral iron treatment for aproximately 8 weeks. Also in Group 2, intra venous ferric carboxymaltose iron replacement was performed twice with 1 week intervals. Late postpartum (8 weeks after delivery) hemoglobin levels were collected and recorded.Findings: Difference between the group 1 and group 2 in terms of late postpartum maternal haemoglobin levels was found statistically significant (respectively, 10,52±1,26 g/dl vs 11,82±1,06 g/dl, P value 0,021). After intravenous treatment, increase rate of hemoglobin in group 2 was found statistically significant when comparing with of in group 2. (1,51±0,72 g/dl vs 2,90±0,46 g/dl, P value ˂0,01 ).Conclusion: Although clinical evaluations and stability of anemic postpartum cases are part of our overall treatment and patient follow-up approaches, moderate and deeper anemic cases should be treated with 3rd generation intravenous iron forms for rapid hematologic improvement.Amaç: Çalışmamızda, orta düzey postpartum anemisi olan ve klinik olarak stabil olan hastalarda intravenöz ve oral demir tedavi seçeneklerinin hematolojik etkinliğini karşılaştırmayı amaçladık.Materyal ve Metod: Doğum sonrası taburcu olmadan hemen önce hemoglobin değerlerini 8-9.5 gr/dl olan 180 postpartum olgu iki gruba ayrıldı. 1. grup oral fe +2 (n = 127, %70.6), diğer grup ise intravenöz ferrik karboksmaltoz (n = 53, %29.4) ile tedavi edildi. Grup 1 oral demir tedavisi yaklaşık 8 hafta aldı. Grup 2'ye 1 hafta ara ile iki kez intravenöz ferrik karboksmaltoz replasmanı yapıldı. Geç postpartum (doğumdan 8 hafta sonra) hemoglobin düzeyleri toplandı ve kaydedildi.Bulgular: Postpartum maternal hemoglobin seviyeleri açısından grup 1 ve grup 2 arasındaki fark istatistiksel olarak anlamlı bulunmuştur (sırasıyla, 10,52±1,26 g/dl, 11,82±1,06 g/dl, P değeri 0,021). Grup 2’ de intravenöz demir tedavisi sonrası hb seviyelerinde artış oranı grup 1’le karşılaştırıldığında anlamlı olarak yüksek bulunmuştur. (sırasıyla, 1,51±0,72 g/dl, 2,90±0,46 g/dl, P değeri ˂0,01 ). Sonuç: Anemik postpartum vakaların klinik değerlendirmeleri genel tedavi ve hasta takip yaklaşımlarımızın bir parçası olsada, orta ve derin anemik olgular hızlı hematolojik iyileşme için 3. jenerasyon intravenöz demir formları ile tedavi edilmelidir.

48. Purification of phycocyanin from isolated and identified hot spring cyanobacteria

Abstract

EgeUn###, Phycocyanin is a phycobiliprotein used as a healthy food additive as well as an ingredient in cosmetic dye preparation. In this study, we attempted molecular and morphological identification of three thermophilic filamentous cyanobacteria and also explored a method to separate and purify thermostable C-phycocyanin of analytical grade from them for biotechnological applications. Cyanobacteria strains were collected from hot springs in Karakoc-Seferihisar, Gulbahce-Urla and Sifne-Cesme in Izmir, Turkey. The samples were identified both by morphological observations and genetic assay. Filamentous cyanobacteria DNA from the collected samples was isolated and extracted, and were analyzed using,a 16S rRNA cyanobacteria-specific PCR and a denaturing-gradient gel electrophoresis (DGGE). Extraction and purification of phycocyanin was performed in two stages, ammonium sulfate saturation and dialysis, gel filtration and ion exchange chromatography. Conventional separation of Geitlerinema sp., Halospirulina sp. and Phormidium animale was successfully performed yielding C-phycocyanin at 493.760 +/- 3.610, 89.060 +/- 3.209, 32.978 +/- 0.350, 4.046 +/- 0.193, 8.303 +/- 0.511 and 4.196 +/- 0.090 mg/g purity from gel filtration and ion exchange chromatography processes, respectively. The SDS-PAGE demonstrated that the P. animale, Halospirulina sp. and Geitlerinema sp. were purified phycocyanin. Abstract

Phycocyanin is a precious, natural, blue coloured pigment-protein complex that has commercial value and wide application in cosmetics, food, and pharmaceutical industries. In the present study, we performed various cell disruption methods (ultrasonication, homogenization, freeze/thaw and CaCl2 extraction) for phycocyanin extraction from different forms of biomass of a thermophilic Pseudanabaena sp. that has a high potential to produce high-quality phycocyanin. Using potassium phosphate buffer and ultrasonic bath method, we achieved the highest phycocyanin yield (345 mgPC.g-biomass) from freeze-dried biomass and we obtained increased yield as the duration of application increases. Phycocyanin yields were calculated as 345 mgPC.g-biomass, 255 mgPC.g-biomass and 220 mgPC.g-biomass for 5, 10 and 15 min, respectively. In this study, cell disruption methods have determined significantly more effective on freeze-dried biomass rather than frozen biomass. Phycocyanin content of freeze-dried biomass was analysed after six months of storage and dramatic decrement was observed in the phycocyanin content of the cells. [ABSTRACT FROM AUTHOR]

Abstract

The aim of this study is to determine different culture media and temperature values on the growth rate of Desmodesmus protuberans (F.E. Fritsch & M.F.Rich) E. Hegewald, and to investigate antimicrobial and antioxidant potency of the crude extracts. the microalga isolates from Lake Eğirdir in Isparta (Turkey) was identified as D. protuberans and included in Ege University Microalgae Culture Collection. D. protuberans was cultivated at two temperature regimes (22 and 28ºC) and growth media (BBM, BG-11 and RD medium) under the light intensity of 75 ?mol m-2s-1 for testing the impact of different environmental conditions on the growth rate. All of the productions of D. protuberans biomass were extracted with methanol and investigated the antimicrobial and antioxidant activity. Antimicrobial activities of the extracts of alga were tested by disc diffusion assay against five Gram-positive and four Gram-negative bacterial strains and against the yeast Candida albicans. the methanol extract of the biomass from BBM medium (28°C) showed the highest growth inhibition effect on C. albicans, while RD medium (22°C) extract showed the highest antioxidant activity. These results demonstrated that the differences in the bioactivity of biomasses are stemmed from different temperature and media conditions.
