Gerçekleştirilen tez çalışmasında Türkiye'de endüstriyel üretimi bulunan nar kabukları ve fermente edilmiş şalgam suyu atıklarının biyoetanol üretim potansiyelleri araştırılmıştır. Fermentasyon deneylerinde Saccharomyces cerevisiae, Kluyveromyces marxianusve Pichia stipitis kullanılmıştır. Her iki hammaddeden biyoetanol üretimine ön-işlem tipi, başlangıç hammadde konsantrasyonu, organik ve inorganik azot kaynakları ve mineral tuzları gibi önemli parametrelerin etkisi belirlenmiştir. Ek olarak nar kabukları enzimatik hidrolize tabi tutulmuş ve farklı tarımsal atıkların fermente edilmiş şalgam suyu atıklarından biyoetanol üretimine etkisi belirlenmiştir. Her iki hammadde ve denenen her üç mayada da en yüksek etanol üretimine 100 g/L başlangıç hammadde konsantrasyonunda ulaşılmıştır. Bununla beraber azot kaynakları, mineral tuzları ve enzimatik hidrolizin nar kabuklarından etanol üretimini belirgin şekilde arttırdığı saptanmıştır. Şalgam suyu atıklarında ise farklı besiyeri bileşenlerinin ve farklı tarımsal atıkların etanol üretimine olumlu etkileri gözlenmiştir. Nar kabuklarından elde edilen en yüksek biyoetanol konsantrasyonu 14.4 FPU selülaz varlığında S. cerevisiae, K. marxianus ve P. stipitis için sırasıyla 12.69 g/L, 14.35 g/L ve 4.23 g/L olarak bulunmuştur. Tez çalışmasında en yüksek teorik etanol verimi, QP ve YP/S değerleri enzimatik hidrolize uğratılan yıkanmış nar kabuğu hidrolizatlarından elde edilmiştir. Teorik etanol verimi sırasıyla sırasıyla %97.8, %98,7, %35.5, Qp değerleri sırasıyla 0.98, 0.99, 0.04 g/L.h ve YP/S değerleri sırasıyla 0.50, 0.50 ve 0.30 g/g olarak saptanmıştır. Fermente edilmiş şalgam suyu atıklarında ise en yüksek etanol üretiminin 24 g/L melas ilave edilmesi sonucunda olduğu belirlenmiştir. Bu ortamda S. cerevisiae, K. marxianus ve P. stipitis için sırasıyla 7.32, 7.25 ve 1.55 g/L etanol üretimi gözlenmiştir. Bu koşullarda S. cerevisiae, K. marxianus ve P. stipitis teorik etanol veriminin sırasıyla %65, %64 ve %18'ine ulaşmıştır. QP değerleri aynı mikroorganizmalar için sırasıyla 0.61, 0.60 ve 0.05 g/L.h şeklinde belirlenirken, YP/S değerleri 0.46, 0.45 ve 0.37 g/g olarak hesaplanmıştır. In this thesis, bioethanol potentials of turnip juice discards and pomegranate peel waste which are industrial by-products in Turkey were investigated. Saccharomyces cerevisiae, Kluyveromyces marxianusand Pichia stipitis were used in fermentation experiments. The effects of important parameters such as pre-treatment type, initial raw material concentration, organic and inorganic nitrogen sources and mineral salts on bioethanol production from both raw materials were determined. In addition, pomegranate peels were subjected to enzymatic hydrolysis and the effect of different agricultural wastes on bioethanol production from fermented turnip juice discards was determined. The highest ethanol production was achieved at the initial raw material concentration of 100 g/L in both raw materials and all three yeasts tested. The same microorganisms were used for ethanol production from turnip juice wastes, and the pretreatment type, initial raw material concentration and the effect of different media components were investigated. The effect of different agricultural wastes on bioethanol production from turnip juice wastes was also determined. In the study, the 100 g/L initial raw material concentration yielded the highest ethanol production for both raw material types. However, nitrogen sources, mineral salts and enzymatic hydrolysis significantly increased ethanol production in pomegranate peels. In fermented turnip juice discards, positive effects of different media components and different agricultural wastes on ethanol production were observed. The highest bioethanol concentration obtained from pomegranate peels was found to be 12.69 g/L, 14.35 g/L and 4.23 g/L for S. cerevisiae, K. marxianus and P. stipitis, respectively, in the presence of 14.4 FPU cellulase enzyme. In the thesis study, the highest kinetic parameters were obtained from washed pomegranate peel hydrolysates subjected to enzymatic hydrolysis. In the washed pomegranate peel hydrolyzate, the highest theoretical ethanol yields were 97.8%, 98.7%, 35.5%, Qp values were 0.98, 0.99, 0.04 g / Lh and YP/S values were 0.50, 0.50 and 0.30 g/g, respectively. The highest ethanol concentration was determined as a result of the addition of 24 g/L molasses to the turnip juice wastes. S. cerevisiae, K. marxianus and P. stipitis produced 7.32, 7.25 and 1.55 g/L ethanol, respectively in the presence of fermented turnip juice discard supplemented with 24 g/L molasses. Under these conditions, S. cerevisiae, K. marxianus and P. stipitis reached 65%, 64% and 18% of theoretical ethanol yield, respectively. In these conditions, QP values were determined as 0.61, 0.60 and 0.05 g/L.h for the same microorganisms, while YP/S values were calculated as 0.46, 0.45 and 0.37 g/g, respectively. 174

The lignocellulosic substances such as agricultural wastes are promising feedstocks for bioethanol production. Because they are cost effective, renewable, abundant and not having primary value for food and feed. The current study suggests that improvements in polysaccharide hydrolysis of under-utilized biomass of carrot pomace may find practical use in its conversión to bioethanol by Saccharomyces cerevisiae and Pichia stipitis fermentation. Some important parameters for bioethanol production such as pretreatment procedures (CaO and activated charcoal treatments), nitrogen sources ((NH)2SO4, soy wheat, cheese whey), and pomace loading amount (15-120 g/L) were optimized in the study. The highest bioethanol production could be achived when the saccharification and fermentation conditions were optimized in order to increase monosaccharide yield and fermentation of both six-carbon and five-carbon monosaccharides. The bioethanol production was 1.9 -fold higher for S. cerevisiae and 4.6 -fold higher for P. stipitis when (NH4)2SO4 was added in addition to the trace nitrogen substances, vitamins and minerals present in carrot pomace. The highest bioethanol production values were obtained as 6.91 and 2.66 g/L in the presence of 120 g/L pomace loading, 1 g/L (NH)2SO4 at the end of 72 hours incubation time at pH 6 by S. cerevisiae and P. stipitis, respectively.

Bu tezde yeni benzofuran, naftofuran veya siklobütan halkası içeren keton ve ketoksim türevi bileşiklerin antibakteriyel ve antifungal aktiviteleri Gram (+) ve Gram (-) bakteriler ve 4 Candida türü üzerinde araştırılmıştır. Çalışmada bileşiklerin Minimum İnhibisyon Konsantrasyonları (MİK) ve Minimum Bakterisidal Konsantrasyonları (MBC) belirlenmiş ve etkili bulunan bileşiklerin zamana bağlı öldürme eğrileri çıkartılmıştır. Bileşiklerden 4, 21, 41 ve 45 bazı bakteriler üzerinde, 18 ise hem Gram (+) bakteriler hem de Candida türleri üzerinde aktivite göstermiştir. MBC ve öldürme eğrileri 18 ve 21 nolu bileşiklerin bakteriyosidal olduklarını göstermiştir. Candida türleri üzerindeki MBC ve MİK değerleri 18 nolu bileşik için aynıdır.18 numaralı bileşiğin bakteriler için MİK değerleri Staphylococcus aureus ATCC 29213?te 32-64 µg/mL, Metisiline dirençli Staphylococcus aureus (MRSA)?da 16-32 µg/mL, Enterococcus faecalis ATCC 29212?de 32-64 µg/mL, Vankomisine dirençli Enterococcus faecalis (VRE)?de 16-32 µg/mL olarak bulunmuştur. Mayalarda ise Candida krusei ATCC 6258?de 32 µg/mL, Candida albicans ATCC 10231?de 64 µg/mL, Candida glabrata ATCC 90030?da 128 µg/mL, Candida parapsilosis ATCC 22019?de 128 µg/mL olarak saptanmıştır. 21 numaralı bileşiğin Enterococcus faecalis ATCC 29212?de MİK değeri 32-64 µg/mL, VRE?ye karşı ise 32-64 µg/mL olarak tesbit edilmiştir. In this thesis new keton and benzofuran, naphthofuran or cyclobutane ring containing ketoxime derivatives were investigated for Gram (+) and Gram (-) antibacterial and antifungal activities for 4 Candida specieses. Minimum İnhibition Concentration (MIC) and Minimum Bactericidal Concentration (MBC) of the compounds were determined and time kill studies were carried out for the active compounds. Compounds 4,, 21, 41 and 45 were active against some of the bacteria while compound 18 was active on some of the bacteria as well as the yeasts tested. The results from the MBC and time kill studies suggested that compounds number 18 and 21 are bacteriocidal. MIC and MBC results for the Candida specieses were the same indicating sidal activity. MIC values of compound 18 were as follow: Staphylococcus aureus ATCC 29213 32-64 µg/mL, Methicillin Resistant Staphylococcus aureus (MRSA) 16-32 µg/mL, Enterococcus faecalis ATCC 29212 32-64 µg/mL, Vancomycin Resistant Enterococcus faecalis (VRE) 16-32 µg/mL. Teh MIC values of compound 18 for yeasts were: Candida krusei ATCC 6258 32 µg/mL, Candida albicans ATCC 10231 64 µg/mL, Candida glabrata ATCC 90030 128 µg/mL and Candida parapsilosis ATCC 22019 128 µg/mL.
