Abstract

Solid cancers like pancreatic ductal adenocarcinoma (PDAC), a type of pancreatic cancer, frequently exploit nerves for rapid dissemination. This neural invasion (NI) is an independent prognostic factor in PDAC, but insufficiently modeled in genetically engineered mouse models (GEMM) of PDAC. Here, we systematically screened for human-like NI in Europe's largest repository of GEMM of PDAC, comprising 295 different genotypes. This phenotype screen uncovered 2 GEMMs of PDAC with human-like NI, which are both characterized by pancreas-specific overexpression of transforming growth factor [alpha] (TGF-[alpha]) and conditional depletion of p53. Mechanistically, cancer-cell-derived TGF-[alpha] upregulated CCL2 secretion from sensory neurons, which induced hyperphosphorylation of the cytoskeletal protein paxillin via CCR4 on cancer cells. This activated the cancer migration machinery and filopodia formation toward neurons. Disrupting CCR4 or paxillin activity limited NI and dampened tumor size and tumor innervation. In human PDAC, phospho-paxillin and TGF-[alpha]-expression constituted strong prognostic factors. Abstract

The autonomic nervous system plays a relevant role in development and progression of pancreatic cancer (PCa). In the presented dissertation, we investigated the role of indirect cholinergic activation in PCa progression through MTT-viability, invasion, immunohistochemistry, immunoblotting, flow-cytometric-cell-cycle assays, phospho-kinase arrays and xenografted mice. Inhibition of acetylcholinesterase diminished PCa cell growth and invasiveness in vitro and in vivo and led to immune modulation. We did not however see a significant survival benefit in vivo., Das autonome Nervensystem spielt eine wichtige Rolle in der Entwicklung und Progression des Pankreaskarzinoms (PCa). In der vorliegenden Dissertation untersuchten wir durch Viabilität- und Invasionsassays, Immunhistochemie, Western Blot, Zellzyklus, Phosphokinase Arrays und ein Xenograft Mausmodell die Rolle der cholinergen Aktivierung in der Progression des PCa. Die Inhibierung der Acetylcholinesterase hemmte das Tumor-Zellwachstum und die Invasion in vitro und in vivo und führte zu einer Immunmodulation. Wir sahen jedoch keinen signifikanten Effekt auf das Überleben in vivo.

Abstract

In dieser Arbeit konnte gezeigt werden, dass die stereochemische Inhibition der CX3CR1/Fraktalkine-Achse keine signifikanten Änderung der auf DRG-Neurone gerichteten Migration der Pankreaskarzinom(PDAC)-Zellliinen und keine zytotoxische Auswirkung auf die PDAC-Zelllinien aufweist. Eine Behandlung mit CX3CR1 Antikörpern zeigte hingegen einen stark antiproliferativen Effekt auf diese Zelllinien. Der ERK1/2-Signaltransduktionsweg scheint in diesen Prozessen keine direkte Rolle zu spielen., In the present work it was shown that the stereochemical inhibition of the CX3CR1/fractalkine axis does not have a significant influence on the migration of pancreatic cancer (PDAC) cell lines toward the DRG neurons. It exerted no cytotoxic effect on the PDAC cell lines. Treatment with a Cx3Cr1-antibody, on the other hand, showed a strong antiproliferative effect on these cell lines. The ERK1/2 signaling pathway does not seem to play a direct role in these processes.

Abstract

Die vorliegende Arbeit befasst sich mit der Rolle von humanen Perizyten für die neurale Invasion und Angiogenese im duktalen Adenokarzinom des Pankreas (PDAC). Es zeigte sich, dass insbesondere CD146+Perizyten im PDAC ausgeprägter vorkommen als im gesunden Pankreasgewebe. In vitro weisen pankreaskarzinomzell-aktivierte Perizyten eine veränderte Zellmorphologie auf, sowie ein verändertes Genexpressionsprofil. Die Ergebnisse der vorliegenden Arbeit deuten daraufhin, dass eine Aktivierung von Perizyten in der Tumormikroumgebung des PDAC stattfindet., Pancreatic ductal adenocancinoma (PDAC) exhibits an overproportional degree of neural invasion (NI). Here, we investigated the role of human pericytes in PDAC-associated NI and angiogenesis. We found that CD146+pericytes were increased around microcapillaries when compared to normal pancreas. Upon co-culture with PDAC cells, pericytes show altered cell morphology with increased cell surface area and increased gene expression of pro-inflammatory and metabolic pathways. This indicates that there might be an activation of pericytes in the tumor microenvironment of human PDAC.

34. Neuronal glutamate promotes pancreatic cancer cell migration through neuro-cancer synapses that fuel the GRIN2D-EZH2-E2F1-RB pathway

Abstract

Pancreatic ductal adenocarcinoma or pancreatic cancer (PDAC) exhibits a strong propensity to neural invasion (NI), but the molecular mechanisms of NI are unclear. Recent studies showed that central nervous system tumors like gliomas can make glutamatergic synapses with neurons, which they exploit for their growth. Purpose: We explored whether a similar glutamatergic neuronal input over the glutamate-NMDAR signaling into the cancer cells fosters their growth and promotes NI. Method: Analysis of the glutamate receptor profile of human and murine PDAC cell lines. Cultured PDAC cell lines were treated with L-Glutamate or DRG (dorsal root ganglion) conditioned medium (CM) or cocultured with DRG, and the effects of GluN2D (encoded by GRIN2D) and the transduction molecules of Glutamate-GluN2D-NMDAR signaling were quantified in the neuroinvasive and non-neuroinvasive pancreatic cancer cells. Results: Here we first profiled the glutamate receptor profile of human and murine PDAC cell lines on the CCLE database and correlated this pattern to the neuro-invasiveness of our cell lines. We show that the glutamate receptor subtype GluN2D (encoded by GRIN2D) was specifically involved in cell migration and invasion of the neuroinvasive pancreatic cancer cells mediated by L-Glutamate or DRG (dorsal root ganglion) conditioned medium (CM). Importantly, these phenotypic features could be completely reversed after GluN2D antagonist and GRIN2D siRNA treatment, however, not in non-neuroinvasive pancreatic cancer cells. The transduction molecules of Glutamate-GluN2D-NMDAR signaling were be activated and up-regulated by co-culture with DRG or through DRG CM. In co-cultures of neurons and PDAC cancer cells, GRIN2D, GRIN1, and the NMDAR-interaction partners PSD95 were upregulated in neuro-invasive PDAC cells exposed to neurons whereas synaptobrevin-1 and vGlut-2 were notably on axons that extended from neurons and synapsed on PDAC cells.
