Abstract

Cells respond to biophysical and biochemical signals. We developed a composite filament from collagen and silica particles modified to interact with collagen and/or present a laminin epitope (IKVAV) crucial for cell-matrix adhesion and signal transduction. This combines scaffolding and signaling and shows that local tuning of collagen organization enhances cell differentiation.

Abstract

We developed a microfluidic system for vibrational polariton studies, which consists of two microfluidic chips: one for solution mixing and another for tuning an infrared cavity made of a pair of gold mirrors and a PDMS (polydimethylsiloxane) spacer. We show that the cavity of the system can be accurately tuned with either piezoelectric actuators or microflow-induced pressure to result in resonant coupling between a cavity mode and a variational mode of the solution molecules. Acrylonitrile solutions were chosen to prove the concept of vabriational strong coupling (VSC) of a C≡N stretching mode with light inside the cavity. We also show that the Rabi splitting energy is linearly proportional to the square root of molecular concentration, thereby proving the relevance and reliability of the system for VSC studies. Abstract

Bioinorganic chemistry and inorganic chemical biology are multidisciplinary fields dealing with metal ions in biological systems. In this article we will describe a research journey in these fields. This journey will take us fromchemical design of inorganicmetal complexes with a biological activity (catalytic antioxidants) or a biological interest (metal-based probes) and their characterization out of cellular context, typical of bioinorganic chemistry, to their studies in cells, typical of chemical biology. First, we will focus on the description of a bio-inspired strategy to design metal-based catalytic antioxidants. Starting from the description of themain features of an antioxidant cell-protecting metalloenzyme, the superoxide dismutase (SOD), we will delineate the main parameters to be controlled to design low-molecular weight (LMW) metal complexes meant to mimic its activity: choice of the metal cation, tuning of the redox potential, and modulation of the charge. The effect of the hydration level and inertness with regard to metal ion release have also been explored. Methods to quantify the SOD-like activity, out of cellular context and in cells, and analysis of the integrity of the metal-based antioxidant in cells (through ion mobility coupled with mass spectrometry) will be described. Strategies to develop peptide-based metal complexes through conjugation of a LMW complex with peptides aimed at targeting specific organelle will be presented. Peptides can also be used to coordinate the metal ion: a combinatorial approach, with an activity-based screening, or and a rationale design using three-stranded coil-coiled, will also be presented. The cellular activity of a molecule is strongly dependent on its sub-cellular location. A second focus of this article will be X-ray fluorescence imaging and infra-red microscopy, as techniques to image metal cations and metal-carbonyls in cells using the metallic core (M and M(CO)x). After a brief overview, we will describe how metal-based probes can be developed, as trackers of organelles or tags, with application to mitochondria tracking and to the imaging of the Mn-based SOD mimics previously described. Overall, we hope this this story will provide an insight of what are bioinorganic chemistry and inorganic chemical biology, showing how artificial metal-based systems can be chemically designed, associated with their direct study in cells. Abstract

Catalase mimics are low molecular weight metal complexes that reproduce the activity of catalase, an antioxidant metalloprotein that participates in the cellular regulation of H2O2 concentration by catalyzing its dismutation. H2O2 is a reactive oxygen species that is vital for the normal functioning of cells. However, its overproduction contributes to oxidative stress, which damages cells. Owing to their biocompatibility, peptidyl complexes are an attractive option for clinical applications to regulate H2O2 by enzyme mimics. We report here the synthesis and characterization of four new peptidyl di-copper complexes bearing two coordinating sequences. Characterization of the complexes showed that, depending on the linker used between the two coordinating sequences, their catalytic activity for H2O2 dismutation, their thermodynamic stability and their resistance to H2O2 degradation are very different, with (CATm2)Cu2 being the most promising catalyst. Abstract

Sensitive detection of proteins by X-ray fluorescence microspectroscopy using the Re core of a single core multimodal probe for imaging., Bio-imaging techniques alternative to fluorescence microscopy are gaining increasing interest as complementary tools to visualize and analyze biological systems. Among them, X-ray fluorescence microspectroscopy provides information on the local content and distribution of heavy elements (Z ≥ 14) in cells or biological samples. In this context, similar tools to those developed for fluorescence microscopy are desired, including chemical probes or tags. In this work, we study rhenium complexes as a convenient and sensitive probe for X-ray fluorescence microspectroscopy. We demonstrate their ability to label and sense exogenously incubated or endogenous proteins inside cells.

27. Probing helix propensity of monomers within a helical oligomer

Abstract

A simple strategy is proposed to assess the propensity of a given monomer to follow or not follow a particular helical scheme and to study helix reversal phenomena within helical oligomers. It consists of placing a monomer having a low helix propensity between two conformationally stable helical segments. Helix reversion then occurs preferentially at the site of this monomer, leading to the formation of isomers having P (right-handed) or M (left-handed) helicities at each of the two helical segments. The proportion between the P-P/M-M and P-M isomers is indicative of the stereochemical relations between the inserted monomer and the helical frame. Thus, xylylene or carboxylic acid anhydride spacers have been introduced between two helical oligoamides of 8-amino-2-quinolinecarboxylic acid. Both these spacers presumably lack some of the structural features that confer quinoline units with a high helix propensity. Only one species is observed in solution in the case of an anhydride spacer. This species was shown by x-ray crystallography to be a racemic mixture of P-P and M-M helices. Unexpectedly, the anhydride is consistently incorporated within helical oligoamides. For the xylylene spacer, the P-P/M-M racemate and P-M meso compound are in equal proportions in chloroform, showing that this spacer does not have a propensity to adopt any helical conformation in this solvent. However, the equilibria between the various isomers are shifted in toluene, where one species largely prevails. This species was shown by x-ray crystallography to be the P-P/M-M racemate. Abstract

The synthesis and characterization of four quinoline-derived foldamers with increasing oligomeric length, dimer, tetramer, pentamer, and nonamer, functionalized with on one end an oligo(p-phenylene vinylene) (OPV) and on the other end a perylene bisimide (PB) chromophore is described. The results have shown when a helical bridge is separating the charge transfer couple, hampering the usefulness of a uniform description of the charge-separation phenomena.

Abstract

Abstract: Alzheimer's disease and oxidative stress are connected. In the present communication, we report the use of a MnII‐based superoxide dismutase (SOD) mimic ([MnII(L)]+, 1+) as a pro‐drug candidate to target CuII‐associated events, namely, CuII‐induced formation of reactive oxygen species (ROS) and modulation of the amyloid‐β (Aβ) peptide aggregation. Complex 1+ is able to remove CuII from Aβ, stop ROS and prevent alteration of Aβ aggregation as would do the corresponding free ligand LH. Using 1+ instead of LH in further biological applications would have the double advantage to avoid the cell toxicity of LH and to benefit from its proved SOD‐like activity. [ABSTRACT FROM AUTHOR]
