Abstract

Background/Objectives: Vaccines may improve the control and eradication of bovine tuberculosis. However, the evaluation of experimental candidates requires the assessment of the protection, excretion, transmission and biosafety. A natural transmission trial among likely infected animals was conducted. Methods: Seventy-four male heifers were randomly distributed (five groups) and vaccinated subcutaneously with attenuated strains (M. bovis Δmce2 or M. bovis Δmce2-phoP), a recombinant M. bovis BCG Pasteur (BCGr) or M. bovis BCG Pasteur. Then, they cohoused with a naturally infected bTB cohort under field conditions exposed to the infection. Results: A 23% of transmission of wild-type strains was confirmed (non-vaccinated group). Strikingly, first vaccination did not induce immune response (caudal fold test and IFN-gamma release assay). However, after 74 days of exposure to bTB, animals were re-vaccinated. Although their sensitization increased throughout the trial, the vaccines did not confer significant protection, when compared to the non-vaccinated group, as demonstrated by pathology progression of lesions and confirmatory tools. Besides, the likelihood of acquiring the infection was similar in all groups compared to the non-vaccinated group (p > 0.076). Respiratory and digestive excretion of viable vaccine candidates was undetectable. To note, the group vaccinated with M. bovis Δmce2-phoP exhibited the highest proportion of animals without macroscopic lesions, compared to the one vaccinated with BCG, although this was not statistically supported. Conclusions: This highlights that further evaluation of these vaccines would not guarantee better protection. The limitations detected during the trial are discussed regarding the transmission rate of M. bovis wild-type, the imperfect test for studying sensitization, the need for a DIVA diagnosis and management conditions of the trials performed under routine husbandry conditions. Re-vaccination of likely infected bovines did not highlight a conclusive result, even suggesting a detrimental effect on those vaccinated with M. bovis BCG. [ABSTRACT FROM AUTHOR]

Abstract

Population-based data on COVID-19 are urgently needed. We report on three rounds of probability sample household surveys in the state of Rio Grande do Sul (Brazil), carried out in nine large municipalities using the Wondfo lateral flow point-of-care test for immunoglobulin M and G antibodies against SARS-CoV-2 ((https://en.wondfo.com.cn/product/wondfo-sars-cov-2-antibody-test-lateral-flow-method-2/)). Before survey use, the assay underwent four validation studies with pooled estimates of sensitivity (84.8%; 95% confidence interval (CI) = 81.4-87.8%) and specificity (99.0%; 95% CI = 97.8-99.7%). We calculated that the seroprevalence was 0.048% (2/4,151; 95% CI = 0.006-0.174) on 11-13 April (round 1), 0.135% (6/4,460; 95% CI = 0.049-0.293%) on 25-27 April (round 2) and 0.222% (10/4,500; 95% CI = 0.107-0.408) on 9-11 May (round 3), with a significant upward trend over the course of the surveys. Of 37 family members of positive individuals, 17 (35%) were also positive. The epidemic is at an early stage in the state, and there is high compliance with social distancing, unlike in other parts of Brazil. Periodic survey rounds will continue to monitor trends until at least the end of September, and our population-based data will inform decisions on preventive policies and health system preparedness at the state level. 38. Cloning and expression of foreign genes in mycobacteria

Abstract

In this thesis, the construction and use of integrative vectors to express foreign genes in mycobacteria is described. These vectors are based on the transposition properties of the IS900 insertion sequence. Transformation by stable integration of multiple copies of the vector into the chromosome of Mycobacterium smegmatis, M. bovis BCG and M. vaccae was achieved. The M. leprae gene encoding the 18-kDa antigen was used to drive the expression of foreign antigens in mycobacteria. The major immunogenic site of Foot-and-Mouth disease virus (FMDV) comprising amino acids 140-160 of the virus protein 1 (VP1) was inserted in the 18kD gene and expressed as a fusion protein. Guinea pigs were vaccinated with the recombinant BCG expressing the FMDV epitope and immune responses specific for the FMDV were detected. A system for testing mycobacterial promoter strength in both extracellularly and intracellularly growing mycobacteria, as well as in E. coli was developed. It consists of a shuttle vector containing a promoterless reporter gene (lacZ). A promoter library was constructed using M. bovis BCG chromosomal DNA and screened in M. smegmatis and E. coli and a number of clones containing DNA inserts with promoter activity were isolated. The activity of each individual clone was measured in both, E. coli and M. smegmatis. Several previously characterized mycobacterial promoters including those of the Mycobacterium bovis BCG hsp60, the M. leprae 18kD and the putative iron regulated M. leprae 28kD gene were also cloned in front of the promoterless lacZ gene and the promoter strength was determined. To study intracellular expression, rBCG was used to infect murine macrophages and the activity of the reporter gene was measured using a fluorescent substrate and a FACS can system. Relative differences in the expression of the reporter gene during intracellular and extracellular growth were detected for some promoters. The BCG hsp60 promoter was shown to be the strongest of the characterized promoters also during intracellular growth. To obtain a higher level of expression of the 18kD/FMDV fusion, the 18kD promoter was replaced by the hsp60 promoter. A level of expression of the fused gene that in vitro was at least 10 fold higher was obtained with the hsp60 promoter in BCG.

Abstract

The OX513A strain of Aedes aegypti, which was developed by the British company Oxitec, expresses a self-limiting transgene that prevents larvae from developing to adulthood. In April 2014, the Brazilian National Technical Commission on Biosafety completed a risk assessment of 0X513A and concluded that the strain did not present new biological risks to humans or the environment and could be released in Brazil. At that point, Brazil became the first country to approve the unconstrained release of a genetically modified mosquito. During the assessment, the commission produced a comprehensive list of--and systematically analysed--the perceived hazards. Such hazards included the potential survival to adulthood of immature stages carrying the transgene--should the transgene fail to be expressed or be turned off by exposure to sufficient environmental tetracycline. Other perceived hazards included the potential allergenicity and/or toxicity of the proteins expressed by the gene, the potential for gene flow or increased transmission of human pathogens and the occupation of vacant breeding sites by other vector species. The Zika epidemic both elevated the perceived importance of Ae. aegypti as a vector--among policy-makers and regulators as well as the general public--and increased concerns over the release of males of the 0X513A strain. We have therefore reassessed the potential hazards. We found that release of the transgenic mosquitoes would still be both safe and of great potential value in the control of diseases spread by Ae aegypti, such as chikungunya, dengue and Zika. La souche OX513A d'Aedes aegypti, qui a ete creee par la societe britannique Oxitec, exprime un transgene autolimitant qui empeche les larves de se developper et de devenir adultes. En avril 2014, la Commission technique nationale de biosecurite du Bresil a procede a une evaluation des risques lies a la souche OX513A et conclu qu'elle ne presentait pas de nouveaux risques biologiques pour les etres humains ou l'environnement et pouvait etre lachee au Bresil. Le Bresil est donc devenu le premier pays a approuver le lacher non contraint d'un moustique genetiquement modifie. Au cours de l'evaluation, la commission a etabli une liste exhaustive des risques percus, qu'elle a par ailleurs systematiquement analyses. Ces risques incluaient la survie potentielle a l'age adulte des larves immatures porteuses du transgene--si le transgene ne s'exprime pas ou est desactive par une exposition a la tetracycline suffisante dans l'environnement. Les autres risques percus incluaient les potentielles proprietes allergisantes et/ ou la toxicite des proteines exprimees par le gene, l'eventualite d'un flux de genes ou d'une transmission accrue d'agents pathogenes pour l'homme et l'occupation de sites de reproduction vacants par d'autres especes vectrices. L'epidemie d'infections a virus Zika a accentue l'importance accordee par les responsables politiques, les organismes de reglementation ainsi que le grand public a Ae. aegypti en tant que moustique vecteur, et a accru l'Inquietude relative au lacher de males de la souche OX513A. Nous avons donc reevalue les risques potentiels. Nous estimons que le lacher de moustiques transgeniques serait a la fois sans danger et extremement utile pour lutter contre les maladies transmises par Ae. aegypti, telles que le chikungunya, la dengue et le virus Zika. La cepa OX513A de Aedes aegypti, que desarrollo la empresa britanica Oxltec, expresa un transgen autollmitado que Impide que las larvas se desarrollen hasta la edad adulta. En abril de 2014, la Comision Nacional Tecnica de Bioseguridad de Brasil realizo una evaluacion de riesgos de 0X513Ay concluyo que la cepa no presentaba nuevos riesgos biologicos para los humanos o el medloambiente y que podria liberarse en Brasil. En ese momento, Brasil se convirtio en el primer pais en aprobar la liberacion limitada de un mosquito modificado geneticamente. A lo largo de la evaluacion, la comision redacto una lista completa, y analizada sistematicamente, de las posibles contingencias. Entre dichos peligros se encontraba la posible supervivencia hasta la edad adulta de etapas inmaduras que portan el transgen, en caso de que este no consiga expresarse o se inutilice debido a la exposicion a la suficiente tetraciclina medioambiental. Otras posibles contingencias eran la alergia y/o toxicidad de las proteinas expresadas por el gen, la posibilidad de un flujo genetico o el aumento de la transmision de patogenos humanos y la ocupacion de lugares de cria desocupados por parte de otras especies vectores. La epidemia por el virus de Zlka aumento la importancia de Ae. aegypti como vector, entre los responsables y reguladores politicos, asi como entre el publico general, y aumento las preocupaciones acerca de la liberacion de machos de la cepa 0X513A. Por lo tanto, se han vuelto a evaluar los posibles riesgos.
