Abstract

International audience; Collybia fusipes is the cause of a root rot of oaks in Europe. Infection by the pathogen is site dependent and inconsistently correlated with crown deterioration. This study aimed at clarifying the relationship between soil factors and C. fusipes impact on tree health. The database of the Département de la Santé des Forêts, the forest health survey service, collected over the French forest during the last 12 years allowed us to perform a large scale study dealing with soil textures in which C. fusipes was reported to induce problems on Quercus robur and/or Q. petraea. Furthermore, a specific survey from 30 plots in north eastern France was carried out to determine which of the soil factors, such as texture, degree of waterlogging, presence and nature of a layer limiting rooting, carbon/nitrogen ratio, cation exchange capacity, or pH could be related to an increased risk of decline of pedunculate oaks infected by C. fusipes. The main results were that C. fusipes is preferentially distributed on coarse textured soils and that its impact on tree crown appearance also increased with the soil sand content.

10. Demonstration of direct bioanalysis of drugs in plasma using nanoelectrospray infusion from a silicon chip coupled with tandem mass spectrometry

Abstract

Quantitative bioanalysis by direct nanoelectrospray infusion coupled to tandem mass spectrometry has been achieved using an automated liquid sampler integrated with an array of microfabricated electrospray nozzles allowing rapid, serial sample introduction (1 min/ sample). Standard curves prepared in human plasma for verapamil (r(2) = 0.999) and its metabolite norverapamil (r(2) = 0.998) were linear over a range of 2.5-500 ng/ mL. Based on the observed precision and accuracy, a lower limit of quantitation of 5 ng/mL was assigned for both analytes. Sample preparation consisted of protein precipitation with an organic solvent containing the structural analogue gallopamil as an internal standard. Protein precipitation was selected both to maximize throughput and to test the robustness of direct nanoelectrospray infusion. Aliquots of supernatant (10 muL) were transferred to the back plane of the chip using disposable, conductive pipet tips for direct infusion at a flow rate of 300 nL/min. Electrospray ionization occurred from the etched nozzles (30-mum o.d.) on the front of the chip, initiated by a voltage applied to the liquid through the pipet tip. The chip was positioned near the API sampling orifice of a triple quadrupole mass spectrometer, which was operated in selected reaction monitoring mode. Results are presented that document the complete elimination of system carry-over, attributed to lack of a redundant fluid path. This technology offers potential advantages for MS-based screening applications in drug discovery by reducing the time for methods development and sample analysis.

Abstract

Both (R)- and (S)-4-hydroxypentylaminoacetamide have been synthesized by reductive amination of glycinamide on the gamma-valerolactols corresponding to (R)- and (S)-gamma-valerolactone, respectively. These enantiomeric lactones were readily obtained in high enantiomeric excess (ee) by enzymic porcine pancreatic lipase (PPL) kinetic resolution of rac-methyl gamma-hydroxyvalerate.

Abstract

The article presents an inoculation method for early assessment of Larch resistance to Lachnellula willkommii. Efficiency of inoculation with mycelium and ascospores was compared on 4 to 13 year-old Larix decidua. Only mycelium induced high infection rates. Mycelial inoculum was contained in colonized sorghum-straw pieces and was introduced in 1 mm diameter holes drilled through the bark in April/May. The main infection was detectable as early as 18 months after inoculation and infection rate increased only until the 28th month after inoculation. The inoculation method was effective for detecting differences in susceptibility among trees.

37. Genetics of somatic incompatibility in <e1>Collybia fusipes</e1>

Abstract

The genetics of somatic incompatibility in tetrapolar Collybia fusipes was studied using eight dikaryotic isolates collected from the wild and their experimentally derived progeny. Monokaryons from each isolate were all paired with the same unrelated monokaryon and also paired together in all combinations. The somatic compatibility of the two resulting sets of dikaryons was studied. Two different types of somatic incompatible interaction were observed, lightly or heavily pigmented lines developing between the two isolates. The dikaryons that had one nuclear type in common and one coming from sibling monokaryons were compatible in 7–27% of the cases, incompatible with a lightly pigmented interaction in 30–93% and incompatible with a heavily pigmented interaction in 0–53%. The results suggest that at least three to four loci control the somatic incompatibility in C. fusipes, one of them alone controlling the heavily pigmented interaction.

38. Structure of <e1>Collybia fusipes</e1> populations in two infected oak stands (B. Mar ais, F. Martin and C. Delatour)

Abstract

Collybia fusipes is the cause of a root rot of oak trees. We studied the structure of C. fusipes populations in two infected oak stands by using somatic incompatibility and DNA amplification. Isolates were obtained from different oak root systems or from within the same root system and somatic incompatibility groups (SIG) were identified. Many small SIGs that seldom encompassed more than one root system were present in both stands. More than one SIG was usually present on an individual root system: there were 3·1±1·3 SIGs on the pedunculate oaks and 2·2±0·6 on the red oaks. The largest SIG contained more than 70% of the isolates obtained from the root system of 14 of the 20 trees studied. Isolates that belonged to the same SIG usually had the same ribosomal intergenic spacer. It is concluded that C. fusipes spreads poorly from tree to tree by vegetative means.

Abstract

The article reports on the method, which is described to collect wood samples axenically from standing trees by the use of a Pressler borer in conjunction with protective sterile jackets. Wood samples from living trees in the forest are collected with a Pressler borer. The wound is contaminated by the borer itself during tree penetration and the wood core is contaminated as soon as air contact occurs. The wood core is extracted into the extractor's jacket which is closely maintained at the borer distal end during extraction.
