International audience; In order to improve the diagnosis of environmental contamination of freshwater, multi-biomarkers approaches have been developed to better reflect the physiological state of an organism and potentially detect exposures to xenobiotics. Genotoxic biomarkers seem to be quite relevant due to the multitude of substances interacting with the DNA molecule which can affect its integrity in somatic and germ cells. Among the existing tools used to highlight the genotoxicity on fish erythrocytes, the alkaline comet assay (SCGE) is the most widely used method to detect primary DNA strand breaks and flow cytometry (FCM) is becoming a fast and convenient tool to notice chromosomal damages for aquatic biomonitoring. The present work proposes to evaluate the relevance to include two genotoxic biomarkers (primary DNA strand breaks and chromosomal damages) in the existing battery of multi-biomarkers developed on three-spined stickleback (Gasterosteus aculeatus). After 21 days of exposure, biological samples were recovered from sticklebacks caged in six sites in Northern France to constitute the battery. The inclusion of genotoxicity biomarkers seems to bring a little improvement to the discriminating power of sites as obtained by a Principal Component Analysis (PCA) explaining 30.8% of the variance, although they provide a complementary information on the toxicity of sites. The “Rhonelle” river was the most affected by primary DNA strand breaks measured in fish but this parameter was not well represented on axis, in opposition to fish caged in “Deule” and “Sensée Bouchain” sites showing lower levels of chromosomal damages, in accordance with the first component axis of the PCA.

Abstract

Regulations, such as the EU Water Framework Directive, for the protection of the aquatic environment, encourage the development of new sensitive and predictive tools to evaluate the effects of contaminants on aquatic living organisms. In this context, energy metabolism biomarkers are of interest given the central place of energy in all physiological processes (i.e. defense, growth and reproduction). In this study, we addressed for the first time three digestive enzymes (amylase, trypsin and intestinal alkaline phosphatase) and evaluated their interest as novel biomarkers in the threespine stickleback (Gasterosteus aculeatus), a model species in multidisciplinary fields of biology, and recently in ecotoxicology. The interest of the two digestive enzymes as biomarkers was evaluated in comparison with other parameters, usually applied in ecotoxicology studies: biometric parameters, reproduction parameters, antioxidant parameters, and other energy metabolism parameters. For that, juvenile sticklebacks were exposed for 3 months in semi-static conditions to cadmium at 0.5 μg.L-1 (chemical stress), and two water temperatures 16°C and 21°C (physical stress). Results showed that the high temperature (21 °C) was the major stress factor on stickleback physiology. The very low cadmium concentration used in this study had no effect on most of the studied parameters (i.e. growth parameters, antioxidant parameters and parents' reproduction parameters). However, digestive enzymes showed an interesting sensitivity to cadmium exposure which was emphasis at 21 °C (significant decrease). Although cadmium had no effect on parental reproduction, negative consequences on their offspring survival were observed after in vitro fertilization trials (decreased survival capacity). Compared to conventional measurements, digestive enzymes responded rapidly, which could be an important advantage for their use as early warning tool, to reflect organism’s health.

Abstract

The measure of DNA integrity by the single cell gel electrophoresis (SCGE) or comet assay is especially recommended for its sensitivity and its capacity for detecting different types of damages. Therefore, it has been applied in environmental genotoxicity in a variety of organisms. It appears today necessary to define both reference and threshold levels of DNA damage, for their application in in situ biomonitoring. However, little is known about the influence of both biological (sex, reproduction status) or external (temperature…) confounding factors on the measure of DNA damage by the comet assay. These variables need to be taken into account if the robustness of the assay is to be established (Jha, 2008). In the zebra mussel Dreissena polymorpha (recommended as a sentinel species in the evaluation of freshwater quality) the measure of DNA damage by the comet assay is mainly performed on hemocytes, which are circulating cells involved in key physiological functions such as immunity, homeostasis, detoxication…. This communication will present and discuss results from an innovative study about the variability of the baseline level of DNA damage in hemocytes of mussels encaged for one year in the canal de l’Aisne à la Marne (Reims), according to their sex and their reproductive status. The sensitivity and the suitability of hemocytes in the evaluation of environmental genotoxicity will also be discussed, referring to observations during a 6 month-exposure of mussels in mesocosms to environmentally realistic concentrations of carbamazepine.

Abstract

communication par affiche; International audience; The growing global consumption of drugs by humans in association with a deficiency of water treatment plants to totally eliminate them resulted in the contamination of aquatic environment by pharmaceutical residues. Today, our knowledge on ecotoxicity and biological effects of these organic pollutants on aquatic biota is still scarce. Our survey focused on the evaluation of the genotoxicity of two pharmaceutical compounds: diclofenac and carbamazepine, a non-steroidal anti-inflammatory and an anti-epileptic agent respectively, which are frequently measured in surface and ground-waters at concentrations below 10 µg/L. The measure of the DNA damage by the comet assay (or SCGE assay) was studied in the zebra mussel (Dreissena polymorpha) as biomarker of genotoxicity. This assay was applied on two cell types: hemocytes and sperm cells after short term ex vivo exposures (1-6h) in laboratory experiments to the two tested contaminants and after a long-term in situ exposure (2 and 5 months) of mussels in mesocosms contaminated with diclofenac. Concentrations tested for both experiments (laboratory/mesocosms) were environmentally relevant (from 0.1 to 10 µg/L). Results showed a significant increase of DNA damage in hemocytes (after 6h) and sperm cells (from 1h to 6h) at all concentrations tested after ex vivo exposure to carbamazepine. No genotoxic effect of diclofenac was observed on hemocytes but a low genotoxicity in sperm cells after 1h and 3h exposure, with a return to a baseline level of DNA damage after 6h (no cytotoxic effect was observed in parallel). However, long-term exposure of mussels in mesocosms revealed the genotoxicity of diclofenac for both hemocytes and sperm cells, with a positive relationship in the degree of DNA damage for the two cell types. Our study is the first to reveal in the zebra mussel the interest to evaluate DNA integrity in sperm cells. Sperm cells appeared as a sensitive cell type with a faster response to a genotoxic stress than hemocytes. A possible repair of DNA damage by sperm cells was observed, but seems rapidly limited with the intensity of the genotoxic stress. Our results will be discussed according to the litterature data as well as mechanisms underlying genotoxicity of these pharmaceuticals, which still need to be elucidated.

Abstract

The global dynamic spread of chemical contamination through the aquatic environment calls for the development of biomarkers of interest. Reproduction is a key element to be considered because it is related to the sustainability of species. Spermatogenesis is a complex process that leads to the formation of mature germ cells, whose steps and impairments need to be finely described in ecotoxicological analyses. The physiological process has been commonly described by histological analyses of gonads in different taxa. In the present paper, we describe the development of a novel technique to characterize spermatogenesis based on the analysis of the DNA content of germ cells by flow cytometry, using a DNA-intercalating agent. This new biomarker, referred to as an index of sexual maturity, proved relevant to describe the seasonal reproductive cycle of the zebra mussel, Dreissena polymorpha (Pallas, 1771), used as a sentinel species in the biomonitoring of continental waters and sensitive to highlight the reprotoxicity of carbamazepine (an anti-epileptic pharmaceutical) tested under ecosystemic conditions (mesocosms). Abstract

The degradation in the quality of silver eel and their health could have been a major factor in the collapse of the European eel (Anguilla anguilla) population. However, the health status of the spawners has been poorly studied until now. This study evaluated the quality of migrant male silver eels from four Mediterranean habitats in France presenting different degrees of contamination: Canet-Saint-Nazaire, Salses-Leucate and Bages-Sigean lagoons and La Berre River. We considered pathogens including Anguillicoloides crassus and EVEX virus and the concentration of chemical contaminants including PCBs, OCs and heavy metals. Our study results revealed different patterns of pollution and infection in the four habitats, with high individual variability. No single silver eel was free of pollution. Total dichloro-diphenyl-trichloroethane (DDT) and copper contaminations, as well as the Swim bladder Degenerative Index (induced by parasitism), were remarkably high in eels from Canet lagoon, while eels from Salses lagoon showed lower levels of contaminants and parasite infection. A non-negligible proportion of eels were strongly impacted with levels of contaminants/parasites that could potentially impair their migration and reproduction. Our study revealed low to moderate contamination levels compared with the other Mediterranean sites previously reported, except for high concentrations of DDTs and Cu in Canet lagoon. We discuss the contribution of these results in the context of possible implications for silver eels reproductive success and local eel population management.

Abstract

Human activities have led to increased levels of various pollutants including metals in aquatic ecosystems. Increase of metallic concentrations in aquatic environments represents a potential risk to exposed organisms, including fish. The aim of this study was to characterize the environmental risk to fish health linked to a polymetallic contamination from former uranium mines in France. This contamination is characterized by metals naturally present in the areas (manganese and iron), uranium, and metals (aluminum and barium) added to precipitate uranium and its decay products. Effects from mine releases in two contaminated ponds (Pontabrier for Haute-Vienne Department and Saint-Pierre for Cantal Department) were compared to those assessed at four other ponds outside the influence of mine tailings (two reference ponds/department). In this way, 360 adult three-spined sticklebacks ( Gasterosteus aculeatus) were caged for 28 days in these six ponds before biomarker analyses (immune system, antioxidant system, biometry, histology, DNA integrity, etc.). Ponds receiving uranium mine tailings presented higher concentrations of uranium, manganese and aluminum, especially for the Haute-Vienne Department. This uranium contamination could explain the higher bioaccumulation of this metal in fish caged in Pontabrier and Saint-Pierre Ponds. In the same way, many fish biomarkers (antioxidant and immune systems, acetylcholinesterase activity and biometric parameters) were impacted by this environmental exposure to mine tailings. Abstract

This study investigates cadmium effects on key messenger RNA (mRNA) expression (MT, MnSOD, CuZnSOD, CAT, ABCB1, HSP70, and CO1) by qPCR in the cockle Cerastoderma glaucum after chronic exposure to two high but environmentally relevant concentrations of CdCl (50 μg/L and 5 mg/L) for 12 h to 18 days. Cd accumulation measured in cockles' tissues is significantly higher in both treatment conditions compared to controls and in a dose-dependent manner. Stress on stress tests performed at different times of the experiment clearly demonstrated that exposure to both concentrations of Cd significantly affects cockle survival time in air. Important changes in gene transcription were also highlighted. In particular, MT, HSP70, CAT, and CuZnSOD seem to be relevant biomarkers of Cd exposure because (1) their mRNA levels increase upon exposure and (2) they are highly correlated to Cd accumulation in tissues.
