In the tumor microenvironment (TME), ROS production affects survival, progression, and therapy resistance in colorectal cancer (CRC). H2O2-mediated oxidative stress can modulate Wnt/β-catenin signaling and metabolic reprogramming of the TME. Currently, it is unclear how mild/moderate oxidative stress (eustress) modulates Wnt/β-catenin/APC and JNK signaling relationships in primary and metastatic CRC cells. In this study, we determined the effects of the H2O2 concentration inducing eustress on isogenic SW480 and SW620 cells, also in combination with JNK inhibition. We assessed cell viability, mitochondrial respiration, glycolysis, and Wnt/β-catenin/APC/JNK gene and protein expression. Primary CRC cells were more sensitive to H2O2 eustress combined with JNK inhibition, showing a reduction in viability compared to metastatic cells. JNK inhibition under eustress reduced both glycolytic and respiratory capacity in SW620 cells, indicating a greater capacity to adapt to TME. In primary CRC cells, H2O2 alone significantly increased APC, LEF1, LRP6, cMYC and IL8 gene expression, whereas in metastatic CRC cells, this effect occurred after JNK inhibition. In metastatic but not in primary tumor cells, eustress and inhibition of JNK reduced APC, β-catenin, and pJNK protein. The results showed differential cross-regulation of Wnt/JNK in primary and metastatic tumor cells under environmental eustress conditions. The body of work split into two volumes, offers the reader a view both organic and thorough, delving deeply into the thoughts and actions of the prior of Barbiana. The critical apparatus who add to the edition is particularly accurate in offering writings that deepen the foresight of Milani on themes still crucial today: education, inclusion, justice and the dignity of the human person. The designation of the Extraordinary Jubilee of Mercy offers a chance to our author to have a deeper appreciation of the aspects of mercy, in particular of its close link with politics and welfare. In particular, mercy as fraternity and solidarity for the construction of the common good. Through reference to institutions that more specifically are always interested in practical charity, mercy, the author wants to underline that its practice should be common to the whole world and lay the basis for the social and civil organization of a community that give particular attention to the end result. The author, through a socio-political and cultural analysis of the reality in which we live and looking at, also, the Social doctrine of the Church, comes to the conclusion that it is necessary to construct a Community where everyone is disposed to recognize each other as a depository of this right. From the 47th Social week in Turin of Italian Catholics a deepned and sharp reflection on the family today and in its future starts. This journey, among history, sociology, philosophy and theology, in the light of the social magisterium, arrives at the concept of Christian family that bring us up, not only to the “good life of the Gospel” but also to “the social life and politics”. The author starts from the transformation of the family model that today is the mirror of the social chaos to set then against the cultural, social, political and pastoral terms. He dwells upon the attention that the economic and social politics of the governments keep for the family and he would like the passage from welfare state to a welfare society. The reflection ended by defining the connotations of a family. The conclusion: a strong appeal to the laity to engage in politics and to address issues in the light of the Gospel; a strong appeal to the role of the parish, a place to welcome its families and to train of its new builders. The Year of faith causes many reflections on the human subject. The author reflects on both its current and complex aspect: that of the commitment of Christians in its institutions and for its institutions. Today there is a widespread attitude of apathy and indifference. The starting point is to discover that faith is the basis not only of the relationship with God, but the relationship between all men to trust and rely on each other. This is the dynamic of human coexistence. The focal point is the coherence between faith and life, that man must be faithful to his commitments and must be able to not disappoint. There needs to be consistency between the Gospel and culture. Another crucial point is the comparison of the reasons for faith and the reasons why we have faith. The possible relationship between faith and politics is a dialogue of mutual recognition. The Christian is called to give through his commitment and testimony, the face of God revealed in Jesus Christ, the model of charity and justice, not only for individuals but for society as a whole.
