Abstract

Autistic individuals experience higher rates of externalising and internalising symptoms that may vary with environmental factors. However, there is limited research on variation across settings that may highlight common factors with globally generalisable effects. Data were taken from two cohorts: a multinational European sample (n = 764; 453 autistic; 311 non-autistic; 6-30 years), and a South African sample (n = 100 non-autistic; 3-11 years). An exploratory factor analysis aggregated clinical (Verbal Comprehension and Perceptual Index), adaptive traits (Vineland Adaptive Behaviour Scale) and socio-economic variables (parental employment and education, home and family characteristics) in each cohort separately. With regression, we investigated the effect of these factors and autistic traits on internalising and externalising scores (measured with the Strengths and Difficulties Questionnaire). Cohorts showed similar four-factor structures (Person Characteristics, Family System, Parental and Material Resources). The 'Family System' factor captured family size and maternal factors and was associated with lower internalising and externalising symptoms in both cohorts. In the European cohort, high autistic traits reduced this effect; the opposite was found in the South Africa cohort. Our exploratory findings from two separate analyses represent consistent evidence that Family System is associated with internalising and externalising symptoms, with a context-specific impact in persons with high autism traits. Abstract

Mechanical function of the heart during open-chest cardiac surgery is exclusively monitored by echocardiographic techniques. However, little is known about local kinematics, particularly for the reperfused regions after ischemic events. We report a novel imaging modality, which extracts local and global kinematic parameters from videos of $\textit{in situ}$ beating hearts, displaying live video cardiograms of the contraction events. A custom algorithm tracked the movement of a video marker positioned $\textit{ad hoc}$ onto a selected area and analyzed, during the entire recording, the contraction trajectory, displacement, velocity, acceleration, kinetic energy and force. Moreover, global epicardial velocity and vorticity were analyzed by means of Particle Image Velocimetry tool. We validated our new technique by i) computational modeling of cardiac ischemia, ii) video recordings of ischemic/reperfused rat hearts, iii) videos of beating human hearts before and after coronary artery bypass graft, and iv) local Frank-Starling effect. In rats, we observed a decrement of kinematic parameters during acute ischemia and a significant increment in the same region after reperfusion. We detected similar behavior in operated patients. This modality adds important functional values on cardiac outcomes and supports the intervention in a contact-free and non-invasive mode. Moreover, it does not require particular operator-dependent skills.

Abstract

BACKGROUND:Systemic and local immune suppression plays a significant role in glioma progression. Glioma microenvironment contains both brain-resident microglial cells (MG) and bone marrow-derived macrophages (BMDM), but the study of their functional and immune regulatory activity has been hampered until now by the lack of markers allowing a proper identification and isolation to collect pure populations. METHODS:Myeloid and lymphoid infiltrate were characterized in grade II, III and IV gliomas by multicolor flow cytometry, along with the composition of the cell subsets of circulating myeloid cells. Macrophages were sorted and tested for their immunosuppressive ability. Moreover, following preoperative administration of 5-aminolevulinic acid to patients, distinct areas of tumor lesion were surgically removed and analyzed, based on protoporphyrin IX fluorescence emission. RESULTS:The immune microenvironment of grade II to grade IV gliomas contains a large proportion of myeloid cells and a small proportion of lymphocytes expressing markers of dysfunctional activity. BMDM and resident MG cells were characterized through a combination of markers, thus permitting their geographical identification in the lesions, their sorting and subsequent analysis of the functional characteristics. The infiltration by BMDM reached the highest percentages in grade IV gliomas, and it increased from the periphery to the center of the lesion, where it exerted a strong immunosuppression that was, instead, absent in the marginal area. By contrast, MG showed little or no suppression. Functional differences, such as iron metabolism and phagocytosis, characterized resident versus blood-derived macrophages. Significant alterations in circulating monocytes were present in grade IV patients, correlating with accumulation of tumor macrophages. CONCLUSIONS:Grade IV gliomas have an alteration in both circulating and tumor-associated myeloid cells and, differently from grade II and III gliomas, show a significant presence of blood-derived, immune suppressive macrophages. BMDM and MG have different functional properties.

Abstract

This article presents a real-world observational study on the effectiveness and toxicity of cemiplimab in patients with advanced cutaneous squamous cell carcinoma (CSCC). The study enrolled 90 patients in four Italian centers, showing an overall response rate of 47.8% and a disease control rate of 73.4%. Patients with head and neck primary tumors had better overall survival compared to those with tumors in other locations. The study confirmed the safety profile of cemiplimab, with treatment-related adverse events occurring in 51.1% of patients, but none were Grade 4 or 5. Further research is needed to personalize therapy for this patient population. [Extracted from the article]

Abstract

In this paper the initiation and propagation of delamination in thin-walled green composite structures were studied. In particular, laminates of flax and hemp fibers with epoxy resin were investigated from experimental and numerical points of view, using the finite element code LS-DYNA. First, an experimental campaign was conducted using DCB and 4ENF tests to obtain the values of the interlaminar fracture modes, required for the cohesive fracture modeling. In addition, in-plane crashworthiness tests were performed on composite plates to analyze the damage of the specimens, under both quasi-static and dynamic conditions. A trigger mechanism was considered to help delamination to start and to ensure the most progressive crushing possible. The predominant damage mode was splaying, but some ply fragmentation in the middle layers and some buckling phenomena were also observed. In general, more stable progressive crushing was observed under dynamic conditions than under quasi-static ones, even if a reduction in terms of absorbed energy was obtained by moving to the dynamic condition. The numerical predictions showed a good agreement with the experimental reference results, validating the current framework for crashworthiness analysis of green composite structures.
