The first International Conference on Unconventional Animal Models of Alzheimers Disease and Aging (UAMAA) took place on December 13-16, 2023, in Santiago, Chile. The Alzheimers disease (AD) research field is currently in search for new and unconventional models that could hold greater translational potential than transgenic mouse models. Thus this UAMAA conference is timely and significant. The event consisted of 6 sessions with talks from 28 world-class scientists from all over the world. These animal models of interest include the degu (Octodon degu), the dog (Canis familiaris), and certain species of nonhuman primates that may better recapitulate neuropathology and cognitive impairments in human AD. Our conference has provided a formal forum to discuss and highlight new research directions, alternative animal models, and innovative approaches for the AD and aging research field.

Degus (Octodon degus) are a highly gregarious species of caviomorph rodent native to South America. Kinship does not appear to play a role in degu social structure, and alloparenting is often observed between unrelated females. We hypothesize that female degus readily establish new, cooperative peer relationships. Here, we examined changes in dyadic behavior as individuals became more familiar, testing the prediction that interactions between female strangers would quickly resemble those of cagemates. Adult degus underwent a several week series of 20 min "reunion" social exposures, interleaving reunions with initial strangers and, as a control, familiar cagemates. Males showed initially higher levels of interaction with strangers that converged with cagemate levels over experience. Females could be split into two groups: those that consistently interacted more with strangers (SC‐HIGH) and those that did not (SC‐LOW); however, unlike males, the higher interaction levels observed between strangers did not change with familiarity. Following 10 reunion sessions female strangers were housed together to create "new cagemates." Even after co‐housing, SC‐HIGH (but not SC‐LOW) females continued to interact more with the relatively unfamiliar peer than their prior cagemate, particularly in face‐to‐face and rear‐sniffing interactions. A final set of reunions with new strangers found that individual differences in female responses to social novelty were preserved. These results reveal sex differences in the rules relating familiarization to social relationships in degus, and that female predispositions toward cooperation may be due to inherent responses to new individuals more than to how they negotiate relationships over time. Individuals experience changes in spatiotemporal group cohesion during social foraging, leading to groups splitting up and remerging as smaller subgroups. These temporal changes represent fission–fusion dynamics, which is hypothesized to adjust foraging group size to ecological and social settings. While available evidence supports the importance of ecological drivers, social factors and the joint effects of ecological and social conditions remain relatively less clear. In the present study, we address both components of context to examine whether the stability of foraging groups is driven by socioecological conditions. Specifically, we examined the extent to which stability in group membership by female and male foragers is enhanced by increasing predation risk, food abundance, group kinship and adult opposite-sex ratio. Additionally, we determined whether foraging group stability during mating and offspring rearing seasons was contingent on the sex of individuals. We tested these effects in the degu, Octodon degus , a diurnal and group-living rodent that forages socially and rears offspring communally. We found that degus foraged in small groups of mixed composition, where social stability increased with increasing abundance of high-quality food, increasing refuge density, decreasing interindividual distance and in the presence of predators, and was higher during spring than during winter. In addition, the stability of foraging groups was differentially sensitive to opposite-sex ratio in females and males. The minor secreted proteinase of B. pumilus 3-19 MprBp classified as the unique bacillary adamalysin-like enzyme of the metzincin clan. The functional role of this metalloproteinase in the bacilli cells is not clear. Analysis of the regulatory region of the mprBp gene showed the presence of potential binding sites to the transcription regulatory factors Spo0A (sporulation) and DegU (biodegradation). The study of mprBp activity in mutant strains of B. subtilis defective in regulatory proteins of the Spo- and Deg-systems showed that the mprBp gene is partially controlled by the Deg-system of signal transduction and independent from the Spo-system. When establishing new peer relationships, animals may explore different modes of interaction, testing-out dominance roles, reciprocation of affiliation, and responses to investigation. This exploration is potentially risky, as higher variability may be counterproductive to establishing expectations and trust. There is therefore a tradeoff between exploration within a new social relationship and maintaining predictable, 'safe' behaviours, raising questions about how animals differ in how they engage with strangers. The Chilean degu offers an opportune case study to investigate novel social situations, as females form relationships relatively rapidly with unrelated peers. We presented degu dyads with a series of 20 min 'reunion' sessions and found that session-to-session variability in stranger females is, in fact, lower than in cagemates, and lower than stranger or cagemate males. Reduced variability was observed only after an initial social exposure, suggesting it was a feature of new relationships rather than novelty. There was no evidence that groups differed in predictability of behaviours within a reunion. It is known that in the wild, female degus differ from males by readily forming cooperative relationships with unrelated individuals. The data therefore raise the possibility that animals predisposed to cooperation might also show reduced behavioural variability across encounters with new individuals. Abstract Background Global transcription machinery engineering (gTME) is an effective approach employed in strain engineering to rewire gene expression and reshape cellular metabolic fluxes at the transcriptional level. Results In this study, we utilized gTME to engineer the positive transcription factor, DegU, in the regulation network of major alkaline protease, AprE, in Bacillus pumilus. To validate its functionality when incorporated into the chromosome, we performed several experiments. First, three negative transcription factors, SinR, Hpr, and AbrB, were deleted to promote AprE synthesis. Second, several hyper-active DegU mutants, designated as DegU(hy), were selected using the fluorescence colorimetric method with the host of the Bacillus subtilis ΔdegSU mutant. Third, we integrated a screened degU(L113F) sequence into the chromosome of the Δhpr mutant of B. pumilus SCU11 to replace the original degU gene using a CRISPR/Cas9 system. Finally, based on transcriptomic and molecular dynamic analysis, we interpreted the possible mechanism of high-yielding and found that the strain produced alkaline proteases 2.7 times higher than that of the control strain (B. pumilus SCU11) in LB medium. Conclusion Our findings serve as a proof-of-concept that tuning the global regulator is feasible and crucial for improving the production performance of B. pumilus. Additionally, our study established a paradigm for gene function research in strains that are difficult to handle.

Sociality-i.e., life in social groups-has evolved many times in rodents, and there is considerable variation in the nature of these groups. While many species-typical behaviors have been described in field settings, the use of consistent behavioral assays in the laboratory provides key data for comparisons across species. The preference for interaction with familiar or novel individuals is an important dimension of social behavior. Familiarity preference, in particular, may be associated with more closed, less flexible social groups. The dimension from selectivity to gregariousness has been used as a factor in classification of social group types. Laboratory tests of social choice range from brief (10 minutes) to extended (e.g., 3 hours). As familiarity preferences typically need long testing intervals to manifest, we used 3-hour peer partner preference tests to test for the presence of familiarity preferences in same-sex cage-mates and strangers in rats. We then conducted an aggregated analysis of familiarity preferences across multiple rodent species (adult male and female rats, mice, prairie voles, meadow voles, and female degus) tested with the same protocol. We found a high degree of consistency within species across data sets, supporting the existence of strong, species-typical familiarity preferences in prairie voles and meadow voles, and a lack of familiarity preferences in other species tested. Sociability, or total time spent near conspecifics, was unrelated to selectivity in social preference. These findings provide important background for interpreting the neurobiological mechanisms involved in social behavior in these species.

Group-living animals vary in social behavior across multiple dimensions, including in the selectivity of social interactions with familiar versus unfamiliar peers. Standardized behavioral tests can be used to tease apart different dimensions of behavior. These serve a dual function-on one hand, helping to isolate behavioral factors that may support collective behavior in natural habitats, and, on another, providing a basis for comparative approaches to understanding physiological mechanisms of behavior. Degus (Octodon degus) are South American caviomorph rodents that nest and forage in groups with relatively low genetic relatedness. Flexibility in group membership is likely supported by gregariousness toward strangers, but the relative preference for strangers compared with familiar individuals has not been systematically tested. We assessed the specificity of social preferences in female degus using a same-sex partner preference test. Degus huddled extensively with both familiar and unfamiliar peers, with no average preference for one over the other. Detailed analysis of social interactions demonstrated an effect of familiarity on social investigation and aggressive behaviors, indicating that degus distinguished between familiar and unfamiliar conspecifics, even though it did not impact huddling. This behavioral profile is thus far unique to degus; in similar tests, meadow and prairie voles exhibit strong partner preferences for known peers, while mice exhibit low social huddling and spend relatively less time in social chambers. Understanding how group-living species differ in specific aspects of social behavior such as familiarity/novelty preference and propensity for social contact will offer a foundation to interpret differences in neural systems supporting sociality.

Abstract The long-lived Chilean rodent (Octodon degus) has been reported to show spontaneous age-dependent neuropathology and cognitive impairments similar to those observed in human AD. However, the handful of published papers on degus of differing genetic backgrounds yield inconsistent findings about sporadic AD-like pathological features, with notably differing results between lab in-bred degus versus outbred degus. This motivates more extensive characterization of spontaneously occurring AD-like pathology and behavior in degus. In the present study, we show AD-like neuropathological markers in the form of amyloid deposits and tau abnormalities in a cognitively impaired subset of aged outbred degus. Compared to the aged degus that show normal burrowing behavior, the age-matched degus with burrowing behavior deficits correlatively exhibit detectable human AD-like Aβ deposits and tau neuropathology, along with neuroinflammatory markers that include enhanced microglial activation and higher numbers of reactive astrocytes in the brain. This subset of cognitively impaired aged degus also exhibits cerebral amyloid angiopathy and tauopathy. We find robust neurodegenerative features in behaviorally deficient aged degus, including hippocampal neuronal loss, altered parvalbumin and perineuronal net staining in the cortex, and increased c-Fos neuronal activation in the cortex that is consistent with the neural circuit hyperactivity reported in human AD patients. By focusing on the subset of aged degus that show AD-like behavioral deficits and correlative neuropathology, our findings establish outbred degus as a natural model of sporadic AD and demonstrate the potential importance of wild-type outbred genetic backgrounds for AD pathogenesis.

We report a case of ventricular septal defect causing congestive heart failure in a two-year-old, male common degu (Octodon degus). The patient presented for anorexia and dental disease, and a grade 4/6 holosystolic cardiac murmur was detected on physical exam. Thoracic radiographs showed cardiomegaly and a diffuse interstitial pulmonary pattern, consistent with congestive heart failure. Echocardiography was supportive of a perimembranous ventricular septal defect exhibiting low-velocity left-to-right shunting, and biatrial enlargement. These diagnoses were confirmed on post-mortem exam, along with pulmonary edema and cardiomyocyte hypertrophy, degeneration, and regeneration. This is the first published account of a ventricular septal defect and congestive heart failure in a degu.

The degu (Octodon degus) is a rodent that normally constructs burrows for nesting and rearing. To navigate inside these burrows, degus may use idiothetic and/or sensory cues more than visual information, which is less effective in burrows. Spatial information for navigation is processed in several key brain regions including the retrosplenial cortex (RS). However, the structural characteristics of the degu RS have not been previously reported. The present study measured the sizes of the RS and constituent areas 29 and 30 in the degu, and compared these to those found in the rat, which is a terrestrial rodent. The proportion of the rostrocaudal length of the entire RS relative to that of the entire cortex was significantly larger in degus versus rats. The proportion of the rostrocaudal length of the RS at levels rostral to the splenium of the corpus callosum relative to that of the entire cortex was also significantly larger in degus versus rats. Furthermore, the ratio of the estimated volume of area 29 relative to that of area 30 was significantly larger in degus versus rats. These results show that the degu has a rostrocaudally longer rostral RS with a larger area 29 compared to the rat, which suggests that these structural features may be relevant to differences in spatial information processing between the fossorial degu and terrestrial rat. Lichenysin, producted by Bacillus licheniformis , is an important cyclic lipopeptide biosurfactant, which has potential applications in oil exploitation, drug development, biological control of agriculture and bioremediation. While studies are lacking on metabolism regulation of lichenysin biosynthesis, which limits metabolic engineering and large-scale production of lichenysin. In this study, the yield of lichenysin was improved obviously by 13.6 folds to 2.18 ± 0.03 g/L in degU deletion strain (WX02△ degU) compared with the wild-type strain (WX02) and completely inhibited in degU overexpressed strain (WX02/pHY- degU). We further proved that DegU, a transcription factor plays a significant role in multicellular behavior, is a key negative regulator of lichenysin synthesis lchA operon. But interestingly, lichenysin yield was still inhibited by overexpressing DegU in the promoter-substituted strain (WX02-P P43 lch), in which promoter of lchA operon cannot be controlled by DegU. Thus, through 13C-metabolic flux analysis, we found that deletion of degU also enhanced glucose uptake, branched chain amino acid synthesis, and fatty acid synthesis, while decrease acetoin synthesis, which is beneficial for the supply of lichenysin precursors. Further experiments demonstrate that DegU regulates these pathways by binding to the promoter regions of related genes. Overall, we systematically investigated the multi-pathway regulation network mediated by DegU on lichenysin biosynthesis, which not only contributes to the further metabolic engineering for lichenysin high-production, but sheds light on studies of transcription factor regulation. • The multi-pathway regulation network of DegU on lichenysin biosynthesis was systematically explored by 13C-MFA. • DegU is a key transcriptional inhibitor of lichenysin synthesis operon lchA. • DegU negatively regulates glucose uptake, BCAA and fatty acid synthesis, while positively regulates acetoin synthesis. • Lichenysin yield was increased by 13.6 folds to 2 176 mg/L by knocking out degU. Physiological responses to inhaled anesthetics vary among species. Therefore, a precise anesthetic technique is important for each individual species. In this study, we focused on the degu (Octodon degus), a small herbivorous rodent. Degus have recently begun to be used as laboratory models for brain research because of certain human-like characteristics, such as spontaneous development of Alzheimer's disease. In this study, we evaluated appropriate induction and maintenance anesthesia conditions for isoflurane and sevoflurane in degus by a stimulation test, electroencephalography (EEG), minimum alveolar concentration (MAC), and vital signs. During induction, more rapid time to loss of the righting reflex and deeper anesthesia in degus were observed in isoflurane. The MAC value for degus were 1.75 ± 0.0% in isoflurane and 2.25 ± 0.27% in sevoflurane. Whereas some degus were awake during maintenance anesthesia using both anesthetics at concentrations of ≤2%, no rats were awake when using sevoflurane at a concentration of 2%. The duration of the total flat EEG, a measure of the depth of maintenance anesthesia, was longer for isoflurane than for sevoflurane. Furthermore, higher concentrations of both anesthetics suppressed the respiratory rate in degus. These new findings regarding inhalation anesthesia in degus will contribute to future developments in the fields of laboratory animals and veterinary medicine.

Alkaline protease AprE, produced by Bacillus licheniformis 2709 is an important edible hydrolase, which has potential applications in nutrient acquisition and medicine. The expression of AprE is finely regulated by a complex transcriptional regulation system. However, there is little study on transcriptional regulation mechanism of AprE biosynthesis in Bacillus licheniformis , which limits system engineering and further enhancement of AprE. Here, the severely depressed expression of aprE in degU and degS deletion mutants illustrated that the regulator DegU and its phosphorylation played a crucial part in AprE biosynthesis. Further electrophoretic mobility shift assay (EMSA) in vitro indicated that phosphorylated DegU can directly bind to the regulatory region though the DNase I foot-printing experiments failed to observe protected region. The plasmid-mediated overexpression of degU32 (Hy) obviously improved the yield of AprE by 41.6 % compared with the control strain, which demonstrated the importance of phosphorylation state of DegU on the transcription of aprE in vivo. In this study, the putative binding sequence of aprE (5′-TAAAT......AAAAT.......AACAT...TAAAA-3′) located upstream −91 to −87 bp, −101 to −97 bp, −195 to −191 bp, −215 to −211 bp of the transcription start site (TSS) in B. licheniformis was computationally identified based on the DNA-binding sites of DegU in Bacillus subtilis. Overall, we systematically investigated the influence of the interplay between phosphorylated DegU and its cognate DNA sequence on expression of aprE , which not only contributes to the further AprE high-production in a genetically modified host in the future, but also significantly increases our understanding of the aprE transcription mechanism. Listeria monocytogenes is more heat-resistant than most other non-spore-forming foodborne pathogens, posing a severe threat to food safety and human health, particularly during chilled food processing. The DegU orphan response regulator is known to control heat resistance in L. monocytogenes ; however, the underlying regulatory mechanism is poorly understood. Here, we show that DegU contributes to L. monocytogenes exponential growth under mild heat-shock stress. We further demonstrate that DegU directly senses heat stress through autoregulation and upregulates the hrcA-grpE-dnaK-dnaJ operon, leading to increased production of heat-shock proteins. We also show that DegU can directly regulate the expression of the hrcA-grpE-dnaK-dnaJ operon. In conclusion, our results shed light on the regulatory mechanisms underlying how DegU directly activates the hrcA-grpE-dnaK-dnaJ operon, thereby regulating heat resistance in L. monocytogenes. The degu has drawn increasing attention for use as an experimental animal in stress response studies due to its physiological features, such as diurnality and seasonal breeding, which differ from conventional laboratory rodents. Stress response is elicited by steroid hormones secreted by the adrenal gland, whose functions are controlled by pituitary hormones reaching through the adrenal arteries. However, knowledge of the arterial anatomy of the degu adrenal gland remains insufficient. To address this issue, we observed adrenal arteries in 20 male degus injected with red-colored latex. Adrenal arterial branching patterns were classified into Types 1-4, which respectively have 1 to 4 parent arteries that give rise to the adrenal arteries. Based on the combination of the parent arteries, Types 2 and 3 were categorized into subtypes a to c, while Type 4 was categorized into subtypes a and b. On the left side, Type 2 (45%) and Type 3 (45%) were predominant, whereas Type 1 (5%) and Type 4 (5%) were infrequent. On the right side, Type 2 (50%) and Type 3 (45%) were predominant, whereas Type 4 (5%) was infrequent. Type 1 was not present. There were 0 to 4 cranial, 1 to 4 middle and 1 to 4 caudal adrenal arteries, with the total number varying from 2 to 9. The present observation provides knowledge of comparative anatomical features of the degu adrenal arteries, which can serve as an anatomical basis for comparative endocrinological studies. A 7-year-and-8-month-old, male degu (Octodon degus) with anorexia, depression, and labored breathing was found to have a thoracic effusion and enlargement of the right testis on radiographic examination. Despite treatment, the animal died. At necropsy, hepatomegaly, splenomegaly, and multifocal nodules on the intestinal serosa and mesentery were observed. Histologically, the foci were densely cellular invasive neoplasms composed of sheets of round to polygonal cells, with occasional multinucleated giant cells. Immunohistochemically, the neoplastic cells were immunopositive for ionized calcium-binding adapter molecule 1, human leukocyte antigen-DR, and CD204. These findings were consistent with disseminated histiocytic sarcoma.

This article reviews the development, hand-rearing, feeding, housing, and social behavior of common pet rodent species (rats, mice, hamsters, gerbils, guinea pigs, chinchillas, and degus). In addition, common gastrointestinal, respiratory, cardiovascular, dermatologic, musculoskeletal, neurologic, and ophthalmic disorders in pediatric pet rodents are reviewed. We then conducted an aggregated analysis of familiarity preferences across multiple rodent species (adult male and female rats, mice, prairie voles, meadow voles, and female degus) tested with the same protocol. We found a high degree of consistency within species across data sets, supporting the existence of strong, species-typical familiarity preferences in prairie voles and meadow voles, and a lack of familiarity preferences in other species tested. Sociability, or total time spent near conspecifics, was unrelated to selectivity in social preference. Sociality—i.e., life in social groups—has evolved many times in rodents, and there is considerable variation in the nature of these groups. While many species-typical behaviors have been described in field settings, the use of consistent behavioral assays in the laboratory provides key data for comparisons across species. The preference for interaction with familiar or novel individuals is an important dimension of social behavior. Familiarity preference, in particular, may be associated with more closed, less flexible social groups. The dimension from selectivity to gregariousness has been used as a factor in classification of social group types. Laboratory tests of social choice range from brief (10 minutes) to extended (e.g., 3 hours). As familiarity preferences typically need long testing intervals to manifest, we used 3-hour peer partner preference tests to test for the presence of familiarity preferences in same-sex cage-mates and strangers in rats. Most nonhuman animals do not show selective preference for types of music, but researchers have typically employed only Western classical music in such studies. Thus, there has been bias in music choice. Degus (Octodon degus), originally from the mountain areas of Chile, have highly developed vocal communication. Here, we examined music preference of degus using not only Western classical music (music composed by Bach and Stravinsky), but also South American folk music (Chilean and Peruvian). The degus preferred the South American music to the Western classical music but did not show selective preference between the two Western classical music choices. Furthermore, the degus preferred the Chilean to the Peruvian music to some extent. In the second experiment, we examined preference for music vs. silence. Degus overall showed a preference for Chilean music over silence, but preferred silence over Western music. The present results indicate that the previous negative data for musical preference in nonhuman animals may be due to biased music selection (Krause, 2012). Our results suggest the possibility that the soundscape of an environment influences folk music created by native peoples living there and the auditory preference of other resident animals there.

In recent years, degus (Octodon degus), rodents native to South America, have been becoming increasingly popular as pet animals. Data about neoplastic diseases in this species are still sparse and mainly limited to single-case reports. The aim of this study was to present neoplastic and non-neoplastic proliferative changes in 16/100 pet degus examined at the Veterinary Faculty University of Ljubljana from 2010 to 2015 and to describe the clinic-pathological features of these lesions. Twenty different lesions of the integumentary, musculoskeletal, genitourinary and gastrointestinal systems were diagnosed: amongst these were 13 malignant tumors, six benign tumors, and one non-neoplastic lesion. Cutaneous fibrosarcoma was the most common tumor (7/16 degus). It was detected more often in females (6/7 degus) and lesions were located mainly in hind limbs. The gastrointestinal tract was frequently affected, namely with two malignant neoplasms - an intestinal lymphoma and a mesenteric mesothelioma, four benign tumors - two biliary cystadenomas, an oral squamous papilloma and a hepatocellular adenoma, and a single non-neoplastic proliferative lesion. The minor secreted proteinase of B. pumilus 3-19 MprBp classified as the unique bacillary adamalysin-like enzyme of the metzincin clan. The functional role of this metalloproteinase in the bacilli cells is not clear. Analysis of the regulatory region of the mprBp gene showed the presence of potential binding sites to the transcription regulatory factors Spo0A (sporulation) and DegU (biodegradation). The study of mprBp activity in mutant strains of B. subtilis defective in regulatory proteins of the Spo- and Deg-systems showed that the mprBp gene is partially controlled by the Deg-system of signal transduction and independent from the Spo-system.

Introduction: A large variety of pet feeds is commercially available for pet herbivores; these feeds may differ in nutrient composition from the natural diet, and may trigger different feeding behaviours than observed in natural habitats. Here, we surveyed literature on the natural diet and activity budget of rabbits (Oryctolagus cuniculus), guinea pigs (Cavia porcellus), chinchilla (Chinchilla lanigera) and degus (Octodon degus), as well as 260 compound feeds commercially available for these species between 1982 and 2020. The species are classified as herbivores, with available data on natural diets suggesting a crude fibre (CF) level of approximately 250 g/kg dry matter (DM), natural feeding activity at a magnitude of 4-7 h per day, with distinctively shorter feeding when fed on compound pet feeds. Only for a minority of feeds did the first 5 listed ingredients not include a starchy ingredient, or only ingredients not considered suitable for human consumption. The percentage of feeds whose CF level was less than 150 g/kg DM was 22 % for production rabbits, 18 % for pet rabbits, 26 % for guinea pigs, 18 % for chinchillas and 14 % for degus; in other words, a majority of currently marketed products have CF levels in accord with published recommendations, even if that means a discrepancy to natural diets. Screening the producers' feeding instructions suggests they should generally not be followed uncritically. No temporal trends in the CF levels of these diets was evident. There is no consensus whether only products resembling natural diet items should be included in pet feeds, or whether vegetable byproducts not edible by humans should also be included. The observed practice of pet feed composition largely did not follow either concept, questioning the general rationale of pet feed composition. Whole forage like hay is recommended as the main diet component for these species, and 76 % of pet feed products noted that hay should be fed in the feeding instructions.

The wild strain of B. subtilis 168, the protease-deficient strain of B. subtilis BRB14, and the regulatory mutants of B. subtilis 168 degU and B. subtilis 168 ccpA were examined for their ability to form biofilms under stress conditions—with elevated concentrations of divalent metal ions (Ca2+, Mg2+, Mn2+, and Zn2+), as well as in the presence of ethylenediaminetetraacetic acid (EDTA). It was shown that regulatory mutants are more resistant to the addition of divalent metal ions than wild and protease-deficient strains. Abstract A 6-year-old female degu (Octodon degus) in good body condition was presented with a mass at the base of the tail. Following presentation and the owners' approval, the tail of the degu was surgically amputated. The mass was submitted for pathological evaluation following the surgical procedure. Histologically, the mass was comprised of lobules and sheets of polygonal cells displaying moderate amounts of basophilic foamy cytoplasm (physaliferous cells). The pathological results of the tail mass were consistent with chordoma. Approximately 3 months following the tail amputation the patient died. [ABSTRACT FROM AUTHOR]

Alzheimer's disease (AD) is a multifactorial progressive neurodegenerative disease. Despite decades of research, no disease modifying therapy is available and a change of research objectives and/or development of novel research tools may be required. Much AD research has been based on experimental models using animals with a short lifespan that have been extensively genetically manipulated and do not represent the full spectrum of late-onset AD, which make up the majority of cases. The aetiology of AD is heterogeneous and involves multiple factors associated with the late-onset of the disease like disturbances in brain insulin, oxidative stress, neuroinflammation, metabolic syndrome, retinal degeneration and sleep disturbances which are all progressive abnormalities that could account for many molecular, biochemical and histopathological lesions found in brain from patients dying from AD. This review is based on the long-lived rodent Octodon degus (degu) which is a small diurnal rodent native to South America that can spontaneously develop cognitive decline with concomitant phospho-tau, β-amyloid pathology and neuroinflammation in brain. In addition, the degu can also develop several other conditions like type 2 diabetes, macular and retinal degeneration and atherosclerosis, conditions that are often associated with aging and are often comorbid with AD. DegS and DegU make up a two component system belonging to a class of signal transduction systems that play important roles in a broad range of bacterial responses to the environment. However, little study has been done to explore the physiological functions of DegS-DegU in mosquitocidal Lysinibacillus sphaericus . In this study, it was found that deletion of degU or degS-degU inhibited the swarming motility, biofilm formation, sporulation and binary toxin production through regulating the related genes, and phosphorylation was necessary for the functions of DegU. Based on the findings, a regulation network mediated by DegU was delineated. Both DegU-pi and Spo0A-pi positively regulates genes which are linked with the transition from stage Ⅱ to the end of the sporulation process and also influences the production of binary toxins via regulation on sigE . Both DegU-pi and Spo0A-pi negatively regulate abrB/sinR and influence the biofilm formation. DegU-pi can positively regulate the motility via the regulation on sigD. Whether the regulations are directly or indirectly need to be explored. Moreover, Spo0A-pi may indirectly regulate the swarming motility through negatively regulating DegU. The brain arteries derived from 50 adult degu individuals of both sexes were injected with synthetic latex introduced with a syringe into the left ventricle of the heart under constant pressure. After fixation in 5% formalin and brain preparation, it was found that the sources of the brain's supply of blood are vertebral arteries and the basilar artery formed as a result of their anastomosis. The basilar artery gave rise to caudal cerebellar arteries and then divided into two branches which formed the arterial circle of the brain. The internal carotid arteries in degus, except for one case, were heavily reduced and did not play an important role in the blood supply to the brain. The arterial circle of the brain in 48% of the cases was open from the rostral side. Variation was identified in the anatomy and the pattern of the arteries of the base of the brain in the degu which involved an asymmetry of the descent of caudal cerebellar arteries (6.0%), rostral cerebellar arteries (8%) as well as middle cerebral arteries (12%). In 6% of the individuals double middle cerebral arteries were found. In one out of 50 cases there was observed a reduction in the left vertebral artery and the appearance of the internal carotid artery on the same side. In that case the left part of the arterial circle of the brain was supplied with blood by an internal carotid artery, which was present only in that animal.

The K-state in the model bacterium Bacillus subtilis is associated with transformability (competence) as well as with growth arrest and tolerance for antibiotics. Entry into the K-state is determined by the stochastic activation of the transcription factor ComK and occurs in about ~15% of the population in domesticated strains. Although the upstream mechanisms that regulate the K-state have been intensively studied and are well understood, it has remained unexplained why undomesticated isolates of B. subtilis are poorly transformable compared to their domesticated counterparts. We show here that this is because fewer cells enter the K-state, suggesting that a regulatory pathway limiting entry to the K-state is missing in domesticated strains. We find that loss of this limitation is largely due to an inactivating point mutation in the promoter of degQ. The resulting low level of DegQ decreases the concentration of phosphorylated DegU, which leads to the de-repression of the srfA operon and ultimately to the stabilization of ComK. As a result, more cells reach the threshold concentration of ComK needed to activate the auto-regulatory loop at the comK promoter. In addition, we demonstrate that the activation of srfA transcription in undomesticated strains is transient, turning off abruptly as cells enter the stationary phase. Thus, the K-state and transformability are more transient and less frequently expressed in the undomesticated strains. This limitation is more extreme than appreciated from studies of domesticated strains. Selection has apparently limited both the frequency and the duration of the bistably expressed K-state in wild strains, likely because of the high cost of growth arrest associated with the K-state. Future modeling of K-state regulation and of the fitness advantages and costs of the K-state must take these features into account.

The tongue of an adult degu was examined by light and scanning electron microscopy. It consists of an apex, corpus, and radix and contains a lingual prominence. The aim of this study was to describe the course of muscle fascicles of the proper lingual muscle, the presence and nature of the lingual salivary glands, and particularly the appearance and distribution of the lingual papillae. Three major types of papillae have been observed: filiform, conical, and vallate. The dorsal surface of the lingual apex extends in caudally bent filiform papillae with two spines. The lingual corpus bears long filiform papillae with a single tip. The lingual radix contains crown-like papillae in the region of the prominence and conical papillae in the remaining areas. Two oval vallate papillae were discovered caudally on the lingual radix. This first description of the lingual structures in a degu could be used for comparative studies or as basic data for differentiation of lingual morphology in this species. [ABSTRACT FROM AUTHOR]

Some animals learn to fear a situation after observing another individual come to harm, and this learning is influenced by the animals’ social relationship and history. An important but sometimes overlooked factor in studies of observational fear learning is that social context not only affects observers, but may also influence the behavior and communications expressed by those being observed. Here we sought to investigate whether observational fear learning in the degu ( Octodon degus ) is affected by social familiarity, and the degree to which vocal expressions of alarm or distress contribute. ‘Demonstrator’ degus underwent contextual fear conditioning in the presence of a cagemate or stranger observer. Among the 15 male pairs, observers of familiar demonstrators exhibited higher freezing rates than observers of strangers when returned to the conditioning environment one day later. Observer freezing during testing was, however, also related to the proportion of short- versus long- inter-call-intervals (ICIs) in vocalizations recorded during prior conditioning. In a regression model that included both social relationship and ICI patterns, only the latter was significant. Further investigation of vocalizations, including use of a novel, directed k-means clustering approach, suggested that temporal structure rather than tonal variations may have been responsible for communicating danger. These data offer insight into how different expressions of distress or fear may impact an observer, adding to the complexity of social context effects in studies of empathy and social cognition. The cranial mesenteric artery exhibits a species-specific ramification pattern that adapts to the morphology of the intestinal tract. The degu is a strictly herbivorous rodent with a well-developed large intestine with a spiral loop in the ascending colon; therefore, the cranial mesenteric artery likely demonstrates a degu-specific ramification pattern. Thus, we traced the cranial mesenteric artery to establish the detailed ramification pattern of the branches. Eighteen male degus were injected with 0.3–0.8 ml of a latex mixture and water at a 1:1 ratio in conjunction with red acrylic paint coloring using a catheter inserted into the thoracic aorta. The cranial mesenteric artery was traced using a surgical microscope and photographed using a digital camera. The arteries emerging from the cranial mesenteric artery exhibited frequent variations in number, distribution area, anastomosis pattern, and branching order. In the most frequent cases (22%), the cranial mesenteric artery sequentially gave rise to caudal pancreaticoduodenal, middle colic, right colic, jejunal, and ileocolic arteries. The right and middle colic arteries exhibited four different ramification patterns. In the most common cases (67%), the middle and right colic arteries emerged independently from the cranial mesenteric artery. The former was distributed to the transverse and descending colon, whereas the latter sent branches to the spiral loop of the ascending colon. The complex ramification pattern of the right colic artery in the degu may be an adaptation to the characteristic running pattern of the ascending colon. Thus, we present the first comprehensive report of the arterial branching pattern of the cranial mesenteric artery in the degu. An adult, 2-year-old, 173 g, intact male degu ( Octodon degus ) was presented to the authors’ clinic with a 2-week history of reduced food intake, weight loss, epiphora, and dyspnea. Physical examination revealed lethargy, cachexia, bilateral serous ocular discharge, dehydration, inspiratory dyspnea, and severe dental disease. Diagnostic investigation showed extensive apical elongation of premolars causing partial nasal cavity obstruction by masses of increased opacity, and loss of conchal detail. The animal was euthanized due to general poor condition and the severity of disease. Postmortem micro–computed tomography showed marked reserve crown elongation of all premolars and molars. All cheek teeth were abnormally curved and had widened interdental spaces. The apex of the maxillary left premolar nearly penetrated the left nasal bone. Both maxillary premolars penetrated into the nasal cavity and caused complete left and partial right nasal cavity obstruction. Apices of all premolars were dysplastic. [ABSTRACT FROM AUTHOR]

Chromatin organization and the management of transcription and splicing are fundamental to the correct functioning of every cell but, in particular, for highly active cells such as photoreceptors, the sensory neurons of the retina. Rod photoreceptor cells of nocturnal animals have recently been shown to have an inverted chromatin architecture compared with rod photoreceptor cells of diurnal animals. The heterochromatin is concentrated in the center of the nucleus, whereas the genetically active euchromatin is positioned close to the nuclear membrane. This unique chromatin architecture suggests that the transcription and splicing machinery is also subject to specific adaptations in these cells. Recently, we described the protein Simiate, which is enriched in nuclear speckles and seems to be involved in transcription and splicing processes. Here, we examine the distribution of Simiate and nuclear speckles in neurons of mouse retinae. In retinal neurons of the inner nuclear and ganglion cell layer, Simiate is concentrated in a clustered pattern in the nuclear interior, whereas in rod and cone photoreceptor cells, Simiate is present at the nuclear periphery. Further staining with markers for the transcription and splicing machinery has confirmed the localization of nuclear speckle components at the periphery. Comparing the distribution of nuclear speckles in retinae of the nocturnal mouse with the diurnal degu, we found no differences in the arrangement of the transcription and splicing machinery in their photoreceptor cells, thus suggesting that the organization of these machineries is not related to the animal's lifestyle but rather represents a general characteristic of photoreceptor organization and function. During lactation, both the nutritional and energetic requirements of suckling change gradually. These changes normally are accompanied by modifications in chemical composition of the milk. We investigated the temporal course of milk composition during lactation in a precocial caviomorph rodent, the "degu" (Octodon degus) under laboratory condition. Female degus were kept in laboratory during gestation and lactation and fed with commercial food pellets. Milk was collected at three stages of lactation: early (days 5-8, n = 12), middle (days 15-21, n = 7) and late (days 26-40, n = 6), and analyzed for protein, carbohydrates, lipids, ash, total solids and energy. On average, carbohydrates decreased from 3.1 ± 0.3 % (early) to 1.1 ± 0.3 % (late) during lactation; lipids, protein, ash, total solids and energy remained about the same. Lipids, the main component of the milk, were 17.3 % and protein remained near 4.4 %. Over lactation, total energy concentration of milk remained near 4.0 kJ mL-1. La leche fue colectada en tres estados de lactancia: temprana (días 5-8, n = 12), media (días 15-21, n = 7) y tardía (días 26-40, n = 6), y analizadas para proteínas, hidratos de carbono, lípidos, ceniza, sólidos totales y energía. Understanding the nature of faunal assembly and community structure remains central to ecology. Research in North American deserts and some tropical forests provides evidence of energetic compensation and zero-sum dynamics, suggesting that species in some natural assemblages may be replaced with limited impact on ecosystem function. Experimental removal of a dominant small mammal (degu, Octodon degus) from replicate plots in semiarid coastal thorn-scrub habitat in north-central Chile revealed no evidence for energetic or functional compensation; energy consumption remained significantly lower on degu exclusions relative to control plots after 17 years of exclusion. This occurred in spite of the fact that the geographic species pools for South American sites generally are similar in size to those of most North American sites (mean and median number of species, 16.3 and 21.5 vs. 21.0 and 20, respectively). A macroecological assessment of energetically equivalent species at 394 arid sites in North America, the Gobi Desert, and South America indicated that the number of potentially equivalent species was lower than (Gobi) or similar to (South America) that found in North America, but when segregated by trophic groups, these faunas differed markedly. North American sites included large numbers of granivorous species whereas South American sites were dominated by omnivores. The more general trophic strategy in the latter sites would be expected to facilitate compensatory responses within local faunas, suggesting either that our site is anomalous or that other factors are governing local dynamics. Further research is needed to understand the generality of compensatory dynamics within natural systems, as this mechanism has direct relevance to discussions on ecological resilience in the face of ongoing environmental change. Background: Two-component signal transduction pathways allow bacteria to sense and respond to the environment. Typically such pathways comprise a sensor histidine kinase and a response regulator. Phosphorylation of the response regulator commonly results in its activation, allowing the protein to bind to target promoter elements to regulate transcription. Several mechanisms are used to prevent inappropriate phosphorylation of the response regulator, thereby ensuring a specific response. In Bacillus subtilis, the DegS-DegU two-component system controls transcription of target genes in a manner dependent on the level of the phosphorylated response regulator, DegU. Previous work has tentatively indicated that DegU, and DegU H12L, a DegU variant which displays enhanced stability of the phosphoryl moiety, can be phosphorylated in the absence of the kinase, DegS. Results: The data presented here reveal that DegU H12L requires aspartic acid 56 (D56), the identified DegU phosphorylation site, for its activity. By indirectly measuring the level of DegU ~ Pin the cell by assessment of several well recognised DegU regulated processes it was shown that DegU H12L retains its activity in the absence of DegS, and that mutation of D56 produced an inactive protein. Further experiments designed to raise the level of acetyl phosphate within the cell suggest that DegU can be phosphorylated by acetyl phosphate in the absence of degS. Additionally, the phenotypic and biochemical experiments presented indicate that DegU H12L can reliably mimic high levels of phosphorylated DegU. Conclusions: The ability of acetyl phosphate to modify DegU, and indeed DegU H12L, reveal an additional layer of regulation for DegU phosphorylation that will be relevant when the level of DegS is low or in the absence of degS. Given the number of processes that DegU can activate or inhibit, extensive regulation at a number of levels is required to ensure that the system is not inappropriately stimulated. DegS has both kinase and phosphatase activity and our findings demonstrate that the phosphatase activity of DegS is essential to control the level of DegU phosphate. The brain arteries derived from 50 adult degu individuals of both sexes were injected with synthetic latex introduced with a syringe into the left ventricle of the heart under constant pressure. After fixation in 5% formalin and brain preparation, it was found that the sources of the brain's supply of blood are vertebral arteries and the basilar artery formed as a result of their anastomosis. The basilar artery gave rise to caudal cerebellar arteries and then divided into two branches which formed the arterial circle of the brain. The internal carotid arteries in degus, except for one case, were heavily reduced and did not play an important role in the blood supply to the brain. The arterial circle of the brain in 48% of the cases was open from the rostral side. Variation was identified in the anatomy and the pattern of the arteries of the base of the brain in the degu which involved an asymmetry of the descent of caudal cerebellar arteries (6.0%), rostral cerebellar arteries (8%) as well as middle cerebral arteries (12%). In 6% of the individuals double middle cerebral arteries were found. In one out of 50 cases there was observed a reduction in the left vertebral artery and the appearance of the internal carotid artery on the same side. In that case the left part of the arterial circle of the brain was supplied with blood by an internal carotid artery, which was present only in that animal. [ABSTRACT FROM AUTHOR]
