Despite extensive sorting, packaging waste often contains a mixture of different materials that make high-quality recycling difficult, especially in the case of flexible packaging. This is partly due to the widespread use of multi-layer laminates and packaging consisting of different inseparably combined materials. To improve the post-consumer recyclate quality and develop optimised recycling processes, it is important to generate a comprehensive understanding of the composition of the sorted packaging waste streams. Therefore, in this study, polyolefin sorting fractions for flexible packaging waste from three European countries are analysed in detail. By selective extraction of the different plastics, their mass fractions in the waste streams are determined. This shows that the PE-rich sorting fractions for flexible packaging are made up of 85–90% of PE, but also contain a certain proportion of foreign materials. A detailed analysis of the layer structures of various types of packaging also provides information on the prevalence of multi-layer packaging and the polymer and non-polymer materials used therein. This shows that particularly in food packaging, with 63–84% of multi-layer and 50–70% of multi-material packaging, a high proportion of foreign materials is used and introduced into the sorting fractions. These insights provide a valuable contribution to the development of recyclable packaging, potential sorting streams and recycling processes, especially with regard to the challenges of the closed-loop recycling of food packaging.

Abstract Aging is the main risk factor for Alzheimer’s disease (AD) and other neurodegenerative pathologies, but the molecular and cellular changes underlying pathological aging of the nervous system are poorly understood. AD pathology seems to correlate with the appearance of cells that become senescent due to the progressive accumulation of cellular insults causing DNA damage. Senescence has also been shown to reduce the autophagic flux, a mechanism involved in clearing damaged proteins from the cell, and such impairment has been linked to AD pathogenesis. In this study, we investigated the role of cellular senescence on AD pathology by crossing a mouse model of AD-like amyloid-β (Aβ) pathology (5xFAD) with a mouse model of senescence that is genetically deficient for the RNA component of the telomerase (Terc−/−). We studied changes in amyloid pathology, neurodegeneration, and the autophagy process in brain tissue samples and primary cultures derived from these mice by complementary biochemical and immunostaining approaches. Postmortem human brain samples were also processed to evaluate autophagy defects in AD patients. Our results show that accelerated senescence produces an early accumulation of intraneuronal Aβ in the subiculum and cortical layer V of 5xFAD mice. This correlates with a reduction in amyloid plaques and Aβ levels in connecting brain regions at a later disease stage. Neuronal loss was specifically observed in brain regions presenting intraneuronal Aβ and was linked to telomere attrition. Our results indicate that senescence affects intraneuronal Aβ accumulation by impairing autophagy function and that early autophagy defects can be found in the brains of AD patients. Together, these findings demonstrate the instrumental role of senescence in intraneuronal Aβ accumulation, which represents a key event in AD pathophysiology, and emphasize the correlation between the initial stages of amyloid pathology and defects in the autophagy flux.

Abstract Background Recently, periostin (POSTN), a gene encoding a protein with similarity to the fasciclin family and involved in cell survival and angiogenesis, has emerged as a promising marker for tumor progression in various types of human cancers. There is some controversy regarding both POSTN expression levels and the nature of periostin-producing cells within tumors. In this study, we used quantitative RT-PCR to assess periostin gene expression in normal tissues, primary cell cultures, tumor tissues and tumor cell lines. Results Periostin expression levels are highly variable in both normal tissues and tumors and strong POSTN overexpression is mostly detected in tumors from pancreas and liver. POSTN is not expressed in blood cancers. In melanoma samples, average periostin expression is not increased in primary tumors whereas POSTN overexpression was detected in about 60% of melanoma metastatic tumors in the liver or lymph nodes. Identification of the cellular source of periostin production in melanoma metastases -cancer cells or stroma- was assessed by comparing periostin expression in 23 newly-established melanoma cell lines and matched tumors. In contrast to the reduction by more than 99% of COL6A3 stromal marker mRNA in all cell lines, significant POSTN transcription was maintained in some melanoma cell lines, suggesting that both stromal cells and melanoma cells express periostin. The high level of periostin expression in primary cultures of skin fibroblasts suggests that fibroblasts may contribute for a large part to periostin production in melanoma-associated stroma. On the other hand, periostin expression in melanoma cells is probably acquired during the tumorigenic process as 1) normal melanocytes do not express POSTN and 2) melanoma cells from distinct metastases of the same patient were associated with very different levels of periostin expression. Conclusion Our comparative analysis suggests that, although periostin overexpression is clearly detected in some cancers, it is not a general feature of tumors. In melanoma, our study identifies both stromal and melanoma cells as sources of periostin production and correlates POSTN expression levels with increased primary tumor thickness and metastatic process development.

A spatial analysis of the mudflat community of Bourgneuf Bay, France before and after experimental oyster reef removal reveals the role of oyster reefs in influencing microphytobenthos via predator facilitation.

Abstract The vast majority of adult cancer cells achieve cellular immortality by activating a telomere maintenance mechanism (TMM). While this is mostly achieved by the de-silencing of hTERT telomerase gene expression, an alternative homologous recombination-based and telomerase-independent mechanism, known as ALT (Alternative Lengthening of Telomeres), is frequently activated in a subset of tumors, including paediatric cancers. Being absent from normal cells, the ALT mechanism offers interesting perspectives for new targeted cancer therapies. To date, however, the development of better translationally applicable tools for ALT detection in tumor sections is still needed. Here, using a newly derived ALT-positive cancer cell mouse xenograft model, we extensively examined how the previously known ALT markers could be used as reliable tools for ALT diagnosis in tumor sections. We found that, together with the detection of ultra-bright telomeric signals (UBS), an ALT hallmark, native telomeric FISH, that detects single-stranded C-rich telomeric DNA, provides a very sensitive and robust tool for ALT diagnosis in tissues. We applied these assays to paediatric tumor samples and readily identified three ALT-positive tumors for which the TMM was confirmed by the gold-standard C-circle amplification assay. Although the latter offers a robust assay for ALT detection in the context of research laboratories, it is more difficult to set up in histopathological laboratories and could therefore be conveniently replaced by the combination of UBS detection and native telomeric FISH.

Este trabalho trata de uma sequência de ensino por investigação sobre o sistema respiratório utilizando a metodologia Social and Emotional Learning-SEL para ensino de Ciências, com vistas a apresentar uma proposta de ensino que propicie desenvolvimento de habilidades cognitivas e socioemocionais. O processo de ensino aprendizagem foi embasado na teoria histórico-cultural de Vygotsky e no campo didático metodológico, o ensino por investigação. Trata-se de uma pesquisa qualitativa de estudo de caso, que contou com a participação de 11 professores com titulação de mestres em Ensino de Ciências. Os resultados apresentaram o desenvolvimento de habilidades socioemocionais, em especial, o autogerenciamento e a autogestão. A atividade proposta, mostrou-se como uma rica ferramenta para professores ampliarem seus objetivos de aprendizagem com os alunos.

Abstract PARN, poly(A)‐specific ribonuclease, regulates the turnover of mRNAs and the maturation and stabilization of the hTR RNA component of telomerase. Biallelic PARN mutations were associated with Høyeraal–Hreidarsson (HH) syndrome, a rare telomere biology disorder that, because of its severity, is likely not exclusively due to hTR down‐regulation. Whether PARN deficiency was affecting the expression of telomere‐related genes was still unclear. Using cells from two unrelated HH individuals carrying novel PARN mutations and a human PARN knock‐out (KO) cell line with inducible PARN complementation, we found that PARN deficiency affects both telomere length and stability and down‐regulates the expression of TRF1, TRF2, TPP1, RAP1, and POT1 shelterin transcripts. Down‐regulation of dyskerin‐encoding DKC1 mRNA was also observed and found to result from p53 activation in PARN‐deficient cells. We further showed that PARN deficiency compromises ribosomal RNA biogenesis in patients' fibroblasts and cells from heterozygous Parn KO mice. Homozygous Parn KO however resulted in early embryonic lethality that was not overcome by p53 KO. Our results refine our knowledge on the pleiotropic cellular consequences of PARN deficiency.

The marine diatom Haslea ostrearia is known for its ability to produce marennine, a blue-green water-soluble pigment, which is responsible for the oyster greening phenomenon in the French Atlantic Coast. Marennine can be considered a bioactive extracellular compound (BEC) as it exhibits activities against detrimental pathogens in bivalve aquaculture. However, marennine could also be toxic to animals. The aim of this experimental study was to assess the effect of marennine produced by H. ostrearia upon three commercially important bivalve species, the American oyster Crassostrea virginica, the blue mussel Mytilus edulis and the giant scallop Placopecten magellanicus. Effect of this BEC was also assessed on different ontogenic stages of scallops. The animals were exposed to 3 mg L−1 of marennine and physiological response associated with clearance rates (CR), oxygen consumption and condition index (CI) were measured after 16 h of exposure (day 0), and after one subsequent week of recovery (day 7). In the short term, CR and oxygen consumption on mussels and oysters were significantly altered by marennine but not for scallops. CR recovery was observed in oysters after seven days, but no depuration was observed in mussels. Nevertheless, oxygen consumption in both mussels and oysters recovered on day 7. CI in the marennine-exposed group for both adult oysters and scallops was not significantly different, yet CI of mussels was lowered by 18.2 % compared to control. Scallop adults seemed largely resistant to marennine, but juveniles were more susceptible with a reduced CR and oxygen consumption by 71.9 % and 24.5 %, respectively, and no depuration activity observed until day 7. Additionally, CI in marennine-exposed juvenile scallops was 27 % lower than that of control. This study demonstrates species- and age-specific effect of marennine on bivalves, thus highlighting the importance of further assessment on the utilization of this pigment as a natural product decreasing pathogenicity of bacteria in shellfish aquaculture systems.

Satellite remote sensing (RS) is routinely used for the large-scale monitoring of microphytobenthos (MPB) biomass in intertidal mudflats and has greatly improved our knowledge of MPB spatio-temporal variability and its potential drivers. Processes operating on smaller scales however, such as the impact of benthic macrofauna on MPB development, to date remain underinvestigated. In this study, we analysed the influence of wild Crassostrea gigas oyster reefs on MPB biofilm development using multispectral RS. A 30-year time series (1985–2015) combining high-resolution (30 m) Landsat and SPOT data was built in order to explore the relationship between C. gigas reefs and MPB spatial distribution and seasonal dynamics, using the normalized difference vegetation index (NDVI). Emphasis was placed on the analysis of a before–after control-impact (BACI) experiment designed to assess the effect of oyster killing on the surrounding MPB biofilms. Our RS data reveal that the presence of oyster reefs positively affects MPB biofilm development. Analysis of the historical time series first showed the presence of persistent, highly concentrated MPB patches around oyster reefs. This observation was supported by the BACI experiment which showed that killing the oysters (while leaving the physical reef structure, i.e. oyster shells, intact) negatively affected both MPB biofilm biomass and spatial stability around the reef. As such, our results are consistent with the hypothesis of nutrient input as an explanation for the MPB growth-promoting effect of oysters, whereby organic and inorganic matter released through oyster excretion and biodeposition stimulates MPB biomass accumulation. MPB also showed marked seasonal variations in biomass and patch shape, size and degree of aggregation around the oyster reefs. Seasonal variations in biomass, with higher NDVI during spring and autumn, were consistent with those observed on broader scales in other European mudflats. Our study provides the first multi-sensor RS satellite evidence of the promoting and structuring effect of oyster reefs on MPB biofilms.

Neodymium-Iron-Boron (NdFeB) based permanent magnets are indispensable in today’s technology-driven society. Moreover, their use is likely to increase since they are key in clean energy applications such as wind turbines, hybrid/electric vehicles, and electric bikes. They contain critical raw materials as rare earth elements are used. Indeed, permanent magnets are considered strategic materials by the EU, and their recycling represents a potential secondary supply to decrease the import dependence. The VALOMAG project is developing a technical solution to recover rare earth (RE) based permanent magnets by dismantling end-of-life (EoL) products such as computer hard disc drives, electric motors, and generators from electric vehicles and wind turbines. It also assesses two short loop recycling technologies: Hydrogen Decrepitation (HD) or Hydrogenation–Disproportionation–Desorption–Recombination (HDDR) and strip-casting for high and medium quality magnet wastes; and hydrometallurgical processes for EoL low-quality magnets. Moreover, Life Cycle Assessment (LCA) and Process Integration with a Flowsheet simulation tool will integrate the whole recycling value chain (collection, dismantling, physical and chemical treatment options, and re-manufacturing) and assess the environmental impact and processes efficiency. A market study on the types and expected future quantities for the scrap magnets and the characterisation of the EoL magnets from hard disc drives (HDD) will be presented as preliminary results. Pre-treatment and sorting of 2.5 tons of NdFeB magnets scraps were carried out, and the two short loop recycling routes and the hydrometallurgical route are under investigation at the lab and pilot scale. The results will be used to develop a process integration and to assess the three routes through LCA.

Summary: Unlimited replicative potential is one of the hallmarks of cancer cells. In melanoma, hTERT (telomerase reverse transcriptase) is frequently overexpressed because of activating mutations in its promoter, suggesting that telomerase is necessary for melanoma development. We observed, however, that a subset of melanoma metastases and derived cell lines had no telomere maintenance mechanism. Early passages of the latter displayed long telomeres that progressively shortened and fused before cell death. We propose that, during melanoma formation, oncogenic mutations occur in precursor melanocytes with long telomeres, providing cells with sufficient replicative potential, thereby bypassing the need to re-activate telomerase. Our data further support the emerging idea that long telomeres promote melanoma formation. These observations are important when considering anticancer therapies targeting telomerase. : Viceconte et al. report that metastatic cutaneous melanoma cells display long telomeres and do not always have an active telomere maintenance mechanism, showing that the ability to maintain telomeres is not an absolute requirement for metastasis formation. Some tumors acquire replicative immortality by activating an ALTernative telomerase-independent telomere maintenance mechanism. ALT offers interesting therapeutic perspectives but lacks any known specific targets. We discovered a crucial role for TSPYL5 (Testis-Specific Y-encoded-Like Protein 5) in keeping ALT cell viability by protecting POT1 (Protection Of Telomeres 1) from proteasomal degradation.

Marennine, the blue-green pigment excreted by the pennate diatom Haslea ostrearia has potential for applications as a natural antimicrobial agent in bivalve aquaculture. However, utilization of this pigment should be assessed further prior to its application in bivalve hatcheries. The present study aims to investigate the effect of the culture supernatant of H. ostrearia containing marennine (Blue Water, BW) on physiological rates of adult mussel Mytilus edulis. The hypothesis tested in this study was that BW could alter clearance and respiration rates of adult M. edulis as indicative physiological traits. Furthermore, the effect of BW on veligers and pediveligers larvae was assessed in physiological experiments to determine whether the fixation of marennine by the larvae can influence larviphagy in this species. Our results showed that BW significantly lowered CR by 43% and reduced the oxygen consumption by 30% in adult mussels, as compared to control. In larviphagy experiments, mussels ingested veliger and pediveliger larvae without discrimination between greened or non-greened larvae. Moreover, fixation of BW in bivalve gills at 3 mg L−1 affected the physiological rate but not intensity of larviphagy in adult M. edulis. This study thus provides an insight into the necessity to determine the potential effect of marennine on adult bivalve for the future utilization of this pigment in bivalve hatcheries.
