Abstract

The chemical burden on the environment and human population is increasing. Consequently, regulatory risk assessment must keep pace to manage, reduce, and prevent adverse impacts on human and environmental health associated with hazardous chemicals. Surveillance of chemicals of known, emerging, or potential future concern, entering the environment-food-human continuum is needed to document the reality of risks posed by chemicals on ecosystem and human health from a one health perspective, feed into early warning systems and support public policies for exposure mitigation provisions and safe and sustainable by design strategies. The use of less-conventional sampling strategies and integration of full-scan, high-resolution mass spectrometry and effect-directed analysis in environmental and human monitoring programmes have the potential to enhance the screening and identification of a wider range of chemicals of known, emerging or potential future concern. Here, we outline the key needs and recommendations identified within the European Partnership for Assessment of Risks from Chemicals (PARC) project for leveraging these innovative methodologies to support the development of next-generation chemical risk assessment.

Abstract

A reconstructed human epidermal model (RHE) colonized with human microbiota and sebum was developed to reproduce the complexity of the skin ecosystem in vitro. The RHE model was exposed to simulated solar radiation (SSR) with or without SPF50+ sunscreen (with UVB, UVA, long‐UVA, and visible light protection). Structural identification of discriminant metabolites was acquired by nuclear magnetic resonance and metabolomic fingerprints were identified using reverse phase‐ultra high‐performance liquid chromatography–high resolution mass spectrometry, followed by pathway enrichment analysis. Over 50 metabolites were significantly altered by SSR (p < 0.05, log2 values), showing high skin oxidative stress (glutathione and purine pathways, urea cycle) and altered skin microbiota (branched‐chain amino acid cycle and tryptophan pathway). 16S and internal transcribed spacer rRNA sequencing showed the relative abundance of various bacteria and fungi altered by SSR. This study identified highly accurate metabolomic fingerprints and metagenomic modifications of sun‐exposed skin to help elucidate the interactions between the skin and its microbiota. Application of SPF50+ sunscreen protected the skin ecosystem model from the deleterious effects of SSR and preserved the physiological interactions within the skin ecosystem. Abstract

Obesity, which is a worldwide public health issue, is associated with chronic inflammation that contribute to long-term complications, including insulin resistance, type 2 diabetes and non-alcoholic fatty liver disease. We hypothesized that obesity may also influence the sensitivity to food contaminants, such as fumonisin B1 (FB1), a mycotoxin produced mainly by the Fusarium verticillioides. FB1, a common contaminant of corn, is the most abundant and best characterized member of the fumonisins family. We investigated whether diet-induced obesity could modulate the sensitivity to oral FB1 exposure, with emphasis on gut health and hepatotoxicity. Thus, metabolic effects of FB1 were assessed in obese and non-obese male C57BL/6J mice. Mice received a high-fat diet (HFD) or normal chow diet (CHOW) for 15 weeks. Then, during the last three weeks, mice were exposed to these diets in combination or not with FB1 (10 mg/kg body weight/day) through drinking water. As expected, HFD feeding induced significant body weight gain, increased fasting glycemia, and hepatic steatosis. Combined exposure to HFD and FB1 resulted in body weight loss and a decrease in fasting blood glucose level. This co-exposition also induces gut dysbiosis, an increase in plasma FB1 level, a decrease in liver weight and hepatic steatosis. Moreover, plasma transaminase levels were significantly increased and associated with liver inflammation in HFD/FB1-treated mice. Liver gene expression analysis revealed that the combined exposure to HFD and FB1 was associated with reduced expression of genes involved in lipogenesis and increased expression of immune response and cell cycle-associated genes. These results suggest that, in the context of obesity, FB1 exposure promotes gut dysbiosis and severe liver inflammation. To our knowledge, this study provides the first example of obesity-induced hepatitis in response to a food contaminant.

Abstract

Humans are exposed to a mixture of pesticides through diet as well as through the environment. We conducted a suspect-screening based study to describe the probability of (concomitant) exposure to a set of pesticide profiles in five European countries (Latvia, Hungary, Czech Republic, Spain and the Netherlands). We explored whether living in an agricultural area (compared to living in a peri-urban area), being a a child (compared to being an adult), and the season in which the urine sample was collected had an impact on the probability of detection of pesticides (-metabolites). In total 2088 urine samples were collected from 1050 participants (525 parent-child pairs) and analyzed through harmonized suspect screening by five different laboratories. Fourty pesticide biomarkers (either pesticide metabolites or the parent pesticides as such) relating to 29 pesticides were identified at high levels of confidence in samples across all study sites. Most frequently detected were biomarkers related to the parent pesticides acetamiprid and chlorpropham. Other biomarkers with high detection rates in at least four countries related to the parent pesticides boscalid, fludioxonil, pirimiphos-methyl, pyrimethanil, clothianidin, fluazifop and propamocarb. In 84% of the samples at least two different pesticides were detected. The median number of detected pesticides in the urine samples was 3, and the maximum was 13 pesticides detected in a single sample. The most frequently co-occurring substances were acetamiprid with chlorpropham (in 62 urine samples), and acetamiprid with tebuconazole (30 samples). Some variation in the probability of detection of pesticides (-metabolites) was observed with living in an agricultural area or season of urine sampling, though no consistent patterns were observed. Abstract

Background and AimsThe constitutive androstane receptor (CAR) is a nuclear receptor able to recognize a large panel of xenobiotics leading to the modulation of the expression of its target genes involved in xenobiotic detoxication and energy metabolism. While CAR hepatic activity is thought to be higher in women than in men, its response to an acute pharmacological activation has never been investigated in both sexes.MethodsHepatic transcriptome, plasma and hepatic metabolome, have been analyzed inCar+/+andCar-/-male and female mice treated either with the CAR-specific agonist, 1,4-bis[2-(3,5-dichloropyridyloxy)]benzene (TCPOBOP), or with vehicle.ResultsWhile 90% of TCPOBOP-sensitive genes were modulated in a sex- independent way, the remaining 10% were almost exclusively impacted in female liver specifically. These female-specific CAR-sensitive genes were mainly involved in xenobiotic metabolism, inflammation and extracellular matrix organization. CAR activation also induced higher hepatic oxidative stress and hepatocyte cytolysis in females than in males. Data mining on human data confirmed that CAR activation may be involved in sexually-dimorphic drug-induced liver injury. Hepatic expression of flavin monooxygenase 3(Fmo3)was almost abolished and associated with a decrease of hepatic trimethylamine-N-oxide (TMAO) concentration in TCPOBOP-treated females. In line with a possible role in the control of TMAO homeostasis, CAR activation decreased platelet hyperresponsiveness in female mice supplemented with dietary choline.ConclusionsOur results demonstrate that more than 10% of CAR-sensitive genes are sex-specific and influence hepatic and systemic response such as platelet aggregation. Also, CAR activation may be an important mechanism of sexually- dimorphic drug-induced liver injury.

Abstract

International audience; Introduction Omics approaches including metabolomics generate huge datasets requiring powerful processing tools. In the field of non-targeted mass spectrometry metabolomics, HRMS data mining has been the subject of much work, with the development of powerful interactive tools, including Workflow4Metabolomics (W4M). One of the main bottlenecks of current workflows lies in the complexity of the interpretation of results, in particular a lack of reliable MS/MS spectra reference databases to be used for confident identification (level 1 and 2 according to the metabolomics standard initiative). Creating reliable and well curated DB remains a laborious and challenging task, but necessary to ensure the quality of the reference spectra. In this work, we present a tool inspired by manual curation allowing the export of MS/MS data from targeted and data dependent (DDA) fragmentation methods to laboratory spectral databases, named “MS2Snoop”. Method Already integrated in W4M pipelines, MS2Snoop works with R and uses as starting point the peak list extracted from LC-MS and MS/MS analyses with XCMS 3.0 using W4M. Then, lists of precursors and fragment ions are processed with MSPurity using W4M, to assign fragmentation spectra to XCMS features, and to filter data on the purity of precursor ions. From these data, MS2Snoop has been developed to allow: • Retrieval of precursor and fragment ions data (m/z, RT, scan-by-scan intensities). • Matching of precursor ions with a list of expected compounds. • Establishing relationships between a precursor and its potential fragment ions via grouping, to obtain a raw spectrum. • Selecting a reference ion in the spectrum (precursor ion if still present, otherwise the most abundant ion). • Determining correlation between the reference ion and the other ions of the fragmentation spectrum using all scans. • Constructing a curated spectrum by eliminating ions with a correlation value lower than a threshold defined by the user. Results MS2Snoop was assessed on targeted MSMS analyses for about 30 compounds (pesticides and some of their metabolites) by performing the steps presented above. To validate these spectra, a manual curation was performed on the same data to compare the m/z and relative abundances of raw and MS2snoop treated spectra. The majority of the automatically extracted spectra using MS2Snoop showed similar results as spectra curated by an expert (fragment ions list, relative abundances), showing that MS2Snoop deconvolution process was efficient to provide automatically cleaned MS/MS spectra without interfering ions on all tested compounds. Conclusion MS2Snoop already allows extracting and cleaning MS/MS datasets using common data processing steps for metabolomics studies as a starting point, and then combined with inputs of a list of expected compounds and MS/MS raw data deconvoluted by MSPurity. Developed and validated with targeted MS/MS spectra, MS2Snoop is compatible with DDA spectra in the limit of the number of MS/MS scans acquired. This package will allow high throughput extraction of MS/MS spectra in order to feed the PeakForest project. Preliminary results obtained with MS2Snoop allowed considering it as a valuable tool for confident annotation of metabolites on chromatographic methods coupled to non-targeted mass spectrometry. A beta version is already publicly available on W4M.

Abstract

This deliverable describes the results of the joint pesticide survey ’SPECIMEn’ in five partner countries: Czech Republic, Hungary, Latvia, Spain and the Netherlands. It also contains a report on the implementation of a study conducted in Switzerland, which has a different design, but data was harmonised as much as possible. In all participating partner countries, urine samples have been collected in two seasons, i.e., winter 2019/2020 and summer 2020. Per country at least 100 locations have been included, of which about half are located in so-called hotspot areas. On the collected urine samples, a harmonised suspect screening method was applied to detect multiple pesticide-related markers. The deliverable describes the results obtained with this analysis.

Abstract

Within the Human Biomonitoring for Europe initiative (HBM4EU), a study to determine new biomarkers of exposure to pesticides and to assess exposure patterns was conducted. Human urine samples (N = 2,088) were collected from five European regions in two different seasons. The objective of the study was to identify pesticides and their metabolites in collected urine samples with a harmonized suspect screening approach based on liquid chromatography coupled to high resolution mass spectrometry (LC-HRMS) applied in five laboratories. A combined data processing workflow included comprehensive data reduction, correction of mass error and retention time (RT) drifts, isotopic pattern analysis, adduct and elemental composition annotation, finalized by a mining of the elemental compositions for possible annotations of pesticide metabolites. The obtained tentative annotations (n = 498) were used for acquiring representative data-dependent tandem mass spectra (MS2) and verified by spectral comparison to reference spectra generated from commercially available reference standards or produced through human liver S9 in vitro incubation experiments. 14 parent pesticides and 71 metabolites (including 16 glucuronide and 11 sulfate conjugates) were detected. Collectively these related to 46 unique pesticides. For the remaining tentative annotations either (i) no data-dependent MS2 spectra could be acquired, (ii) the spectral purity was too low for sufficient matching, or (iii) RTs indicated a wrong annotation, leaving potential for more pesticides and/or their metabolites being confirmed in further studies. Thus, the reported results are reflecting only a part of the possible pesticide exposure.
