Identifying the source of seafood is critical for combatting seafood fraud, but current tools are predominantly developed and applied on a species-specific basis. This study investigates how multiple marine taxa could be geolocated at global scales by exploiting stable oxygen isotope compositions in carbonate biominerals (δ 18O biomin), where we expect to see universally expressed and predictable spatial variation in δ 18O biomin values across taxa. We constructed global ocean isoscapes of predicted δ 18O biomin values specific to fish (otoliths), cephalopod (statoliths) and shellfish (shells), and a fourth combined “universal” isoscape, and evaluated their capacity to derive δ 18O biomin values among known-origin samples. High correspondence between isoscape-predicted δ 18O biomin values and a compiled database of measured, georeferenced values (3954 datapoints encompassing 68 species) indicated that this δ 18O biomin approach works effectively, particularly in regions with highly resolved projections of seawater δ 18O composition. When compared to taxon-specific isoscapes, the universal isoscape demonstrated similar accuracy, indicating exciting potential for universal provenance applications. We tested the universal framework via a case study, using machine-learning models to identify sample origins amongst regions of divergent (Tropical Asia vs Temperate Australasia) and similar (Temperate Asia vs Temperate Australasia) climates and latitudes. Classification accuracy averaged 75.3% between divergent regions, and 66% between similar regions. When endothermic tuna species were excluded from the analysis, the accuracy between divergent regions increased up to 90% between divergent regions. This study presents the first empirical step towards developing universal chemical markers, which have the potential to support a more inclusive and global approach of validating provenance of seafood.

Environmental flow releases in lowland Australian rivers are currently timed to avoid high-carbon production on floodplains. Moreover, return flows (water draining from floodplains back into rivers) are avoided if there exists a risk of introducing deoxygenated “blackwater” into the main channel. This concern has restricted the range of possible watering scenarios being considered by environmental flow managers. We utilised a series of blackwater flows in the lower Murrumbidgee floodplain, Australia, in 2016 and 2017 to determine the origin and trophic contribution of blackwater dissolved organic carbon (DOC) in a floodplain wetland. We demonstrate a consistent difference in the isotope signature of blackwater DOC compared to both dissolved inorganic carbon (DIC) and river water DOC, explained by the greater contribution of floodplain vegetation (including the river red gum Eucalyptus camaldulensis) to blackwater DOC. Stable carbon isotope signatures suggest a contribution of blackwater to algal production, whereby microbial-mediated conversion of blackwater DOC into DIC may create opportunities for primary autotrophic productivity. This carbon signature was incorporated by the common yabby Cherax destructor. In the main river channel, C. destructor, the native gudgeon Hypseleotris spp. and the introduced European carp Cyprinus carpio may utilise the same basal carbon source. The use of small to moderate floodplain inundation with return flow to the river, properly monitored, would ameliorate the risk of hypoxia while providing the benefit of floodplain-derived DOC and associated increases to in-stream productivity.

Background Globally, food provenance has become a concern for government authorities, the seafood industry and consumers due to increasing food safety and authenticity requirements. Wild-catch fisheries and aquaculture are both important industries; aquaculture is seen as an opportunity to strengthen food security for the growing global population. However, unregulated aquaculture can expose consumers to health risks from pathogens, antibiotics and banned chemicals. Consumers and retailers, and the reputation of the global seafood industry, is affected by food fraud through species substitution and the exchange of aquaculture produce with wild-caught product and vice versa. To ensure consumer confidence and to allow authorities to effectively enforce regulations and contain risks, methods to determine the species, production methods and geographic origin of seafood need to be readily available. Scope and approach This review summarises the currently available and emerging methodologies to determine the provenance and authenticity of seafood. The main focus of this review is to give an overview of the methods that could potentially be used by authorities to enforce regulations and to contain risks, and for the seafood industry to self-regulate and protect itself from food fraud. Key findings and conclusions The most common methods used are DNA profiling, fatty acid profiling, different methods of inductively coupled plasma spectrometry and stable isotope analysis. Additionally, methods such as blockchain, radio frequency identification and x-ray fluorescence through Itrax are currently being tested for their effectiveness in determining seafood provenance. However, these methods have drawbacks and it is likely that a combination of methods would be best suited to determine the provenance of seafood considering its complex supply chain.

The range expansion of tropical fish into temperate waters is increasing markedly in response to climate change. Range-expanding fish encounter novel diets and environments, but we know little about how these conditions facilitate or hinder distribution shifts. Here, we quantified relative growth rate, morphometric condition and trophic niche of juvenile Acanthurus triostegus, a dominant range-expanding tropical surgeonfish, at four locations across 10° of latitude off eastern Australia. We related these metrics to differences in temperature and nutritional quality of dominant seaweeds and the epilithic algal matrix. Temperate food sources were richer in nitrogen than tropical diets. Stable isotope analysis (δ13 carbon and δ15 nitrogen) of fish muscle revealed a large trophic niche breadth at the highest latitude indicating a generalist foraging strategy, and more nitrogen-enriched isotopic signatures compared to tropical regions. Fish length was strongly correlated to δ13C in all regions, suggesting an ontogenetic shift in diet independent of latitude. Despite temperature differences of 4°C, fish growth and body condition were similar across tropical and temperate regions. These results suggest that more nutritious temperate diets may compensate for the effects of cooler water temperatures. Neither summer water temperatures nor dietary factors appear to limit the success of juvenile tropical vagrants as they continue to expand their range along eastern Australia.

Mangrove ecosystems store large quantities of organic carbon for long periods of time. This study explores organic carbon stock change through the first comparative study of radiometric analysis and repeat field measures over a multi-decadal period in a mangrove system. Examining one tall gallery forest of Avicennia marina, and an adjacent interior scrub mangrove of mixed Avicennia marina and Aegiceras corniculatum, radiometric analysis estimated a soil organic carbon accumulation rate of 4.3 ± 0.6 Mg C ha−1 y−1 in the tall gallery forest and 2.2 ± 0.5 Mg C ha−1 y−1 in a stunted mangrove. Repeat measures of root carbon separated by 30 years estimated an increase of 5.06 Mg C ha−1 y−1 in the tall forest and 6.63 Mg C ha−1 y−1 in the stunted forest—suggesting an underestimate of carbon accumulation by radiometric dating of 15% and 67% in the tall and stunted forest, respectively. A higher carbon stock in the interior forest was attributed to root mass increase, associated with landward mangrove encroachment. Extrapolated to the entire region of NSW we estimate that mangrove encroachment has contributed at least about 1.8 Tg C sequestration over the 70 years for which this has been observed in New South Wales, Australia.

Saltmarshes provide many valuable ecosystem services including storage of a large amount of ‘blue carbon’ within their soils. To date, up to 50% of the world’s saltmarshes have been lost or severely degraded primarily due to a variety of anthropogenic pressures. Previous efforts have aimed to restore saltmarshes and their ecosystem functions, but the success of these efforts is rarely evaluated. To fill this gap, we used a range of metrics, including organic carbon stocks, root production, soil respiration and microbial communities to compare natural and a 20-year restoration effort in saltmarsh habitats within the Sydney Olympic Park in New South Wales, Australia. We addressed four main questions: (1) Have above- and belowground plant biomass recovered to natural levels? (2) Have organic carbon stocks of soils recovered? (3) Are microbial communities similar between natural and regenerated saltmarshes? and (4) Are microbial communities at both habitats associated to ecosystem characteristics? For both soil organic carbon stocks and belowground biomass, we found no significant differences between natural and regenerated habitats (F(1,14) = 0.47, p = 0.5; F(1,42) = 0.08, p = 0.76). Demand for seafood, farmed or wild-caught, is growing globally. Consequently, seafood provenance is increasingly important to regulatory bodies, market chain actors and consumers. The limitations of current seafood provenance methods can be overcome using complementary or standalone nuclear techniques. This study focuses on determining the production method and geographic origin of Asian seabass (Lates calcarifer) using Stable Isotope Analysis (SIA) and X-ray fluorescence (XRF) through Itrax. The data were analysed using three different statistical methods; univariate and multivariate analysis, randomForest and LDA. The SIA model had accuracy of 84% when distinguishing the production methods and geographic origin of the L. calcarifer. The model using elemental analysis from the XRF returned an accuracy of 72%, and a combined SIA and elemental model was 81% accurate in determining provenance. However, the SIA model had two incorrect predictions compared to one incorrect prediction in the elemental model, while the combined model had no incorrectly predicted samples. The results of this study highlight that a combination of both SIA and elemental profiling through Itrax is ideal for seafood provenance.

Coastal wetlands (mangrove, tidal marsh and seagrass) sustain the highest rates of carbon sequestration per unit area of all natural systems1,2, primarily because of their comparatively high productivity and preservation of organic carbon within sedimentary substrates3. Climate change and associated relative sea-level rise (RSLR) have been proposed to increase the rate of organic-carbon burial in coastal wetlands in the first half of the twenty-first century4, but these carbon–climate feedback effects have been modelled to diminish over time as wetlands are increasingly submerged and carbon stores become compromised by erosion4,5. Here we show that tidal marshes on coastlines that experienced rapid RSLR over the past few millennia (in the late Holocene, from about 4,200 years ago to the present) have on average 1.7 to 3.7 times higher soil carbon concentrations within 20 centimetres of the surface than those subject to a long period of sea-level stability. This disparity increases with depth, with soil carbon concentrations reduced by a factor of 4.9 to 9.1 at depths of 50 to 100 centimetres. We analyse the response of a wetland exposed to recent rapid RSLR following subsidence associated with pillar collapse in an underlying mine and demonstrate that the gain in carbon accumulation and elevation is proportional to the accommodation space (that is, the space available for mineral and organic material accumulation) created by RSLR. Our results suggest that coastal wetlands characteristic of tectonically stable coastlines have lower carbon storage owing to a lack of accommodation space and that carbon sequestration increases according to the vertical and lateral accommodation space6 created by RSLR. Such wetlands will provide long-term mitigating feedback effects that are relevant to global climate–carbon modelling. Wetlands exposed to rapid sea-level rise over the late Holocene contain more soil carbon than those that experienced a long period of sea-level stability.

The centres of mangrove biodiversity and productivity have been under-represented in studies of fish diet and habitat utilization, particularly in relation to environmental changes between wet and dry seasons. Feeding habits of five dominant fish species (Thryssa kammalensis, Ambassis gymnocephalus, Escualosa thoracata, Stolephorus baganensis and Johnius belangerii) were investigated from two sites in the Matang Mangrove ecosystem, Perak Malaysia, with sampling encompassing both wet and dry seasons. The stomach fullness of the fish was on average significantly higher in the wet season (48%) than in the dry season (32%) with isopods and copepods forming a more important component of the fish diet during the wet season than the dry season. The stomach contents of each species were, on average: T. kammalensis {(Animal based (A) = 50.11%, Plant based (P) = 38.24%)}, A. gymnocephalus (A = 50.35%, P = 37.99%), E. thoracata (A = 42.09%, P = 44.4%), S. baganensis (A = 38.17%, P = 46.55%) and J. belangerri (A = 25.35%, P = 58.86%). Therefore, all five fish species can be considered omnivorous. The similarity among diets varied from 60-80% during the wet and dry season. During the dry season, stable carbon and nitrogen isotope values of the samples had less variation, indicating narrow dietary sources compared to the wet season where the distribution of their isotopic values was larger. Seasons should be considered for planning mangrove management, as results of this study found contracted trophic breadth for commercially valuable fish in the dry season.

Waterbirds are highly mobile, moving over large distances to access resources. Although consistent migration routes are observed in highly seasonal and predictable environments, movement patterns to utilize ephemeral resources in dryland environments are largely unknown. This makes conservation planning and water policy challenging as the relative importance of widely dispersed wetlands is difficult to rank. We addressed this challenge by combining a citizen science project with the novel application of X‐ray fluorescence of feathers to detect continental scale movement of waterbirds using elemental signatures. By doing so, we gained important insight into the movements of 24 waterbird species, including the significance of the Murray–Darling basin as a key source of waterbirds across the continent. Our approach highlights the benefits of elemental signatures to identify key areas of habitat use and priorities for wetland management.

Food fraud is an economically motivated act of deception which can impact consumers and the reputation of actors along a food supply chain. Food fraud includes adulteration, substitution, mislabeling, and other actions that might reduce the quality of food, deceive consumers, and other actors in the food supply chain, while increasing financial gains for the perpetrators. These actions can harm human health, thus raising food safety concerns, as demonstrated by outbreaks of disease caused by the adulteration of food. Food fraud occurs within global and national food systems and with a complex of intersecting food supply chains and networks, food fraud is increasingly challenging to combat. Many governments now have food monitoring programs, food safety strategies, and legal instruments to minimize opportunities for food fraud. This chapter provides an overview of common acts of food fraud and discusses some of their implications. Case studies on the adulteration of infant milk powder and horse meat substitution are used to describe how acts of food fraud can be perpetuated, how technology played a role in identifying the fraud, and how governments and the public responded. The technological advances and emerging analytical methods that are critically important to face the evolving challenges of food fraud are outlined. Specifically this chapter discusses emerging technologies, advantages and limitations of DNA, fatty acid, and elemental profiling, and stable isotope analysis used as analytical tools for detecting food fraud and determining the provenance of food.

Global demand for seafood is rising, with a commensurate increase in supply from farmed and wild-caught products. Determining seafood provenance is important to reduce food fraud, and food safety and biosecurity risks. DNA and fatty acid profiling cannot independently distinguish between farmed, wild-caught and geographic origins of seafood. This study applied stable isotope analysis (SIA) and X-ray fluorescence (XRF), using Itrax, to test their effectiveness as tools to distinguish the origin and production methods of black tiger prawns (Penaeus monodon) from a range of Asia-Pacific locations. Isotopic and elemental data (31 elements) were analysed using multivariate methods, linear discriminant analysis (LDA), and randomForest. LDA and randomForest had consistent results: XRF effectively distinguished the production method and geographic origin of P. monodon (up to 100% accuracy), while SIA had a lower accuracy (up to 95% accuracy). However, SIA and XRF are effective complementary methods for determining provenance of black tiger prawns.

Estuaries are one of the most impacted coastal environments globally, subjected to multiple stressors from urban, industry and coastal development. With increasing anthropogenic activity surrounding estuarine systems, sewage inputs have become a common concern. Stable isotope analysis provides a well-established tool to investigate the incorporation of nitrogen into marine organisms and identify major nutrient sources. Benthic macroinvertebrate communities are often used as bioindicators in ecological studies as they typically display predictable responses to anthropogenic pressures, however have a suite of limitations and costs associated with their use. 16S rDNA amplicon sequencing techniques allow for investigation of the microbial communities inhabiting complex environmental samples, with potential as a tool in the ecological assessment of pollution. These communities have not yet been adequately considered for ecological studies and biomonitoring, with a need to better understand interactions with environmental stressors and implications for ecosystem function. This study used a combination of stable isotope analysis to trace the uptake of anthropogenic nitrogen in biota, traditional assessment of benthic macroinvertebrate communities, and 16S rDNA genotyping of benthic microbial communities. The ratio of stable isotopes of carbon (δ13C) is commonly used to track the flow of energy among individuals and ecosystems, including in mangrove forests. Effective use of this technique requires understanding of the spatial variability in δ13C among primary producer(s) as well as quantification of the isotopic fractionations that occur as C moves within and among ecosystem components. In this experiment, we assessed δ13C variation in the cosmopolitan mangrove Avicennia marina across four sites of varying physico-chemical conditions across two estuaries. We also compared the isotopic values of five distinct tissue types (leaves, woody stems, cable roots, pneumatophores and fine roots) in individual plants. We found a significant site effect (F3, 36 = 15.78; P The within-plant δ13C fractionation we report for A. marina is greater than that reported in most other ecosystems. This has implications for studies of estuarine carbon cycling. The consistent and large size of the fractionation from leaf to woody stem (∼2.0‰) and mostly consistent fractionation from leaf to root tissues (>3.0‰) means that it may now be possible to partition the individual contributions of various mangrove tissues to estuarine food webs. Similarly, the contributions of mangrove leaves, woody debris and belowground sources to blue carbon stocks might also be quantified. Above all, however, our results emphasize the importance of considering appropriate mangrove tissue types when using δ13C to trace carbon cycling in estuarine systems.

Nile tilapia (Oreochromis niloticus) is currently one of the most farmed freshwater fish and contributes significantly to total global aquaculture production. The genetically improved strain of O. niloticus (GIFT) was introduced to Papua New Guinea (PNG) in 1999 to improve food and income security. The high cost and low availability of commercial fish feed hinder the growth of GIFT farming in PNG. Stable carbon and nitrogen isotopes were used to determine the role of supplementary and natural food sources in the diet of GIFT in pond-based aquaculture. Two treatments were used: treatment 1 was daily feeding, and treatment 2 was weekly feeding, each with three replicates. Isotopic analysis of muscle tissue and all potential food sources showed that pellet feed contributed 7% to the growth of GIFT in daily-fed ponds and 33% in the weekly-fed ponds. Highly enriched δ15N values for chicken manure, compared to depleted values for GIFT and other natural food sources in both treatments, clearly indicate insignificant contributions of this input to production. After 90 days of cultivation, the average final body weight of GIFT receiving daily feed inputs was 134 g (average 19 cm), while for weekly-fed it was 92 g (17 cm). The feed conversion ratio (FCR) was poor (6.4:1) in the daily-fed GIFT ponds compared to a better, and preferable, FCR (1:1) in the weekly-fed ponds. The findings of this study show that pelleted feed was not the major contributor to the growth of GIFT. Genetically improved farmed tilapia aquaculture should focus on enhancing natural food availability for fish production.

The Indo-Australian Summer Monsoon (IASM) is the dominant climate feature of northern Australia, affecting rainfall/runoff patterns over a large portion of the continent and exerting a major control on the ecosystems of Australia's Top End, including the viability of wetland ecosystems and the structure of the woody savanna. We examined the behaviour the IASM from 35 kyr using proxy data preserved in the sediments of Table Top Swamp, a small seasonal swamp in northern Australia. Elemental data, stable C and N isotopes, pollen and sedimentary data were combined to develop a picture of monsoon activity and landscape and ecosystem response. Results demonstrated that between 35 and 25 ka conditions were drier and more stable than present, with a more grass dominated savanna and limited wetland development, implying reduced IASM activity. After ~25 ka, there is evidence of increased moisture at the study site, but also increased IASM variability. However, despite evidence of at least periodic increases in moisture, including periods of wetland establishment, the IASM displayed a subdued response to peak precession insolation forcing by comparison to the other global monsoon systems. Instead, the greatest change occurred from ~10 ka when the continental shelf flooded, increasing moisture advection to the study site and resulting in establishment of a quasi-permeant wetland. Whereas the early Holocene was marked by both the onset of pollen preservation and a wetter vegetation mosaic, indicative of a consistently active IASM, the mid-late Holocene was marked by drier vegetation, increased fire, but also increased C3 vegetation and runoff, implying increased IASM variability. Holocene changes in ecosystem dynamics occur coincident with an expansion in human population, which likely also influenced vegetation and landscape response at the study site.

Two independent eight-week experiments were conducted to evaluate the effect of dietary choline content and water temperature on the contribution of major raw materials to the muscle of juvenile yellowtail kingfish (YTK; Seriola lalandi) using stable isotopes and a mixing model. Experiment 1 (dose-response design) focused on rearing fish (mean ± SD; initial weight = 156.3 ± 15.3 g) at 16 °C. Fish were fed six semi-purified research diets with increasing levels of feed-grade choline chloride (CC; 70% CC). The diets contained measured choline content ranging from 0.59 to 6.22 g/kg diet. Except for the control diet, all semi-purified diets contained 0.3% of 2-amino-2-methyl-1-propanol (AMP; de novo choline synthesis inhibitor). Three major raw materials assessed in the semi-purified diets were fishmeal 1, sodium caseinate, and gelatine. In experiment 2 (2 × 3 factorial design), fish (157.3 ± 11.9 g) were reared at 16 °C and 24 °C, and fed three practical diets supplemented with 0.0, 3.0, or 6.0 g CC/kg diet. The six major raw materials assessed in the practical diets were fishmeal 2, poultry meal, meat meal, blood meal, lupin, and wheat flour. Stable isotope ratios of carbon (δ13C) and nitrogen (δ15N) were used as natural tracers and the stable isotope mixing model (SIMM) was based on a Bayesian framework (R package ‘MixSIAR’). In general, the calculated raw material contributions were within the 95% credible intervals range of the SIMM estimates. Based on the SIMM estimate, experiment 1 showed that increasing dietary choline content elevated the mean proportional contribution of sodium caseinate (56.8–81.1%) to the muscle of juvenile YTK. In comparison, fishmeal 1 and gelatine contribution decreased with increasing dietary choline. Unlike experiment 1, experiment 2 showed that choline supplementation and water temperature had little effect on raw material contribution to the muscle of YTK when fed practical diets. In addition, the SIMM estimated contribution of fishmeal 2 (20.7–45.9%) was lower than that of the calculated (47.6%). The study indicated that raw material contributions to muscle of YTK was more profoundly affected by severe choline-deficiency than temperature, at 16 °C and 24 °C. Furthermore, the study indicated that fishmeal inclusion in YTK diet could be reduced. The present study demonstrated the novel use of naturally-occurring stable isotopes and SIMM to evaluate aquafeed down to ingredient level. The findings can guide feed formulation for YTK and other species by demonstrating the potential micronutrient-raw material-environment interactions.

Previous studies on the relationship between ecosystem productivity, size and food-chain length have been restricted to comparisons between locations. We examined the effect of temporal variability in productivity on trophic structure of a floodplain ecosystem, hypothesising that during the wet-flood pulses, the increased resource availability might lead to higher food-chain lengths. We examined multiple common sampling locations and species during a severe El Nino drought which followed a consecutive series of historically wet La Nina years, comparing trophic position and dietary sources. While carbon stable isotopes showed no significant difference between the two phases, nitrogen stable isotopes indicated that most species were feeding higher in the food chain in the wet phase. The results suggest that oscillations in climate phase-driven changes have effects on food-chain lengths through changes in productivity, without the introduction of new sources of carbon or changes to the composition of higher-order predators.

We review links between autotrophic production, crab herbivory and estuarine food chains in southeastern Australia, a region where mangrove and saltmarsh co-exist towards mangrove poleward limits. Studies in estuaries and embayments across the region have demonstrated a consistent and important role of crab larvae released in synchronised spawning during the spring tide in supplementing the diet of zooplanktivorous fishes. These numerically dominant fishes have been linked through isotopic studies to commercially important predator species. The role of macrophytic vegetation (mangrove and saltmarsh) in the diet of crabs remains controversial, though isotopic analysis has demonstrated the contribution of the C4 grass Sporobolus virginicus to crabs feeding in saltmarsh. Other dietary sources include microphytic algae, and potentially stranded zooplankton. The link between sediment organic matter, crab and zooplanktivore isotopic composition remained tightly coupled in spite of variation in anthropogenic N between estuaries and the isotopic signature of mangroves. Long-term threats to these trophic linkages include coastal squeeze associated with sea-level rise, and the fragmentation and disconnection of habitats.

Stable carbon and nitrogen isotope analyses were used to evaluate the influence of microalgal diet on growth and survival of hatchery-reared Pacific oysterCrassostrea gigas larvae.D-veliger larvae were fedmonospecific diets of Pavlova lutheri (P), Tahitian Isochrysis aff. galbana (T), and Chaetoceros calcitrans (C), and a standard hatchery trispecific diet of 25P:25T:50C (PTC). The dietary effects on nutrient assimilation, isotopic turnover rates, and discrimination factors were assessed using exponential models. Of the four dietary treatments evaluated over the 15-day feeding trial, larvae fed C. calcitrans and PTC had the best growth and high survival. Larvae in all dietary treatments had relatively high isotopic turnover rates (0.075–0.327/day), although some discrimination factors found in this study were out of the normal range for aquatic invertebrates (Δ13C: 0.4‰–4.1‰ and δ15N: 0.1‰–5.3‰). Quantifying carbon storage in coastal wetland environments is important for identifying areas of high carbon sequestration value that could be targeted for conservation. Dams constitute a major threat to aquatic ecosystems world-wide by modifying habitat and impairing opportunities for many freshwater fish. Subsequently, there can be a loss in fish diversity and change in the species distributions and population structure that ultimately contribute to changes in trophic structure and ecosystem function. In this study, stable isotope analysis (SIA) was used to compare trophic interaction and niche space of five fish species residing upstream and downstream of a significant barrier (Tallowa Dam) on the Shoalhaven River in south-eastern Australia. Significant reduction in niche space was found among predator/prey species residing in upstream habitat, implying limited dietary opportunity. Mixing calculations for Macquaria novemaculeata confirmed ontogenetic differences upstream and downstream of the dam. Causal mechanisms for this variability in trophic structure include habitat modification and/or discontinuity in river connectivity. SIA provides a useful tool for demonstrating the effect of barriers and/or impoundments on aquatic ecosystems, and for developing future monitoring programmes to evaluate restoration strategies.

Detailed description of each analytical technique used in the 18 year study

Subterranean ecosystems host highly adapted aquatic invertebrate biota which play a key role in sustaining groundwater ecological functioning and hydrological dynamics. However, functional biodiversity studies in groundwater environments, the main source of unfrozen freshwater on Earth, are scarce, probably due to the cryptic nature of the systems. To address this, we investigate groundwater trophic ecology via stable isotope analysis, employing δ13C and δ15N in bulk tissues, and amino acids. Specimens were collected from a shallow calcrete aquifer in the arid Yilgarn region of Western Australia: a well-known hot-spot for stygofaunal biodiversity. Sampling campaigns were carried out during dry (low rainfall: LR) and the wet (high rainfall: HR) periods. δ13C values indicate that most of the stygofauna shifted towards more 13C-depleted carbon sources under HR, suggesting a preference for fresher organic matter. Conversion of δ15N values in glutamic acid and phenylalanine to a trophic index showed broadly stable trophic levels with organisms clustering as low-level secondary consumers. However, mixing models indicate that HR conditions trigger changes in dietary preferences, with increasing predation of amphipods by beetle larvae. Overall, stygofauna showed a tendency towards opportunistic and omnivorous habits-typical of an ecologically tolerant community-shaped by bottom-up controls linked with changes in carbon flows. This study provides baseline biochemical and ecological data for stygofaunal trophic interactions in calcretes. Further studies on the carbon inputs and taxa-specific physiology will help refine the interpretation of the energy flows shaping biodiversity in groundwaters. This will aid understanding of groundwater ecosystem functioning and allow modelling of the impact of future climate change factors such as aridification.

Carbon mitigation services provided by coastal wetlands are not spatially homogeneous, nevertheless are commonly described on the basis of vegetation distribution within the intertidal zone. Distribution of mangrove and saltmarsh varies in response to frequency of tidal inundation, resulting in environmental gradients in edaphic factors that influence vegetation structure, and subsequently affect sedimentary carbon additions by vegetation and carbon losses by decomposition. Current sampling approaches and reporting do not adequately account for variability of carbon storage within a wetland, and assessments need to capture spatial variation associated with carbon storage to improve estimates of potential carbon mitigation services by natural ecosystems. This study quantifies the variation in near-surface carbon storage (i.e. upper 30 cm) across an intertidal gradient using a stratified sampling approach that recognises vegetation structure. Vegetation distribution and structure, as well as sedimentary controls on carbon content, explained variation in carbon storage. Saltmarsh near-surface carbon storage varied considerably between structural form. This was less evident for mangrove structural forms (i.e. tall, shrub, dwarf), which may be due to mangrove roots extending to depths beyond 30 cm. Sedimentary characteristics correlated with carbon content, demonstrating considerable influence on near-surface carbon storage within a wetland. The principal finding of this study was that variation within a wetland corresponds to the variation between sites. Stable carbon isotopes offer a means to identify previous vegetation contributions to sediment, associated with an earlier stage of wetland development, likely reflecting previous environmental conditions. A stratified sampling approach that recognises vegetation structure provides the capacity to account for variability of carbon within a wetland that is inadequately described by current sampling protocols.

Groundwaters provide the vast majority of unfrozen freshwater resources on the planet, but our knowledge of subsurface ecosystems is surprisingly limited. Stygofauna, or stygobionts -subterranean obligate aquatic animals - provide ecosystem services such as grazing biofilms and maintaining water quality, but we know little about how their ecosystems function. The cryptic nature of groundwaters, together with the high degree of local endemism and stygofaunal site-specific adaptations, represent major obstacles for the field. To overcome these challenges, and integrate biodiversity and ecosystem function, requires a holistic design drawing on classical ecology, taxonomy, molecular ecology and geochemistry. This study presents an approach based on the integration of existing concepts in groundwater ecology with three more novel scientific techniques: compound specific stable isotope analysis (CSIA) of amino acids, radiocarbon analysis (14C) and DNA analyses of environmental samples, stygofauna and gut contents. The combination of these techniques allows elucidation of aspects of ecosystem function that are often obscured in small invertebrates and cryptic systems. Carbon (δ13C) and nitrogen (δ15N) CSIA provides a linkage between biogeochemical patterns and ecological dynamics. It allows the identification of stygofaunal food web structures and energy flows based on the metabolic pathway of specific amino groups. Concurrently, 14C provides complementary data on the carbon recycling and incorporation within the stygobiotic trophic webs. Changes in groundwater environmental conditions (e.g. aquifer recharge), and subsequent community adaptations, can be pinpointed via the measurementof the radiocarbon fingerprint of water, sediment and specimens. DNA analyses are a rapidly expanding approach in ecology. eDNA is mainly employed as a biomonitoring tool, while metabarcoding of individuals and/or gut contents provides insight into diet regimes. In all cases, the application of the approaches in combination provides more powerful data than any one alone. By combining quantitative (CSIA and 14C) and qualitative (eDNA and DNA metabarcoding) approaches via Bayesian Mixing Models (BMM), linkages can be made between community composition, energy and nutrient sources in the system, and trophic function. This suggested multidisciplinary design will contribute to a more thorough comprehension of the biogeochemical and ecological patterns within these undervalued but essential ecosystems.

Despite being the fourth largest criminal market in the world, no forensic tools have been sufficiently developed to accurately determine the legal status of seized animals and their parts. Although legal trading is permissible for farmed or captive-bred animals, many animals are illegally removed from the wild and laundered by masquerading them as captive bred. Here we present high-resolution x-ray fluorescence (XRF) as a non-invasive and cost-effective tool for forensic classification. We tested the efficacy of this technique by using machine learning on a training set of zoo specimens and wild-caught individuals of short-beaked echidnas (Tachyglossus aculeatus), a small insectivorous monotreme in Australia. XRF outperformed stable isotope analysis (δ13C, δ15N), reducing overall classification error below 4%. XRF has the added advantage of providing samples every 200 μm on a single quill, enabling 100% classification accuracy by taking the consensus of votes per quill. This accurate and cost-effective forensic technique could provide a much needed in situ solution for combating the illegal laundering of wildlife, and conversely, assist with certification of legally bred animals.

Environmental water allocations are used to improve the ecological health of wetlands. There is now increasing demand for allocations to improve ecosystem productivity and respiration, and enhance carbon sequestration. Despite global recognition of wetlands as carbon sinks, information regarding carbon dynamics is lacking. This is the first study estimating carbon sequestration for semiarid Phragmites australis reedbeds. The study combined aboveground biomass assessments with stable isotope analyses of soils and modeling of biomass using Normalized Digital Vegetation Index (NDVI) to investigate the capacity of environmental water allocations to improve carbon storage. The study considered relationships between soil organic carbon (SOC), carbon sources, and reedbed persistence in the Macquarie Marshes, a regulated semiarid floodplain of the Murray-Darling Basin, Australia. SOC storage levels to 1 m soil depth were higher in persistent reedbeds (167 Mg ha−1) than ephemeral reedbeds (116–138 Mg ha−1). In situ P. australis was the predominant source of surface SOC at persistent reedbeds; mixed sources of surface SOC were proposed for ephemeral reedbeds. 13C enrichment with increasing soil depth occurred in persistent and ephemeral reedbeds and may not relate to flow characteristics. Despite high SOC at persistent reedbeds, differences in the rate of accretion contributed to significantly higher rates of carbon sequestration at ephemeral reedbeds (approximately 554 and 465 g m−2 yr−1) compared to persistent reedbeds (5.17 g m−2 yr−1). However, under current water regimes, rapid accretion at ephemeral reedbeds cannot be maintained. Effective management of persistent P. australis reedbeds may enhance carbon sequestration in the Macquarie Marshes and floodplain wetlands more generally.

Coastal wetlands (mangrove, tidal marsh and seagrass) sustain the highest rates of carbon sequestration per unit area of all natural systems

The uptake, depuration, and organ distribution of the radioisotope 109Cd were used to explore the internal kinetics of this nonessential metal following accumulation from waterborne cadmium by the freshwater decapod crustacean Macrobrachium australiense. Short- (6 h) and long-term (7 to 14 days) exposures to the radioisotope in solutions of 0.56 μg Cd/L were followed by depuration in metal- and isotope-free water for up to 21 days. The anatomical distribution of the radionuclide was visualized using autoradiography at predefined time points. The gills did not become saturated with cadmium after 14 days of exposure and demonstrated a greater rate of cadmium uptake relative to the hepatopancreas. Cadmium concentrations decreased rapidly during depuration from both gills and hepatopancreas after short exposures but slowly following long-term exposures.
