The purpose of this study was to compare zinc and copper biochemical indices of antioxidant status and their relationship in elite athletes of different modalities: aerobic with high-impact (triathletes, n = 10 and long-distance runners, n = 12), anaerobic with high-impact (short-distance runners, n = 9), and anaerobic with low-impact (short-distance swimmers, n = 13). The influence of recent dietary intake and body composition was also evaluated. A venous blood sample was drawn 16-20 hr after competition for the following measurements: packed-cell volume and hemoglobin in blood; copper and zinc in plasma and erythrocytes; ceruloplasmin in plasma; superoxide dismutase activity and metal-lothionein in erythrocytes; and erythrocyte osmotic fragility. Zinc and copper intakes were not different in the athlete groups and did not affect the biochemical indices measured. Athletes of the long-distance high-impact aerobic modalities had higher indices of antioxidant protection (erythrocyte zinc, superoxide dismutase activity, and metallothionein) than those of the short-distance low-impact modalities, suggesting that there is adaptation of the antioxidant capacity to the specific training. Significant correlations were observed in all athletes between erythrocyte zinc, superoxide dismutase activity, and metallothionein consistent with the importance of an adequate zinc status in the response of antioxidant mechanisms to intense exercise. Abstract: Introduction: Endometrial cancer patients with high grade tumours, deep myometrial invasion or advanced stage disease have a poor prognosis. Randomised studies have demonstrated the prevention of loco-regional relapses with radiotherapy (RT) with no effect on overall survival (OS). The possible additive effect of chemotherapy (CT) remains unclear. Two randomised clinical trials (NSGO-EC-9501/EORTC-55991 and MaNGO ILIADE-III) were undertaken to clarify if sequential combination of chemotherapy and radiotherapy improves progression-free survival (PFS) in high-risk endometrial cancer. The two studies were pooled. Methods: Patients (n =540; 534 evaluable) with operated endometrial cancer International Federation of Obstetrics and Gynaecology (FIGO) stage I–III with no residual tumour and prognostic factors implying high-risk were randomly allocated to adjuvant radiotherapy with or without sequential chemotherapy. Results: In the NSGO/EORTC study, the combined modality treatment was associated with 36% reduction in the risk for relapse or death (hazard ratio (HR) 0.64, 95%confidence interval (CI) 0.41–0.99; P =0.04); two-sided tests were used. The result from the Gynaecologic Oncology group at the Mario Negri Institute (MaNGO)-study pointed in the same direction (HR 0.61), but was not significant. In the combined analysis, the estimate of risk for relapse or death was similar but with narrower confidence limits (HR 0.63, CI 0.44–0.89; P =0.009). Neither study showed significant differences in the overall survival. In the combined analysis, overall survival approached statistical significance (HR 0.69, CI 0.46–1.03; P =0.07) and cancer-specific survival (CSS) was significant (HR 0.55, CI 0.35–0.88; P =0.01). Conclusion: Addition of adjuvant chemotherapy to radiation improves progression-free survival in operated endometrial cancer patients with no residual tumour and a high-risk profile. Arbovirus surveillance in marmosets (Callithrix spp.) that live close to humans helps identify viral circulation in the environment and contributes to public health. We investigated the exposure to arboviral infections in 47 captive and free-living Callithrix from urban and peri-urban areas in the semiarid region of northeastern Brazil (SNB) in 2018. The samples were tested for antibodies against 23 arbovirus antigens and the presence of Orthoflavivirus zikaense (ZIKV), Orthoflavivirus denguei, Alphavirus chikungunya, and Orthoflavivirus flavi using hemagglutination inhibition (HI) and RT-qPCR assays. HI highlighted three samples seropositivity for Flavivirus (3/47; 6,4%). One of these samples displayed monotypic antibodies to Alphavirus eastern (formerly known as Eastern equine encephalitis virus; genus Alphavirus). RT-qPCR revealed that one sample (1/41; 2.4%) of C. jacchus captured in a commercial area of Petrolina-PE was positive for ZIKV. Our results indicated that marmosets living close to humans in the SNB were exposed to arboviruses, with the identification of ZIKV, even after the largest epidemic of the virus in the country and the subsequent human cases reduction. This work aimed to evaluate the potential of Trichoderma spp. in the bioremediation of herbicides and biostimulation of plants in herbicide-contaminated soils. In the first phase, the experiment followed a completely randomized design in a 4 × 3 × 4 factorial scheme with five replications, four strains of Trichoderma spp., three herbicides, and four herbicide doses. The mycelial growth speed index (MGSI) of Trichoderma spp. was obtained by growing it in a Petri dish with PDA associated with different doses of herbicides and evaluated every 24 h for 10 days. Results indicated the selection of Trichoderma harzianum T1A to continue the experiment, as it showed an increase in MGSI with metsulfuron-methyl herbicide. The herbicides indaziflam and atrazine reduced the development of Trichoderma spp. The second phase evaluated cucumber cultivated at the scheme corresponding to three doses of the herbicide metsulfuron-methyl and with and without application of T. harzianum T1A in six pots containing two plants per pot, totaling 12 plants/replications per treatment. The parameters chlorophyll a, chlorophyll b, chlorophyll total index, plant height, phytotoxicity, leaf area index, aerial part dry mass, root length, and root dry mass was assessed in cucumber plants. Herbicide application caused phytotoxicity in cucumber plants and reduced all development parameters evaluated in the first cultivation. The phytotoxic effect was still observed in the second cultivation, leading to reduced root and shoot dry mass using metsulfuron-methyl. The results showed that T. harzianum T1A resulted in significant beneficial effects on cucumber development, increasing plant height by 36%, leaf area index by 71%, root length by 23%, shoot dry mass by 54%, and root dry mass by 21% in the first crop, across all herbicide doses used. In the second cultivation, no significant effect of Trichoderma application was observed. Several technological approaches have been used to develop vaccines against COVID-19, including those based on inactivated viruses, viral vectors, and mRNA. This study aimed to monitor the maintenance of anti-SARS-CoV-2 antibodies in individuals from Brazil according to the primary vaccination regimen, as follows: BNT162b2 (group 1; 22) and ChAdOx1 (group 2; 18). Everyone received BNT162b2 in the first booster while in the second booster CoronaVac, Ad26.COV2.S, or BNT162b2. Blood samples were collected from 2021 to 2023 to analyze specific RBD (ELISA) and neutralizing antibodies (PRNT50). We observed a progressive increase in anti-RBD and neutralizing antibodies in each subsequent dose, remaining at high titers until the end of follow-up. Group 1 had higher anti-RBD antibody titers than group 2 after beginning the primary regimen, with significant differences after the 2nd and 3rd doses. Group 2 showed a more expressive increase after the first booster with BNT162B2 (heterologous booster). Group 2 also presented high levels of neutralizing antibodies against the Gamma and Delta variants until five months after the second booster. In conclusion, the circulating levels of anti-RBD and neutralizing antibodies against the two variants of SARS-CoV-2 were durable even five months after the 4th dose, suggesting that periodic booster vaccinations (homologous or heterologous) induced long-lasting immunity.

Mangaba is a fruit native to Brazil, rich in bioactive compounds. To evaluate physicochemical composition, bioactive compounds, antioxidant and antifungal activity of mangaba fruit pulp. Moisture, ash, protein, lipid, energy values and phenolic compounds were determined. Antioxidant activity was determined by capture of 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical. Evaluation of antifungal activity was performed by Minimum Inhibitory Concentration, according to protocols M07-A9 and M27-S3, and minimum fungicidal concentration. Freeze-dried mangaba pulp presented high levels of carbohydrates, low levels of lipids, and high energy density. Phenolic analysis demonstrated that chlorogenic acid was found in the highest concentration, followed by p-coumaric acid and ferulic acid. Mangaba extract showed antioxidant activity like BHT. Mangaba extract inhibited the growth of Candida albicans (ATCC 90028), Cryptococcus gattii (AFLP4), Candida guilliermondii (ATCC 6260) and Candida albicans (MYA 2876). Freeze-dried mangaba inhibited fungal activity associated with antioxidant effect due to presence of phenolic compounds.

Septic shock is a leading cause of hospitalisation, morbidity, and mortality for children worldwide. In 2020, the paediatric Surviving Sepsis Campaign (SSC) issued evidence-based recommendations for clinicians caring for children with septic shock and sepsis-associated organ dysfunction based on the evidence available at the time. There are now more trials from multiple settings, including low-income and middle-income countries (LMICs), addressing optimal fluid choice and amount, selection and timing of vasoactive infusions, and optimal monitoring and therapeutic endpoints. In response to developments in adult critical care to trial personalised haemodynamic management algorithms, it is timely to critically reassess the current state of applying SSC guidelines in LMIC settings. In this Viewpoint, we briefly outline the challenges to improve sepsis care in LMICs and then discuss three key concepts that are relevant to management of children with septic shock around the world, especially in LMICs. These concepts include uncertainties surrounding the early recognition of paediatric septic shock, choices for initial haemodynamic support, and titration of ongoing resuscitation to therapeutic endpoints. Specifically, given the evolving understanding of clinical phenotypes, we focus on the controversies surrounding the concepts of early fluid resuscitation and vasoactive agent use, including insights gained from experience in LMICs and high-income countries. We outline the key components of sepsis management that are both globally relevant and translatable to low-resource settings, with a view to open the conversation to the large variety of treatment pathways, especially in LMICs. Between 2016 and 2018, Brazil experienced the largest sylvatic epidemic of yellow fever virus (YFV). Despite to the magnitude and rapid spread of the epidemic, little is known about YFV dispersion. The study evaluated whether the squirrel monkey is a good model for yellow fever (YF) studies. Methods: Ten animals were infected with 1 × 10 6 PFU/mL of YFV, with one negative control. Blood samples were collected daily during the first 7 days and at 10, 20 and 30 days post infection (dpi) for detection of viral load and cytokines by RT-qPCR; measurements of AST, ALT, urea and creatinine were taken; IgM/IgG antibodies were detected by ELISA, and hemagglutination inhibition and neutralization tests were performed. The animals exhibited fever, flushed appearance, vomiting and petechiae, and one animal died. Viremia was detected between 1 and 10 dpi, and IgM/IgG antibodies appeared between 4 and 30 dpi. The levels of AST, ALT and urea increased. The immune responses were characterized by expression of S100 and CD11b cells; endothelial markers (VCAM-1, ICAM-1 and VLA-4), cell death and stress (Lysozyme and iNOS); and pro-inflammatory cytokines (IL-8, TNF-α, and IFN-γ) and anti-inflammatory cytokines (IL-10 and TGF-β). The squirrel monkeys showed changes similar to those described in humans with YF, and are a good experimental model for the study of YF.

Oxidative stress plays a key role in the initiation and progression of metabolic diseases, including obesity. Preventing the accumulation of reactive oxygen species and oxidative damage to macromolecules is a beneficial strategy for reducing comorbidities associated with obesity. Fruits from the Spondias genus are known for their antioxidant activity, but they are not available year-round due to their seasonality. In this context, we investigated the antioxidant activity and identified the chemical constituents of the aqueous extract of the stem bark of Spondias purpurea L. (EBSp). Additionally, we evaluated the effect of EBSp consumption on metabolic parameters in mice with obesity induced by a high-fat diet. Chemical analyses revealed 19 annotated compounds from EBSp, including flavan-3-ols, proanthocyanidins, methoxylated coumarin, and gallic and ellagic acids, besides other phenolic compounds. In vitro, EBSp showed antioxidant activity through the scavenging of the free radicals and the protection of macromolecules against oxidative damage. Cellular assays revealed that EBSp reduced the levels of malondialdehyde produced by erythrocytes exposed to the oxidizing agent AAPH. Flow cytometry studies showed that EBSp reduced reactive oxygen species levels in human peripheral blood mononuclear cells treated with hydrogen peroxide. Obese mice treated with EBSp (400 mg.kg -1 ) for 60 days showed reduced levels of malondialdehyde in the heart, liver, kidneys, and nervous system. The total cholesterol levels in mice treated with EBSp reached levels similar to those after treatment with the drug simvastatin. Together, the results show that the combination of the different phenolic compounds in S. purpurea L. bark promotes antioxidant effects in vitro and in vivo, resulting in cytoprotection in the context of oxidative stress associated with obesity and a reduction in hypercholesterolemia. Erythrina poeppigiana belongs to Fabaceae family (subfamily Papillionoideae) and is commonly found in tropical and subtropical regions in Brazil. Herein, we described the purification and characterization of a new Kunitz-type inhibitor, obtained from E. poeppigiana seeds (EpTI). EpTI is composed by three isoforms of identical amino-terminal sequences with a molecular weight ranging from 17 to 20 kDa. The physicochemical features showed by EpTI are common to Kunitz inhibitors, including the dissociation constant (13.1 nM), stability against thermal (37-100 °C) and pH (2-10) ranging, and the presence of disulfide bonds stabilizing its reactive site. Furthermore, we investigated the antimicrobial, anti-adhesion, and anti-biofilm properties of EpTI against Gram-positive and negative bacteria. The inhibitor showed antimicrobial activity with a minimum inhibitory concentration (MIC, 5-10 µM) and minimum bactericidal concentration (MBC) of 10 µM for Enterobacter aerogenes, Enterobacter cloacae, Klebsiella pneumoniae, Staphylococcus aureus, and Staphylococcus haemolyticus. The combination of EpTI with ciprofloxacin showed a marked synergistic effect, reducing the antibiotic concentration by 150%. The increase in crystal violet uptake for S. aureus and K. pneumoniae strains was approximately 30% and 50%, respectively, suggesting that the bacteria plasma membrane is targeted by EpTI. Treatment with EpTI at 1x and 10 x MIC significantly reduced the biofilm formation and prompted the disruption of a mature biofilm. At MIC/2, EpTI decreased the bacterial adhesion to polystyrene surface within 2 h. Finally, EpTI showed low toxicity in animal model Galleria mellonella. Background: Data on the prevalence and mortality of paediatric sepsis in resource-poor settings are scarce. We aimed to assess the prevalence and in-hospital mortality of severe sepsis and septic shock treated in paediatric intensive care units (PICUs) in Brazil, and risk factors for mortality., Methods: We performed a nationwide, 1-day, prospective point prevalence study with follow-up of patients with severe sepsis and septic shock, using a stratified random sample of all PICUs in Brazil. Patients were enrolled at each participating PICU on a single day between March 25 and 29, 2019. All patients occupying a bed at the PICU on the study day (either admitted previously or on that day) were included if they were aged 28 days to 18 years and met the criteria for severe sepsis or septic shock at any time during hospitalisation. Patients were followed up until hospital discharge or death, censored at 60 days. Risk factors for mortality were assessed using a Poisson regression model. We used prevalence to generate national estimates., Findings: Of 241 PICUs invited to participate, 144 PICUs (capacity of 1242 beds) included patients in the study. On the day of the study, 1122 children were admitted to the participating PICUs, of whom 280 met the criteria for severe sepsis or septic shock during hospitalisation, resulting in a prevalence of 25·0% (95% CI 21·6-28·8), with a mortality rate of 19·8% (15·4-25·2; 50 of 252 patients with complete clinical data). Increased risk of mortality was associated with higher Pediatric Sequential Organ Failure Assessment score (relative risk per point increase 1·21, 95% CI 1·14-1·29, p<0·0001), unknown vaccination status (2·57, 1·26-5·24; p=0·011), incomplete vaccination status (2·16, 1·19-3·92; p=0·012), health care-associated infection (2·12, 1·23-3·64, p=0·0073), and compliance with antibiotics (2·38, 1·46-3·86, p=0·0007). The estimated incidence of PICU-treated sepsis was 74·6 cases per 100 000 paediatric population (95% CI 61·5-90·5), which translates to 42 374 cases per year (34 940-51 443) in Brazil, with an estimated mortality of 8305 (6848-10 083)., Interpretation: In this representative sample of PICUs in a middle-income country, the prevalences of severe sepsis or septic shock and in-hospital mortality were high. Several factors affect the vegetative growth of fungi, such as temperature, pH, and culture medium. In addition to mycelial growth, these factors affect metabolite production. There are limited studies that have identified the metabolites produced by the fungus Bjerkandera adusta , which have potential biotechnological applications. Here, we evaluated the effects of temperature, culture medium, and incubation time on the production of mycelial mass and metabolites of B. adusta isolated from Pinus taeda . The highest mycelial mass was obtained at 24 °C, in the potato dextrose and malt extract media, upon incubation for 28 and 35 days. The disaccharide α-α-trehalose was for the first time isolated and identified by X-ray diffraction in this fungal genus.

Sepsis, defined as infection with associated organ dysfunction, accounts for most childhood deaths due to infection globally. Evidence for the optimal support of children with septic shock refractory to the initial sepsis management bundle remains minimal. There is an urgent need for more effective interventions. Administration of hydrocortisone in children with septic shock might fasten shock resolution, and has been shown to dampen the systemic host immune response, augment adrenergic effects, and support the stress response. Ascorbic acid (vitamin C) is one of the most powerful naturally occurring antioxidants and has beneficial effects on multiple pathways which are severely deranged during septic shock. A regimen combining hydrocortisone, ascorbic acid, and thiamine termed "metabolic resuscitation" or "HAT therapy" has been tested in large trials in critically ill adults with sepsis with conflicting results. Available information on intravenous ascorbic acid indicates an excellent safety profile even at very high doses both in adults and children. Given the pharmacological properties and beneficial effects shown both in vitro and in animal studies, and its safety profile, ascorbic acid either as a single therapy or as part of HAT treatment represents a promising candidate for future pediatric sepsis treatments. While pediatric age groups may be more susceptible to ascorbic acid deficiency during sepsis, there is a lack of high-quality trial data on HAT therapy in this age group. A single centre retrospective study identified potential for mortality benefit in children with septic shock, and the results from a randomized controlled pilot trial are being awaited. Zootherapy is a traditional secular practice among the Guarani-Kaiowá indigenous ethnic group living in Mato Grosso do Sul, Brazil. My people use the oil extracted from larvae of the snout beetle Rhynchophorus palmarum (Linnaeus, 1758) to treat and heal skin wounds and respiratory diseases. Based on this ethnopharmacological knowledge, the chemical composition and antioxidant, antimicrobial, and healing properties of R. palmarum larvae oil (RPLO) were investigated, as well as possible toxic effects, through in vitro and in vivo assays. The chemical composition of the RPLO was determined using gas chromatography coupled with mass spectrometry. The antioxidant activity of RPLO was investigated through the direct 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging assay, and the antimicrobial activity was evaluated against Gram-positive and Gram-negative bacteria that are pathogenic to humans. The healing properties of RPLO were investigated by performing a cell migration assay using human lung fibroblasts (MRC-5), and the toxicity was analyzed, in vivo, using a Caenorhabditis elegans model and MRC-5 cells, in vitro. RPLO contains 52.2% saturated fatty acids and 47.4% unsaturated fatty acids, with palmitic acid (42.7%) and oleic acid (40%) representing its major components, respectively. RPLO possesses direct antioxidant activity, with a half-maximal inhibitory concentration (IC50) of 46.15 mg.ml-1. The antimicrobial activity of RPLO was not observed at a concentration of 1% (v/v). RPLO did not alter the viability of MRC-5 cells and did not exert toxic effects on C. elegans. Furthermore, MRC-5 cells incubated with 0.5% RPLO showed a higher rate of cell migration than that of the control group, supporting its healing properties. Taken together, RPLO possesses direct antioxidant activity and the potential to aid in the healing process and is not toxic toward in vitro and in vivo models, corroborating the safe use of the oil in traditional Guarani-Kaiowá medicine., Competing Interests: The authors have declared that no competing interests exist.

Background: Staphylococcus haemolyticus is one of the most frequently coagulasenegative staphylococci isolated from healthcare-associated infections, mainly those related to implanted medical devices., Objectives: This study aimed to determine the antimicrobial susceptibility profile and biofilm forming capacity of S. haemolyticus isolated from bloodstream infections., Methods: A total of 40 S. haemolyticus isolates were characterized according to their genetic relatedness by repetitive element sequence based-PCR (REP-PCR), antimicrobial susceptibility profile, SCCmec typing, ability to form biofilm on abiotic surface and occurrence of putative genes related to biofilm formation., Results: One S. haemolyticus was susceptible to all antimicrobials. The other isolates (n=39) were resistant to cefoxitin; and among them 34 (87.2%) harbored the mecA gene into the SCCmec type I (5.9%), type III (29.4%), type IV (5.9%) and type V (20.6%); and 38.2% isolates were designated as NT. Apart from cefoxitin, 94.9% of the isolates were resistant to at least four antimicrobial classes, and 32.5% displayed minimal inhibitory concentration (MIC) values higher than 4.0 μg/mL for vancomycin. All isolates formed biofilm on polystyrene surface and were classified as strong biofilm-producers, except for one isolate. All isolates were negative for icaA gene, and the prevalence of the other genes was as follows: atl, 100%; fbp, 92.5%; aap, 90.0%; and bap, 20.0%., Conclusion: This study reports a high prevalence of methicillin-resistant S. haemolyticus displaying decreased susceptibility to vancomycin with the ability to form strong biofilms on abiotic surface. Introduction: End-stage renal disease (ESRD) patients are known to have a high risk of developing cancer-related inflammation. Elevated serum levels of tumor markers in ESRD/hemodialysis patients makes analysis and interpretation difficult. Aim: To verify the possible relationship between chronic low-grade systemic inflammation serum levels determined by C-reactive protein (CRP) and the tumor biomarkers in patients on hemodialysis. Materials and methods: A prospective study of prevalence was conducted in the Hemodialysis Sector of the University Hospital of the University of Brasília between July 2016 and December 2016 in men aged 18 to 60 years without clinically detectable cancer. We assessed inflammation by serum high-sensitivity CRP test (hs-CRP) and serum tumor in the case groups and controls. The hemodialysis group was split into two subgroups: group 1: patients with inflammation (CRP > 5 mg/L, n=27), and group 2: patients without inflam-mation (CRP ≤5 mg/L, n=33)., Results: There was no significant difference in age mean levels between case groups and controls (44.00±08.00 vs. 41.00±07.00, p=0.08). Background: The dental surgeon and dental health assistant experience risk situations and must be prepared for the prevention of diseases and accidents at work., Objective: Evaluating the prevalence of occupational infectious diseases and prevention measures used by the DS and dental health assistants., Methods: Cross-sectional study carried out with professionals working at the Northwestern region of Rio Grande do Sul. We carried out an interview using a structured questionnaire containing variables related to sociodemographic factors and prevention of infectious diseases. Subjects were submitted to blood collection for serological test for infectious diseases., Results: The use of goggles and cap was higher among the dental health assistant. The dental surgeon reported frequent use of mask: 45.5% replaced the used mask at each new patient. All participants reported using gloves and changing them for each new patient. 78.3% of workers have attended patients with infectious diseases. Among professionals evaluated, dental surgeons reported the highest number of accidents with needle stick instruments (59.1%). Post-exposure prophylaxis procedures are known by 68.2% of dental surgeon and 62.5% of dental health assistant. We did not find positive results for hepatitis B, hepatitis C and human immunodeficiency virus (HIV)., Conclusions: We did not found cases of occupational infectious disease. Dental professionals adopt some preventive actions, but a few professionals, even knowing about the risks they will be exposed during work, do not use some protective equipment.

Background: Staphylococcus aureus can asymptomatically colonize the human anterior nares and skin, and nasal colonization by this bacterium represents a potential risk for development of invasive infections. The aim of this study was to determine the prevalence of S. aureus nasal carriage among healthcare workers and students attending a university hospital and to characterize the isolates phenotypically and molecularly., Methods: A cross-sectional study was performed with 324 volunteers. Cultures from nasal samples were obtained and S. aureus isolates were characterized according to their antimicrobial susceptibility profile and four virulence factors-encoding genes. MRSA isolates were characterized regarding their oxacillin/cefoxitin susceptibility, SCC mec, and REP-PCR types. Potential risks for S. aureus and MRSA carriage were analyzed., Results: Of 324 nasal samples, 42.9% were identified as S. aureus , of which 28.8% were MRSA. S. aureus carriers were significantly higher in males and students (OR = 2.898, 95%CI 1.553-5.410); however, no variables were associated with MRSA carriage. All isolates were susceptible to vancomycin and the highest rate of resistance was observed for penicillin (90.6%). All isolates harbored the coa gene, and 97.8%, the icaA gene; 15.8% and 6.5% were positive for tst and lukS-PV/lukF-PV genes, respectively. Among MRSA isolates, 45% carried the mecA gene but were phenotypically susceptible to oxacillin/cefoxitin; two harbored the tst and none had lukS-PV/lukF-PV genes. All MRSAs were distributed into six SCC mec types and type I (62.5%) was the most frequent. REP-PCR typing identified four main clusters among MRSA isolates., Conclusion: High prevalence of healthcare workers and students were identified as nasal carriers of S. aureus exhibiting different antimicrobial resistance profiles, including mecA -positive oxacillin-susceptible S. aureus (OS-MRSA) and the presence of virulence-encoding genes. Objectives: To develop evidence-based recommendations for clinicians caring for children (including infants, school-aged children, and adolescents) with septic shock and other sepsis-associated organ dysfunction., Design: A panel of 49 international experts, representing 12 international organizations, as well as three methodologists and three public members was convened. Panel members assembled at key international meetings (for those panel members attending the conference), and a stand-alone meeting was held for all panel members in November 2018. A formal conflict-of-interest policy was developed at the onset of the process and enforced throughout. Teleconferences and electronic-based discussion among the chairs, co-chairs, methodologists, and group heads, as well as within subgroups, served as an integral part of the guideline development process., Methods: The panel consisted of six subgroups: recognition and management of infection, hemodynamics and resuscitation, ventilation, endocrine and metabolic therapies, adjunctive therapies, and research priorities. We conducted a systematic review for each Population, Intervention, Control, and Outcomes question to identify the best available evidence, statistically summarized the evidence, and then assessed the quality of evidence using the Grading of Recommendations Assessment, Development, and Evaluation approach. We used the evidence-to-decision framework to formulate recommendations as strong or weak, or as a best practice statement. In addition, "in our practice" statements were included when evidence was inconclusive to issue a recommendation, but the panel felt that some guidance based on practice patterns may be appropriate., Results: The panel provided 77 statements on the management and resuscitation of children with septic shock and other sepsis-associated organ dysfunction. Overall, six were strong recommendations, 49 were weak recommendations, and nine were best-practice statements. For 13 questions, no recommendations could be made; but, for 10 of these, "in our practice" statements were provided. In addition, 52 research priorities were identified., Conclusions: A large cohort of international experts was able to achieve consensus regarding many recommendations for the best care of children with sepsis, acknowledging that most aspects of care had relatively low quality of evidence resulting in the frequent issuance of weak recommendations. Despite this challenge, these recommendations regarding the management of children with septic shock and other sepsis-associated organ dysfunction provide a foundation for consistent care to improve outcomes and inform future research.

Background: Work might cause severe physical stress associated with emotional overload, especially among hospital employees., Objective: To identify risk factors for cardiovascular diseases and occupational stress among employees of a teaching hospital., Method: Cross-sectional study conducted with 45 employees of a philanthropic health institution in the central area of Rio Grande do Sul, Brazil. Data collection was performed to analyze anthropometric variables, blood pressure, biochemical markers and indicators of health and occupational stress., Results: 60.0% of the participants reported sedentary behavior. Analysis of non-modifiable risk factors showed that 55.6% of the sample had family history (mother and father) of hypertension, 22.2% of myocardial infarction and stroke and 13.3% of diabetes. Body mass index categories overweight and obesity predominated (55.5%); 73.4% of the sample was categorized as with moderate-to-high risk to health based on the waist-to-hip ratio; the body fat percentage was above normal or indicated tendency to obesity for 73.3% of the participants. About 71.1% of the sample exhibited excellent or normal blood pressure. Total cholesterol was high or borderline for 88.9% of the sample. On assessment of occupational risk, 55.5% of the participants were categorized as with intermediate degree of exposure., Conclusion: The results point to the relevance of health policies to promote lifestyle changes in and outside the workplace with consequent impact on the physical and mental state of workers.

Pizzuto, F, Fonseca de Oliveira, C, Amorim Soares, TS, Rago, V, Silva, G, and Oliveira, J. Relationship between running economy and kinematic parameters in long-distance runners. J Strength Cond Res 33(7): 1921-1928, 2019-The purpose of this study was to explore the relationship between running economy (RE) and sagittal, frontal, and transverse plane kinematic parameters in long-distance runners. A secondary purpose was to identify the kinematic predictors of RE during running at the lowest RE value, representing an individual's most efficient running intensity. Twenty recreational long-distance runners ran 3 submaximal stages on a treadmill (65, 75, and 85% of velocity at maximum oxygen consumption). Respiratory data were collected using a portable gas analysis system. Kinematics were gathered using passive retroreflective markers and 8 high-resolution infrared cameras to collect the respective trajectories. Hip, knee, and ankle angles at foot strike and stance phase, as well as spatio-temporal parameters were calculated during each gait cycle. Knee flexion/extension range of motion (ROM), knee ab/adduction ROM, and hip ab/adduction ROM during the stance phase of the gait cycle showed positive moderate to large correlations with RE (r ± 90% confidence intervals = 0.51 ± 0.29; 0.49 ± 0.30; 0.53 ± 0.28, respectively). Knee and hip ab/adduction ROMs during the stance phase are predictors of RE, accounting for 44% of RE variance. Therefore, sagittal and frontal plane kinematics affect RE-inducing alterations in running performance. Coaches, athletic trainers, and anyone involved in running training prescription should consider a relationship between these parameters to ensure optimal technique and, consequently, to improve RE in recreational long-distance runners.

The ability of Diplodia pinea to inhibit Armillaria sp., Bjerkandera adusta, Botrytis cinerea, and Rhizoctonia sp. mycelium growth was analyzed using the double-culture method. Wild-type fungal strains were incubated in a biochemical oxygen demand incubator using potato agar dextrose medium at 24 ± 2 °C for 35 days in darkness. D. pinea significantly inhibited the growth of all fungi species tested (30.75 to 98.37% inhibition) and showed moderate antagonistic activity (antagonistic index, 14.5). Chemical analysis of D. pinea culture broth extracts revealed steroids, triterpenes, and phenolic compounds. Alkaloids were qualitatively detected in the mycelium crude extract. The presence of these compounds may be related to the antagonistic activity observed. The inhibition ability of D. pinea is due to competition with the tested fungi for substrate and space.

Antimicrobial resistance is a critical health problem, and pathogens responsible for common infections have developed resistance to antimicrobials, posing a threat to global health and placing a huge burden on health services. During the past two decades, the search for new bioactive agents in nature has become extremely important for promoting health and in the development of more efficient antimicrobials. The genus Eugenia is one of the largest in the Myrtaceae family, comprising approximately 1000 species from Mexico to Argentina, with a few species distributed in Australia and Africa. Eugenia species are used in folk medicine, with antidiabetic, antirheumatic, antipyretic, anti-inflammatory, antidiarrheal, antifungal, and antibacterial properties. This study systematically reviews the Eugenia species to compile the phytochemical composition and antimicrobial effects. In addition, we provide information regarding the traditional uses and cytotoxic activity of Eugenia species. We conducted a systematic literature search of specialized databases (Web of Science, Scielo, Lilacs, Pubmed, Science Direct, Scopus) and selected articles published between 1973 and 2015 using Eugenia and antimicrobial activity, Eugenia and toxicity, and Eugenia and chemical composition as key words. Ninety-three studies were included, and the phytochemical analyses from these studies show that Eugenia species are a rich source of flavonoids, tannins, triterpenes, and sesquiterpenes. Chemical constituents play an apparent role in the antimicrobial effects and reinforce the known antimicrobial potential of the Eugenia genus. A novel Kunitz-type inhibitor from Platypodium elegans seeds (PeTI) was purified and characterized. The mass spectrometry analyses of PeTI indicated an intact mass of 19 701 Da and a partial sequence homologous to Kunitz inhibitors. PeTI was purified by ion exchange and affinity chromatographies. A complex with a 1:1 ratio was obtained only for bovine trypsin, showing a K i = 0.16 nM. Stability studies showed that PeTI was stable over a wide range of temperature (37-80 °C) and pH (2-10). The inhibitory activity of PeTI was affected by dithiothreitol (DTT). Bioassays of PeTI on Spodoptera frugiperda showed negative effects on larval development and weight gain, besides extending the insect life cycle. The activities of digestive enzymes, trypsin and chymotrypsin, were reduced by feeding larvae with 0.2% PeTI in an artificial diet. In summary, we describe a novel Kunitz inhibitor with promising biotechnological potential for pest control.

Objective: To estimate the potential impact of the nonavalent HPV vaccine for high-grade cervical lesions and invasive cervical cancer (ICC) in Portugal., Methods: The present secondary analysis used data collected in the CLEOPATRE II study on the prevalence of HPV 6/11/16/18/31/33/45/52/58 among female patients aged 20-88 years. The prevalence of HPV types in patients with cervical intraepithelial neoplasia (CIN) grades 2/3 and ICC was examined., Results: Data were included from 582 patients. There were 177, 341, and 64 patients with CIN2, CIN3, and ICC, respectively, and 169 (95.5%), 339 (99.4%), and 62 (96.9) of them had HPV infections. Of patients with HPV infections, HPV 16, 18, 31, 33, 45, 52, and 58 infections were identified in 150 (88.8%), 329 (97.1%), and 60 (96.8%) patients with CIN2, CIN3, and ICC, respectively. HPV genotypes 6, 11, 16, 18, 31, 33, 45, 52, and 58 were identified in 540 (94.7%) of the patients with HPV infections., Conclusion: The addition of the five HPV genotypes included in the nonavalent HPV vaccine (HPV 31/33/45/52/58) could result in the new HPV vaccine preventing 94.7% of CIN2/3 and ICC occurrences., (© 2017 International Federation of Gynecology and Obstetrics.)

The frequent use of synthetic pesticides to control Aedes aegypti population can lead to environmental and/or human contamination and the emergence of resistant insects. Linalool and methyl cinnamate are presented as an alternative to the synthetic pesticides, since they can exhibit larvicidal, repellent and/or insecticidal activity and are considered safe for use. The aim of this study was to evaluate the larvicidal activity of methyl cinnamate, linalool and methyl cinnamate/linalool in combination (MC-L) (1:4 ratio, respectively) against Aedes aegypti. The in vitro preliminary toxicity through brine shrimp lethality assay and hemolytic activity, and the phytotoxic potential were also investigated to assess the safety of their use as larvicide. Methyl cinnamate showed significant larvicidal activity when compared to linalool (LC 50 values of 35.4µg/mL and 275.2µg/mL, respectively) and to MC-L (LC 50 138.0µg/mL). Larvae morphological changes subjected to the specified treatments were observed, as the flooding of tracheal system and midgut damage, hindering the larval development and survival. Preliminary in vitro toxicity through brine shrimp showed the high bioactivity of the substances (methyl cinnamate LC 50 35.5µg/mL; linalool LC 50 96.1µg/mL) and the mixture (MC-L LC 50 57.7µg/mL). The results showed that, despite the higher larvicidal activity of methyl cinnamate, the use of MC-L as a larvicide seems to be more appropriate due to its significant larvicidal activity and low toxicity., (Copyright © 2017 Elsevier Inc. Purpose: The relatively recent advent of polyphenol supplement for exercise studies has been tested in a variety of forms and doses. However, the dose-response on adipokines and oxidative stress biomarker effect remains unknown. The aim of the present study was to assess the effect of intense, long-duration (48-h) exercise, and a single dose of proanthocyanidin, on plasma leptin, adiponectin, and electronegative low-density lipoprotein (LDL(-)) concentrations., Methods: Fifty-four healthy male army cadets (22 ± 2 years) participated in a double-blind, randomized, placebo-controlled study and were distributed between control (CG; n = 27) and supplemented groups (SG; n = 27). Immediately before the start of the exercise, both CG and SG groups received a capsule containing starch (200 mg) or proanthocyanidin (dry Vitis vinifera extract, 200 mg), respectively. Following a 12-h fasting period, the plasma adiponectin, leptin, and LDL(-) concentrations were measured prior to the start of the exercise after 24 and 48 h of military training, and after 24 h of rest. The effects of the proanthocyanidin (supplement), exercise (time), and their interaction were investigated using factorial two-way ANOVA., Results: Plasma leptin concentration was only influenced by exercise (p = 0.001). Plasma adiponectin concentration was influenced by exercise (p = 0.037), and by the exercise x supplement interaction (p = 0.033). LDL(-) was influenced by the supplement (p = 0.001), exercise (p = 0.001), and their interaction (p = 0.001)., Conclusions: A single dose of proanthocyanidin (200 mg) was able to reduce LDL(-) concentration and increase plasma adiponectin concentration after 24 h of rest in SG group, indicating its potential protective action.

Introduction: Infections caused by multidrug-resistant bacteria are increasingly common and represent a serious problem for public health. Staphylococcus aureus is one of the major agents of infections, and methicillin-resistant S. aureus (MRSA) has spread worldwide. The aim of this study was to phenotypically and genotypically characterize 55 MRSAs isolated in the University Hospital of Londrina, Paraná, Brazil, during 2010., Methodology: Bacterial isolates were characterized based on their antimicrobial susceptibility profile, biofilm production capacity, and staphylococcal chromosome cassette mec (SCCmec) type. Determination of clonal groups was performed by polymerase chain reaction using the RW3A, JB1, and BOX A1R primers and high-resolution melting (HRM) analysis., Results: The majority of isolates harbored SCCmec type II. SCCmec III, characteristic of the Brazilian endemic clone, was observed in four strains. Only two isolates harbored SCCmec type IV, which is common in community-acquired MRSA strains. Most isolates also showed resistance to more than four of the tested antimicrobials, and 30 isolates exhibited the ability to produce biofilm. DNA polymorphism analysis showed a higher discriminatory power for the JB1 primer, but RW3A revealed several clonal groups of MRSA with similar genotypic and phenotypic characteristics. HRM analysis showed eight different sequence types., Conclusions: These results are important for epidemiological studies involving MRSA infections.

Candida is a common fungus with the capacity to cause infections in humans. However, most studies have concentrated on clinical isolates and little is known about the identity, ecology and drug resistance of free living species/strains. Here, we isolate eight strains of Candida haemulonii and four strains of Cutaneotrichosporon dermatis from three marine cnidarian zoanthids species (Palythoa caribaeorum, Palythoa variabilis and Zoanthus sociatus) collected from Brazilian coral reefs. Strains were identified by sequencing of the D1/D2 domain LSU rDNA and ITS region. We tested these environmental isolates for their capacity to grow in media with increasing concentration of NaCl, capacity to grow in different temperatures, enzymatic activity and antifungal susceptibility. For C. haemulonii, all strains strongly produced gelatinase, esterase and albuminase and were either able to express lipase, phospholipase and keratinase, but not express urease and DNase. The strains were able to grow at 37 °C, but not at 39 °C, and except for LMS 40, all of them could grow in a 10 % NaCl medium. All isolates were resistant to all antifungals tested, with exception for ketoconazole and tioconazole (MIC = 2 µg/mL). For C. dermatis, all strains could grow at 39 °C and could not express phospholipase, keratinase or gelatinase. However, all were capable of expressing urease, lipase and esterase. Three out of four strains could grow in a 10 % NaCl medium, but none grew in a 30 % NaCl medium. The strains showed high values of minimal inhibitory concentration. LMPV 90 was resistant to tioconazole, terbinafine, fluconazole and posaconazole, and LMS 38 was resistant to all antifungal agents tested. We discuss the characterization of C. haemulonii and C. dermatis as a possible emerging pathogen due to its animal-related enzymatic arsenal and antifungal resistance.

Our aim is to investigate two major tendencies in nineteenth-century Brazilian alienism: mentalism and organicism, by conducting a descriptive study of original Brazilian documents on medical health treatments in the 1830s, 1840s and 1850s. Primary sources of Brazilian alienism were theses, memoirs, official reports, and documents written by clinicians and asylum directors. We analysed early mental treatment in Brazilian lunatic asylums, exploring the relative contributions of two main theoretical orientations: moral treatment (based on Pinel and Esquirol) and 'medical-organicist therapeutic orientation'. Intertextuality was used to assess reports of medical organicist treatment in Brazil. We concluded that contemporaneous textual sources indicate that mid-nineteenth-century alienism in Brazil was predominantly influenced by organicism exported from European countries. Trypanosoma cruzi trypomastigotes are able to resist lysis via the complement system, which involves many surface proteins including the complement regulatory protein (CRP). To examine the diversity in CRP recognition among strains of T. cruzi, the expression levels of the translated protein on trypomastigote surfaces were analyzed by flow cytometry, and associations between protein expression and the biological behavior of these strains, especially the ability to induce lytic antibodies in animal models, were assessed. The highly virulent T. cruzi strains Ninoa, INC-5, and Colombiana and the less virulent strains CL-Brener, LGB-231, and JG were used in the experiments. An expression profile analysis showed that the Colombiana and INC-5 strains have higher translated protein levels and induced higher production of antibodies in mice than the other strains. Our results indicated that there are differences in the surface expression of CRP between parasite strains, with a tendency for the most virulent strains to have higher expression levels. Combined, these results contribute to a better understanding of CRP functions and the complexity of host-parasite interactions, considering the large number of virulence factors involved in the process., (Copyright © 2016 Elsevier Inc. Background: Eugenol (EUG) is a major phenolic compound present in clove whose anti-cancer properties have been demonstrated previously. These anti-cancer properties may involves the modulation of different mechanisms, including α-estrogen receptor (αER) in luminal breast cancer cells, COX-2 inhibition in melanoma cells or p53 and caspase-3 activation in colon cancer cells., Hypothesis: EUG promotes a burst in ROS production causing cell-cycle perturbations, mitochondria toxicity and clastogenesis triggering apoptosis in melanoma breast- and cervix-cancer cells in vitro., Methods: Morphological changes were evaluated through the light- and electronic- microscopy. Cell-cycle, ROS, PCNA and Apoptosis was detected by flow cytometry and clastogenicity was evaluated by Comet-assay., Results: The results obtained herein pointed out that EUG promotes, increasing ROS production leading to abrogation of G2/M of phase of cell-cycle, and consecutively, clastogenesis in vitro. In addition, EUG induces Proliferation Cell Nuclear Antigen (PCNA) downregulation and decreasing in mitochondria potential (ΔΨm). Of note, a Bax up-regulation was also observed on cells treated with EUG. All of these findings cooperate in order to induce apoptosis in cancer cells., Conclusion: These promising results presented herein shed new light on the mechanisms of action of EUG suggesting a possible applicability of this phenylpropanoid as adjuvant in anti-cancer therapy., (Copyright © 2016 Elsevier GmbH. RNA interference (RNAi) is a powerful tool in entomology and shows promise as a crop protection strategy, but variability in its efficiency across different insect species limits its applicability. For oral uptake of the double-stranded RNA (dsRNA), the RNAi trigger, two different mechanisms are known: systemic RNA interference deficient-1 (Sid-1) transmembrane channel-mediated uptake and clathrin-mediated endocytosis. So far, a wide range of experiments has been conducted, confirming the involvement of one of the pathways in dsRNA uptake, but never both pathways in the same species. We investigated the role of both pathways in dsRNA uptake in the Colorado potato beetle, Leptinotarsa decemlineata, known to have an efficient RNAi response. Through RNAi-of-RNAi experiments, we demonstrated the contribution of two different sid-1-like (sil) genes, silA and silC, and clathrin heavy chain and the 16kDa subunit of the vacuolar H(+) ATPase (vha16), elements of the endocytic pathway, to the RNAi response. Furthermore, the sid-1-like genes were examined through phylogenetic and hydrophobicity analysis. This article reports for the first time on the involvement of two pathways in dsRNA uptake in an insect species and stresses the importance of evaluating both pathways through a well-devised reporter system in any future experiments on cellular dsRNA uptake., (© 2016 The Royal Entomological Society.)

Objectives: The aim of this study is to evaluate the distribution of the filler size along with the zeta potential, and the integrity of silane-bonded filler surface in different types of restorative dental composites as a function of the material age condition., Materials and Methods: Filtek P60 (hybrid composite), Filtek Z250 (small-particle filled composite), Filtek Z350XT (nanofilled composite), and Filtek Silorane (silorane composite) (3M ESPE) were tested at different stage condition (i.e., fresh/new, aged, and expired). Composites were submitted to an accelerated aging protocol (Arrhenius model). Specimens were obtained by first diluting each composite specimen in ethanol and then dispersed in potassium chloride solution (0.001 mol%). Composite fillers were characterized for their zeta potential, mean particle size, size distribution, via poly-dispersion dynamic light scattering. The integrity of the silane-bonded surface of the fillers was characterized by FTIR., Results: The material age influenced significantly the outcomes; Zeta potential, filler characteristics, and silane integrity varied both after aging and expiration. Silorane presented the broadest filler distribution and lowest zeta potential. Nanofilled and silorane composites exhibited decreased peak intensities in the FTIR analysis, indicating a deficiency of the silane integrity after aging or expiry time., Conclusion: Regardless to the material condition, the hybrid and the small-particle-filled composites were more stable overtime as no significant alteration in filler size distribution, diameter, and zeta potential occurred. A deficiency in the silane integrity in the nanofilled and silorane composites seems to be affected by the material stage condition., Clinical Significance: The materials conditions tested in this study influenced the filler size distribution, the zeta potential, and integrity of the silane adsorbed on fillers in the nanofilled and silorane composites. Thus, this may result in a decrease of the clinical performance of aforementioned composites, in particular, if these are used after inappropriate storage conditions.

This study aimed to compare the biochemical markers of muscle and liver injury and total antioxidant capacity in army cadets after a traditional army physical training program and to correlate these effects with aerobic conditioning. Male army cadets (n = 87; age, 20 ± 2 years) were evaluated 12 hours before the start of training (T0), 12 hours after a 30-km march (T1), and 48 hours after military training (T2). Creatine kinase (CK), aspartate aminotransferase (AST), and alanine aminotransferase (ALT) serum levels were measured using an autoanalyzer. Total antioxidant capacity was determined by 2,2-diphenyl-1-picrylhydrazyl radical scavenging activity (Sc%). Seventy-four percent of army cadets were classified as having excellent aerobic conditioning (53.9 ± 3.0 ml · kg(-) · min(-1) predicted VO2max from the Cooper test). The median serum concentration of all enzymes increased 12 hours after marching (T1: CK 675%; ALT 59%; AST 336%; AST/ALT ratio 85%; p = 0.001) and 48 hours after the end of training (T2: CK 878%; ALT 256%; AST 418%; AST/ALT ratio 180%; p = 0.001). Sc% was higher in T2 (31.1 ± 9.8%; p = 0.01) than in T0 (3.4% change). Maximal oxygen consumption (ml · kg(-1) · min(-1)) correlated negatively with CK (r = -0.25; p = 0.009) and ALT (r = -0.21; p = 0.03) serum levels, and positively with the change in Sc% (r = 0.22; p = 0.04) at T2. The results indicate that intense military training can cause liver and muscle injury and that aerobic conditioning can be considered as a protective factor for these injuries.

Objectives: To determine the effect of material condition (new, aged, and expired) on the polymerization kinetics and polymerization stress of different classifications of dental composites., Materials and Methods: Specimens were obtained according to the following factors: Composites: (3M ESPE) Filtek P60, Filtek Z250, Filtek Z350XT, and Filtek Silorane; and Material conditions: new, aged, and expired. The syringe composites underwent an accelerated aging protocol (Arrhenius model) representing approximately 9 months of aging. Infrared (IR) spectra were obtained kinetically and were analyzed for: maximum conversion rate (%/s), time into exposure when maximum rate occurred (s), conversion at maximum rate (%), and total conversion (%) at 90 s by comparison of absorption IR peak ratios before and after polymerization. Polymerization was evaluated at the bottom surface of 2.0 mm-thick specimens. Polymerization stress was determined in a tensilometer, inserting the composite between acrylic rods fixed to clamps in a universal test machine and dividing the maximum load recorded by the rods cross-sectional area. Polymerization stress (MPa) was calculated at 300 s. Data were statistically analyzed by two-way ANOVA and Tukey's post hoc test (α=0.05)., Results: The majority of the polymerization kinetic parameters were not influenced by the material condition. Silorane composite presented significantly lower conversion rate and lower conversion at the maximum rate when expired (p<0.05). The nanofilled composite (Filtek Z350XT) presented a significantly higher total conversion when aged and expired compared to the new one (p>0.05). In all conditions, Filtek Z350XT and Filtek Silorane presented significantly lower conversion rates (p < 0.05). Filtek Silorane also exhibited the lowest stress, irrespective of the material condition (p<0.05). The polymerization stress was not influenced by the material condition (p>0.05)., Conclusions: Most of the kinetic parameters are not influenced by the material condition. Filtek P60 and Filtek Z250 are more stable as both composites present similar polymerization kinetic results, irrespective of the material condition. Silorane composite presents lower stress values among the tested materials in all conditions. Aging does not affect stress development in restorative composites., (Copyright © 2015 Elsevier Ltd. Introduction: Methicillin-resistant Staphylococcus aureus (MRSA) is one of the leading causes of infections acquired in both community and hospital settings. In this study, MRSA isolated from different sources of hospitalized patients was characterized by molecular and phenotypic methods., Methodology: A total of 123 S. aureus isolates were characterized according to their genetic relatedness by repetitive element sequence based-PCR (REP-PCR), in vitro antimicrobial susceptibility profile, SCCmec typing and presence of seven virulence factor-encoding genes., Results: REP-PCR fingerprinting showed low relatedness between the isolates, and the predominance of one specific lineage or clonal group was not observed. All isolates were susceptible to teicoplanin and linezolide. All isolates were resistant to cefoxitin and penicillin, and the majority were also resistant to one or more other antimicrobials. Fifty isolates (41.7%) were intermediately resistant to vancomycin. Most isolates harbored SCCmec type II (53.7%), followed by type I (22.8%), type IV (8.1%) and type III (1.6%). All isolates harbored at least two virulence factor-encoding genes, and the prevalence was as follows: coa, 100%; icaA, 100%; hla, 13.0%; hlb, 91.1%, hld, 91.1%; lukS-PV and lukF-PV, 2.4%; and tst, 34.1%. A positive association with the presence of hla and SCCmec type II, and tst and SCCmec type I was observed., Conclusion: This study showed the high virulence potential of multidrug-resistant MRSA circulating in a teaching hospital. A high prevalence of MRSA showing intermediate vancomycin resistance was also observed, indicating the urgent need to improve strategies for controlling the use of antimicrobials for appropriate management of S. aureus infections.

The aim of this study was to characterize the hematogeneous spread, in vivo, of breast cancer (BC) cell lines that express hormonal receptors (HR) comparing with triple-negative (TN) BC, particularly considering the lung and liver. Female Balb/c nu nu mice (n=30) were injected with two breast cancer cell lines (MCF7 and HCC1806). Nuclear medicine imaging with Technetium ((99m)Tc)-hydroxymethylene diphosphonate ((99m)Tc-HMDP) and (99m)Tc-Hexakis 2-methoxy-2-methylpropylisonitrile (MIBI) were performed between the 7th and 8th weeks after injection. The histological metastatic foci were analyzed by morphometric and immunohistochemistry studies regarding estrogen receptor (ER), progesterone receptor (PR), human epidermal growth factor receptor 2 (ERBB2) and cytokeratin (CK)-5/6. The mean area of lung metastasis in MCF7 cases was significantly higher (p=0.023), although the number of liver foci was higher in the HCC1806 group (p=0.006). Logistic regression revealed a potentiating model for liver metastasis with HCC1806 cells (odds ratio=16; p=0.03). The number and area of lung-metastatic foci were not predictive of liver dissemination. Lung metastasis study showed ER positivity in 57.1% of the MCF7 group, compared to 80% of the HCC1806 group. PR was positive in 42.9% of MCF7 cases and negative in 60% of HCC1806 cases. HR-positive cells developed massive lung metastization. TN cells seem to potentiate liver metastasis. ER, PR, ERBB2 and basal-like CK expression in metastases was not uniformly correlated with that of primary tumor cells., (Copyright © 2015 International Institute of Anticancer Research (Dr. John G.
