1. Does overconfident CEO lead to corporate environmental misconducts? Evidence from China.

Abstract

Enterprises are drawing growing criticism for violating environmental rules. The research examines whether and how top executives' mental bias leads to corporate environmental misconduct (CEI). Drawing on upper echelon theory (UET) and agency theory, we link CEO overconfidence with CEI, and explore the boundary conditions from the perspective of management discretion at the governance level. Using a data set covering the Chinese listed enterprises from 2004 to 2016, the empirical results demonstrate that CEO overconfidence positively and markedly influenced CEI. Moreover, shareholder concentration and CEO duality reinforce the relationship between overconfidence and CEI, whereas board independence is the opposite. The findings clarify ecological outcomes of CEO overconfidence and have remarkable significance in theory and practice.

2. How Chief Executive Officers’ first-hand experience of the Great Chinese Famine affects risk-taking?

Abstract

Abstract To understand the influence of Chief Executive Officers’ (CEOs) early life experiences on their firms’ strategic decision-making, here we explore the impacts of CEOs’ experience of the Great Chinese Famine (1959–1961) on their cognitions and firm risk-taking. Using longitudinal data from publicly listed companies in China from 2006 to 2017, we found that CEOs who experienced the 3-year-long famine are more risk-averse, but that this relationship only exists among CEOs who experienced the famine during their formative years (between 6 and 18 years old). In addition, we found that the negative relationship between CEOs’ early experiences of famine and firms’ risk-taking is weaker when the firm is state-owned and stronger when the firm is facing fiercely competitive markets. We also take a step further to explore the impact of the severity of the famine on individuals, and we find that there is a positive relationship between the severity of the famine and firm risk-taking only within a moderate level. When the severity exceeds a certain level, CEOs are reluctant to take risks with the increase in the severity of the famine. Our findings provide a new theoretical and practical perspective in studying the relationship between CEOs’ early experiences with disaster and firm risk-taking.

3. Born or not: A moderated mediation model of the relationship between work-family conflict and female employees' wellbeing based on fertility intention during the COVID-19 pandemic

The previous academic research on work-family conflict mainly focused on the relevant elements in the work field. This study concludes that elements of the family domain have a significant impact on the relationship between work-family conflict and employee wellbeing. Female employees' perceptions of wellbeing largely depend on their willingness to have children when they take on family roles. During COVID-19, employees had more time to fulfill both work and family roles in the family sphere due to the epidemic blockade, the contribution of the female employee's significant other (husband) in family matters had a significant impact on Fertility intention. This study using SPSS 24.0 AMOS 20.0 and M plus 7.4 statistical analysis tools to test the proposed hypotheses. In the paired data of 412 working female employees and husbands of Chinese dual-earner families with different occupational backgrounds, hypothesis testing results support that female employees' work → family conflict is negatively related to female employees' fertility intentions, and female employees' fertility intentions are positively related to wellbeing; female employees' family → work conflict is negatively related to female employees' wellbeing; husband's flexible work stress is negatively related to husband's share of housework; husband's share of housework moderated the front, rear and overall mediating effects by the fertility intention. When formulating policies, the managers should consider not only the direct effects of policies, but also the indirect effects that policies may have on other family members of employees. Managers should develop management policies during an epidemic that are more responsive to the actual needs of employees during an epidemic. The management of female employees should give due consideration to the family status of female employees and the enterprises should recognize the importance of childcare for female employees.

4. A Coordinated Revenue-Sharing-Based Pricing Decision Model for Remanufactured Products in Carbon Cap and Trade Regulated Closed-Loop Supply Chain

Abstract

The pricing strategy of remanufactured products has been widely regarded as a significant issue due to the associated economic benefits for both suppliers and retailers awareness worldwide. However, the overall profit determined based on centralized pricing strategy or decentralized pricing strategy is not in line with the actual profit of the manufacturer and the retailer, especially when taking the cost of carbon emission under the system of cap and trade into account. This paper proposes a revenue sharing contract model to coordinate the optimal pricing strategies for both manufacturers and retailers who engages in remanufacturing. The model considers the different Willingness to Pay (WTP) for remanufactured products, and profit distribution of manufacturers and retailers under the Cap and Trade policy. The optimal retail and wholesale price of new products and remanufactured products are derived by solving a convex programming model. Our results identify that: (1) The carbon emission cap is negatively correlated with the retail price of both new products and remanufactured products when the established profits function of the closed-loop supply chain is set to be a constant. The retail price of remanufactured products is also negatively correlated with the value of WTP, while the retail price of new products is positively correlated with WTP. (2) The profit of retailers is positively correlated with carbon emission cap, while the relation of the manufacturer's profit and carbon limit present an inverted U shape, which indicates the optimal carbon emission limit for manufacturers achieving the maximum profit. Moreover, the profit of the retailer and manufacturer and the coefficient of WTP is positively correlated. Numerical experiments are conducted to examine the feasibility of the proposed pricing strategies and gain optimal managerial insights for carbon cap and trade policy.

Abstract

Moisture and thermal are the key factors for influencing methane desorption during CBM exploitation. Using high-pressure water injection technology into coalbed, new fractures and pathways are formed to transport methane. A phenomenon of water-inhibiting gas flow existed. This study is focused on various water pressures impacted on gas-adsorbed coal samples, and then the desorption capacity could be revealed under different conditions. And the results are shown that methane desorption capacity was decreased with the increase in water pressure at room temperature and the downtrend would be steady until water pressure was large enough. Heating could promote gas desorption capacity effectively, with the increasing of water injection pressures, and the promotion of thermal on desorption became more obvious. These results are expected to provide a clearer understanding of theoretical efficiency of heat water or steam injection into coalbed, and they can provide some theoretical and experimental guidance on CBM production and methane control.

Abstract

Spontaneous coronary artery dissection (SCAD) is a rare cause of acute myocardial infarction (AMI). We are presenting a case of a young woman with a history of untreated dyslipidemia presented with AMI secondary to left anterior descending coronary artery dissection during postpartum period. Physicians should be aware of this rare etiology of AMI which occurs during pregnancy and postpartum, since early diagnosis and treatment play a key role in saving both the mother and the baby. It is important to screen for other possible causes such as collagen vascular diseases, blunt injury to the chest, or cocaine abuse.

Abstract

BACKGROUND: Among patients with stable coronary disease and moderate or severe ischemia, whether clinical outcomes are better in those who receive an invasive intervention plus medical therapy than in those who receive medical therapy alone is uncertain. METHODS: We randomly assigned 5179 patients with moderate or severe ischemia to an initial invasive strategy (angiography and revascularization when feasible) and medical therapy or to an initial conservative strategy of medical therapy alone and angiography if medical therapy failed. The primary outcome was a composite of death from cardiovascular causes, myocardial infarction, or hospitalization for unstable angina, heart failure, or resuscitated cardiac arrest. A key secondary outcome was death from cardiovascular causes or myocardial infarction. RESULTS: Over a median of 3.2 years, 318 primary outcome events occurred in the invasive-strategy group and 352 occurred in the conservative-strategy group. At 6 months, the cumulative event rate was 5.3% in the invasive-strategy group and 3.4% in the conservative-strategy group (difference, 1.9 percentage points; 95% confidence interval [CI], 0.8 to 3.0); at 5 years, the cumulative event rate was 16.4% and 18.2%, respectively (difference, -1.8 percentage points; 95% CI, -4.7 to 1.0). Results were similar with respect to the key secondary outcome. The incidence of the primary outcome was sensitive to the definition of myocardial infarction; a secondary analysis yielded more procedural myocardial infarctions of uncertain clinical importance. There were 145 deaths in the invasive-strategy group and 144 deaths in the conservative-strategy group (hazard ratio, 1.05; 95% CI, 0.83 to 1.32). CONCLUSIONS: Among patients with stable coronary disease and moderate or severe ischemia, we did not find evidence that an initial invasive strategy, as compared with an initial conservative strategy, reduced the risk of ischemic cardiovascular events or death from any cause over a median of 3.2 years. The trial findings were sensitive to the definition of myocardial infarction that was used. Abstract

Abstract Background Angiostrongyliasis cantonensis is a severe yet rare parasitic infection caused by the larvae of Angiostrongylus cantonensis. The primary characteristic feature of this foodborne illness in humans is eosinophilic meningitis. Recently, there has been a gradual increase in reported cases globally. Due to the lack of typical clinical symptoms, signs, and specific laboratory tests, early diagnosis of this disease poses significant challenges. Failure to diagnose and treat this condition promptly can result in fatalities. Methods We present the case of a 13-year-old male patient who initially presented with fever and headache. The patient was preliminarily diagnosed with bacterial meningitis and received treatment with antibacterial drugs. However, the patient’s condition worsened, and he developed progressive consciousness disturbances. Eventually, metagenomic next-generation sequencing (mNGS) testing of cerebrospinal fluid samples indicated Angiostrongylus cantonensis infection. Following treatment with albendazole and prednisone, the patient made a full recovery. We include this case report as part of a literature review to emphasize the potential applications of mNGS in the early diagnosis of Angiostrongyliasis cantonensis. Conclusion mNGS technology plays a crucial role in the diagnosis of angiostrongyliasis cantonensis. As this technology continues to evolve and be applied, we believe it will play an increasingly important role in diagnosing, treating, and monitoring angiostrongyliasis cantonensis.

Abstract

This study aims to analyze the expression level and correlation of miR-182-5p and its target gene PAPPA in coronary atherosclerosis (CAD).Real time PCR, ELISA, and Western blotting methods were used to detect the expression levels. Dual-luciferase reporter gene assays were used to analyze the interaction between the 3'-UTR of PAPPA and miR-182-5p.The expression level of miR-182-5p in CAD was significantly lower than that in normal population, while the content of serum PAPPA was significantly increased, and the expression level of miR-182-5p was negatively correlated with the PAPPA content. The expression level of miR-182-5p decreased, while the expression level of PAPPA increased significantly in the ox-LDL treated HA-VSMC cells. Researchers found that PAPPA could promote the activation of IGF signaling pathway in HA-VSMC cells treated by ox-LDL, further activate NF-kB, PI3K/AKT and ERK signaling pathway, and promote cell proliferation. However, miR-182-5p could inhibit the expression of PAPPA, block the activation of IGF signal pathway, and inhibit the proliferation of HA-VSMC cells induced by ox-LDL. miR-182-5p had a targeted action site in the 3'-UTR of PAPPA by bioinformatics prediction. The analysis of luciferase reporter gene further confirmed that miR-182-5p could target the 3'-UTR of PAPPA to inhibit its expression.miR-182-5p demonstrated a protective effect on atherosclerosis and may be a potential therapeutic target for atherosclerosis.

Abstract

There is a long-lasting debate on whether environmental regulation fosters or hinders corporate green innovation. To address this debate, this study differentiates two types of regulations: mandatory environmental regulations (MERs) and voluntary environmental programs (VEPs), and explores whether MERs and VEPs, separately and jointly, affects corporate green innovation. This study also investigates the moderating effect of organizational slack on the focal relationship. With the data of top 100 listed companies in China, the results show that MERs significantly negatively affect green innovation, while VEPs significantly positively influence green innovation, and VEPs mitigate the negative impact of MERs on green innovation. Moreover, organizational slack strengthens the positive relationship between VEPs and green innovation, while no significant moderating effect on the MER–green innovation relationship. This study focuses on the influence of Chief Executive Officer (CEO) characteristics on environmental information disclosure (EID) in the annual reports of companies listed on the Stock Exchange of Thailand. A regression analysis method is used to explore the relationship between CEO characteristics and corporate environmental disclosure. Pollution-intensive companies are taken as the samples in this study. Results show that financial expertise, educational level, and tenure of CEOs are positively correlated with corporate EID. By contrast, age or gender of CEOs is insignificantly related to EID.

Abstract

By employing China's provincial panel data covering period 2001-2016, the present study empirically investigates the impact of ICT on CO2 emission intensity. Specifically, this paper utilizes Internet penetration and mobile phone penetration as proxies to measure ICT respectively and employs quantile regression method to estimate the benchmark model at five quantiles (0.1, 0.25, 0.5, 0.75, and 0.9). It is demonstrated that Internet penetration at the national level has a significant negative effect on the CO2 emission intensity for all quantiles. In addition, Internet penetration has a significant negative effect on CO2 emission intensity at all quantiles except for 0.1 quantile for China's eastern provinces and has a significant negative effect on CO2 emission intensity at all quantiles for China's central provinces, whereas Internet penetration has no significant negative impact on CO2 emission intensity at all quantiles for China's western provinces. By comparison, the reduction effect of Internet penetration on CO2 emission intensity in China's eastern and central provinces is more obvious and in contrast with the reduction effect of Internet penetration on CO2 emission intensity in China's eastern provinces, it is greater in China's central provinces. Finally, the impact of mobile phone penetration on CO2 emission intensity is generally consistent with the impact of Internet penetration. This study provides further evidence that developing countries can simultaneously achieve economic development and reduce carbon emissions through ICT.

Abstract

Liquid nitrogen, an ultra-low-temperature fluid, can cause fissures in coal, especially water-bearing coal. Several coal samples with different water saturation levels were used in this study and compared with a dry sample. The surface changes of each sample were photographed using metallographic microscopy to investigate the effects of liquid nitrogen soaking. Our results show that: 1) For multiple liquid nitrogen freeze–thaw cycles, the porosity of the water-bearing coal samples increased with an increase in the saturation level. Due to the effects of liquid nitrogen circulation freezing and thawing, the porosity can be increased effectively. 2) After being dipped into liquid nitrogen, the length and the width of fissures in the coal samples increased, and some new fissures were simultaneously produced. Liquid nitrogen soaking caused the fissures to expand, which led to an increase in the fissure proportion. The combined effects of (1) and (2) lead us to conclude that the desired cracking effect was achieved. 3) From percolation theory, the connectivity of pore fissures effectively increased due to the effects of the liquid nitrogen, which is directly related to the water-bearing percentage. The more saturated the coal sample, the more connected the pore fissures become. It can be seen that liquid nitrogen is beneficial for improving coal seam permeability and modifying coal properties.

This study focuses on CEO hubris and its effect on corporate unethical behaviour—pollution in particular, and in addition examines critical institutional contingencies [state ownership (SO), political connection (PC) and industrial competition] which may moderate this effect. With data from over-polluting listed firms based on the real-time pollution monitoring system in transitional China from 2015 to 2017, we find that CEO hubris is significantly positively related to firm pollution, and that the moderating role of SO is not significant, that PC positively moderates the hubris–pollution relationship and that industrial competition negatively moderates this relationship. These findings contribute to research on the upper echelon theory, institutional theory and the growing literature on emerging economies.

The value of green innovation on achieving sustainable development is increasingly recognized in recent years. This paper explores whether and how external institutional pressure (including coercive pressure, normative pressure and mimetic pressure, though the last was not discussed in depth) promote green innovation and investigates the moderating effect of internal organizational slack by combining institutional theory and resource-based view. With the sample of China's top 100 listed companies from 2008 to 2014 and generalized estimating equations (GEE) approach, the findings supports the Porter Hypothesis that both coercive pressure and normative pressure have significant positive effects on corporate green innovation. Organizational slack positively moderates the relationship between coercive pressure and green innovation, but has no significant impact on the relationship between normative pressure and green innovation. Accordingly, the scientific value of this research is that it extends the debates on Porter Hypothesis and the role of organizational slacks. The results suggest that the government should strengthen the implementation of coercive tools, the media should play roles of “muckraking”, “catalyst” and the “vanguard” of public inquiry to insert normative pressure, and firms should rationally allocate slacks to improve green innovation.

Abstract

The improvement of regional eco-efficiency is a significant approach to achieve coordinated development of China's economic development and environmental protection, and an effective environmental regulation system is a crucial driver to the improvement of regional eco-efficiency. Based on the panel data of China's 30 provinces during 2000–2013, this paper divides environmental regulation into three types: command-and-control regulation, market-based regulation and voluntary regulation. STIRPAT model is employed to test the effects of these three environmental regulations on eco-efficiency in the eastern, central and western regions of China. Research results turn out that: (1) in the eastern region, market-based and voluntary environmental regulation have a positive impact on eco-efficiency improvement while command-and-control environmental regulation has no significant impact, and (2) in the central region, command-and-control and market-based environmental regulation can promote eco-efficiency more significantly compared with voluntary environmental regulation, and (3) in the western region, command-and-control environmental regulation plays a positive role in eco-efficiency improvement whilst market-based and voluntary environmental regulation have no significant impacts. These findings indicate that the effects of different types of environmental regulation on eco-efficiency apparently differ in regions. Finally, to optimize and adjust the environmental regulation tools, policy recommendations targeting different regions are concluded.

Abstract

Accompanied with rapid economic growth in the emerging economies is the severe ecological deterioration. As major pollution makers and main energy consumers, enterprises are expected to be responsible to the natural environment. However, it's still in dispute whether and when it pays to be green. Based on institutional theory and agency theory, this paper explores the impact of corporate environmental responsibility (CER) on corporate financial performance (CFP) and studies the moderating effect, as well as the co-moderating effect, of external government regulation and internal organizational slack on the relationship. With the data of 1179 observations of Chinese energy-intensive listed companies in 2012–2014, empirical results show that CER significantly positively influences CFP, and that the moderating effect of government regulation is significantly positive, i.e., stringent government regulation significantly reinforces the positive influence of CER on CFP, while organizational slack has a negative moderating effect, i.e., the firms with abundant organization slack are less likely to gain a higher financial performance from undertaking environmental responsibility. In addition, the moderating effect of organizational slack also depends on the degree of government regulation, that is to say, stringent government regulation weakens the negative moderating effect of organizational slack between CER and CFP. The results demonstrate that it pays to be green, especially those lean firms with few slacks under stringent government regulation.

Abstract

Green innovation is an important approach for achieving sustainable development. Most research on determinants of corporate green innovation has focused on either external or internal drivers. Combining institutional theory and resource-based view, the scientific value of this study lies in simultaneously exploring the influence of external legitimacy pressure and internal corporate profitability, and their interaction on green innovation. Samples of the top 100 listed companies in China, from 2008 to 2012, were used and the results demonstrated that legitimacy pressure from stakeholders has a significantly positive influence on both corporate green product innovation and process innovation. The results also revealed that corporate profitability positively affects green product innovation, while there was found to be no significant influence on green process innovation. Moreover, corporate profitability positively moderates the relationship between legitimacy pressure and green product innovation. The results show that not only the single factor of external legitimacy pressure and internal profitability, but also their interaction, affects corporate green innovation practices. This offers an integrating perspective on how corporations can be more innovative in sustainability.

Alongside the ever-increasing development of the global economy, firms worldwide have gradually realized the importance of corporate social responsibility (CSR) in business practices. But there are hot debates on whether CSR pays off, and how does CSR contribute to firm value remains unclear. The direct test of the relationship between CSR and firm value seems to be imprecise and spurious since it may be influenced by many indirect factors. This study aims to demonstrate whether corporate social responsibility is positively or negatively associated with firm value, and to specify the mechanisms of the relationship from a media attention perspective. The manufacturing industry in China provides the background for this study. To test the proposed hypotheses, data was gathered for manufacturing firms listed on the Shanghai Stock Exchange from 2011 to 2013. Empirical results indicate that CSR performance is positively related to firm value, and that media attention has a mediating effect between CSR and firm value.

Firms worldwide are increasingly required to disclose (and make efforts to reduce) their carbon emissions due to the environmental damage associated with climate change. Because there has been no previous literature focusing on the determinants of corporate carbon disclosure integrating environmental legitimacy and green innovation, the present study attempted to develop an original framework to fill the research gap. This study explored the influence of environmental legitimacy (an external informal mechanism) on corporate carbon disclosure, and investigated the role of green innovation (an internal formal mechanism) as a mediator. With the samples of Carbon Disclosure Project (CDP) in China from 2008 to 2012, the results demonstrate that environmental legitimacy significantly negatively influences the likelihood of corporate carbon disclosure, and that green process innovation mediates the relationship, while green product innovation has no significant mediating effect. It means that environmental legitimacy not only directly affects the likelihood of corporate carbon disclosure, but also indirectly affects it via green process innovation. Hence, companies must increase both informal and formal mechanisms, i.e., external environmental legitimacy and internal green process innovation, to engage in carbon information disclosure and ensure sustainability.

Moisture and thermal are the key factors for influencing methane desorption during CBM exploitation. Using high-pressure water injection technology into coalbed, new fractures and pathways are formed to transport methane. A phenomenon of water-inhibiting gas flow existed. This study is focused on various water pressures impacted on gas-adsorbed coal samples, and then the desorption capacity could be revealed under different conditions. And the results are shown that methane desorption capacity was decreased with the increase in water pressure at room temperature and the downtrend would be steady until water pressure was large enough. Heating could promote gas desorption capacity effectively, with the increasing of water injection pressures, and the promotion of thermal on desorption became more obvious. These results are expected to provide a clearer understanding of theoretical efficiency of heat water or steam injection into coalbed, and they can provide some theoretical and experimental guidance on CBM production and methane control.

Abstract

Purpose – The purpose of this paper is to demonstrate the relationship between intellectual capital and innovative performance, and to specify the boundary conditions and mechanisms of the relationship from a knowledge-based dynamic capability perspective. Design/methodology/approach – This study empirically analyzes the impact of intellectual capital on innovative performance and the role knowledge-based dynamic capability plays with a sample of 217 firms in China. To test the research hypotheses, regression analysis is applied. Findings – The results show that intellectual capital positively affects innovative performance, and knowledge-based dynamic capability is a mediator rather than a moderator which partly mediates the relationship between intellectual capital and innovative performance. Practical implications – The findings suggest that realizing superior innovative performance is dependent on a firm’s intellectual capital and its ability to sense opportunities and threats, to make timely and correct decisions, and to facilitate necessary changes efficiently. Originality/value – This study is the first to clarify whether knowledge-based dynamic capability plays a moderating role or a mediating role between intellectual capital and innovative performance. The present study thus helps move forward the understanding on the conditions and mechanisms of the effects of intellectual capital.

Abstract

Moisture and thermal are the key factors for influencing methane desorption during CBM exploitation. Using high pressure water injection technology into coalbed, new fractures and pathways are formed to methane transport. It is existed a phenomenon of water inhibiting gas flow. This study is focused on various water pressures impacted on gas adsorbed coal samples, then the desorption capacity could be revealed under different conditions. And the results are shown that methane desorption capacity was decreased with water pressure increased at room temperature and the downtrend would be steady until water pressure was large enough. Heating could promote gas desorption capacity effectively, with the increasing of water injection pressures, the promotion of thermal on desorption became more obvious. There are the others effects on methane desorption capacity influenced by water injection and thermal.

Abstract

The problem of object localization in image appear ubiquitously in computer vision applications including image classification, object detection and visual tracking. Recently, it is shown that multiple-instance learning(MIL) which is regarded as the fourth machine learning framework compared with supervised learning, unsupervised learning and reinforce learning has been verified that will get good effect in object localization in images. In this paper, we propose a novel method to solve the classical MIL problem, named Cross-Validated Smooth Multi-Instance learning (CVS-MIL). We treat the positiveness of instance as a continuous variable. The softmax model is used to bring a bridge between instances and bags and jointly optimize the bag label and instance label in a unified framework. The extensive experiments demonstrate that CVS-MIL consistently achieves superior performance on various MIL benchmarks. Moreover, we simply applied CVS-MIL to a challenging vision task, common object discovery. The state-of-the-art results of object discovery on Pascal VOC datasets further confirm the advantages of the proposed method.

Abstract

Background and Objectives: Myocardial hypertrophy and fibrosis are important determinants of congestive heart failure. Previous work has shown that hepatocyte growth factor (HGF) can reduce acute myocardial injury and tissue fibrosis. This study was designed to examine the effects of HGF on myocardial remodeling following sustained hypertension. Methods and Results: There were 4 experimental groups (n = 6) that included spontaneously hypertensive rats (SHRs) injected with 0.1 mL of adenovirus (Ad)-null into the left ventricular (LV) free wall, SHR injected with 0.1 mL of Ad-HGF gene (5 × 109 pfu/mL), and SHR injected with 0.1 mL of normal saline, and Wistar Kyoto rats injected with 0.1 mL of Ad-null served as control. At 4 weeks after injection, rats were sacrificed, and HGF expression, myocardial fibrosis, and LV function were determined. We observed that HGF protein expression was reduced in the hearts of SHR ( P < .05 vs normal control) and it was markedly increased in SHR injected with Ad-HGF ( P < .01 vs SHR injected with Ad-null). Myocardial fibrosis, collagen I, LV mass index (LVMI), and LV end-diastolic pressure (LVEDP) were increased and −dP/dtmax was decreased in SHR injected with Ad-null or normal saline ( P < .01 vs normal control). Upregulation of myocardial HGF expression in SHR significantly suppressed myocardial fibrosis, collagen I content, LVMI, LVEDP, and increased −dP/dtmax (all P < .05 vs SHR-Ad-null, n = 6). Conclusions: These findings indicate that HGF expression is attenuated in hypertrophic and fibrotic myocardium of SHR. The forced increase in HGF exerts a salutary effect on myocardial fibrosis, collagen I expression, and hemodynamic parameters.

Abstract

LOX-1 is a multifunctional membrane receptor that binds and internalizes oxidized LDL (oxLDL). We tested the hypothesis that blockade of LOX-1 with an anti-LOX-1 antibody limits nephropathy in male rats with diabetes and dyslipidemia (ZS rats; F1hybrid product of Zucker fatty diabetic rats and spontaneous hypertensive heart failure rats). Lean ZS rats were controls, while untreated obese ZS (OM), ZS obese rats injected with nonspecific rabbit IgG (OM-IgG; 2 μg intravenous injection given weekly), and obese ZS rats given anti-LOX-1 rabbit antibody (OM-Ab; 2 μg intravenous injection given weekly) were the experimental groups. The rats were treated from 6 to 21 wk of age. All obese groups had severe dyslipidemia and hyperglycemia. Kidneys of obese rats expressed LOX-1 in capillaries and tubules, were larger, accumulated lipid, had intense oxidative stress, leukocyte infiltration, depressed mitochondrial enzyme level and function, and peritubular fibrosis (all P < 0.05 vs. lean ZS rats). Injections with LOX-1 antibody limited these abnormalities ( P < 0.01 vs. data in OM or OM-lgG rats). In vitro, renal epithelial LOX-1 expression was verified in a cultured proximal tubule cell line. Our study indicates that anti-LOX-1 (vascular and epithelial) therapy may effectively reverse critical pathogenic elements of nephropathy in diabetes and dyslipidemia.

Abstract

Transforming growth factor beta(1) (TGFbeta(1)) has been purported to protect tissues from ischemia-reperfusion (I-R) injury. This study was designed to examine if overexpression of TGFbeta(1) using adeno-associated virus type 2 (AAV) protects cardiomyocytes from reoxygenation injury. TGFbeta(1) was overexpressed in cultured HL-1 mouse cardiomyocytes by transfection with AAV/TGFbeta(1)(Latent) or with AAV/TGFbeta(1)(ACT) (active TGFbeta(1)). TGFbeta(1) upregulation reduced cardiomyocyte apoptosis and necrosis induced by 24 h of hypoxia followed by 3 h of reoxygenation concomitant with reduction in reactive oxygen species release, activation of nicotinamide adenine dinucleotide phosphate (NADPH) oxidase and NF-kappaB expression. Transfection with AAV/TGFbeta(1)(ACT) was superior to that with AAV/TGFbeta(1)(Latent). To determine if AAV/TGFbeta(1)(ACT) upregulation in vivo would induce cardioprotection from I-R injury, rat hearts were injected with AAV/TGFbeta(1)(ACT) or phosphate-buffered saline (PBS). Abstract

Human urotensin II (hU-II) is a potent vasoconstrictor, highly expressed in cardiac tissues and blood vessels. Recent studies indicate that hU-II participates in vascular and myocardial remodeling after injury. This study was designed to study the role of hU-II in cell DNA synthesis and apoptosis in human umbilical vein endothelial cells (HUVECs) and underlying intracellular signaling mechanisms.Cultured HUVECs were incubated with hU-II (10(-10)-10(-8)M) for 24h. Cell DNA synthesis was examined by 3H thymidine incorporation. Apoptosis was detected by flow cytometry and TUNEL. hU-II increased the 3H thymidine incorporation into DNA in a concentration-dependent manner. hU-II inhibited endothelial apoptosis induced by serum withdrawal (5.74+/-0.64% versus 13.20+/-1.96%, P0.01) and TNFalpha (6.76+/-0.70% versus 13.80+/-1.02%, P0.01). The data from flow cytometry and TUNEL are consistent. Further studies showed that hU-II caused the phosphorylation of mitogen-activated protein kinasep42/44 (MAPKp42/44) in a concentration-dependent manner and this effect of hU-II was inhibited by pretreatment of cells with the MEK inhibitor (PD98059, 10muM). In addition, the use of PD98059 also attenuated 3H thymidine incorporation and anti-apoptotic effect elicited by hU-II (both P0.01 versus hU-II alone).Our observations provide evidence that hU-II promotes cell proliferation and inhibits apoptosis in HUVECs, and MAPKp42/44 activation may play a signal transduction role in this process.

Abstract

TGFbeta(1) deficiency has been attributed to the development of atherosclerosis. There is, however, little direct evidence for this concept. To examine this hypothesis, low-density lipoprotein receptor knockout (LDLR(-/-)) mice were injected via tail vein with recombinant adeno-associated virus type 2 (rAAV) carrying a bioactive TGFbeta(1) mutant (AAV/TGFbeta1ACT, n=10) or granulocyte-macrophage-colony stimulating factor (AAV/GM-CSF, n=10, a negative control) or saline (n=9, control), and then put on a high cholesterol diet. At 18 weeks, blood lipids were found to be similarly elevated in all LDLR(-/-) mice. TGFbeta1ACT and GM-CSF (DNA, mRNA, and protein) were highly expressed in the tissues of mice given TGFbeta1ACT or AAV/GM-CSF, respectively, showing sustained transfection following gene delivery by the systemic route. Saline-treated and AAV/GM-CSF-treated LDLR(-/-) mice showed extensive areas of atherosclerotic lesion formation. There was evidence of intense oxidative stress (nitrotyrosine staining), inflammation (CD68 staining), and expression of adhesion molecules and the ox-LDL receptor LOX-1 (gene array analysis) in the atherosclerotic tissues. Importantly, atherosclerotic lesion formation was markedly inhibited in the LDLR(-/-) mice given AAV/TGFbeta1ACT. Abstract

There is increasing evidence of cross-talk between dyslipidemia and renin–angiotensin system (RAS) in atherogenesis. Both dyslipidemia and RAS activation enhance the expression of a newly described receptor for oxidized-low density lipoprotein (ox-LDL), lectin-like ox-LDL receptor-1 (LOX-1). We postulated that the blockade of dyslipidemia with rosuvastatin, a 3-hydroxy-3-methylglutaryl coenzyme A reductase inhibitor and RAS with candesartan, an angiotensin II type 1 receptor blocker, would have a synergistic inhibitory effect on LOX-1 expression and atherogenesis. Apo-E knockout mice were fed a high-cholesterol diet (1% cholesterol, HC-diet) alone, or HC-diet with rosuvastatin (1 mg/(kg d)), candesartan (1 mg/(kg d)) or with both. Twelve weeks later the extent of atherosclerosis was determined by Sudan IV staining. Apo-E knockout mice on HC-diet had extensive atherosclerosis. Both rosuvastatin and candesartan decreased the extent of atherosclerosis (by 23 and 26%, respectively), despite the HC-diet; however, the combination of rosuvastatin and candesartan reduced atherosclerosis further (by 67%). Rosuvastatin decreased plasma levels of total cholesterol by over 50%, whereas candesartan had no effect. LOX-1 protein expression was found to be markedly up-regulated in HC-diet-fed apo-E knockout mice. While rosuvastatin and candesartan each had a small inhibitory effect on the expression of LOX-1 in the atherosclerotic tissues, the combination totally blocked the up-regulation of LOX-1. P38 mitogen-activated protein kinase (MAPK) expression and phosphorylation were increased in apo-E knockout mice, attenuated by rosuvastatin or candesartan alone, and completely blocked by the combination of the two agents. P44/42 MAPK expression and phosphorylation were not affected by the HC-diet, rosuvastatin, candesartan, or their combination. This study demonstrates the potent effect of rosuvastatin and candesartan on atherogenesis, as well as on the expression of LOX-1 and on the activation of p38 MAPK, but not p44/42 MAPK.

Abstract

The endothelial lectinlike, oxidatively (ox-) modified LDL receptor LOX-1 is a critical player in the pathogenesis of atherosclerosis and myocardial ischemia. Ox-LDL binding of LOX-1 results in the expression of various adhesion molecules, which attract monocytes to endothelial cells, an initial step in atherogenesis. We wished to examine the role of the ox-LDL/LOX-1 signaling pathway in fibroblasts, which naturally express low levels of LOX-1. Rat cardiac fibroblasts were transfected with either cytomegalovirus (CMV)-LOX-1 wt (amino acids [aa] 1 to 273) or CMV-LOX-1 1-261 (an ox-LDL–binding negative mutant, aa 1 to 261) plasmid. Western blots showed that LOX-1 protein expression was increased significantly in cells transfected with CMV-LOX-1 wt or CMV-LOX-1 1-261 plasmid ( P wt showed ox-LDL binding, whereas fibroblasts without transfection and those transfected with CMV-LOX-1 1-261 did not bind ox-LDL. Compared with untransfected cells, ox-LDL treatment (50 μg/mL, 24 hours) markedly induced the expression of the leukocyte adhesion molecules intercellular adhesion molecule-1 (ICAM-1), and vascular cell adhesion molecule-1 (VCAM)-1 as well as matrix metalloproteinase (MMP)-1 in cells transfected with CMV-LOX-1 wt ( P 1-261 . Concurrently, ox-LDL treatment enhanced the phosphorylation of p38 mitogen-activated protein kinase (MAPK) ( P wt –transfected cells. These data suggest that in cardiac fibroblasts, ox-LDL binds to LOX-1 and activates p38 MAPK, followed by the expression of ICAM-1, VCAM-1, and MMP-1. Thus, fibroblasts transform into an endothelial phenotype on transfection with CMV-LOX-1 wt and subsequent exposure to ox-LDL. This study provides a useful model system (plasmid-transfected fibroblasts) to study the molecular biology of LOX-1.

Background : Aspirin is thought to exert salutary effects in vascular disease states by inhibiting platelet aggregation. Endothelial activation, accumulation of oxidized low-density lipoprotein (ox-LDL) and intense inflammation also characterize atherosclerotic plaque in acute myocardial ischemia. Ox-LDL induces expression of lectin-like receptors (LOX-1) on endothelial cells and leads to the expression of matrix metalloproteinases (MMPs), which destabilize the atherosclerotic plaque.
