Abstract

Pharmacologic inhibitors of cyclin-dependent kinases 4 and 6 (CDK4/6) were designed to induce cancer cell cycle arrest. Recent studies have suggested that these agents also exert other effects, influencing cancer cell immunogenicity, apoptotic responses, and differentiation. Using cell-based and mouse models of breast cancer together with clinical specimens, we show that CDK4/6 inhibitors induce remodeling of cancer cell chromatin characterized by widespread enhancer activation, and that this explains many of these effects. The newly activated enhancers include classical super-enhancers that drive luminal differentiation and apoptotic evasion, as well as a set of enhancers overlying endogenous retroviral elements that is enriched for proximity to interferon-driven genes. Mechanistically, CDK4/6 inhibition increases the level of several Activator Protein-1 (AP-1) transcription factor proteins, which are in turn implicated in the activity of many of the new enhancers. Our findings offer insights into CDK4/6 pathway biology and should inform the future development of CDK4/6 inhibitors.

Abstract

We publish an extension of openQCD-1.6 with AVX-512 vector instructions using Intel intrinsics. Recent Intel processors support extended instruction sets with operations on 512-bit wide vectors, increasing both the capacity for floating point operations and register memory. Optimal use of the new capabilities requires reorganising data and floating point operations into these wider vector units. We report on the implementation and performance of the AVX-512 OpenQCD extension on clusters using Intel Knights Landing and Xeon Scalable (Skylake) CPUs. In complete HMC trajectories with physically relevant parameters we observe a performance increase of 5% to 10%., Comment: 9 pages, 4 figures and 4 tables. Abstract

The 'asymmetry thesis', articulated by Fritz Scharpf, holds that EU governance is characterised by an asymmetry between positive and negative integration. The EU has well-developed capacities for negative integration but only limited capacities for positive integration. The present paper challenges the orthodoxy that this thesis has become in EU law and political science scholarship. It argues that the asymmetry thesis no longer accurately depicts European integration, revisiting its key legal and institutional assumptions. Taking the internal market as the most likely case to test the thesis, we show that negative integration has become weaker, positive integration has gained in strength, and both developments have had an impact on the substance of EU law and policymaking, which is promoting non-economic concerns and market-correcting policies to a greater extent than it used to. 31. Exploring Staff Attitudes to Distance Learning -- What Are the Opportunities, Challenges and Impacts on Engineering Academics and Instructional Designers

Abstract

Higher Education Institutions often see distance learning as a means of expanding student numbers and increasing global reach and reputation. Much of the academic literature, however, remains focused on the impact of distance learning on students and the technologies that support it, rather than considering the impact on those staff that are tasked with designing and delivering it. We describe a qualitative study across two engineering departments in a research-intensive UK university, which examines staff perceptions of the impact of converting programmes from successful on-campus ones to distance learning. The findings provide a rich picture of the practical concerns that individual academics have over the impact of distance learning on "pedagogy," on "technology," on their "institution," on "students" and on "themselves." This is an important contribution to the literature that should benefit other engineering departments around the globe who are also grappling with the opportunities and challenges of distance learning.

As a theologically inspired action that has captured the attention of a wide public, Fair Trade action by churches can be viewed as a successful form of Public Theology. Despite the popularity of Fair Trade, and the considerable Church involvement in its promotion, little attention has been paid by academic theology to the public impact of church support for Fair Trade. This research sets out to correct this oversight by bringing together the study of Fair Trade and Public Theology for the greater illumination of both. Using a qualitative study, consisting of twenty-five in depth interviews of community stakeholders and churchgoers, this research asks to what extent, and in what ways, this action is ‘public’ and examines the nature and content of the theology of the churchgoers in regard to Fair Trade. This research contributes to the better understanding of the nature of support for Fair Trade. My investigation of the theology of the churchgoers indicates that their beliefs shape their understanding of what Fair Trade is. They view Fair Trade as an act of justice and as demonstration of Christian principles in action. The churchgoers’ desire for Fair Trade to ‘live up’ to these theologically inspired ideals leads them to defend forms of Fair Trade practice that offer a distinctive alternative to mainstream trade. My analysis of Fair Trade action by churches complements the existing body of knowledge on Public Theology, by offering detailed example of the practical outworking of some of the key points of discussion within the discourse. Using Fair Trade action by churches as a case study of a specific form of Public Theology in practice, I indicate the ways in which engagement between the Church and a wider audience can be balanced with the exercise of a distinctive theological voice.

Drawing on my research of Luton Fair Trade Town, in this article I argue that faith communities, working in partnership with Fair Trade Towns, are well-placed to raise awareness of sustainable development. I will discuss the ability of faith communities to raise awareness and bring about change towards sustainable development, utilizing both social and spiritual capital. An analysis focusing on social capital highlights the ability of faith communities to bridge different sections of society, issues and geographies. Spiritual capital concentrates on the motivation to action and this can provide a challenge to the status quo and a vision of how society can work differently, offering a necessary contribution to working towards a sustainable future. 43. Algorithmic Culture, Networked Learning and the Technological Horizon of Theory

Abstract

Going via Bernard Stiegler's theorisation of technology, and his response to Chris Anderson's claim that the era of hyper-networked, algorithmically driven digital technologies signals the end of theory, this paper aims to place the educational practice of networked learning as a space to think the edge, excess or limit of this proposed algorithmic dominance. The author discusses how networked learning can negotiate the border between educational theory, the practice of teaching and learning, and the processes and systems of educational technology, but suggests that to do this it must engage these disciplines through a thinking of technology which does not decide upon its status in advance. He argues that affirming this particular relation to technology is increasingly urgent given we are at a moment in which educational institutions are asking how to prepare our students for an age of continuing technological disruption.
