To tease apart differences between conventional cage (CC) and cage-free (CF) housing systems, this study focuses on the effects of excreta exposure and age by comparing microbial communities, intestinal permeability, and corticosterone in hens in enriched colonies (EC) and CF housing systems during early- and late-lay. Hens were randomly selected from two rooms of CF (n = 20) and EC (n = 20) at 35 and 76 weeks of age. One hour following an oral gavage of fluorescein isothiocyanate dextran (FITC-D), hens were euthanized, and ileal contents and blood were collected. Serum FITC-D using a fluorescent spectrophotometer and corticosterone using a commercial competitive ELISA kit were analyzed. Following DNA isolation from the ileum contents, the V4 region of the 16S rRNA gene was sequenced. Sequence data were filtered in Mothur v1.43.0, followed by de novo operational taxonomic unit (OTU) clustering and classifying with the SILVA SSU v138 reference database. Serum FITC-D was altered by housing type, age of hens, and the interaction between housing type and age of hens (p < 0.001), with 76-week-old hens housed in EC having the highest FITC-D. Corticosterone increased with age (p = 0.023). Microbial community diversity measurements favored hens housed in the CF housing system as ileal contents tended to have increased species evenness (p = 0.008) and greater alpha diversity (p = 0.006). The majority of the over-representation of OTUs were associated with peak lay.

Two trials were conducted to determine interactive effects between lipid source (palm oil, PO versus soybean oil, SO) and emulsifier addition (none versus glycerol monolaurate-GML) on apparent total tract digestibility (ATTD) of gross energy (GE) in broilers and growth performance in poults. In trial 1, 0.05 % GML addition had no impact on the ATTD of GE of SO but improved the ATTD of PO from 77.11 % to 88.21 % (interaction, P=0.03). Without GML addition, PO had a lower ATTD of GE (77.11 %) compared to SO (96.48 %) resulting in an AME of 7,259 versus 9,092 kcal/kg for PO and SO, respectively. In trial 2, the addition of 0.10 % GML reduced ADFI in poults fed diets containing 5 % PO compared to poults fed 0 or 0.05 % GML, while the addition of either 0.05 or 0.10 % GML reduced ADFI in poults fed diets containing 5 % SO compared to poults fed no GML (P=0.01). There was a similar response with ADG (P=0.01) where the addition of either 0.05 or 0.10 % GML reduced ADG in poults fed diets containing SO compared to poults fed no GML, while the addition of GML was largely without effect in poults fed diets containing PO. There was no interaction between lipid source and emulsifier addition on feed efficiency (P>0.10). Poults fed diets containing PO had a poorer feed efficiency compared to birds fed diets containing SO (P=0.01). The main effect of emulsifier was inconsistent in that poults fed the diets containing 0.10 % GML had the greatest feed efficiency compared to poults fed the diets containing 0.05 % GML, with poults fed diets containing no emulsifier being intermediate (P=0.10). In conclusion, addition of GML improved the ATTD of GE for PO but had no effect on the ATTD of GE for SO. This improvement in energy digestibility, did not however, translate to an improvement in poult performance. Broilers and poults fed diets containing SO had a greater feed efficiency compared to birds fed diets containing PO.

ABSTRACT: Amino acids (AA) are an expensive nutritional components of poultry diets. Distillers dried grains with solubles (DDGS) is the primary co-product produced by the dry grind bioethanol industry, although new technologies are being implemented to produce high protein distillers dried grains (HP-DDG) and corn fermented protein (CFP), but data on their nutritive value in poultry are lacking. Two experiments (EXP) were conducted to determine the energy and AA digestibility of DDGS, HP-DDG, and CFP in poults in addition to a feeding trial to evaluate increasing dietary levels of HP-DDG and CFP on growth performance and intestinal characteristics. In EXP 1, 6 different DDGS sources were evaluated using poults to determine their nitrogen-corrected apparent metabolizable energy (AMEn) concentrations, and cecectomized roosters were used to determine their standardized ileal (SID) AA digestibility (SID-AA). In EXP 2, AMEn and SID-AA for HP-DDG and CFP were determined in young poults, and a feeding trial was conducted to evaluate growth performance and intestinal morphology and permeability of poults fed diets containing 7.5 and 15% HP-DDG or CFP from 1 to 32 d of age. In EXP 1, the AMEn concentration among the DDGS samples ranged from 2,530 to 3,573 kcal/kg DM but was not different (P = 0.57) among the samples, with an average SID for LYS of 66.6%. In EXP 2, different (P = 0.001) AMEn concentrations for HP-DDG and CFP were observed (3,114 and 3,760 kcal/kg DM, respectively), with the SID for LYS being 66.55 and 77.00% for HP-DDG and CFP, respectively. Including HP-DDG or CFP into the diet at 7.5 and 15% had no effect (P > 0.05) on growth, feed intake, or feed conversion. Neither co-product nor its inclusion rate affected intestinal morphology and permeability (P > 0.05). Overall, DDGS, HP-DDG, and CFP are excellent sources of AMEn and digestible AA, with dietary inclusion rates of up to 15% of HP-DDG or CFP having no impact on growth or intestinal characteristics.

The adoption of methods detecting intestinal permeability in poultry has been slow due to the lack of urine availability in avian species. The objective of this study was to examine intestinal permeability assays in broilers using serum. Fluorescein isothiocyanate-dextran (FITC-D) and lactulose/mannitol/sucralose (LMS), indigestible sugars, were used to detect intestinal permeability across two fed states (fed or fasted) and four sugar treatments (Control, FITC-D, LMS, or FITC-D+LMS). Broilers housed in pens were assigned one of eight treatments and sampled on 14, 28, and 42 days of age. Data were analyzed using PROC Glimmix for fed state, sugar treatment, age, and all interactions. Serum lactulose and FITC-D increased in fasted compared to fed birds (p < 0.006), whereas mannitol increased in fed compared to fasted birds (p < 0.001). Serum lactulose and FITC-D decreased on day 28 compared to other timepoints (p < 0.003). Serum FITC-D only had a significant sugar by fed state interaction (p < 0.05) with elevated concentrations in fasted and fed birds that received FITC-D. Serum lactulose was significant for all interactions with elevated concentrations in broilers provided lactulose and fasted (p < 0.001). The ability to detect a three-way interaction with serum lactulose suggests an increased sensitivity; however, additional studies are needed.

Animal health and feed intake are closely interrelated, with the latter being an important indicator of an animal’s health status. Automated sensors for dairy cattle have been developed to detect changes in indicators of health, such as decreased rumination or activity. Previous studies have identified associations between sensor measurements and feed intake. Thus, the objective of this study was to determine if health disorders impact the associations identified between sensors and dry matter intake (DMI), and to measure the impact of health disorders on DMI. A total of 934 cows with health disorders (lameness, mastitis, and other), of which 57, 94, and 333 cows had observations for a rumen bolus and one of two ear tags, were analyzed to determine how health disorders impact the association of sensors with DMI. Eleven sensor measurements were collected across the three sensors, including total and point-in-time activity, rumination time, inner-ear temperature, rumen pH and rumen temperature. Associations of health disorders and sensor measures with DMI were evaluated when accounting for systematic effects (i.e., contemporary group, parity, and days in milk) and energy sinks accounted for in determination of feed efficiency (e.g., milk production, body weight and composition). In order to determine if inclusion of health disorders or sensor measures improved model fit, model AICs were assessed. Health disorders were significantly associated with all sensor measurements (P< 0.0001), with the direction of association dependent on sensor measure and health disorder. Moreover, DMI decreased with all health disorders, with larger impacts observed in animals in third and higher lactations. Numerous sensor measurements were associated with DMI, including when DMI was adjusted for energy sink variables and health. Inclusion of rumen bolus temperature, rumination or activity with health data reduced model AIC when evaluating DMI as the dependent variable. Some sensor measures, including measurements of activity, temperature and rumination, accounted for additional variation in feed intake when adjusted for health disorders. Results from the study indicate that feed intake and sensor measures are impacted by health disorders. These findings may have implications for use of sensors in genetic evaluations and precision feeding of dairy cattle.

The gastrointestinal health of poultry can be impacted by a variety of factors including their environment. As egg production moves from conventional cage housing (CC) toward cage-free housing (CF), it is important to understand this impact on intestinal health. This study was conducted to determine if housing type impacted intestinal permeability, morphology, and microbial communities in commercial hens across housing systems. Hens were randomly selected from 2 rooms of CC (n = 25) and CF (n = 25) at a commercial facility. Birds were given fluorescein isothiocyanate dextran (FITC-D) by oral gavage to measure intestinal permeability. Jejunal and ileal samples were collected to evaluate villus height, crypt depth, and their ratio. Ileal contents were collected for bacterial DNA isolation and 16S rRNA gene sequencing. This report demonstrates that high levels of lameness can be induced by a limited bacterial challenge in drinking water for birds raised on litter flooring, comparable with lameness induced by the gold standard for inducing lameness, growth on suspended wire flooring. The bacterium used in the challenge was cultured from lesions in birds induced for bacterial chondronecrosis with osteomyelitis (BCO) in the wire-flooring model so the epidemiology appears similar. The litter-flooring model could better approximate broiler operations. Furthermore, the work demonstrates that 2 commercial probiotics (GalliProTect and GalliProMax) can reduce lameness in the bacterial challenge litter-flooring model. Lameness attributable to BCO is one of the most significant animal welfare issues for broiler production. The wire-flooring and litter-flooring models afford alternatives for understanding the etiology, and epidemiology of BCO, and development of management strategies to reduce lameness. Probiotics afford a promising management strategy. The results suggest that the probiotic protection may extend beyond just intestinal health and intestinal barrier function.

Abstract Objective To determine the effect of bacitracin methylene disalicylate (BMD) and feed changes on gastrointestinal integrity, endotoxin permeability, and morphometric parameters in the duodenum of broilers. Results Birds were raised on a starter diet without growth promoting antibiotics for 31 days then switched to a grower diet. Four of the pens including 50 g/ton of BMD while 4 pens remained antibiotic free. Eight birds per treatment were sampled prior to the feed change and at 3 and 7 days following the feed change. Gastrointestinal integrity and endotoxin permeability in the duodenum were determined using a modified Ussing Chamber and an adjacent section fixed in 10% formalin for morphometric analysis. Data were analyzed using Proc Glimmix of SAS with the model fitting BMD treatment, time, and the interaction of BMD treatment and time as fixed effects. Intestinal integrity increased at d 3 and 7 compared to prior to the feed change and addition of BMD (P > 0.001) and villus height was decreased with BMD supplementation (P = 0.049). All other tested effects similar (P > 0.1). In conclusion, the practice of changing feed had a greater effect on intestinal health than addition of BMD. However, the factors driving these differences 42 are unclear.

The objective of this study was to evaluate the of intestinal permeability and liver bacterial translocation (BT) across a modern commercial broiler, a commercial broiler of 1995 genetics, and an unselected Jungle Fowl line. Modern 2015 (MB2015) broiler chicken, random bred line initiated from 1995 (RB1995), and the Giant Jungle fowl (JF). Chickens were randomly allocated to four different dietary treatments. Dietary treatments were (1) a control corn-based diet throughout the trial [corn-corn (C-C)]; (2) an early phase malnutrition diet where chicks received a rye-based diet for 10 days, and then switched to the control diet [rye-corn (R-C)]; (3) a malnutrition rye-diet that was fed throughout the trial [rye-rye (R-R)]; and (4) a late phase malnutrition diet where chicks received the control diet for 10 days, and then switched to the rye diet for the last phase [corn-rye (C-R)]. Paracellular permeability was evaluated using fluorescein isothiocyanate dextran (FITC-D). Liver BT was also evaluated. MB2015 and RB1995 consuming the rye-based diet showed increase serum levels of FITC-D when compared to the corn-fed chickens (P < 0.05). Overall, MB2015 appeared to have higher enteric permeability than the JF. To our knowledge, this would be the first paper to evaluate the effect of compensatory growth on intestinal permeability and liver BT. Further studies to evaluate microbiome and inflammatory markers in these chicken models are currently being evaluated.

Beef cattle phenotypes are affected by the consumption of toxic fescue. Toxic fescue’s impact is dependent on heat stress and breed composition, with genetic variability for robustness to toxin exposure believed to exist within and across breeds. The study objective was to characterize the effect of fescue toxicosis across breeds for known and novel heat and fescue stress-associated phenotypes. One-hundred crossbred fall-calving Charolais- and Hereford-sired cows of parities 1–3 were allocated to graze either toxic fescue (n = 50), non-toxic fescue (n = 25), or a rotation between toxic and non-toxic fescue (n = 25) for 156 days. Phenotypes impacted by breed (genetics) included hair coat score (p < 0.0001), hair reduction/shedding rate (p < 0.05), rectal temperature (RT) (p < 0.0001), vaginal temperature (p < 0.05), serum phosphorus concentration (p < 0.02) and respiration rate (RR) (p < 0.003). Cows on toxic fescue experienced reduced hair shedding efficacy (p < 0.0001), higher vaginal temperatures (p < 0.0001), increased systolic blood pressure (p < 0.04), increased RR (p < 0.0001) and reduced average daily gain (p < 0.0001), compared to cows grazing non-toxic fescue. Calves born from cows with higher RT during the last third of gestation had higher RT at weaning (p < 0.02), indicating potential physiological effects of in utero heat stress. The study indicates that beef cows exhibit variable responses to toxic fescue within and across breeds which may impact future calf phenotypes.

This article is the first in a series of manuscripts to evaluate nutritional rehabilitation in chickens as a model to study interventions in children malnutrition (Part 1: Performance, Bone Mineralization, and Intestinal Morphometric Analysis). Inclusion of rye in poultry diets induces a nutritional deficit that leads to increased bacterial translocation, intestinal viscosity, and decreased bone mineralization. However, it is unclear the effect of diet on developmental stage or genetic strain. Therefore, the objective was to determine the effects of a rye diet during either the early or late phase of development on performance, bone mineralization, and intestinal morphology across three diverse genetic backgrounds. Modern 2015 (Cobb 500) broiler chicken, 1995 Cobb broiler chicken, and the Giant Jungle Fowl were randomly allocated into four different dietary treatments. Dietary treatments were (1) a control corn-based diet throughout the trial (corn–corn); (2) an early phase malnutrition diet where chicks received a rye-based diet for 10 days, and then switched to the control diet (rye–corn); (3) a malnutrition rye-diet that was fed throughout the trial (rye–rye); and (4) a late phase malnutrition diet where chicks received the control diet for 10 days, and then switched to the rye diet for the last phase (corn–rye). At 10 days of age, chicks were weighed and diets were switched in groups 2 and 4. At day 20 of age, all chickens were weighed and euthanized to collect bone and intestinal samples.
