To determine the quality changes and shelf life of grass carp fillets with sizing (GCFS), the aerobic bacterial count, microbial succession, water holding capacity (WHC), total volatile basic nitrogen (TVB-N), thiobarbituric acid reactive substances (TBARS), biogenic amines, ATP-related compounds and K value of GCFS were analyzed during storage at 4 ℃ and 10 ℃. Results showed that the aerobic bacterial count of GCFS stored at 4 ℃ and 10 ℃ approached or exceeded the limit on 7 d and 3 d, respectively, suggesting that low temperature significantly affects microbial amplification and microflora composition in GCFS, especially retarding the growth of Acinetobacter, Shewanella and Aeromonas. During storage, no significant difference (P>0.05) of WHC in samples was observed. The pH of GCFS reached the lowest level on 3 d, and the contents of TBARS, TVB-N and the specific biogenic amines (cadaverine, putrescine, histamine and tyramine) increased continuously. Furthermore, the changes in nucleotide degradation were greatly affected by storage temperature. The K value of GCFS exceeded the recommended limit value (60%) after 7 d of storage at 4 ℃ or 3 d of storage at 10 ℃, meaning that the samples entered the stale state. The results of sensory evaluation showed a negative correlation between the quality and storage time, with the phenomena of textural softening, flesh discoloration and fishy-smell aggravation in raw/cooked GCFS stored at 10 ℃ for 3 d and 4 ℃ for 5 d. Considering the results of various indicators and the pursuit of high edible quality, the suggested shelf life of GCFS at 4 ℃ and 10 ℃ was 3 d and 1 d, respectively.

Nickel-based single-crystal superalloys are designed for extreme conditions due to their superior corrosion and creep resistance properties. However, these pose challenges in the subsequent recycling after reaching their end-of-life. Molten magnesium (Mg) can rapidly corrode the stable spent nickel-based superalloys and selectively dissolve nickel (Ni). This waste-free process represents an effective method for recycling spent superalloys and accomplishing metal regeneration. This study investigates the mechanism of selectively dissolving Ni from DD5, a nickel-based single-crystal superalloy, by optimizing process temperature, time, and Mg content in an inert atmosphere. Vacuum distillation was employed to separate the resulting Mg, residual superalloy (i.e., the material left post-extraction), and Ni-rich alloy (i.e., the metal product selectively extracted). The findings revealed that the residual superalloy after selective Ni dissolution is characterized by a porous skeleton structure with pore sizes predominantly ranging from 2 to 30 nm and a low compressive strength which is 1/10 of the original DD5 superalloy.

The impact of eating behaviors on physical fitness and overall health is widely recognized. However, the specific effect of late-night eating on physical fitness and the relationship between unhealthy eating habits and muscle strength in adolescents have not been explored yet. This study was designed to investigate the relationship between eating behaviors and muscle strength among Chinese male adolescents. The eating habits of 1916 high school students (mean age = 16.7 ± 0.8) were assessed by a questionnaire comprising items related to the consumption of breakfast and frequency of late-night food intake. Muscle strength was measured by grip strength (upper body strength) and standing long jump (lower body strength) tests. To discern the relationship between eating behaviors and muscle strength, a covariance analysis (ANCOV) was performed by making adjustments for potential confounding variables. The final adjusted model suggested a significant relationship between grip strength and the frequency of breakfast consumption (a higher frequency was related with better grip strength; p < 0.001) and late-night eating (a lower frequency was linked with better grip strength; p = 0.002). Few unhealthy eating behaviors were associated with better grip strength and standing long jump performance (p < 0.001 and p = 0.026, respectively). These findings suggest that consumption of breakfast, reduction of the late-night eating, and decreased unhealthy eating habits may improve muscle strength and function in male adolescents.

Abstract Background Although some studies have examined the association between exercise and falls, most have focused on specific exercises, and the results have been inconsistent. In addition, there is a lack of evidence on elderly Chinese women who have different living and exercise habits compared to those in other countries. Therefore, this study aimed to investigate whether physical exercise is associated with falls in elderly Chinese women. Methods This cross-sectional study included 1429 elderly Chinese women with a mean age of 69.2 years. Information on physical exercise habits and fall experiences was collected using a self-report questionnaire. Logistic regression models were used to analyze the association between physical exercise habits and falls. Results The results showed that 15% participants had a fall in the past year. After adjusting for confounding factors, the odd ratios (ORs) and 95% Confidence Intervals (CIs) for fall experiences across categories of exercise frequency were as follow: 1 (reference) for no exercise behavior, 0.50 (0.29, 0.85) for exercise 1 to 5 times a week, and 0.37 (0.25, 0.55) for exercise more than 6 times a week. Furthermore, the ORs (95% CIs) across categories of exercise insistence were 1 (reference) for less than 1 year, 0.78 (0.37, 1.65) for 1 to 3 years, and 0.38 (0.20, 0.74) for more than 3 years. In terms of exercise duration, the ORs (95% CIs) for 2 h/day were 1 (reference), 0.85 (0.53, 1.36), and 2.80 (1.30, 6.05). Unlike other variables, longer exercise duration was associated unfavorably with falls. Conclusion Physical exercise habits were associated with falls in elderly Chinese women. Keeping a proper exercise habit may contribute to lower risk of falling in elderly women.

Chitosan oligosaccharides are the degradation products of chitin obtained from the shell extracts of shrimps and crabs. Compared with chitosan, chitosan oligosaccharides have better solubility and a wider application range. In this study, high-molecular-weight chitosan oligosaccharides (COST, chitosan oligosaccharides, MW ≤ 1000) were isolated and purified by a GPC gel column, and the molecular weight range was further reduced to obtain high-purity and low-molecular-weight chitosan (COS46). Compared with COST, COS46 is better at inhibiting CCl4-induced cell death, improving cell morphology, reducing ALT content, and improving cell antioxidant capacity. The effects of COST and COS46 on CCl4-induced acute liver injury were further verified in mice. Both COS46 and COST improved the appearance of the liver induced by CCl4, decreased the levels of ALT and AST in serum, and decreased the oxidation/antioxidant index in the liver. From the liver pathological section, the effect of COS46 was better. In addition, some indicators of COS46 showed a dose-dependent effect. In conclusion, compared with COST, low-molecular-weight COS46 has better antioxidant capacity and a better therapeutic effect on CCl4-induced acute liver injury.

Sulforaphane (SFN), a bioactive phytocompound extracted from cruciferous plants, has received increasing attention due to its vital cytoprotective role in eliminating oxidative free radical through activation of nuclear factor erythroid 2-related factor (Nrf2)-mediated signal transduction pathway. This study aims at a better insight into the protective benefit of SFN in attenuating paraquat (PQ)-caused impairment in bovine in vitro-matured oocytes and the possible mechanisms involved therein. Results showed that addition of 1 μM SFN during oocyte maturation obtained higher proportions of matured oocytes and in vitro-fertilized embryos. SFN application attenuated the toxicological effects of PQ on bovine oocytes, as manifested by enhanced extending capability of cumulus cell and increased extrusion proportion of first polar body. Following incubation with SFN, oocytes exposed to PQ exhibited reduced intracellular ROS and lipid accumulation levels, and elevated T-SOD and GSH contents. SFN also effectively inhibited PQ-mediated increase in BAX and CASPASE-3 protein expressions. Besides, SFN promoted the transcription of NRF2 and its downstream antioxidative-related genes GCLC, GCLM, HO-1, NQO-1, and TXN1 in a PQ-exposed environment, indicating that SFN prevents PQ-caused cytotoxicity through activation of Nrf2 signal transduction pathway. The mechanisms underlying the role of SFN against PQ-induced injury included the inhibition of TXNIP protein and restoration of the global O-GlcNAc level. Collectively, these findings provide novel evidence for the protective role of SFN in alleviating PQ-caused injury, and suggest that SFN application may be an efficacious intervention strategy against PQ cytotoxicity.

Abstract Background Mice with humanized livers are important models to study drug toxicology testing, development of hepatitis virus treatments, and hepatocyte transplantation therapy. However, the huge difference between mouse and human in size and anatomy limited the application of humanized mice in investigating human diseases. Therefore, it is urgent to construct humanized livers in pigs to precisely investigate hepatocyte regeneration and human hepatocyte therapy. CRISPR/Cas9 system and somatic cell cloning technology were used to generate two pig models with FAH deficiency and exhibiting severe immunodeficiency (FAH/RAG1 and FAH/RAG1/IL2RG deficiency). Human primary hepatocytes were then successfully transplanted into the FG pig model and constructed two pigs with human liver. Results The constructed FAH/RAG1/IL2RG triple-knockout pig models were characterized by chronic liver injury and severe immunodeficiency. Importantly, the FG pigs transplanted with primary human hepatocytes produced human albumin in a time dependent manner as early as 1 week after transplantation. Furthermore, the colonization of human hepatocytes was confirmed by immunochemistry staining. Conclusions We successfully generated pig models with severe immunodeficiency that could construct human liver tissues.

Nonalcoholic fatty liver disease (NAFLD) is primarily caused by abnormal lipid metabolism and the accumulation of triglycerides in the liver. NAFLD is also associated with hepatic steatosis and nutritional and energy imbalances and is a chronic liver disease associated with a number of factors. Nuclear receptors play a key role in balancing energy and nutrient metabolism, and the peroxisome proliferator-activated receptor alpha (PPARα) and farnesoid X receptor (FXR) regulate lipid metabolism genes, controlling hepatocyte lipid utilization and regulating bile acid (BA) synthesis and transport. They play an important role in lipid metabolism and BA homeostasis. At present, PPARα and FXR are the most promising targets for the treatment of NAFLD among nuclear receptors. This review focuses on the crosstalk mechanisms and transcriptional regulation of PPARα and FXR in the pathogenesis of NAFLD and summarizes PPARα and FXR drugs in clinical trials, laying a theoretical foundation for the targeted treatment of NAFLD and the development of novel therapeutic strategies.

Hydrogen reduction is becoming a promising method for recycling lithium-ion battery cathode materials. However, the reaction mechanism and kinetics during hydrogen reduction are unclear, requiring further investigation. Therefore, non-isothermal and isothermal reduction experiments were conducted to evaluate the temperature dependence of the hydrogen reduction kinetics using simultaneous thermogravimetric and differential thermal analysis equipped with mass spectrometry. XRD and SEM were used to characterize the reduction products to understand the underlying reduction mechanisms. The hydrogen reduction profile could be divided into three main stages: decomposition of cathode materials, reduction of the resultant nickel and cobalt oxides, and reduction of LiMnO2 and residual nickel and cobalt oxides. The hydrogen reduction rate increased with increasing temperature, and 800°C was the optimum temperature for separating the magnetic Ni-Co alloy from the non-magnetic manganese oxide particles. The apparent activation energy for the isothermal tests in the range of 500–700°C was 84.86 kJ/mol, and the rate-controlling step was the inward diffusion of H2(g) within each particle. There was an downward progression of the reduction through the material bed for the isothermal tests in the range of 700–900°C, with an apparent activation energy of 51.82 kJ/mol.

Correct reprogramming of the DLK1-DIO3 imprinted region is critical for the development of cloned animals. However, in pigs, the imprinting and regulation of the DLK1-DIO3 region has not been systematically analyzed. The objective of this study was to investigate the imprinting status and methylation regulation of the DLK1-DIO3 region in wild-type and cloned neonatal pigs. We mapped the imprinting control region, IG-DMR, by homologous alignment and validated it in sperm, oocytes, fibroblasts, and parthenogenetic embryos. Subsequently, single nucleotide polymorphism-based sequencing and bisulfite sequencing polymerase chain reaction were conducted to analyze imprinting and methylation in different types of fibroblasts, as well as wild-type and cloned neonatal pigs. The results showed that Somatic cell nuclear transfer (SCNT) resulted in hypermethylation of the IG-DMR and aberrant gene expression in the DLK1-DIO3 region. Similar to wild-type pigs, imprinted expression and methylation were observed in the surviving cloned pigs, whereas in dead cloned pigs, the IG-DMR was hypermethylated and the expression of GTL2 was nearly undetectable. Our study reveals that abnormal imprinting of the DLK1-DIO3 region occurs in cloned pigs, which provides a theoretical basis for improving the cloning efficiency by gene editing to correct abnormal imprinting.

The extensive changes in public and private life caused by the ongoing COVID-19 epidemic in China have created a “new normal.” To understand urban residents’ satisfaction with public sports services under these conditions and to identify factors that influence satisfaction, urban residents in several major Chinese cities were randomly sampled with an online questionnaire to measure their level of satisfaction with public sports services in China during the COVID-19 epidemic. With the response rate of 87.9%, 703 valid questionnaires out of 800 questionnaires distributed were analyzed. A structural equation model was constructed where health literacy and the residents’ expectations were independent variables, residents’ participation was a mediating variable, and residents’ satisfaction was the dependent variable. Cronbach’s α test and Kaiser-Meyer-Olkin test confirmed that the questionnaire was reliable and valid. Survey results suggest that young urban residents’ participation in public sports services is influenced by personal health literacy and residents’ expectations. Participation serves a mediating role between health literacy and residents’ satisfaction, but not between residents’ expectations and their satisfaction. Young urban residents’ satisfaction with public sports services may be improved by increasing access to health information, improving hardware and software platforms to support those services, and catering the services to the interests of the population.

Intestinal environment disorder is a potential pathological mechanism of obesity. There is increasing evidence that disorders in the homeostasis of the intestinal environment can affect various metabolic organs, such as fat and liver, and lead to metabolic diseases. However, there are few therapeutic approaches for obesity targeting the intestinal environment. In this review, on the one hand, we discuss how intestinal microbial metabolites SCFA regulate intestinal function to improve obesity and the possible mechanisms and pathways related to obesity-related pathological processes (depending on SCFA-related receptors such as GPCRs, MCT and SMCT, and through epigenetic processes). On the other hand, we discuss dietary management strategies to enrich SCFA-producing bacteria and target specific SCFA-producing bacteria and whether fecal bacteria transplantation therapy to restore the composition of the gut microbiota to regulate SCFA can help prevent or improve obesity. Finally, we believe that it will be of great significance to establish a working model of gut– SCFA– metabolic disease development in the future for the improvement this human health concern.

Background: Functional training is any type of training designed to improve a specific movement or activity for fitness or high performance sports. This study examined the effect of functional training on the strength and power of young tennis players. Methods: 40 male tennis players were assigned to the functional training group (n = 20; age, 16.7 ± 0.4 years) or the conventional training group (n = 20; age, 16.5 ± 0.6 years). The functional training group received three 60 min sessions per week for 12 weeks, while the conventional training group participated in three sessions per week of mono-strength exercise for 12 weeks. Strength and power were measured according to the International Tennis Federation protocol at baseline, 6 weeks after the intervention, and 12 weeks after the intervention. Results: Both forms of training increased (p < 0.05) push-ups, wall squat test, over medicine ball throw, and standing long jump after 6 weeks of training, and the effect improved further as the 12-week mark approached. Except for the wall squat test (left) at 6 weeks, functional training showed no advantage over conventional training. After an additional 6 weeks of training, all measures of strength and power were better (p < 0.05) in the functional training group. Conclusions: Improvements in strength and power could occur after as little as 6 weeks of functional training, and 12-week functional training could outperform conventional training in male adolescent tennis players.

In the development process of the blanket system, assembly design is quite important, and suitable systematic methodologies are required. As we know, the CFETR machine and the associated fusion components are not usually mass-produced large-scale products, but highly customized machines which are still in the design phase. Appropriate assembly methodology plays an important role in fulfilling the function of the fusion machine. This paper has investigated some universal assembly methods for similar complex products. Two preferred methods of design-for-assembly (DFA) and product-process hierarchical modeling (PPHM) have been analyzed and improved taking the fusion blanket system as a study case. The overall process of the blanket system was studied including the stages of design, assembly, and overview of the blanket system hierarchy structure. The two newly proposed methods aim to clarify a probably feasible assembly method for the blanket system, though it is still in the engineering design stage. Case studies of the two favorable assembly methods can be good references to demonstrate and analyze the advantages of DFA and PPHM for decision-making in each product development phase.

Abstract Blastocyst complementation by pluripotent stem cell (PSC) injection is believed to be the most promising method to generate xenogeneic organs. However, ethical issues prevent the study of human chimeras in the late embryonic stage of development. Primate embryonic stem cells (ESCs), which have similar pluripotency to human ESCs, are a good model for studying interspecies chimerism and organ generation. However, whether primate ESCs can be used in xenogenous grafts remains unclear. In this study, we evaluated the chimeric ability of cynomolgus monkey (Macaca fascicularis) ESCs (cmESCs) in pigs, which are excellent hosts because of their many similarities to humans. We report an optimized culture medium that enhanced the anti-apoptotic ability of cmESCs and improved the development of chimeric embryos, in which domesticated cmESCs (D-ESCs) injected into pig blastocysts differentiated into cells of all three germ layers. In addition, we obtained two neonatal interspecies chimeras, in which we observed tissue-specific D-ESC differentiation. Taken together, the results demonstrate the capability of D-ESCs to integrate and differentiate into functional cells in a porcine model, with a chimeric ratio of 0.001–0.0001 in different neonate tissues. We believe this work will facilitate future developments in xenogeneic organogenesis, bringing us one step closer to producing tissue-specific functional cells and organs in a large animal model through interspecies blastocyst complementation.

Valbenazine and deutetrabenazine are the only two therapeutic drugs approved for tardive dyskinesia based on blocking the action of vesicular monoamine transporter 2 (VMAT2). But there exist demethylated inactive metabolism at the nine position for both them resulting in low availability, and CYP2D6 plays a major role in this metabolism resulting in the genetic polymorphism issue. 9-trifluoroethoxy-dihydrotetrabenazine (13e) was identified as a promising lead compound for treating tardive dyskinesia. In this study, we separated 13e via chiral chromatography and acquired R,R,R-13e [(+)-13e] and S,S,S-13e [(−)-13e], and we investigated their VMAT2-inhibitory activity and examined the related pharmacodynamics and pharmacokinetics properties using in vitro and in vivo models (+)-13e displayed high affinity for VMAT2 (Ki = 1.48 nM) and strongly inhibited [3H]DA uptake (IC50 = 6.11 nM) in striatal synaptosomes. Conversely, its enantiomer was inactive. In vivo, (+)-13e decreased locomotion in rats in a dose-dependent manner. The treatment had faster, stronger, and longer-lasting effects than valbenazine at an equivalent dose. Mono-oxidation was the main metabolic pathway in the liver microsomes and in dog plasma after oral administration, and glucuronide conjugation of mono-oxidized and/or demethylated products and direct glucuronide conjugation were also major metabolic pathways in dog plasma. O-detrifluoroethylation of (+)-13e did not occur. Furthermore, CYP3A4 was identified as the primary isoenzyme responsible for mono-oxidation and demethylation metabolism, and CYP2C8 was a secondary isoenzyme (+)-13e displayed high permeability across the Caco-2 cell monolayer, and it was not a P-glycoprotein substrate as demonstrated by its high oral absolute bioavailability (75.9%) in dogs. Thus, our study findings highlighted the potential efficacy and safety of (+)-13e in the treatment of tardive dyskinesia. These results should promote its clinical development.

An anaerobic ceramic membrane bioreactor (AnCMBR) with thermally-hydrolyzed sewage sludge as the inoculum was started up at the ambient temperature (26.97 ± 4.23 °C) for treating simulated domestic wastewater. The AnCMBR was operated for 60 days under 24 h hydraulic retention time (HRT), 50 days sludge retention time (SRT), and the mean organic loading rate (OLR) of 0.42 kgCODm−3d−1. Soluble chemical oxygen demand (SCOD) removal evaluation, volatile fatty acid (VFA), and Alkalinity (ALK) analyses, and the microbial community evaluation by polymerase chain reaction (PCR) were conducted. The reported maximum and mean SCOD removals were 62%, 43.54% ± 11.61 respectively. The total volatile fatty acids/alkalinity (TVFA/ALK) ratio was 0.59 indicating system instability. Microbial community analysis revealed significant shifts during the start-up process. The Bacteroidetes were well acclimatized throughout the start-up. The Firmicutes had suppressed during the start-up while Proteobacteria, Epsilonbacteraeota, Spirochetes phylum newly emerged in the system. The thermally-hydrolyzed inoculum was not established as a successful microbial community for treating domestic wastewater at the ambient conditions in this AnCMBR. The findings provided insights on microbial mechanisms and the basis of anaerobic digestion process instability. In comparison with previous studies, the potential countermeasures to address this were identified as proper selection and frequent addition of inoculum, and process monitoring.

Alterations of apoptosis have notable influences on flesh quality, but the mechanism is still unclear. Thus, apoptotic behaviors and related triggering mechanisms need to be explored. Fish muscle was prepared and stored at 4 °C for 0, 24, 48, 72, 96, and 120 h for apoptosis analysis. Results showed that positive apoptotic nuclei were positively correlated with drop loss and negatively correlated with shear force and water holding capacity (p < 0.05). Results showed that the triggering apoptotic mechanisms were involved with enhanced transcriptional levels of caspase-2, 3, 7, 8, and 9 through mitochondria and death receptor pathways in the muscle of grass carp. The decreased ATP content, changed cytochrome c redox state, increased protein levels of HSP27 and HSP 90, and enhanced activity of cathepsin (B, L, and D), calpain, and serine proteinase were involved in apoptosis activations. Results indicated that caspases, energy metabolism, cytochrome c redox state, heat shock protein expressions, and protease activities played critical roles in apoptosis alterations in carp muscle during refrigerated storage.

This study aims to investigate the effects of CLAUDIN-6 (CLDN6) on cell apoptosis and proliferation of bovine cumulus cells (CCs). Immunofluorescence staining was used to localize CLDN6 protein in CCs. Three pairs of siRNA targeting CLDN6 and one pair of siRNA universal negative sequence as control were transfected into bovine CCs. Then, the effective siRNA was screened by real-time quantitative PCR (RT-qPCR) and Western blotting. The mRNA expression levels of apoptosis related genes (CASPASE-3, BAX and BCL-2) and proliferation related genes (PCNA, CDC42 and CCND2) were evaluated by RT-qPCR in CCs with CLDN6 knockdown. Cell proliferation, apoptosis and cell cycle were detected by flow cytometry with CCK-8 staining, Annexin V-FITC staining and propidium iodide staining, respectively. Results showed that the CLDN6 gene was expressed in bovine CCs and the protein was localized in cell membranes and cytoplasms. After CLDN6 was knocked down in CCs, the cell apoptosis rate significantly decreased and the pro-apoptotic genes BAX and CASPASE-3 were down-regulated significantly, whereas the anti-apoptotic gene BCL-2 was markedly up-regulated (p < 0.05). Additionally, CLDN6 knockdown significantly enhanced cell proliferation of CCs at 72 h after siRNA transfection. The mRNA levels of proliferation-related genes PCNA, CCND2 and CDC42 increased obviously in CCs with CLDN6 knockdown (p < 0.05). After CLDN6 was down-regulated, the percentage of CCs at S phase was significantly increased (p < 0.05). However, there was no remarkable difference in the percentages of cells at the G0/G1 phase and G2/M phase between CCs with or without CLDN6 knockdown (p > 0.05). Therefore, the expression of CLDN6 and its effects on cell proliferation, apoptosis and cell cycle of bovine CCs were first studied. CLDN6 low expression inhibited cell apoptosis, induced cell proliferation and cell cycle arrest of bovine CCs.

Although dissolved air flotation (DAF) plays a crucial role in wastewater treatment, fluid dynamics in the contact zone (CZ) of a DAF tank are still considered black-box processes, which hampers DAF's optimal design and operation. In this article, we perform a modeling study to further unravel the governing mechanisms occurring in a DAF tank. A computational fluid dynamics (CFD) model coupled with a generic population balance model (PBM) for the DAF system was developed in this work. CFD-PBM results showed that the eddy capture is the dominating mechanism for the bubble coalescence in the most volume of the CZ. Simultaneously, the velocity gradient mechanism and turbulent induced mechanism also play a key role in bubble coalescence in the regions with drastic flow transition and in the nozzle downstream. CFD-PBM simulations reveal that increasing the recirculation rate is better than increasing the volume fraction in the smaller recirculated flow to achieve higher bubble number density and smaller bubble size if the same amount of gas flow rate is injected. The baffle with proper transverse corrugate can equalize and reduce the total coalescence rate. By establishing a synergy between fluid dynamics and bubble size to achieve the best CZ efficiency, the generic CFD-PBM approach developed in this study has laid a solid basis for the optimal design and operation of DAF systems.

Abstract Background Inherited factors contribute to lung cancer risk, but the mechanism is not well understood. Defining the biological consequence of GWAS hits in cancers is a promising strategy to elucidate the inherited mechanisms of cancers. The tag-SNP rs753955 (A>G) in 13q12.12 is highly associated with lung cancer risk in the Chinese population. Here, we systematically investigate the biological significance and the underlying mechanism behind 13q12.12 risk locus in vitro and in vivo. Results We characterize a novel p53-responsive enhancer with lung tissue cell specificity in a 49-kb high linkage disequilibrium block of rs753955. This enhancer harbors 3 highly linked common inherited variations (rs17336602, rs4770489, and rs34354770) and six p53 binding sequences either close to or located between the variations. The enhancer effectively protects normal lung cell lines against pulmonary carcinogen NNK-induced DNA damages and malignant transformation by upregulating TNFRSF19 through chromatin looping. These variations significantly weaken the enhancer activity by affecting its p53 response, especially when cells are exposed to NNK. The effect of the mutant enhancer alleles on TNFRSF19 target gene in vivo is supported by expression quantitative trait loci analysis of 117 Chinese NSCLC samples and GTEx data. Differentiated expression of TNFRSF19 and its statistical significant correlation with tumor TNM staging and patient survival indicate a suppressor role of TNFRSF19 in lung cancer. Conclusion This study provides evidence of how the inherited variations in 13q12.12 contribute to lung cancer risk, highlighting the protective roles of the p53-responsive enhancer-mediated TNFRSF19 activation in lung cells under carcinogen stress.

Background: Insulin-like growth factor 2 is a growth-promoting factor that plays an important role in the growth and development of mammals. A nucleotide substitution in intron 3 of IGF2—which disrupts the ZBED6-binding site—affects muscle mass, organ size, and fat deposition in pigs. The ZBED6-binding site is also conserved in cattle. Methods: In the present study, we introduced mutations in the ZBED6-binding site in intron3 of IGF2 in bovine fetal fibroblasts using the CRISPR/Cas9 system, and investigated the effect of disruption of ZBED6 binding on IGF2 expression. Results: Eleven biallelic-mutant single-cell clones were established, three of which contained no foreign DNA residues. Single-cell clones 93 and 135 were used to produce cloned embryos. Dual-luciferase reporter assay in C2C12 cells demonstrated that the mutation in the ZBED6-binding site increases the promoter 3 activity of bovine IGF2. A total of 49 mutant cloned embryos were transplanted into surrogate cows. Unfortunately, all cloned embryos died before birth. IGF2 was found to be hypomethylated in the only fetus born (stillborn), which may have been due to the incomplete reprogramming. Conclusions: We efficiently constructed IGF2-edited cell lines and cloned embryos, which provided a theoretical basis and experimental materials for beef cattle breeding.

Animal cloning is of great importance to the production of transgenic and genome-edited livestock. Especially for multiple gene-editing operations, recloning is one of the most feasible methods for livestock. In addition, a multiple-round cloning method is practically necessary for animal molecular breeding. However, cloning efficiency remains extremely low, especially for serial cloning, which seriously impedes the development of livestock breeding based on genome editing technology. The incomplete reprogramming and failure in oocyte activation of some pluripotent factors were deemed to be the main reason for the low efficiency of animal recloning. Here, to overcome this issue, which occurred frequently in the process of animal recloning, we established a reporter system in which fluorescent proteins were driven by pig OCT4 or SOX2 promoter to monitor the reprogramming process in cloned and recloned pig embryos. We studied the effect of different histone deacetylase (HDAC) inhibitors on incomplete reprogramming. Our results showed that Trichostatin A (TSA) could activate pluripotent factors and significantly enhance the development competence of recloned pig embryos, while the other two inhibitors, valproic acid (VPA) and Scriptaid, had little effect on that. Furthermore, we found no difference in OCT4 mRNA abundance between TSA-treated and untreated embryos. These findings suggest that TSA remarkably improves the reprogramming state of pig recloned embryos by restoring the expression of incompletely activated pluripotent genes OCT4 and SOX2.

Low birth efficiency and developmental abnormalities in embryos derived using round spermatid injection (ROSI) limit the clinical application of this method. Further, the underlying molecular mechanisms remain elusive and warrant further in-depth study. In this study, the embryonic day (E) 11.5 mouse fetuses and corresponding placentas derived upon using ROSI, intracytoplasmic sperm injection (ICSI), and natural in vivo fertilized (control) embryos were collected. Transcriptome and DNA methylation profiles were analyzed and compared using RNA-sequencing (RNA-seq) and whole-genome bisulfite sequencing, respectively. RNA-seq results revealed similar gene expression profiles in the ROSI, ICSI, and control fetuses and placentas. Compared with the other two groups, seven differentially expressed genes (DEGs) were identified in ROSI fetuses, and ten DEGs were identified in the corresponding placentas. However, no differences in CpG methylation were observed in fetuses and placentas from the three groups. Imprinting control region methylation and imprinted gene expression were the same between the three fetus and placenta groups. Although 49 repetitive DNA sequences (RS) were abnormally activated in ROSI fetuses, RS DNA methylation did not differ between the three groups. Interestingly, abnormal hypermethylation in promoter regions and low expression of Fggy and Rec8 were correlated with a crown-rump length less than 6 mm in one ROSI fetus. Our study demonstrates that the transcriptome and DNA methylation in ROSI-derived E11.5 mouse fetuses and placentas were comparable with those in the other two groups. However, some abnormally expressed genes in the ROSI fetus and placenta warrant further investigation to elucidate their effect on the development of ROSI-derived embryos.

Jack-up offshore platform is a type of important marine structure, which is mainly used for satellite launch, oil exploitation, and other engineering tasks in the offshore area. The offshore platform is bound to be subjected to wave loading in the course of use. Whether it can withstand the wave impact is an important engineering problem. To solve this engineering problem, the self-developed fluid–structure–foundation interaction coupling model OlaFlow-ABAQUS is used to explore the dynamic response characteristics of a jack-up offshore platform and its seabed foundation under three conventional wave conditions (wave height is 3, 5, and 7 m, respectively) in a coupled way. The numerical results show that only a small amplitude of periodic sloshing occurs for the jack-up offshore platform under the three conventional wave conditions. The maximum sloshing amplitude is up to 8 cm, and there is no visible residual displacement. It is indicated that there is no plastic deformation zone in the seabed foundation near the pile legs of the jack-up platform. It can thus be concluded that the jack-up platform has excellent stability under conventional wave conditions. Under conventional wave loading, momentary liquefaction occurs in the seabed foundation around the pile legs of the platform, and the maximum liquefaction depth is about 1 m. This study indicates that the coupling model OlaFlow-ABAQUS for the fluid–structure–foundation interaction is feasible, and has some advantages to study the dynamic response and to evaluate the stability of large-scale marine structures and their seabed foundations under ocean waves.

Chitosan obtained from abundant marine resources has been proven to have a variety of biological activities. However, due to its poor water solubility, chitosan application is limited, and the degradation products of chitosan oligosaccharides are better than chitosan regarding performance. Chitosan oligosaccharides have two kinds of active groups, amino and hydroxyl groups, which can form a variety of derivatives, and the properties of these derivatives can be further improved. In this review, the key structures of chitosan oligosaccharides and recent studies on chitosan oligosaccharide derivatives, including their synthesis methods, are described. Finally, the antimicrobial and antitumor applications of chitosan oligosaccharides and their derivatives are discussed.

Swine wastewater is an important reservoir of spread antibiotic resistance to the environment. Intra- and extracellular antibiotic resistance genes (iARGs and eARGs) were quantified during two typical swine wastewater treatment processes including a sequencing membrane bioreactor (SMBR) at pilot-scale and anaerobic-anoxic-oxic (A2O) at full-scale. The concentrations of iARGs and eARGs in raw wastewater were 3.42E+09 and 3.79E+07 copies/mL, respectively. The compositions were different between iARGs and eARGs. SMBR showed 0.63 log higher removals in the concentrations of iARG than A2O, while similar removal effects (3.01–3.44 log copies/mL) of eARGs were performed by the two processes. It suggested that membrane separation had advantages in the concentration removals of iARG rather than eARG. sul1 took the dominance in eARGs in effluent and had positive correlations with intI1, which indicated the risk of horizontal gene transfer of eARGs after wastewater discharge. Microbial community structures were estimated by 16S rRNA gene sequencing with both intra- and extracellular DNA (iDNA and eDNA). Compared between the effluent samples of the two treatment processes, microbial community structures estimated by iDNA had great differences, however which were similar for eDNA. Microbial community and water-quality parameters were the major influencing factors on ARG occurrences during swine wastewater treatment. This study investigated the effects of liquid nitrogen quick-freezing, −35°C air-blast freezing, and −18°C refrigerator direct-freezing on quality retention of frozen giant freshwater prawns using various indicators: microscopic structure, physicochemical, texture, and color properties. The results showed that the shelf lives of prawns treated with liquid nitrogen quick-freezing, −35°C air-blast freezing, and −18°C refrigerator direct-freezing were about 20, 12, and 8 weeks, respectively, in terms of total volatile basic nitrogen content and K-value. Meanwhile, liquid nitrogen quick-freezing could effectively improve texture property of frozen giant freshwater prawn. These findings confirm that liquid nitrogen quick-freezing is a promising method to extend the shelf life of frozen prawns.

Bearing significant concentrations of high value and critical metals, superalloy scraps require comprehensive recycling for metal reclamation. In this study, nickel-based superalloy was treated with molten Mg-Zn for the selective extraction of nickel. The influence of heating temperature, the molar ratio of Mg to Zn in the molten metal, Mg-Zn/superalloy mass ratio, and heating time on metal extraction were investigated. Using the heating temperature of 800 °C, the Mg/Zn molar ratio of 9/1, the Mg-Zn/superalloy mass ratio of 5/1, and heating time of 240 min, the extraction rate of 97.1% was achieved for Ni, and the extraction rates of Fe, Cr and refractory metals (Nb, Mo and Ti) were all less than 1%. In the subsequent vacuum distillation process, nickel with a purity of 98.3 wt% was obtained. Therefore, the proposed method is a short, clean, and efficient process for selectively extracting nickel from the superalloy scraps.

Abstract We propose and experimentally demonstrate spatial rotation manipulation for radially asymmetric modes based on two kinds of polarization maintaining few-mode fibers (PM-FMFs). Theoretical finding shows that due to successful suppression of both polarization and spatial mode coupling, the spatial rotation of radially asymmetric modes has an excellent linear relationship with the twist angle of PM-FMF. Both elliptical core and panda type FMFs are fabricated, in order to realize manageable spatial rotation of LP11 mode within ±360° range. Finally, we characterize individual PM-FMF based spatial orientation rotator and present comprehensive performance comparison between two PM-FMFs in terms of insertion loss, temperature sensitivity, linear polarization maintenance, and mode scalability.

Rock material has different mechanical behaviors under compressive and tensile loading. Correspondingly, there are two types of elastic modulus: compressive elastic modulus Ec and tensile elastic modulus Et, respectively. To distinguish which indirect test methodology, including three-points bending test and Brazilian disc test, is more suitable to measure the tensile elastic modulus Et of rock materials, a series of uniaxial compressive test (UCT), direct tensile test (DTT), three-points bending test, and Brazilian disc test are performed for three typical types of rock: marble, granite, and sandstone. Comparative investigation on the reliability of measurement results of tensile elastic modulus Et is systematically conducted. Finally, it is found that Brazilian disc test could be a suitable method to measure tensile elastic modulus of rock materials, due to the excellent agreement with that measured by DTT and the simplicity of sample preparation, as well as test operation.

A high concentration of suspended solids (SS) in swine wastewater reduces the efficiency of the biological treatment process. The current study developed a short-cut combined magnetic coagulation (MC)–sequence batch membrane bioreactor (SMBR) process to treat swine wastewater. Compared with the single SMBR process, the combined process successfully achieved similarly high removal efficiencies of chemical oxygen demand (COD), total nitrogen (TN), ammonium nitrogen (NH4+-N), and total phosphorous (TP) of 96.0%, 97.6%, 99.0%, and 69.1%, respectively, at dosages of 0.5 g/L of poly aluminium chloride (PAC), 2 mg/L of polyacrylamide (PAM), and 1 g/L of magnetic seeds in Stage II, and concentrations of TN, COD, and NH4+-N in effluent can meet the discharge standards for pollutants for livestock and poultry breeding (GB18596-2001, China). The nitrogen removal loading (NRL) was increased from 0.21 to 0.28 kg/(m3·d), and the hydraulic retention time (HRT) was shortened from 5.0 days to 4.3 days. High-throughput sequencing analysis was carried out to investigate microbial community evolution, and the results showed that the relative abundance of ammonia-oxidizing bacteria (AOB) in the SMBR increased from 0.1% without pre-treatment to 1.78% with the pre-treatment of MC.

A method for predicting the long-term effects of ferric on methane production was developed in an anaerobic membrane bioreactor treating food processing wastewater to provide management tools for maximizing methane recovery using ferric based on a batch test. The results demonstrated the accuracy of the predictions for both batch and long-term continuous operations using a Bayesian network meta-analysis based on the Gompertz model. The prediction bias of methane production for batch and continuous operations was minimized, from 11~19% to less than 0.5%. A biochemical methane potential-based Bayesian network meta-analysis suggested a maximum 2.55% ± 0.42% enhancement for Fe2.25. An anaerobic membrane bioreactor improved the methane yield by 2.27% and loading rate by 4.57% for Fe2.25, operating in the sequenced batch mode. The method allowed for a predictable methane yield enhancement based on the biochemical methane potential. Ferric enhanced the biochemical methane potential in batch tests and the methane yield in a continuously operated reactor by a maximum of 8.20% and 7.61% for Fe2.25, respectively. Copper demonstrated a higher methane (18.91%) and sludge yield (17.22%) in batch but faded in the continuous operation (0.32% of methane yield). The enhancement was primarily due to changing the kinetic patterns for the last period, i.e., increasing the second methane production peak (k71), bringing forward the second peak (λ7, λ8), and prolonging the second period (k62). The dual exponential function demonstrated a better fit in the last three stages (after the first peak), which implied that syntrophic methanogenesis with a ferric shuttle played a primary role in the last three methane production periods, in which long-term effects were sustained, as the Bayesian network meta-analysis predicted.

We propose and experimentally demonstrate an all-fiber tunable LP11 mode rotator with 360° range, based on the bending and twisting of the two-mode fiber (TMF). A theoretical model of TMF perturbation and its effect on the spatial pattern of LP11 mode is put forward. We experimentally characterize the implementation of the LP11 mode rotator by an all-fiber polarization controller configuration. The results agree well with the theoretical investigations. We are able to demonstrate arbitrary LP11 mode rotation with 360° range. The insertion loss is less than 0.4 dB when the operation wavelength is varied from 1540 to 1560 nm.

The effects of solid retention times (SRTs) (100 days, 50 days, 25 days) on the performance, microbial community, and membrane fouling of a lab-scale anaerobic yttria-based ceramic membrane bioreactor (AnCMBR) treating synthetic domestic wastewater at ambient temperature (31.2 ± 2.7 °C) were examined. The soluble chemical oxygen demand (SCOD) removal was higher (89.6%) at 25 days SRT compared with 50 days (39.61%) and 100 days (34.3%) SRT. At 100 days SRT, more Bacteroidetes, Firmicutes, and Proteobacteria were present in the microbial community. At 25 days SRT, more Chloroflexi, Synergistetes, and Pastescibacteria emerged, contributing to the stable performance. The SRT of 25 days has resulted in a more stable microbial community compared with 50 days and 100 days SRT. Both bacterial and archaeal community diversities were higher at 25 days SRT, and the specific production of soluble microbial by-products (SMPs) and extracellular polymeric substances (EPSs) were higher at 25 days SRT as well. Consequently, the membrane flux was lower at 25 days SRT with the increased particle size and the enhanced SMPs and EPSs production. Fourier transform infrared spectroscopy analysis (FTIR) and three-dimensional excitation and emission matrix (3D-EEM) analysis showed that protein and SMPs were the major membrane foulants at all SRT stages. In this study, SRT at 25 days was favorable for the stable operation of an AnCMBR treating domestic wastewater at ambient temperature.

Metallurgical processing of low-grade manganese ore with high iron content is gaining increasing attention due to the gradual depletion of high-grade Mn ores, amid the difficulties in its efficient extraction for both Mn and Fe values in an environmentally-friendly manner. Attempting to tackle the difficulties, this paper describes an innovative process for selectively chlorinating and reducing the high-Fe manganese ore in a simultaneous manner, aiming to produce water-soluble MnCl2 and metallic Fe. After pre-mixing with carbonaceous reductant, CaCl2 and MgCl2 as the chlorinating agent, the Mn ore was heated at 1000 °C. As much as 89.4% Mn can be chlorinated in its water-soluble form, with dissolution of only 3.0% Fe. The presence of CaCl2 during carbothermic reduction resulted in significant promotion in both the Fe reduction rate and formation of large metallic Fe particles due to the segregation effect, facilitating subsequent separation. Selective Mn chlorination by MgCl2 took place with or without the involvement of SiO2, forming MgSiO4 or MgO, respectively.

We propose and experimentally demonstrate a robust mode converter based on liquid crystal-on-silicon (LCOS), which is not placed at the focal plane of the collimating lens. We present theoretical investigations to evaluate the performance of proposed configuration, and our experimental results verify the favorable function of mode-selective conversion with orthogonal linear polarizations, as well as two-mode multiplexing. The robust device configuration with an insertion loss of less than 3 dB, together with cascade capability, can be used for multidimensional fiber-optical transmission system. In particular, in terms of LP11 mode conversion, the off-focus operation allows the range of position offset for input fiber with graded-index (GRIN) lens, LCOS, and few-mode fiber to be 7.5 mm, 151 μm, and 40.4 mm, respectively, indicating easy alignment and maintenance.

The ternary pH buffer system with ammonia-carbonates-volatile fatty acids (VFAs) is essential to anaerobes for bioenergy recovery via anaerobic digestion (AD). However, ammonia and VFAs are recognized as potential inhibitors that depress methanogenesis. In this study, biochemical conversion and the microbial community in batch AD at total solid (TS) from 4% to 14% were investigated to reveal their response to the ternary pH buffer system. The rapid ammonia release, probably promoted by Anaerosphaera and Eubacterium inferred from the concurrent peak of their relative abundance, triggered total ammonia (TAN) inhibition with the accumulation of VFAs in the start-up stage of high solid AD (HSAD, TS ≥ 8%). Along with evolution of the microbial community to resist high TAN and VFAs, methanogenesis recovered with improved degradation of VFAs and reduction of COD. When exposed to 3500 mg·N·L−1 TAN at 8% TS, aceticlastic Methanosarcina became dominant first and then together with hydrogenotrophic Methanoculleus, achieved the optimal biochemical conversion. While in HSAD at 11⁻14% TS, the main pathway of methanogenesis appeared to have shifted from the aceticlastic pathway to the hydrogenotrophic pathway, as inferred by changes in the relative abundance of methanogens, and this could have been induced by the increasing concentration of high free ammonia (FAN, ≥588 mg·N·L−1). Although the anaerobes had acclimatized to high TAN, the propionate-oxidizing bacteria and acetate-oxidizing bacteria might have again been inhibited by high FAN, frustrating the H2 supply for FAN-tolerant Methanoculleus and causing an 8.2⁻11.3% depression of COD reduction (mainly propionate residual).

This paper describes an innovative process for the production of ferrochromium alloy via segregation reduction of chromite at 1300 °C in the presence of calcium chloride. Both charcoal and petroleum coke were used as the reductant. Individual polycrystalline ferrochrome carbide particles were produced, with their particle size and shape resembling that of the starting carbon particles. Interactions among calcium chloride, clinochlore, and chromite resulted in the formation of Ca-bearing chromite, wadalite, and gaseous chlorides. Monocrystalline ferrochrome carbide whiskers were formed only when charcoal in the presence of calcium chloride was used to reduce the chromite fraction containing large amounts of siliceous gangue. Incorporating thermodynamic evaluations, a possible segregation reduction mechanism is suggested based on the characterization of the products using scanning electron microscopy, energy dispersive spectroscopy, X-ray powder diffraction, and electron probe microanalysis, combined with thermogravimetric analysis. Metallization within chromite particles was minimized, which is suggested to be due to the ubiquitous presence of molten and gaseous calcium chloride between the chromite and carbon particles, and especially in the porous rim of the chromite particles during reduction, resulting in restricted metallization only on the carbon particles.

A solid reduction process is described whereby chromite is reduced with the help of calcium chloride to produce ferrochrome alloy powders with high metal recovery. The process involves segregation reduction of chromite using graphite as the reductant and calcium chloride as the segregation catalyst. Experiments were performed in the temperature range of 1200–1400 °C to evaluate the influences of various design parameters using both a thermogravimetric analyzer and an electric tube furnace with continuous off-gas analysis. The reduced products were characterized by scanning electron microscopy, X-ray powder diffraction, synchrotron X-ray absorption spectroscopy, and were subjected to wet chemical analysis. It was concluded that the addition of calcium chloride not only accelerated the carbothermic reduction of chromite but also promoted the formation and growth of individual ferrochrome alloy particles. The alloy formation within chromite particles was minimized, enabling the effective separation of ferrochrome alloy particles from the unwanted gangue without the need for fine grinding. Aberrant expression of special AT-rich binding protein 1 (SATB1), a global genomic organizer, has been associated with various cancers, which raises the question of how higher-order chromatin structure contributes to carcinogenesis. Disruption of apoptosis is one of the hallmarks of cancer. We previously demonstrated that SATB1 mediated specific long-range chromosomal interactions between the mbr enhancer located within 3'-UTR of the BCL2 gene and the promoter to regulate BCL2 expression during early apoptosis. In the present study, we used chromosome conformation capture (3C) assays and molecular analyses to further investigate the function of the SATB1-mediated higher-order chromatin structure in co-regulation of the anti-apoptotic BCL2 gene and the pro-apoptotic NOXA gene located 3.4Mb downstream on Chromosome 18. We demonstrated that the mbr enhancer spatially juxtaposed the promoters of BCL2 and NOXA genes through SATB1-mediated chromatin-loop in Jurkat cells. Decreased SATB1 levels switched the mbr-BCL2 loop to mbr-NOXA loop, and thus changed expression of these two genes. The SATB1-mediated dynamic switch of the chromatin loop structures was essential for the cooperative expression of the BCL2 and NOXA genes in apoptosis. Notably, the role of SATB1 was specific, since inhibition of SATB1 degradation by caspase-6 inhibitor or caspase-6-resistant SATB1 mutant reversed expression of BCL-2 and NOXA in response to apoptotic stimulation. This study reveals the critical role of SATB1-organized higher-order chromatin structure in regulating the dynamic equilibrium of apoptosis-controlling genes with antagonistic functions and suggests that aberrant SATB1 expression might contribute to cancer development by disrupting the co-regulated genes in apoptosis pathways.

Foot-and-mouth disease, one of the most significant diseases of dairy herds, has substantial effects on farm economics, and currently, disease control measures are limited. In this study, we constructed a vector with a human interferon-α (hIFN-α) (without secretory signal sequence) gene cassette containing the immediate early promoter of human cytomegalovirus. Stably transfected bovine fetal fibroblasts were obtained by G418 selection, and hIFN-α transgenic embryos were produced by somatic cell nuclear transfer (SCNT). Forty-six transgenic embryos were transplanted into surrogate cows, and five cows (10.9%) became pregnant. Two male cloned calves were born. Expression of hIFN-α was detected in transfected bovine fetal fibroblasts, transgenic SCNT embryos, and different tissues from a transgenic SCNT calf at two days old. In transfected bovine fetal fibroblasts, expression of intracellular IFN-α induced resistance to vesicular stomatitis virus infection, increased apoptosis, and induced the expression of double-stranded RNA-activated protein kinase gene (PKR) and the 2'-5'-oligoadenylate synthetase gene (2'-5' OAS), which are IFN-inducible genes with antiviral activity. Analysis by qRT-PCR showed that the mRNA expression levels of PKR, 2'-5' OAS, and P53 were significantly increased in wild-type bovine fetal fibroblasts stimulated with extracellular recombinant human IFN-α-2b, showing that intracellular IFN-α induces biological functions similar to extracellular IFN-α. In conclusion, expression of intracellular hIFN-α conferred antiviral properties in transfected bovine fetal fibroblasts and did not significantly affect the full development of SCNT embryos. Thus, IFN-α transgenic technology may provide a revolutionary way to achieve elite breeding of livestock.

Shooting sports require a high level of stability to achieve accuracy and precision in hitting the target. This study presents a technical method for effecting stability in shooting sports by analyzing a case study of the sport. This study examines how stability could improve in shooting sports through technical methods. The research found that selecting the right equipment, maintaining correct body posture, and practicing breathing techniques could enhance stability. Mental preparation and relaxation techniques are also beneficial. Physiological factors like muscle tension and breathing rate influence stability.
