The relevance of the research is due to the need to determine the effectiveness of the project approach from the point of view of network interaction. The purpose of the article is to test the method of network management of physical culture and sports activities of the University developed by the authors within the project approach. The implementation of the project approach has led to increased coverage and an increase in the number of stable connections in the network model. The network model is transformed from a concentric type to a distributed one by increasing the number of projects with active participation of Universities. In the scheme of network interaction, there are connections due to the need to ensure the implementation of training courses, in particular for FSBEI HE USUE connections appear to overcome resource constraints. Much more active is the inclusion of the physical culture and sports activities in projects initiated by state and municipal authorities, the corporate sector, sports organizations and Universities themselves. The number of participants actively using the network strategy has increased.

The article is based on the research materials of the network interaction of universities in the context of physical culture and sports activity. The research is based on the methodology of network theory and the resource approach in the strategic management of organizations. The results of the study show that the formation of a network model can significantly increase the coverage of the population with projects of physical culture and sports activity using already existing resources. The results obtained indicate the effectiveness of strategies for networking organizations.

The realization of the various interests of students in the sports activities of universities requires educational institutions to have a diverse material and technical base: sports facilities of various specifics, qualified personnel, and methodological developments. At the same time, the capabilities of individual higher education institutions are limited. The solution to this problem is possible through the integrated use of the resource base of several educational institutions and municipal facilities for the organization of sports and athletic activities. An effective exchange of resources is based on network models of relationships, the management of which is based on the provisions of the theories of strategic and general management and a project approach. The purpose of this study was the formation of a network model of interaction in the processes of sports activities of universities in the Sverdlovsk region. The study was conducted by the Physical Education and Sports Department in conjunction with the Management and Entrepreneurship Department of the Ural State University of Economics in the period 2019-2020. The information ground of the study is formed based on the analysis of plans for sporting events, tender documentation, as well as the database of contracts concluded by participants in the strategic network. The research methodology is based on a network theory and involves the construction of a directed graph, where the nodes are university subjects, and objects of network interaction are placed on arcs. The results of the study show that the network model is of the concentric type, the main unifying projects in the framework of physical culture and sports activities are such sports events as “Nations Cross”, “Universiade of the Sverdlovsk Region” and “Ski Track of Russia”. There are no links in the network interaction scheme due to the need to implement on a contractual basis the training courses included in the Basic professional educational programs of higher educational institutions, the network is based only on participation in external events initiated by state and municipal authorities, the corporate sector, sports organizations and universities themselves.

The SlipChip platform was tested to perform high-throughput nanoliter multiplex PCR. The advantages of using the SlipChip platform for multiplex PCR include the ability to preload arrays of dry primers, instrument-free sample manipulation, small sample volume, and highthroughput capacity. The SlipChip was designed to preload one primer pair per reaction compartment and to screen up to 384 different primer pairs with less than 30 nanoliters of sample per reaction compartment. Both a 40-well and a 384-well design of the SlipChip were tested for multiplex PCR. In the geometries used here, the sample fluid was spontaneously compartmentalized into discrete volumes even before slipping of the two plates of the SlipChip, but slipping introduced additional capabilities that made devices more robust and versatile. The wells of this SlipChip were designed to overcome potential problems associated with thermal expansion. By using circular wells filled with oil and overlapping them with square wells filled with the aqueous PCR mixture, a droplet of aqueous PCR mixture was always surrounded by the lubricating fluid. In this design, during heating and thermal expansion, only oil was expelled from the compartment and leaking of the aqueous solution was prevented. Both 40-well and 384-well devices were found to be free from cross-contamination, and end point fluorescence detection provided reliable readout. Multiple samples could also be screened on the same SlipChip simultaneously. Multiplex PCR was validated on the 384-well SlipChip with 20 different primer pairs to identify 16 bacterial and fungal species commonly presented in blood infections. The SlipChip correctly identified five different bacterial or fungal species in separate experiments. In addition, the presence of the resistance gene mecA in methicillin resistant Staphylococcus aureus (MRSA) was identified. The SlipChip will be useful for applications involving PCR arrays and lays the foundation for new strategies for diagnostics, point-of-care devices, and immobilization-based arrays. 10.1021/ac1007249

Bacteriophage N4 minivirion RNA polymerase (mini-vRNAP), the RNA polymerase (RNAP) domain of vRNAP, is a member of the T7-1ike RNAP family. Mini-vRNAP recognizes promoters that comprise conserved sequences and a 3-base loop-5-base pair (bp) stem DNA hairpin structure on single-stranded templates. Here, we defined the DNA structural and sequence requirements for mini-vRNAP promoter recognition. Mini-vBNAP binds a 20-nucleotide (nt) N4 P2 promoter deoxyoligonucleotide with high affinity ([K.sub.d] = 2 nM) to form a salt-resistant complex. We show that mini-vRNAP interacts specifically with the central base of the hairpin loop (-11G) and a base at the stem (-8G) and that the guanine 6-keto and 7-imino groups at both positions are essential for binding and complex salt resistance. The major determinant (-11G), which must be presented to mini-vRNAP in the context of a hairpin loop, appears to interact with mini-vRNAP Trp-129. This interaction requires template single-strandedness at positions -2 and -1. Contacts with the promoter are disrupted when the RNA product becomes 11-12 nt long. This detailed description of vRNAP interaction with its promoter hairpin provides insights into RNAP-promoter interactions and explains how the injected vRNAP, which is present in one or two copies, recognizes its promoters on a single copy of the injected genome.
