Animal Emotions: How They Drive Human Behavior gives a concise overview of ancient mammalian emotions deeply rooted in the human brain. Jaak Panksepp, a world-renowned neuroscientist, dedicated his life career to the study of mammalian emotions and he carved out seven distinct emotional systems he called seeking, lust, care, and play (positive emotions), and fear, anger, and sadness (negative emotions), all exerting a tremendous influence on human behavior.Christian Montag, a neuroscientist and psychologist, and a long-time collaborator of Jaak Panksepp, revisits together with Kenneth L. Davis, one of Jaak’s PhD students, Panksepp’s theories and provides the reader with new insights into the nature of emotions and their role as survival tools, both for animals and for humans. They also raise new questions about the background of the research field Jaak Panksepp coined "Affective Neuroscience." How are personality and psychopathology linked to animal emotions? Abstract

Schizophrenia has a heritability of 60-80%1, much of which is attributable to common risk alleles. Here, in a two-stage genome-wide association study of up to 76,755 individuals with schizophrenia and 243,649 control individuals, we report common variant associations at 287 distinct genomic loci. Associations were concentrated in genes that are expressed in excitatory and inhibitory neurons of the central nervous system, but not in other tissues or cell types. Using fine-mapping and functional genomic data, we identify 120 genes (106 protein-coding) that are likely to underpin associations at some of these loci, including 16 genes with credible causal non-synonymous or untranslated region variation. We also implicate fundamental processes related to neuronal function, including synaptic organization, differentiation and transmission. Fine-mapped candidates were enriched for genes associated with rare disruptive coding variants in people with schizophrenia, including the glutamate receptor subunit GRIN2A and transcription factor SP4, and were also enriched for genes implicated by such variants in neurodevelopmental disorders. We identify biological processes relevant to schizophrenia pathophysiology; show convergence of common and rare variant associations in schizophrenia and neurodevelopmental disorders; and provide a resource of prioritized genes and variants to advance mechanistic studies.

Abstract

Water potential directly controls the function of leaves, roots, and microbes, and gradients in water potential drive water flows throughout the soil-plant-atmosphere continuum. Notwithstanding its clear relevance for many ecosystem processes, soil water potential is rarely measured in-situ, and plant water potential observations are generally discrete, sparse, and not yet aggregated into accessible databases. These gaps limit our conceptual understanding of biophysical responses to moisture stress and inject large uncertainty into hydrologic and land surface models. Here, we outline the conceptual and predictive gains that could be made with more continuous and discoverable observations of water potential in soils and plants. We discuss improvements to sensor technologies that facilitate in situ characterization of water potential, as well as strategies for building new networks that aggregate water potential data across sites. We end by highlighting novel opportunities for linking more representative site-level observations of water potential to remotely-sensed proxies. Together, these considerations offer a roadmap for clearer links between ecohydrological processes and the water potential gradients that have the 'potential' to substantially reduce conceptual and modeling uncertainties.

Abstract

Urban regions emit a large fraction of anthropogenic emissions of greenhouse gases (GHG) such as carbon dioxide (CO2) and methane (CH4) that contribute to modern-day climate change. As such, a growing number of urban policymakers and stakeholders are adopting emission reduction targets and implementing policies to reach those targets. Over the past two decades research teams have established urban GHG monitoring networks to determine how much, where, and why a particular city emits GHGs, and to track changes in emissions over time. Coordination among these efforts has been limited, restricting the scope of analyses and insights. Here we present a harmonized data set synthesizing urban GHG observations from cities with monitoring networks across North America that will facilitate cross-city analyses and address scientific questions that are difficult to address in isolation.

Abstract

We present an evaluation of airborne Intensity-Modulated Continuous-Wave (IM-CW) lidar measurements of atmospheric column CO2 mole fractions during the ACT-America project. This lidar system transmits online and offline wavelengths simultaneously on the 1.57111-um CO2 absorption line, with each modulated wavelength using orthogonal swept frequency waveforms. After the spectral characteristics of this system were calibrated through short-path measurements, we used the HITRAN spectroscopic database to derive the average-column CO2 mixing ratio (XCO2) from the lidar measured optical depths. Based on in situ measurements of meteorological parameters and CO2 concentrations for calibration data, we demonstrate that our lidar CO2 measurements were consistent from season to season and had an absolute calibration error (standard deviation) of 0.80 ppm when compared to XCO2 values derived from in situ measurements. By using a 10-second or longer moving average, a long-term stability of 1 ppm or better was obtained. The estimated CO2 measurement precision for 0.1-s, 1-s, 10-s, and 60-s averages were determined to be 3.4 ppm (0.84%), 1.2 ppm (0.30%), 0.43 ppm (0.10%), and 0.26 ppm (0.063%), respectively. These correspond to measurement signal-to-noise ratios of 120, 330, 950, and 1600, respectively. The drift in XCO2 over one-hour of flight time was found to be below our detection limit of about 0.1 ppm. These analyses demonstrate that the measurement stability, precision and accuracy are all well below the thresholds needed to study synoptic-scale variations in atmospheric XCO2., Comment: 20 pages, 5 figures. Abstract

The atmospheric boundary layer mediates the exchange of energy, matter, and momentum between the land surface and the free troposphere, integrating a range of physical, chemical, and biological processes and is defined as the lowest layer of the atmosphere (ranging from a few meters to 3 km). In this review, we investigate how continuous, automated observations of the atmospheric boundary layer can enhance the scientific value of co-located eddy covariance measurements of land-atmosphere fluxes of carbon, water, and energy, as are being made at FLUXNET sites worldwide. We highlight four key opportunities to integrate tower-based flux measurements with continuous, long-term atmospheric boundary layer measurements: (1) to interpret surface flux and atmospheric boundary layer exchange dynamics and feedbacks at flux tower sites, (2) to support flux footprint modelling, the interpretation of surface fluxes in heterogeneous and mountainous terrain, and quality control of eddy covariance flux measurements, (3) to support regional-scale modeling and upscaling of surface fluxes to continental scales, and (4) to quantify land-atmosphere coupling and validate its representation in Earth system models. Adding a suite of atmospheric boundary layer measurements to eddy covariance flux tower sites, and supporting the sharing of these data to tower networks, would allow the Earth science community to address new emerging research questions, better interpret ongoing flux tower measurements, and would present novel opportunities for collaborations between FLUXNET scientists and atmospheric and remote sensing scientists.

Abstract

The FLUXNET2015 dataset provides ecosystem-scale data on CO2, water, and energy exchange between the biosphere and the atmosphere, and other meteorological and biological measurements, from 212 sites around the globe (over 1500 site-years, up to and including year 2014). These sites, independently managed and operated, voluntarily contributed their data to create global datasets. Data were quality controlled and processed using uniform methods, to improve consistency and intercomparability across sites. The dataset is already being used in a number of applications, including ecophysiology studies, remote sensing studies, and development of ecosystem and Earth system models. FLUXNET2015 includes derived-data products, such as gap-filled time series, ecosystem respiration and photosynthetic uptake estimates, estimation of uncertainties, and metadata about the measurements, presented for the first time in this paper. In addition, 206 of these sites are for the first time distributed under a Creative Commons (CC-BY 4.0) license. This paper details this enhanced dataset and the processing methods, now made available as open-source codes, making the dataset more accessible, transparent, and reproducible.

Abstract

Kingdom Authority Christian Center (KACC) is growing more attentive to the changing demographics in the world. It seeks to understand that as other countries and nations continue to diversify by race and ethnicity, change is needed. The problem is that KACC has no training curriculum designed to impact and employ change. A mixed-method approach will be applied to fix KACC's problem and the proposal of correspondence from People of the Dream. This thesis project will provide research from individual interviews, a population group, and an inside viewpoint from one of the people in the dream. This project will also add to the already training analysis provided by the many scholarly articles, peer reviews, and dissertations.

Abstract

Eddy-covariance (EC) flux measurements in Indianapolis were used to quantify the impact of the COVID-19 lockdown on CO and CO2 emissions from a highway and a suburban neighborhood. CO2 fluxes were measured for 6 weeks pre-lockdown (January 22, 2020–March 3, 2020) and during lockdown (March 25, 2020– May 5, 2020) using EC instrumentation at 41 m AGL. Fossil fuel CO2 emissions (CO2ff) were estimated by calculating eddy diffusivity to obtain CO flux and then scaling by the CO:CO2ff emissions ratio (RCO). Flux measurements segregated by wind direction were compared to hourly emissions from the 2020 Hestia inventory model. The lockdown CO2ff average weekday emissions from the highway estimated by EC decreased by 51.5 ± 10.9% (11.2 ± 2.2 µmol m−2 s−1) compared to pre-lockdown, similar to Hestia's estimate 56 ± 7% (12 ± 1 µmol m−2 s−1). The EC measurements detected a significant (2.2 ± 0.7 µmol m−2 s−1) but smaller magnitude decrease in CO2ff emissions from the suburban neighborhood. The daily cycles of CO2ff emissions were significantly correlated with Hestia estimates from the highway but not from the suburbs. This study demonstrates that EC flux towers and high-resolution inventory models in regions with mixed and spatially heterogeneous sources can quantify abrupt changes in sector- and source-specific CO2 fluxes. Abstract

The purpose of this study is to determine whether urban greenhouse gas (GHG) fluxes can be quantified from tower-based mole fraction measurements using Monin–Obukhov similarity theory (MOST). Tower-based GHG mole fraction networks are used in many cities to quantify whole-city GHG emissions. Local-scale, micrometeorological flux estimates would complement whole-city estimates from atmospheric inversions. CO2 mole fraction and eddy-covariance flux data at an urban site in Indianapolis, Indiana, from October 2020 through January 2022 are analyzed. Using MOST flux–variance and flux–gradient relationships, CO2 fluxes were calculated using these mole fraction data and compared to the eddy-covariance fluxes. MOST-based fluxes were calculated using varying measurement heights and methods of estimating stability. The MOST flux–variance relationship method showed good temporal correlation with eddy-covariance fluxes at this site but overestimated flux magnitudes. Fluxes calculated using flux–gradient relationships showed lower temporal correlation with eddy-covariance fluxes but closer magnitudes to eddy-covariance fluxes. Measurement heights closer to ground level produce more precise flux estimates for both MOST-based methods. For flux–gradient methods, flux estimates are more accurate and precise when low-altitude measurements are combined with a large vertical separation between measurement heights. When stability estimates based on eddy-covariance flux measurements are replaced with stability estimates based on the weather station or net radiation data, the MOST-based fluxes still capture the temporal patterns measured via eddy covariance. Based on these results, MOST can be used to estimate the temporal patterns in local GHG fluxes at mole fraction tower sites, complementing the small number of eddy-covariance flux measurements available in urban settings. Abstract

Copy number variants (CNVs) have been strongly implicated in the genetic etiology of schizophrenia (SCZ). However, genome-wide investigation of the contribution of CNV to risk has been hampered by limited sample sizes. We sought to address this obstacle by applying a centralized analysis pipeline to a SCZ cohort of 21,094 cases and 20,227 controls. A global enrichment of CNV burden was observed in cases (odds ratio (OR) = 1.11, P = 5.7 × 10-15), which persisted after excluding loci implicated in previous studies (OR = 1.07, P = 1.7 × 10-6). CNV burden was enriched for genes associated with synaptic function (OR = 1.68, P = 2.8 × 10-11) and neurobehavioral phenotypes in mouse (OR = 1.18, P = 7.3 × 10-5). Genome-wide significant evidence was obtained for eight loci, including 1q21.1, 2p16.3 (NRXN1), 3q29, 7q11.2, 15q13.3, distal 16p11.2, proximal 16p11.2 and 22q11.2. Suggestive support was found for eight additional candidate susceptibility and protective loci, which consisted predominantly of CNVs mediated by nonallelic homologous recombination.

Abstract

Polygenic risk scores have shown great promise in predicting complex disease risk and will become more accurate as training sample sizes increase. The standard approach for calculating risk scores involves linkage disequilibrium (LD)-based marker pruning and applying a p value threshold to association statistics, but this discards information and can reduce predictive accuracy. We introduce LDpred, a method that infers the posterior mean effect size of each marker by using a prior on effect sizes and LD information from an external reference panel. Theory and simulations show that LDpred outperforms the approach of pruning followed by thresholding, particularly at large sample sizes. Accordingly, predicted R(2) increased from 20.1% to 25.3% in a large schizophrenia dataset and from 9.8% to 12.0% in a large multiple sclerosis dataset. A similar relative improvement in accuracy was observed for three additional large disease datasets and for non-European schizophrenia samples. The advantage of LDpred over existing methods will grow as sample sizes increase.

Abstract

For tens of thousands of years, teachers have used stories to promote learning. Today's teachers can do the same. In particular, we can employ Joseph Campbell's "monomyth"--with its stages of separation, initiation, and return--as a model for structuring learning experiences. Within the monomyth, one tempting role for teachers is the sage, but we should resist this temptation. Instead we should acknowledge, and benefit from, our role as tricksters. To do so is to accept and illuminate the dual responsibility of the teacher as both supporter and challenger.

Abstract

Regulatory and coding variants are known to be enriched with associations identified by genome-wide association studies (GWASs) of complex disease, but their contributions to trait heritability are currently unknown. We applied variance-component methods to imputed genotype data for 11 common diseases to partition the heritability explained by genotyped SNPs (hg(2)) across functional categories (while accounting for shared variance due to linkage disequilibrium). Extensive simulations showed that in contrast to current estimates from GWAS summary statistics, the variance-component approach partitions heritability accurately under a wide range of complex-disease architectures. Across the 11 diseases DNaseI hypersensitivity sites (DHSs) from 217 cell types spanned 16% of imputed SNPs (and 24% of genotyped SNPs) but explained an average of 79% (SE = 8%) of hg(2) from imputed SNPs (5.1× enrichment; p = 3.7 × 10(-17)) and 38% (SE = 4%) of hg(2) from genotyped SNPs (1.6× enrichment, p = 1.0 × 10(-4)). Abstract

Schizophrenia is a highly heritable disorder. Genetic risk is conferred by a large number of alleles, including common alleles of small effect that might be detected by genome-wide association studies. Here we report a multi-stage schizophrenia genome-wide association study of up to 36,989 cases and 113,075 controls. We identify 128 independent associations spanning 108 conservatively defined loci that meet genome-wide significance, 83 of which have not been previously reported. Associations were enriched among genes expressed in brain, providing biological plausibility for the findings. Many findings have the potential to provide entirely new insights into aetiology, but associations at DRD2 and several genes involved in glutamatergic neurotransmission highlight molecules of known and potential therapeutic relevance to schizophrenia, and are consistent with leading pathophysiological hypotheses. Independent of genes expressed in brain, associations were enriched among genes expressed in tissues that have important roles in immunity, providing support for the speculated link between the immune system and schizophrenia.
