Abstract

Natural killer (NK) cells are innate lymphoid cells (ILCs) contributing to immune responses to microbes and tumors. Historically, their classification hinged on a limited array of surface protein markers. Here, we used single-cell RNA sequencing (scRNA-seq) and cellular indexing of transcriptomes and epitopes by sequencing (CITE-seq) to dissect the heterogeneity of NK cells. We identified three prominent NK cell subsets in healthy human blood: NK1, NK2 and NK3, further differentiated into six distinct subgroups. Our findings delineate the molecular characteristics, key transcription factors, biological functions, metabolic traits and cytokine responses of each subgroup. These data also suggest two separate ontogenetic origins for NK cells, leading to divergent transcriptional trajectories. Furthermore, we analyzed the distribution of NK cell subsets in the lung, tonsils and intraepithelial lymphocytes isolated from healthy individuals and in 22 tumor types. This standardized terminology aims at fostering clarity and consistency in future research, thereby improving cross-study comparisons.

Abstract

AbstractFast quench detection is a key requirement for the successful implementation of superconducting magnet technology. In high temperature superconductor (HTS) magnets, this issue is especially challenging due to the low quench propagation velocity, and presently represents one of the main factors limiting their application. A new detection technique based on stray-capacitance monitoring is proposed. The capacitance between electrically-insulated magnet elements, such as magnet structure and end parts, is utilized as an indication of local heat deposition in the conductor. In fact, the relative permittivity of helium drops when it changes from the liquid to the gaseous phase. Thus, when heating occurs, part of the helium impregnating the insulation layers boils off, and the monitored stray-capacitance decreases. The proposed technique is successfully demonstrated on three small-scale Bi-2212 magnets manufactured at the Lawrence Berkeley National Laboratory. Results from the detection of thermal runaways and spot-heater induced quenches are reported and discussed. Abstract

Let $p$ be a prime, let $KU_p$ be $p$-complete complex $K$-theory, and let $\mathbb{Z}_p^\times$ denote the group of units in the $p$-adic integers. The $p$-adic Adams operations induce an action of the profinite group $\mathbb{Z}_p^\times$ on $KU_p$, and hence, on the algebraic $K$-theory spectrum $K(KU_p)$. For $p \geq 5$, we give an elementary construction of the continuous homotopy fixed point spectrum $(L_{K(2)}K(KU_p))^{hG}$, where $K(2)$ is the second Morava $K$-theory and $G$ is any closed subgroup of $\mathbb{Z}_p^\times$. Also, for each $G$, we show that there is an associated strongly convergent homotopy fixed point spectral sequence whose $E_2$-term is given by Jannsen's continuous group cohomology, with $E_2^{s,\ast} = 0$, for all $s > 2$. Abstract

Let $n \geq 1$, $p$ a prime, and $T(n)$ any representative of the Bousfield class of the telescope $v_n^{-1}F(n)$ of a finite type $n$ complex. Also, let $E_n$ be the Lubin-Tate spectrum, $K(E_n)$ its algebraic $K$-theory spectrum, and $G_n$ the extended Morava stabilizer group, a profinite group. Motivated by an Ausoni-Rognes conjecture, we show that there are two spectral sequences \[{^{I}}\mspace{-3mu}E_2^{s,t} \Longrightarrow \pi_{t-s}((L_{T(n+1)}K(E_n))^{hG_n}) \Longleftarrow {^{II}}\mspace{-2mu}E_2^{s,t}\] with common abutment $\pi_\ast(-)$ of the continuous homotopy fixed points of $L_{T(n+1)}K(E_n)$, where ${^{I}}\mspace{-3mu}E_2^{s,t}$ is continuous cohomology with coefficients in a certain tower of discrete $G_n$-modules. If the tower satisfies the Mittag-Leffler condition, then there are continuous cochain cohomology groups \[{^{I}}\mspace{-3mu}E_2^{\ast,\ast} \cong H^\ast_\mathrm{cts}(G_n, \pi_\ast(L_{T(n+1)}K(E_n))) \cong {^{II}}\mspace{-2mu}E_2^{\ast,\ast}.\] We isolate two hypotheses, the first of which is true when $(n,p) = (1,2)$, that imply $(L_{T(n+1)}K(E_n))^{hG_n} \simeq L_{T(n+1)}K(L_{K(n)}S^0)$. Abstract

The use of high-field superconducting magnets has furthered the development of medical diagnosis, fusion research, accelerators, and particle physics. High-temperature superconductors enable magnets more powerful than those possible with Nb-Ti (superconducting transition temperature Tc of 9.2 K) and Nb3Sn (Tc of 18.4 K) conductors due to their very high critical field Bc2 of greater than 100 T near 4.2 K. However, the development of high-field accelerator magnets using high-temperature superconductors is still at its early stage. We report the construction of the world's first high-temperature superconducting Bi2Sr2CaCu2Ox (Bi-2212 with Tc of ∼82 K) accelerator dipole magnet. The magnet is based on a canted-cosine-theta design with Bi-2212 Rutherford cables. A high critical current was achieved by an overpressure processing heat treatment. The magnet was constructed from a nine-strand Rutherford cable made from industrial 0.8 mm wires. At 4.2 K, it reached a quench current of 3600 A and a dipole field of 1.64 T in a bore of 31 mm. The magnet did not exhibit the undesirable quench training common in Nb-Ti and Nb3Sn accelerator magnets. It quenched a dozen times without degradation.
