Abstract

The article is published in the memory of Norman Davidson, a physical chemist and a chemistry instructor at the California Institute of Technology who has been on the faculty till his death in February 14, 2002. In this article Mr. Davidson describes different topics of interest including his studies of DNA chemistry, synaptic plasticity and neuronal nitric oxide synthase and trkB receptors.

Abstract

G protein-gated inward rectifier [K.sup.+] (GIRK) channels mediate hyperpolarizing postsynaptic potentials in the nervous system and in the heart during activation of G[Alpha](i/o)-coupled receptors. In neurons and cardiac atrial cells the time course for receptor-mediated GIRK current deactivation is 20-40 times faster than that observed in heterologous systems expressing cloned receptors and GIRK channels, suggesting that an additional component(s) is required to confer the rapid kinetic properties of the native transduction pathway. We report here that heterologous expression of 'regulators of G protein signaling' (RGS proteins), along with cloned G protein-coupled receptors and GIRK channels, reconstitutes the temporal properties of the native receptor [left arrow] GIRK signal transduction pathway. GIRK current waveforms evoked by agonist activation of muscarinic [m.sub.2] receptors or serotonin 1A receptors were dramatically accelerated by coexpression of either RGS1, RGS3, or RGS4, but not RGS2. For the brain-expressed RGS4 isoform, neither the current amplitude nor the steady-state agonist dose-response relationship was significantly affected by RGS expression, although the agonist-independent 'basal' GIRK current was suppressed by [approximately equal to]40%. Because GIRK activation and deactivation kinetics are the limiting rates for the onset and termination of 'slow' postsynaptic inhibitory currents in neurons and atrial cells, RGS proteins may play crucial roles in the timing of information transfer within the brain and to peripheral tissues.

Abstract

The G-alpha subunits of heterotrimeric G proteins suppress the activity of K+ channels of the GIRK family. Free GTP-gamma-S-activated G(sub alpha-i1) subunit suppress the GIRK activity induced by G(sub beta-1-gamma-2) in the membranes of Xenopus oocytes expressing GIRK1. G(sub alpha-i2) and G(sub alpha-i3) have no effect on GIRK in Xenopus. G(sub alpha-s)-GTP-gamma-S inhibits GIRK activity partially. The coupling of G proteins to GIRKs appears to be regulated by antagonistic interactions between G(sub alpha) and G(sub beta-gamma) proteins.

Abstract

The voltage-jump relaxation rate constants of an extended chimeric beta subunit from 94 to 109 beta-4 NH(sub 2)-terminal residues coexpressed with the alpha-3 subunit in Xenopus oocytes were measured. Results indicated that the wild-type alpha-3beta-4 receptors exhibit relaxations that are five to eight times slower than the wild-type alpha-3beta-2. The relaxations in both receptors were also found to contain two exponential components over which the beta subunit has a remarkable effect across a range of acetylcholine concentration.

Abstract

Results indicate G-protein gated inwardly rectifying potassium channels (GIRKs) serve as heteromultimers in neural transmission. Guanine nucleotide-binding proteins (G proteins) subunits Gbeta1 and Ggamma2 can activate GIRK2 and not GIRK3 in Xenopus oocytes. Also, G-protein-mediated inward currents were increased by five to forty times when GRK3 or GRK2 was coexpressed with GIR1. Voltage-jump kinetics of coexpressed channels showed new kinetic properties.
