An improved understanding of the relationships among vegetation, groundwater level, and microtopography is crucial for making well-informed management decisions in areas with shallow groundwater resources. We measured plant species abundance/composition and richness in relation to depth to groundwater (DTW) and microtopography in Owens Valley, California, particularly in areas where DTW ranged from 0 to 4 m. Sampling occurred along 67 vegetation transects across three community types. Relationships between DTW and community composition were evaluated using non-metric multidimensional scaling (NMS), while non-parametric multiplicative regression was used to relate DTW and microtopography to species abundance. The dominant gradient in species composition (NMS Axis 1) explained ~51% of variation in our distance matrix and was most strongly associated (r = 0.55) with DTW. The graminoids Juncus arcticus, Leymus triticoides, and Distichlis spicata had strong affinities toward areas with the shallowest DTW ( 2.0 m). Variation in microtopography affected species abundance and increased species richness for vegetation communities at either extreme of the water table gradient, shallow, and deep DTW but not the intermediate DTW. Findings indicate that desert plant communities from shallow aquifers have adapted to different DTW and microtopography conditions and that considering those adaptations may be important to manage groundwater and vegetation resources in these areas.

Aims Although increases in precipitation variability in arid ecosystems are projected due to climate change, the response of desert shrub communities to precipitation change has not been fully elucidated. Such knowledge is important since drought-adapted plants exhibit varied mechanisms of survival that may contribute to species coexistence. Methods We tested the responses of eight drought-adapted plants, a mix of graminoids, shrubs and forbs to three summer precipitation scenarios (1.3, 2.6 and 3.9 cm per month) in a common garden experiment in the Great Basin (Owens Valley, California). Changes in mineral nutrient uptake (carbon, nitrogen, phosphorus, potassium, calcium, magnesium, manganese, copper, boron, zinc, iron and sodium) and gas exchange parameters (photosynthetic rate and stomatal conductance) were investigated in the studied species. Important Findings Two graminoids (Sporobolus airoides and Leymus triticoides) and one salt tolerant shrub species (Atriplex confertifolia) responded to increased water availability with increases in photosynthetic rate and/or stomatal conductance. There was a significant correlation between water availability and uptake of nutrients for five out of eight species. Artemisia tridentata, with higher rates of photosynthesis, contained greater amounts of potassium, copper and boron, while Juncus arcticus, with higher rates of photosynthesis, contained greater amounts of magnesium and iron, and less sodium. Juncus arcticus and three salt-adapted species (A. confertifolia, Distichlis spicata and S. airoides) exhibited correlations with stomatal conductance and concentrations of nutrients. Results indicate that differential physiological response mechanisms to increased moisture and associated nutrient uptake strategies in drought-adapted species may mediate coexistence under increased summer precipitation.

Focused ultrasound blood-spinal cord barrier opening is a promising new method for improving the non-invasive delivery of therapeutic agents to the spinal cord. A custom 64-element phased array was built, based on the design of a component of a spine-specific array for focusing ultrasound through vertebral laminae and correcting vertebra-induced distortions. Geometric and hydrophone delays were tested in each thoracic vertebra in an ex vivo spine (n=12). Pressure fields were measured in each canal to measure focal aberrations. Geometric delays produced foci shifted by 2.4±1.7 mm from the target, while hydrophone delays produced foci 1.3±1.1 mm from the target. Simulation showed good correlation with hydrophone delays and may be a viable non-invasive method for phase corrections. This work demonstrates the feasibility of transvertebral ultrasound beamforming with a phased-array.

We present final Spitzer trigonometric parallaxes for 361 L, T, and Y dwarfs. We combine these with prior studies to build a list of 525 known L, T, and Y dwarfs within 20 pc of the Sun, 38 of which are presented here for the first time. Using published photometry and spectroscopy as well as our own follow-up, we present an array of color-magnitude and color-color diagrams to further characterize census members, and we provide polynomial fits to the bulk trends. Using these characterizations, we assign each object a $T_{\rm eff}$ value and judge sample completeness over bins of $T_{\rm eff}$ and spectral type. We have developed a chimeric antigen receptor (CAR) platform that functions as a modular system to address limitations of traditional CAR therapies. An inert form of the human NKG2D extracellular domain (iNKG2D) was engineered as the ectodomain of the CAR to generate convertibleCARTM-T cells. These cells were specifically directed to kill antigen-expressing target cells only in the presence of an activating bispecific adapter comprised of an iNKG2D-exclusive ULBP2-based ligand fused to an antigen-targeting antibody (MicAbodyTM). Efficacy against Raji tumors in NSG mice was dependent upon doses of both a rituximab-based MicAbody and convertibleCAR-T cells. We have also demonstrated that the exclusive ligand-receptor partnering enabled the targeted delivery of a mutant form of IL-2 to selectively promote the expansion of convertibleCAR-T cells in vitro and in vivo. By altering the Fv domains of the MicAbody or the payload fused to the orthogonal ligand, convertibleCAR-T cells can be readily targeted or regulated., Landgraf et al. developed a chimeric antigen receptor (CAR) platform that functions as a modular system. convertibleTM-T cells are designed to kill antigen-expressing target cells only in the presence of ligands fused to antigen-targeting antibodies (MicAbodyTM). We evaluated growth and physiology of three wetland graminoids (Distichlis spicata, Leymus triticoides, and Juncus arcticus) as affected by surface and subsurface water. These species are common in western USA and were collected from two areas: Bishop, California and Burns, Oregon. Our hypotheses were: 1) the use of surface water by plants is higher than the use of subsurface water and 2) there are population differences in the response of the species to available water. The species of study were grown in two-layer pots with soil water content in the upper and lower layers independently controlled. The first hypothesis was partially supported by our results, but variability existed among species. However, when all species had equal access to both surface soil water and subsurface water plants tended to preferentially use surface water. We also found that J. arcticus can display reverse hydraulic lift when growing with no subsurface water. The second hypothesis was clearly supported by results. Although the mechanism is not clear, it is possible that an area with periodic and predictable shallow groundwater underlying a dry or saline soil layer, such as the California site, might favor plant populations with high proficiency in water uptake by deeper roots.

Current approaches to reducing the latent HIV reservoir entail first reactivating virus-containing cells to become visible to the immune system. A critical second step is killing these cells to reduce reservoir size. Endogenous cytotoxic T-lymphocytes (CTLs) may not be adequate because of cellular exhaustion and the evolution of CTL-resistant viruses. We have designed a universal CAR-T cell platform based on CTLs engineered to bind a variety of broadly neutralizing anti-HIV antibodies. We show that this platform, convertibleCAR-T cells, effectively kills HIV-infected, but not uninfected, CD4 Tcells from blood, tonsil, or spleen and only when armed with anti-HIV antibodies. convertibleCAR-T cells also kill within 48h more than half of the inducible reservoir found in blood of HIV-infected individuals on antiretroviral therapy. The modularity of convertibleCAR-T cell system, which allows multiplexing with several anti-HIV antibodies yielding greater breadth and control, makes it a promising tool for attacking the latent HIV reservoir.

Recent advances in the deposition of patterned thin film spectral filters have enabled a new class of radically miniaturized spectral sensors. This new technology enables numerically large arrays of spectral bandpass filters with unprecedented manufacturing economy. For example, a 64-channel array occupying two square millimeters and spanning 400-900 nm can be deposited with as few as eight coating steps. Mating this filter array to a photodiode array yields a tiny multispectral sensor with diverse applications. The bandpass filters are single-cavity Fabry-Perot designs with common top and bottom mirrors. The dielectric spacer layer between them determines the passband wavelength and is patterned to differing thicknesses using a binary scheme, i.e., each successive “sub-spacer” layer is half the thickness of the previous one. The technical challenge is uniformly patterning and depositing thinner and thinner sub-spacers, which can be only a few nanometers thick. We have demonstrated 64-channel arrays covering the spectral range of 400-900 nm and 775-1075 nm. These arrays have been mated to high-responsivity 2D silicon detectors, in much the same way that linear variable filters are mated to linear detector arrays. The resulting sensor is less than 3 x 3 x 1 mm in size and ideal for integration into mobile devices, wearable electronics, autonomous aerial vehicles, and countless industrial applications. Sensor performance is currently being evaluated for food quality and freshness measurement, drug identification, fuel quality measurement, explosives detection, colorimetry and illumination measurement, solar flux monitoring, remote sensing, and myriad other applications.

The purpose of this prospective cohort study was to compare multimodal pain management and pain perception after open vs. laparoscopic colorectal surgery within enhanced recovery care. Pain scores at rest and at mobilization were prospectively assessed in consecutive patients using Visual Analog Scales (VAS 0-10) and consumption of different analgesics was recorded daily until 96 hours postoperatively. Uni- and multivariate risk factors for pain peaks (≥ 4/10) were identified by logistic regression and compared between two propensity score matched groups (open vs. laparoscopic). 156 open and 176 laparoscopic procedures were included. Mean VAS scores were consistently < 3 until 96 hours at rest and at mobilization. Patients operated by laparoscopy experienced more pain peaks (≥ 4) within 24 hours (p < 0.05), while patients operated by open approach experienced more pain peaks (≥ 4) during mobilization at 72 hours (p < 0.05). Independent risk factors for insufficient pain control (≥ 4) within 24 hours from surgery were duration of the procedure (OR 3.37, 95%CI 2.03-5.59), emergency surgery (OR 3.01, 95%CI 1.72-5.31), wound infiltration (OR 3.23, 95%CI 0.97-10.70), age < 70 years (OR 2.03, 95% CI 1.18-3.48) and ASA I-II score (OR 2.06, 95% CI 1.19-3.56). The perioperative adding of lidocaine ± ketamine to opioids did not improve postoperative pain perception nor decrease morphine equivalents. In conclusion, overall pain scores were low after colorectal surgery. However, pain peaks remained a concern early after minimally invasive surgery and after epidural removal for open surgery. Multimodal strategies were not superior to opioids alone.

The current report presents a case of an omental fibromatosis discovered incidentally in a 46-year-old woman with no particular medical history and few symptoms. A surgical biopsy was performed initially, and microscopic examination revealed myofibroblastic proliferation. After additional immunohistochemical and molecular analyses, omental fibromatosis was diagnosed. Omental fibromatosis, also called intra-abdominal desmoid, is a rare and benign tumour but can be locally aggressive. Majority of cases are asymptomatic, and difficult to diagnose based on clinical presentation and radiological investigation. Final diagnosis is usually made on histopathology and immunohistochemistry studies. Surgical wide excision is currently the treatment of choice.

When tracking oil drops rising in the ocean, it is important to consider settings as realistically as possible, for example, accounting for surfactant effects and pycnoclines in the ocean. Simulations are presented that quantify the effects of both.

Summary This investigation assessed the usefulness of a limited capacity information processing model for evaluation of stress. The assumption is that the task of anticipating an impending aversive event requires a portion of the limited capacity. Thus, such stress can be measured by a performance decrement in a concurrent task. Stress was manipulated in 14 male and female college students by means of a conditioned emotional response (CER) technique. Two tasks, motor tracking and counting backwards, were systematically studied as both primary and secondary tasks, depending on the presence or absence of reinforcement. The cognitive task of counting backwards was more sensitive to the processing demands imposed by stress than the motor task of tracking.

We analyzed temporal and spatial patterns of water use by a functionally-diverse group of Great Basin plant species and determined their water use rates at the whole-plant and individual-leaf scales under variable summer watering. Species studied were the desert grasses Distichlis spicata and Sporobolus airoides, the desert shrubs Artemisia tridentata, Ericameria nauseosa, and Atriplex confertifolia; the wetland/riparian plants Juncus arcticus, Leymus triticoides, and Salix exigua; and the annual exotic Salsola tragus. Plant species were individually grown in 5.8 m2 plots in a common garden in eastern California. Three irrigation treatments in the form of monthly pulses were applied during the summer: low (1.3 cm), medium (2.6 cm), and high (3.9 cm), in addition to a nonirrigated control. The extended finite element method (XFEM) [1, 2] can approximate the discontinuous displacement field near cracks independently of the finite element mesh through the use of interpolation functions, which can describe the displacement field near cracks in the structure. Therefore, XFEM is appearing as an alternative to conventional contour integral method for crack growth analysis. Most benchmarks for XFEM in published literature were mostly for the through-wall crack or loadings were under tension or bending moment. Subsea equipment provided by FMC Technologies is used under the most extreme conditions of pressure and temperature. Due to the fact that most equipment in subsea is pressure containing, assessment of cracks and crack growth under pressure cycles is an essential part of the integrity assessment. The implementation of fracture mechanic based fatigue to analysis can extend equipment life while improving model accuracy. In this paper, crack growth modelling in a thick wall cylinder was performed by XFEM embedded in Abaqus [3]. The case study of semi-elliptical axial surface crack at internal surface was selected. The fracture and fatigue results based on LCF analysis were compared with contour integral approach and published experimental data [4]. One of the advantages of XFEM is it can easily accommodate changes in the crack propagation direction for mixed mode loading.

Pipe bends are structural components that provide flexibility to accommodate length changes in pipelines while allowing fluid flow. Estimating the collapse load is critical to ensuring the structural integrity of the pipeline. This research discusses modeling and analysis of pipe bends utilizing the finite element method. Three-dimensional models utilizing elbow elements, shell elements, and brick elements are generated to predict the collapse moment of pipe bends subjected to in-plane loading. All simulation was performed using Abaqus. To obtain a more physically-consistent response, material, geometric, and boundary nonlinearities are all included. A MPC user subroutine is utilized to capture the end behavior of the pipe bends correctly when utilizing shell and brick elements. Experimental data from two sources, available in literature, was used to evaluate the effect of the different element types on the predicted structural response. Finally, utilizing the shell, brick, and the elbow elements, parameter sensitivity analyses are performed to identify the key parameters influencing the response of pipe bends. Multiple parameters are varied independently of each other to fully understand and capture their influence on the response. SIMULIA’s Isight software was used to automate the workflow and vary the model parameters about their respective baseline values.

A deformed droplet may leap from a solid substrate, impelled to detach through the conversion of surface energy into kinetic energy that arises as it relaxes to a sphere. Electrowetting provides a means of preparing a droplet on a substrate for lift-off. When a voltage is applied between a water droplet and a dielectric-coated electrode, the wettability of the substrate increases in a controlled way, leading to the spreading of the droplet. Once the voltage is released, the droplet recoils, due to a sudden excess in surface energy, and droplet detachment may follow. The process of drop detachment and lift-off, prevalent in both biology and micro-engineering, has to date been considered primarily in terms of qualitative scaling arguments for idealized superhydrophobic substrates. We here consider the eletrically-induced ejection of droplets from substrates of finite wettability and analyze the process quantitatively. We compare experiments to numerical simulations and analyze how the energy conversion efficiency is affected by the applied voltage and the intrinsic contact angle of the droplet on the substrate. Our results indicate that the finite wettability of the substrate significantly affects the detachment dynamics, and so provide new rationale for the previously reported large critical radius for drop ejection from micro-textured substrates.

Clostridium difficile causes one of the leading nosocomial infections in developed countries, and therapeutic choices are limited. Some strains of C. difficile produce phage tail-like particles upon induction of the SOS response. These particles have bactericidal activity against other C. difficile strains and can therefore be classified as bacteriocins, similar to the R-type pyocins of Pseudomonas aeruginosa . These R-type bacteriocin particles, which have been purified from different strains, each have a different C. difficile -killing spectrum, with no one bacteriocin killing all C. difficile isolates tested. We have identified the genetic locus of these “diffocins” (open reading frames 1359 to 1376) and have found them to be common among the species. The entire diffocin genetic locus of more than 20 kb was cloned and expressed in Bacillus subtilis , and this resulted in production of bactericidal particles. One of the interesting features of these particles is a very large structural protein of ∼200 kDa, the product of gene 1374. This large protein determines the killing spectrum of the particles and is likely the receptor-binding protein. Diffocins may provide an alternate bactericidal agent to prevent or treat infections and to decolonize individuals who are asymptomatic carriers.

Few old-field succession studies have been conducted in arid ecosystems and there is some question if classical secondary succession occurs in arid environments. In order to determine if old-field succession does occur under arid conditions, we sampled 40 previously-cultivated sites abandoned over a period of 29–80 years in the Owens Valley, California. We compared vegetation development at these 40 old fields to vegetation composition on 17 adjacent uncultivated reference sites. We were interested in determining if 1) seral communities could be identified, 2) these communities expressed predictable and progressive patterns, and 3) if so, whether the seral patterns indicated divergent or convergent successional pathways. The 57 sites separated into 6 separate seral communities. Two seres were identified, one progressing toward a big sagebrush late-seral community and the other with fourwing saltbush as the characteristic species. Based on multivariate statistical analysis, the two seres were likely to continue on divergent pathways for 100–140 years, after which the saltbush sere may converge toward the late-seral sagebrush community. Both localized and broad-scale factors were important in defining successional patterns in this arid environment. Our results support the concept that classical old-field succession does occur in arid regions.

AvR2-V10.3 is an engineered R-type pyocin that specifically kills Escherichia coli O157, an enteric pathogen that is a major cause of food-borne diarrheal disease. New therapeutics to counteract E. coli O157 are needed, as currently available antibiotics can exacerbate the consequences of infection. We show here that orogastric administration of AvR2-V10.3 can prevent or ameliorate E. coli O157:H7-induced diarrhea and intestinal inflammation in an infant rabbit model of infection when the compound is administered either in a postexposure prophylactic regimen or after the onset of symptoms. Notably, administration of AvR2-V10.3 also reduces bacterial carriage and fecal shedding of this pathogen. Our findings support the further development of pathogen-specific R-type pyocins as a way to treat enteric infections.

The nonmetal cation polyborate salt of stoichiometry [H2en]2[B11O18(OH)]·7H2O is obtained from the reaction of 1,2-diaminoethane and boric acid (1:5 ratio) in H2O/MeOH. An X-ray crystallographic study of the product reveals that the polyborate moiety is composed of two isolated hydrated polyborate anions: [B4O5(OH)4]2– and [B7O9(OH)5]2–. The structure is templated by the cations with the anions forming a supramolecular H-bonded network, augmented by additional H-bonds involving the waters of crystallization and the cations.

We evaluated invasive exotic weeds and plant species diversity in relation to depth to groundwater (DTW) and microtopography in areas with DTW from 0·3–4 m in Owens Valley, California. Transects dominated by common plant species of the area were read at 1-cm intervals, and species cover was obtained at different scales: 1-m transect portions (microsites), whole transects (68 m average length), and the whole study area. Species richness and the Shannon–Wiener diversity index were obtained in microsites. DTW and microtopographical variation (soil-surface relative elevations) were jointly measured along the vegetation transects. Found in 34% of the sampled transects, the annual Bassia hyssopifolia was the most common exotic weed of the area. This study was designed to better define the nature of the relationship among vegetation, groundwater level and microtopography in an arid area where depth to groundwater (DTW) was 0–4 m. Plant cover, DTW and relative elevations were jointly measured along 67 vegetation transects throughout the Owens Valley, CA, USA. These transects were dominated by major species of the area: Artemisia tridentata, Atriplex torreyi, Ericameria nauseosa, Distichlis spicata, Juncus arcticus, Leymus triticoides, Sarcobatus vermiculatus and Sporobolus airoides. Plant species occurrence was associated with different DTW. J. arcticus and D. spicata occurred more frequently in areas with the shallowest groundwater ( 2·0 m). Species were also linked to different microtopographic positions: L. triticoides and J. arcticus were mainly restricted to depressions whereas A. torreyi and A. tridentata were widely distributed in higher positions on the microtopographical gradient. Only 6% of the variation in vegetation cover was accounted for by DTW throughout the study area (N = 820). Cover of individual species was usually unaffected by DTW variation. Results suggest that species distribution is linked to groundwater conditions, but cover of vegetation is only partially affected by DTW variation. This is possible because water tables in our study area are within the rooting depth of most species and plants appear to be well adapted to shallow DTW variations. The steps on Verner Suomi's path to becoming a research scientist are examined. We argue that his research style—his natural interests in science and engineering, and his methodology in pursuing answers to scientific questions—was developed in his youth on the Iron Range of northeastern Minnesota, as an instructor in the cadet program at the University of Chicago (U of C) during World War II and as a fledgling academician at University of Wisconsin—Madison. We examine several of his early experiments that serve to identify his style. The principal results of this study are 1) despite austere living conditions on the Iron Range during the Great Depression, Suomi benefited from excellent industrial arts courses at Eveleth High School; 2) with his gift for designing instruments, his more practical approach to scientific investigation flourished in the company of world-class scientific thinkers at U of C; 3) his dissertation on the heat budget over a cornfield in the mid-1950s served as a springboard...

Left ventricular hypertrophy (LVH) is due to pressure overload or mechanical stretch and is thought to be associated with remodeling of gap-junctions. We investigated whether the expression of connexin 43 (Cx43) is altered in humans in response to different degrees of LVH. The expression of Cx43 was analyzed by quantitative polymerase chain reaction, Western blot analysis and immunohistochemistry on left ventricular biopsies from patients undergoing aortic or mitral valve replacement. Three groups were analyzed: patients with aortic stenosis with severe LVH (n=9) versus only mild LVH (n=7), and patients with LVH caused by mitral regurgitation (n=5). Cx43 mRNA expression and protein expression were similar in the three groups studied. Furthermore, immunohistochemistry revealed no change in Cx43 distribution. We can conclude that when compared with mild LVH or with LVH due to volume overload, severe LVH due to chronic pressure overload is not accompanied by detectable changes of Cx43 expression or spatial distribution.

Some strains of Pseudomonas aeruginosa produce R-type pyocins, which are high-molecular-weight phage tail-like protein complexes that have bactericidal activity against other Pseudomonas strains. These particles recognize and bind to bacterial surface structures via tail fibers, their primary spectrum determinant. R-type pyocins kill the cell by contracting a sheath-like structure and inserting their hollow core through the cell envelope, resulting in dissipation of the cellular membrane potential. We have retargeted an R-type pyocin to Escherichia coli O157:H7 by fusing a tail spike protein from an O157-specific phage, φV10, to the pyocin tail fiber. The φV10 tail spike protein recognizes and degrades the O157 lipopolysaccharide. This engineered pyocin, termed AVR2-V10, is sensitive and specific, killing 100% of diverse E. coli O157:H7 isolates but no other serotypes tested. AVR2-V10 can kill E. coli O157:H7 on beef surfaces, making it a candidate agent for the elimination of this pathogen from food products. All rare AVR2-V10-resistant mutants isolated and examined have lost the ability to produce the O157 antigen and are expected to have compromised virulence. In addition, E. coli O157:H7 exposed to and killed by AVR2-V10 do not release Shiga toxin, as is often the case with many antibiotics, suggesting potential therapeutic applications. The demonstration that a novel R-type pyocin can be created in the laboratory by fusing a catalytic tail spike from the family Podoviridae to a tail fiber of a member of the family Myoviridae is evidence that the plasticity observed among bacteriophage tail genes can, with modern molecular techniques, be exploited to produce nonnatural, targeted antimicrobial agents.

Demands on groundwater supplies in arid regions have increased interest in the importance of groundwater, relative to precipitation-derived moisture, to vegetation. Depth to water (DTW), precipitation (PPT), and vegetation (total perennial cover, TPC) data have been collected from 30 sites in the Owens Valley, California, since the 1980s. We used these data to construct water budgets for each site, and to calculate precipitation-use efficiencies (PUE) and amount of TPC supported by precipitation and by groundwater. We compared these values among five plant communities over different DTW and PPT regimes to determine relative importance of each water source. Our results confirm that both groundwater and precipitation are important sources of water to Owens Valley vegetation. In general, groundwater becomes more important as DTW decreases, but this relationship varies among communities. TPC increased as DTW decreased up to a point, after which TPC remained constant or declined. Some groundwater usage by shrub- and grass-dominated communities continued to DTW of 6–8 m. PUE was highest at low precipitation and decreased as precipitation increased. Our results indicate that vegetation preferentially used precipitation-derived soil moisture, even with abundant groundwater, and that successful management of groundwater-affected arid ecosystems must account for complex interactions among multiple factors.

Raffinose family oligosaccharides (RFO) accumulate in many developing seeds and are degraded during seed germination. However, acidic α-galactosidase (AGAL) activity and subcellular location do not correlate with raffinose depletion; alkaline α-galactosidases (AGA) may be responsible for RFO hydrolysis in germinating seeds. Three cDNA clones for AGA/SEED IMBIBITION PROTEIN were obtained from the Pioneer Hi-Bred maize expressed sequence database. Two of the clones were expressed in Escherichia coli, and the recombinant proteins, when incubated with naturally occurring galactosides or p-nitrophenyl α-d-galactose, exhibited AGA activity with maximum catalysis at pH 7.5 (ZmAGA1) or pH 8.5 (ZmAGA3). No raffinose biosynthetic capacity was observed with either enzyme. Maximal α-galactosidase activity in mature dehydrated, germinating and germinated maize (Zea mays) seeds occurred at pH 7.5. ZmAGA1 was the sole family member detected in seeds and maize Hi-II, embryo-derived, callus cells. Its transcript accumulated when seed germination was interrupted by heat, cold or dehydration stress, but not in response to NaCl. Tissue prints localized transcripts to the scutellum or the embryo axis, depending on the stress applied. In maize Hi-II callus cells, transcripts accumulated when callus was subjected to heat stress (42 °C), during which ZmAGA1 transcript accumulation was further induced by sucrose. Galactosides in a variety of forms, including raffinose, partially repressed the sucrose-induced accumulation of transcript in heat-stressed callus.

Grounded Theory, a qualitative research method, was used to fully describe the construct of empowerment and its measurement in racially and ethnically diverse urban and rural neighborhoods. Forty-nine grass roots experts, primarily from six communities in Texas, participated via semi-structured interviews and focus groups. Observational data were also collected. Results revealed that the framework of collective empowerment has two main dimensions: ten processes necessary for developing greater empowerment and eight long-term outcomes that result from the process strategies. The two main dimensions of collective empowerment are cyclically, rather than linearly, related. While the results are unique to the communities studied because of the nature of the analysis, the process by which they were elucidated can be replicated in any setting. These findings suggest that the construct of collective empowerment can be both operationalized and evaluated at the community level.

The transpiration coefficient (Kc) method estimates evapotranspiration as a proportion of the evapotranspiration of a reference crop growing in ideal conditions. This approach was designed for irrigated crops and assumes that plants are not subjected to resource limitations. Other assumptions are that plants have high leaf area index and little stomatal resistance to water loss. These conditions are not common for arid-land vegetation. However, mainly due to its simplicity, some studies have proposed the use of transpiration coefficients as a method of determining evapotranspiration in arid environments. In this article, the documented applications of the Kc method in arid environments and their accuracy are reviewed. We also critically discuss the physiological and agronomic concepts that support the Kc method as they relate to water-limited environments. Although changes in depth to groundwater occur naturally, anthropogenic alterations may exacerbate these fluctuations and, thus, affect vegetation reliant on groundwater. These effects include changes in physiology, structure, and community dynamics, particularly in arid regions where groundwater can be an important water source for many plants. To properly manage ecosystems subject to changes in depth to groundwater, plant responses to both rising and falling groundwater tables must be understood. However, most research has focused exclusively on riparian ecosystems, ignoring regions where groundwater is available to a wider range of species. Here, we review responses of riparian and other species to changes in groundwater levels in arid environments. Although decreasing water tables often result in plant water stress and reduced live biomass, the converse is not necessarily true for rising water tables. Initially, rising water tables kill flooded roots because most species cannot tolerate the associated low oxygen levels. Thus, flooded plants can also experience water stress. Ultimately, individual species responses to either scenario depend on drought and flooding tolerance and the change in root system size and water uptake capacity. However, additional environmental and biological factors can play important roles in the severity of vegetation response to altered groundwater tables. Using the reviewed information, we created two conceptual models to highlight vegetation dynamics in areas with groundwater fluctuations. These models use flow charts to identify key vegetation and ecosystem properties and their responses to changes in groundwater tables to predict community responses. We then incorporated key concepts from these models into EDYS, a comprehensive ecosystem model, to highlight the potential complexity of predicting community change under different fluctuating groundwater scenarios. Such models provide a valuable tool for managing vegetation and groundwater use in areas where groundwater is important to both plants and humans, particularly in the context of climate change.

Over the last two decades, several scientists have used Garcia's record of highly reflective cloud (HRC) to study the El Nino–Southern Oscillation (ENSO). Recently Waliser and Zhou removed a satellite bias. This note describes a test for the presence of the ENSO signal in the corrected record. The test treats anomaly values of corrected HRC in a grid that covers the equatorial western Pacific Ocean. For each box in the grid, the HRC anomaly was averaged over El Nino events in the record. From the averages, a map of the El Nino HRC anomaly was made. Similarly, a map of the La Nina HRC anomaly was made. Each map shows a strong, east-southeast– west-northwest trending dipole. The El Nino dipole dips toward the west-northwest, implying excess cloud in the east-southeast; the La Nina dipole dips toward the east-southeast, implying excess cloud in the west-northwest. Accumulation of transcript from three maize (Zea mays L. inbred line B73) GALACTINOL SYNTHASE (ZmGOLS) genes in abscised developing seeds and callus before and after stress was examined. Previous work showed that transcript from ZmGOLS3 predominates during seed development, whereas ZmGOLS2 transcript accumulates when triggered by certain stresses following imbibition. Abscission stimulated accumulation of ZmGOLS transcript in immature kernels. This accumulation was enhanced by increasing durations of dehydration stress. In callus tissue, neither ZmGOLS1 nor ZmGOLS3 was detected regardless of media composition or the nature or duration of the applied stress. Accumulation of ZmGOLS2 transcript in heat- or dehydration-stressed callus was induced by exogenously supplied myo-inositol in low amounts but not by sucrose. However, abundance of transcript was a poor indicator of activity of GOLS or amounts of galactinol/raffinose regardless of type of tissue, composition of media, or imposition of stress. Neither were amounts of galactinol/raffinose tightly associated with activity of GOLS nor amounts of myo-inositol, despite the ability of exogenously supplied raffinose to maintain the growth of dehydration-stressed callus. The abundance of ZmGOLS transcript did not increase in response to exogenously supplied abscisic acid.

Connexin36 (Cx36) is specifically expressed in neurons and in pancreatic beta-cells. Cx36 functions as a critical regulator of insulin secretion and content in beta-cells. In order to identify the molecular mechanisms that control the beta-cell expression of Cx36, we initiated the characterization of the human 5' regulatory region of the CX36 gene. A 2043-bp fragment of the human CX36 promoter was identified from a human BAC library and fused to a luciferase reporter gene. This promoter region was sufficient to confer specific expression to the reporter gene in insulin-secreting cell lines. Within this 5' regulatory region, a putative neuron-restrictive silencer element conserved between rodent and human species was recognized and binds the neuron-restrictive silencing factor (NRSF/REST). This factor is not expressed in insulin-secreting cells and neurons; it functions as a potent repressor through the recruitment of histone deacetylase to the promoter of neuronal genes. The NRSF-mediated repression of Cx36 in HeLa cells was abolished by trichostatin A, confirming the functional importance of histone deacetylase activity. Ectopic expression, by viral gene transfer, of NRSF/REST in different insulin-secreting beta-cell lines induced a marked reduction in Cx36 mRNA and protein content. Moreover, mutations in the Cx36 neuron-restrictive silencer element relieved the low transcriptional activity of the human CX36 promoter observed in HeLa cells and in INS-1 cells expressing NRSF/REST. The data showed that cx36 gene expression in insulin-producing beta-cell lines is strictly controlled by the transcriptional repressor NRSF/REST indicating that Cx36 participates to the neuronal phenotype of the pancreatic beta-cells.

Connexin-36 (Cx36) is a gap junction protein expressed by the insulin-producing beta-cells. We investigated the contribution of this protein in normal beta-cell function by using a viral gene transfer approach to alter Cx36 content in the insulin-producing line of INS-1E cells and rat pancreatic islets. Transcripts for Cx43, Cx45, and Cx36 were detected by reverse transcriptase-PCR in freshly isolated pancreatic islets, whereas only a transcript for Cx36 was detected in INS-1E cells. After infection with a sense viral vector, which induced de novo Cx36 expression in the Cx-defective HeLa cells we used to control the transgene expression, Western blot, immunofluorescence, and freeze-fracture analysis showed a large increase of Cx36 within INS-1E cell membranes. In contrast, after infection with an antisense vector, Cx36 content was decreased by 80%. Glucose-induced insulin release and insulin content were decreased, whether infected INS-1E cells expressed Cx36 levels that were largely higher or lower than those observed in wild-type control cells. In both cases, basal insulin secretion was unaffected. Comparable observations on basal secretion and insulin content were made in freshly isolated rat pancreatic islets. The data indicate that large changes in Cx36 alter insulin content and, at least in INS-1E cells, also affect glucose-induced insulin release.

The height and amount of tropical deep convection are examined for a correlation with the stratospheric quasi-biennial oscillation (QBO). A new 23-yr record of outgoing longwave radiation (OLR) and a corrected 17-yr record of the highly reflective cloud (HRC) index are used as measures of convection. When binned by phase of the QBO, zonal means and maps of OLR and HRC carry a QBO signal. The spatial patterns of the maps highlight the QBO signal of OLR and HRC in typically convective regions. Spectral analysis of zonal mean OLR and HRC near the equator reveals significant peaks at QBO frequencies. Rotated empirical orthogonal function (REOF) analysis is used to determine if ENSO variations of convection are aliased into the observed QBO signals. Some analyses are repeated using the OLR record after ENSO REOF modes have been removed, yielding very similar results compared to the original analyses. In insulin-secreting cells, cytokines activate the c-Jun N-terminal kinase (JNK), which contributes to a cell signaling towards apoptosis. The JNK activation requires the presence of the murine scaffold protein JNK-interacting protein 1 (JIP-1) or human Islet-brain 1(IB1), which organizes MLK3, MKK7 and JNK for proper signaling specificity. Here, we used adenovirus-mediated gene transfer to modulate IB1/JIP-1 cellular content in order to investigate the contribution of IB1/JIP-1 to beta-cell survival. Exposure of the insulin-producing cell line INS-1 or isolated rat pancreatic islets to cytokines (interferon-gamma, tumor necrosis factor-alpha and interleukin-1beta) induced a marked reduction of IB1/JIP-1 content and a concomitant increase in JNK activity and apoptosis rate. This JNK-induced pro-apoptotic program was prevented in INS-1 cells by overproducing IB1/JIP-1 and this effect was associated with inhibition of caspase-3 cleavage. Conversely, reducing IB1/JIP-1 content in INS-1 cells and isolated pancreatic islets induced a robust increase in basal and cytokine-stimulated apoptosis. In heterozygous mice carrying a selective disruption of the IB1/JIP-1 gene, the reduction in IB1/JIP-1 content in happloinsufficient isolated pancreatic islets was associated with an increased JNK activity and basal apoptosis. These data demonstrate that modulation of the IB1-JIP-1 content in beta cells is a crucial regulator of JNK signaling pathway and of cytokine-induced apoptosis.

Infection of baboons with herpesvirus papio 2 (HVP-2) produces a disease that is similar to human infection with herpes simplex viruses (HSV). Molecular characterization of HVP-2 has demonstrated that the virion contains a factor which rapidly shuts off host cell protein synthesis after infection. Reduction of host cell protein synthesis occurs in parallel with the degradation of mRNA species. A homolog of the HSV virion host shutoff (vhs) gene was identified by Southern and DNA sequence analysis. The sequence of the HVP-2 vhs gene homolog had greater than 70% identity with the vhs genes of HSV 1 and 2. Disruption of the HVP-2 vhs open reading frame diminished the ability of the virus to shut off protein synthesis and degrade cellular mRNA, indicating that this gene was responsible for the vhs activity. The HVP-2 model system provides the opportunity to study the biological role of vhs in the context of a natural primate host. Further development of this system will provide a platform for proof-of-concept studies that will test the efficacy of vaccines that utilize vhs-deficient viruses.
